Rapid Tryptophan Assay as a Screening Procedure for Quality Protein Maize.

Tryptophan is an essential amino acid deficient in cereals, especially maize. However, maize (Zea mays L.) is the main source of protein in some developing countries in Africa and Latin America. In general, the nutritional profile of cereals is poor, because they are deficient in essential amino acids such as tryptophan and lysine due to a relatively higher proportion of alcohol-soluble proteins. Quality protein maize (QPM) has been developed through genetic manipulation for the nutritional enrichment of maize to address these problems. Nevertheless, methods for protein, lysine and tryptophan …

Maize root behavior as three-inputs-three-outputs logical gates due to positive gravitropism and nutritropism.

Root growth and their interactions can provide valuable information for the development of asynchronous logic systems. Here, maize root behavior due to positive gravitropism and nutritropism is evaluated as three-inputs-three-outputs logical gates. Using plant roots as the element for unconventional computing, the Boolean functions of each root tropism were constructed through arithmetic-logical operations. One gravity gate (rGG) and two nutrient gates (rNG1 and rNG2) were fabricated using additive manufacturing. The rGG platform was oriented with roots directly pulled down by gravity which co

Clavibacter nebraskensis causing Goss's wilt of maize: Five decades of detaining the enemy in the New World.

UNLABELLED: Goss's bacterial wilt and leaf blight of maize (Zea mays) caused by the gram-positive coryneform bacterium Clavibacter nebraskensis is an economically important disease in North America. C. nebraskensis is included within the high-risk list of quarantine pathogens by several plant protection organizations (EPPO code: CORBMI), hence it is under strict quarantine control around the world. The causal agent was reported for the first time on maize in Nebraska (USA) in 1969. After an outbreak during the 1970s, prevalence of the disease decreased in the 1980s to the early 2000s, before t…

Fusarium mirum sp. nov, intertwining Fusarium madaense and Fusarium andiyazi, pathogens of tropical grasses.

Many species in the Fusarium fujikuroi Species Complex (FFSC) have an affinity for grass species, with whom they live in an endophytic association or cause disease. We recovered isolates of Fusarium from agriculturally important grasses in Africa and Brazil, and characterized them with morphological markers, mating type, and Amplified Fragment Length Polymorphisms (AFLPs). We also conducted multi-locus phylogenetic analyses based on partial DNA sequences of translation elongation factor-1α (TEF1), β-tubulin (TUB), and the second largest subunit of RNA polymerase (RPB2) gene regions. Sexual cro

Leaching and aging decrease nickel toxicity to soil microbial processes in soils freshly spiked with nickel chloride.

Nickel is a trace metal that exhibits pronounced long-term immobilization reactions in soil. It is unknown if the slowly decreasing solubility of Ni in soil on aging correlates with decreased toxicity to soil biota. Three uncontaminated soils (pH 4.5-7.6) were contaminated with NiCl2 and experimentally leached or incubated outdoors with free drainage for up to 15 months. Nickel toxicity was measured for three microbial processes (potential nitrification rate, glucose-induced respiration, and C mineralization of maize residue). Results for leached and aged samples were compared with results for

Maize nitrilases have a dual role in auxin homeostasis and beta-cyanoalanine hydrolysis.

The auxin indole-3-acetic acid (IAA), which is essential for plant growth and development, is suggested to be synthesized via several redundant pathways. In maize (Zea mays), the nitrilase ZmNIT2 is expressed in auxin-synthesizing tissues and efficiently hydrolyses indole-3-acetonitrile to IAA. Zmnit2 transposon insertion mutants were compromised in root growth in young seedlings and sensitivity to indole-3-acetonitrile, and accumulated lower quantities of IAA conjugates in kernels and root tips, suggesting a substantial contribution of ZmNIT2 to total IAA biosynthesis in maize. An additional …

Pratylenchus coffeae Effector PcENG2 Targets the Maize Chitinase ZmCHI5 to Promote Parasitism.

Pratylenchus coffeae, a damaging root-lesion nematode, secretes effectors to manipulate host processes and promote parasitism. Here, we char … lity to P. coffeae when ectopically expressed in Arabidopsis. PcENG2 directly interacts with maize (Zea mays L.) chitinase, a key defense-related protein. This interaction suppresses chitinase enzymatic activity and protects nematode eggs from degradation. Virus-induced gene silencing (VIGS) demonstrated that ZmCHI5 is essential for broad-spectrum nematode …

Studies on liver sulphurylase activity in rats: vitamin-A-deficient and low-protein diets.

Studies on the sulphurylation of phenols by livers from normal and vitamin-A-deficient rats showed a significant reduction in sulphurylating activity in the acute stage of deficiency. At the 'plateau stage' of deficiency, decrease in this activity was marginal and was not significant. Rats consuming a 6% casein diet had a significantly lower liver sulphurylating activity when compared to those fed 12 and 18% casein diets. Comparative data on the sulphurylating activity of rats maintained on diets providing poor-quality protein, i.e. ragi (Eleusine coracana) and maize (Zea mays) revealed a high…

Pollution characteristics, enrichment patterns, and progress in detoxification research of mycotoxins in maize.

Maize is susceptible to mycotoxin contamination throughout harvesting, storage, and processing, which poses potential risks to human health and livestock production. This review summarizes the occurrence, toxicological mechanisms, and contamination dynamics of major mycotoxins commonly detected in maize, including aflatoxins (AFs), ochratoxin A (OTA), zearalenone (ZEN), fumonisins (FBs), and deoxynivalenol (DON). We further examine their distribution and accumulation patterns across the maize production chain, from pre-harvest to processing. During the pre-harvest period, toxin accumulation is

WSD-YOLO: A lightweight YOLO-based model with enhanced feature representation for maize pest detection.

