[Screening of mentally retarded persons for "fragile" X-chromosome].

112 mentally weak persons aged from 2 to 18 with different degree of oligophrenia have been examined. Cultivation of blood lymphocytes was performed on medium 199 containing 5% of cattle serum. 9 persons with "fragile" X-chromosome are revealed. High frequency of autosomes fragility among the examined contingent of patients is found. A supposition is advanced on interrelation between homozygosity of "fragile" parts of autosomes and oligophrenia.

Five polymorphic microsatellite VNTRs on the human X chromosome.

The human genome contains approximately 50,000 copies of an interspersed repeat with the sequence (dT.dG/dA.dC)n, where n = approximately 10-60. We and others have found that several of these repeats have variable lengths in different individuals, with allelic fragments varying in size by multiples of 2 bp. These "microsatellite" variable number of tandem repeats (VNTRs) may be scored by PCR, using unique flanking primers to amplify the repeat-containing regions and resolving the products on DNA sequencing gels. Since few VNTRs have been found on the X chromosome, we screened a flow-sorted X c

[Various hereditary immune deficiencies linked to the X-chromosome].

The organs of immune system in hereditary immunodeficiencies linked to X-chromosome, particularly in Nezelof's syndrome and chronic granulomatosis (4 cases) were studied histologically. The alterations of thymus found in Nezelof's syndrome suggest the disturbance of its morphogenesis at the embryonal stage, those found in chronic granulomatosis suggest the postnatal disturbance of thymus morphogenesis. The children die of polytopic infections foci. Generalized BCG-itis is observed in children with chronic granulomatosis.

X chromosome inactivaton and SV40 transformation of mammalian cells.

Five embryonic mouse cultures and one human fibroblast culture were transformed with SV40. The cultures were studied cytologically to see if the normal pattern of sex chromosome replication was maintained in SV40 transformed cells. Characteristic late replication patterns were observed for both the X and Y chromosomes, and there was no evidence for loss of the inactive X chromosome, even in cells with 4 or more X chromosomes. The human line was heterozygous at two X-linked loci and a clonal analysis showed that the expression of X-linked genes was not affected by SV40 transformation.

X-chromosome linked inheritance of decreased thyroxine-binding globulin.

Partial thyroxine-binding globulin (TBG) deficiency was observed in a euthyroid boy. A family study was conducted in order to trace the defect. Of the 15 members, 3 male subjects were found to be affected, whereas 6 female subjects were shown to be carriers of the trait. The inheritance pattern of the partial TBG deficiency was found to meet the criteria for X chromosome linked transmission. No association with other X-linked characters (colour blindness, glucose-6-phosphate dehydrogenase activity) was present. Karyotype analysis in the female subjects did not show any abnormality. TBG deficie

HIM-8 binds to the X chromosome pairing center and mediates chromosome-specific meiotic synapsis.

The him-8 gene is essential for proper meiotic segregation of the X chromosomes in C. elegans. Here we show that loss of him-8 function causes profound X chromosome-specific defects in homolog pairing and synapsis. him-8 encodes a C2H2 zinc-finger protein that is expressed during meiosis and concentrates at a site on the X chromosome known as the meiotic pairing center (PC). A role for HIM-8 in PC function is supported by genetic interactions between PC lesions and him-8 mutations. HIM-8 bound chromosome sites associate with the nuclear envelope (NE) throughout meiotic prophase. Surprisingly,

Linkage analysis of 62 X-chromosomal loci excludes the X chromosome in an Icelandic family showing apparent X-linked recessive inheritance of neural tube defects.

We report here the findings of a linkage analysis, involving numerous markers from the human X chromosome, in an attempt to localise a putative gene causing apparent X-linked spina bifida and anencephaly (SBA) in a large Icelandic pedigree. Two-point linkage analysis was performed using markers from 62 informative loci in this family. Although small positive lod scores were found at a number of these loci, none reached the significance level for linkage. Haplotypes were extensively analysed and found to exclude linkage to the X chromosome.

Fragile X syndrome in a girl with variant Turner syndrome and an isodicentric X chromosome.

Fragile X (FXS) and Turner (TS) syndromes are X-chromosome-associated disorders. Herein, we report the case of a girl in middle childhood with bicuspid aortic valve in infancy, growth failure, global developmental delay (GDD), visual problems, and coexisting attention-deficit/hyperactivity and anxiety disorders. A high-resolution karyotype in 20 cells revealed 46,X,Idic(X)(p11.21)[19]/45,X[1], suggestive of variant TS. Given her atypical phenotype, subsequent DNA testing was performed. Four FMR1 cytosine-guanine-guanine repeats (30, 410, 580 and 800) were identified, confirming the additional

A new assay for the analysis of X-chromosome inactivation in carriers with an X-linked disease.

Symptoms of X-linked recessive diseases are usually observed in males, but also observed in some female carriers because of nonrandom X inactivation in which the mutated X chromosome is active and the normal X chromosomes is inactive. Therefore, it is important to investigate the patterns of X-chromosome inactivation (XCI) for clinical assessment of carriers with an X-linked disease. We have recently developed a new assay for XCI studies based on a methylation-specific polymerase chain reaction (PCR) technique. The assay involves the chemical modification of DNA with sodium bisulfite and subse

Replication patterns of three isodicentric X chromosomes and an X isochromosome in human lymphocytes.

Chromosomes from four patients with variants of the Turner syndrome were investigated by G- and C-bandind and DNA replication techniques. Their karyotypes were: 1) 46,X,idic(X)(q28), 2) 45,X/46,X,idic(X)(q24), 3) 45,X/46,X,idic(X)(p11), and 4) 46,X,i(Xq). In patients 1, 2, and 3, the abnormal X was isodicentric, with different break-and-fusion points in each case. In each, the G-band pattern on one side of the breakpoint was a mirror image of that on the other side. Each had two distinct C-bands, only one of which was associated with a primary constriction. The fourth patient had an isochromos

The impact of X chromosome inactivation on human health.

X chromosome inactivation (XCI) is a crucial genetic regulatory mechanism in female cells, wherein one of the two X chromosomes is randomly silenced to balance gene expression between sexes. Despite the overall silencing, approximately 15%-30% of human genes escape XCI, leading to their biallelic expression in females. Many studies have highlighted the phenomenon of XCI skewing, where the inactivation is preferentially biased towards one X chromosome. XCI escape and XCI skewing are significantly associated with various health issues, including autoimmune disorders, neurodevelopmental disorders

Role of the X Chromosome in Alzheimer Disease Genetics.

IMPORTANCE: The X chromosome has remained enigmatic in Alzheimer disease (AD), yet it makes up 5% of the genome and carries a high proportion of genes expressed in the brain, making it particularly appealing as a potential source of unexplored genetic variation in AD. OBJECTIVES: To perform the first large-scale X chromosome-wide association study (XWAS) of AD. DESIGN, SETTING, AND PARTICIPANTS: This was a meta-analysis of genetic association studies in case-control, family-based, population-based, and longitudinal AD-related cohorts from the US Alzheimer's Disease Genetics Consortium, the A

Ataxias with autosomal, X-chromosomal or maternal inheritance.

Heredoataxias are a group of genetic disorders with a cerebellar syndrome as the leading clinical manifestation. The current classification distinguishes heredoataxias according to the trait of inheritance into autosomal dominant, autosomal recessive, X-linked, and maternally inherited heredoataxias. The autosomal dominant heredoataxias are separated into spinocerebellar ataxias (SCA1-8, 10-15, 17-23, 25-30, and dentato-rubro-pallido-luysian atrophy), episodic ataxias (EA1-7), and autosomal dominant mitochondrial heredoataxias (Leigh syndrome, MIRAS, ADOAD, and AD-CPEO). The autosomal recessiv

X chromosomal abnormalities in Indian adolescent girls.

In girls of adolescent age, primary amenorrhea is a major problem and it is often suspected as Turner syndrome (TS), with complete or partial absence of one of the two X chromosomes. The girls who are unable to menstruate are primarily investigated by the gynecologists with the help of a physical examination, sonogram of the pelvis, endocrinologic tests, and ultimately cytogenetic analysis. Chromosomal analyses have been carried out in 280 such cases that were referred from different parts of the country. The standard protocol for peripheral blood lymphocyte culture was followed for metaphase

X-chromosome silencing in the germline of C. elegans.

Germline maintenance in the nematode C. elegans requires global repressive mechanisms that involve chromatin organization. During meiosis, the X chromosome in both sexes exhibits a striking reduction of histone modifications that correlate with transcriptional activation when compared with the genome as a whole. The histone modification spectrum on the X chromosome corresponds with a lack of transcriptional competence, as measured by reporter transgene arrays. The X chromosome in XO males is structurally analogous to the sex body in mammals, contains a histone modification associated with hete

Prenatal detection of an inverted X chromosome in a male.

An unusual case of an X chromosome with a pericentric inversion (p11.3q21.3) was detected prenatally in a male fetus. This inversion has not been previously characterized. Although the inverted chromosome was transmitted through the mother, no living males on the maternal side were detected with the aberrant chromosome. Replication studies were performed on cultures of maternal peripheral blood lymphocytes, and it was determined that the inverted X chromosome was early replicating in approximately half of the cells. Following genetic counseling, the pregnancy was continued and a healthy male i