As a major staple crop with global significance, maize is highly vulnerable to pest infestations throughout its growth cycle, which can substantially limit plant development and reduce yield. In practice, accurate detection remains challenging due to pronounced morphological variations across pest life stages, as well as interference from complex field backgrounds, often leading to missed detections. To address these challenges, we propose WSD-YOLO (YOLOv11n with WindmillConv, Single-Scale High-Level Transformer, and Dual-Attention Weighted Aggregation), a high-accuracy maize pest detection mo

Multi-dimensional maize biomass utilization: Mitigating existing agro-waste by meaningful value addition.

Maize stover is an abundantly available biomass across different parts of the world, accounting for about 47-50 % dry mass of the maize plant. Primarily comprising of cobs, stems, leaves, and husks, maize biomass contains significant amount of cellulose, hemicellulose and lignin. Utilization of these lignocellulosic components to produce a suitable spectrum of marketable products such as hydrolytic enzymes, second-generation bioethanol, furfurals, organic acids, biohydrogen, biopolymers, prebiotic xylooligosaccharides, etc., holds promise to generate new markets and complement the economic via

Enzymatic hydrolysis of corn stover lignin by laccase, lignin peroxidase, and manganese peroxidase.

Lignin of high purity and structural integrity was isolated from the enzymatic residue of corn stover. Degradation of the lignin by laccase, lignin peroxidase, and manganese peroxidase was investigated. Structural changes in the lignin after degradation were characterized by scanning electron microscopy, nitrogen adsorption and Fourier transform infrared spectroscopy, and the enzymatic products were systematically analyzed by gas chromatography mass spectrometry. The highest percentage of lignin degradation was obtained with a mixture of three enzymes (25.79%): laccase (Lac), the starting enzy

Impact of dilute acid pretreatment conditions on p-coumarate removal in diverse maize lines.

Prior work has identified that lignins recovered from dilute acid-pretreated corn stover exhibit superior performance in phenol-formaldehyde resins used in wood adhesive applications when compared to diverse process-modified lignins derived from other sources. This improved performance is hypothesized to be due to the higher content of unsubstituted phenolic groups specifically p-coumarate lignin esters. In this work, a diverse set of corn stover samples are employed that exhibit diversity in p-coumarate content and total lignin content to explore the relationship between dilute acid pretreatm

Suitability of Wild Host Plants and Firewood as Hosts of Prostephanus truncatus (Coleoptera: Bostrichidae) in Mozambique.

Prostephanus truncatus (Horn) (Coleoptera: Bostrichidae) populations are maintained in wild host plants such as trees that act as pest reservoirs from where beetle infestation of maize granaries take place. In this study, we assessed the suitability of plant species sold and transported as firewood as well as other plant species in Mozambique as hosts for P. truncatus. Prostephanus truncatus was only recorded from three tree species, which are sold as firewood, i.e., Brachystegia spiciformis, Strychnos spinosa, and Colophospermum mopane. The pest survived and bred in 13 tree and 7 grass specie

Effects of milling methods on the properties of frozen corn dough: Insights into starch structural and physicochemical properties under repeated freeze-thaw cycles.

The effects of dry, semi-dry, and wet milling on the multi-scale structure and physicochemical properties of corn dough under freeze-thaw cycles(F/T) were investigated. The semi-dry milled corn dough with low damaged starch content (1.35%) and uniformly distributed starch granules, readily formed a stable starch-protein matrix. It restricted water migration and accumulation, exhibited the highest relative crystallinity (20.40%), and maintained structural stability during F/T. After F/T, its hardness increased (136.0-224.0 N). High damaged starch (6.03%) and large granules (D50 = 19.14 μm) in d

Comparing In Vivo and In Vitro Pollen Tube Attraction Assays for Arabidopsis and Maize.

Pollen tube attraction is a crucial step in the reproduction of angiosperms, ensuring that the male gametophytes, which carry two sperm cells, accurately target the compatible female gametophytes, or embryo sacs, in order to achieve double fertilization. Various experimental systems have been developed to study this process in vivo and in vitro, particularly in the model plant Arabidopsis thaliana (Arabidopsis). However, fewer methods have been described for other plant species, including grasses, which generate much thicker reproductive tissues and organs. Here, we present and compare optimiz

Replacing maize partially or totally with sorghum alters mycotoxin profile in their silages in microsilos.

Heat- and drought-tolerant sorghum is an attractive alternative to maize, yet its silage mycotoxin trade-offs remain unclear. We compared maize (Zea mays L.), sorghum (Sorghum bicolor (L.) Moench; cultivars Goliath and Sucrosorgo 506) and 1:1 maize-sorghum silages after 60 d ensiling (microsilo scale), with or without a commercial multi-strain lactic acid bacteria (LAB) inoculant. All silages fermented well (pH 4.06 - 4.21; lactic acid 18.1 - 27.9 g/kg dry matter). Maize silage showed the highest Fusarium toxins-deoxynivalenol (DON) 499 - 516 and nivalenol (NIV) 173 - 216 μg/kg dry matter. In …

Root-exuded benzoxazinoids can alleviate negative plant-soil feedbacks.

Plants can suppress the growth of other plants by modifying soil properties. These negative plant-soil feedbacks are often species-specific, suggesting that some plants possess resistance strategies. However, the underlying mechanisms remain largely unknown. Here, we investigated whether benzoxazinoids, a class of dominant secondary metabolites that are exuded into the soil by maize and other cereals, allow maize plants to cope with plant-soil feedbacks. We find that three out of five tested crop species reduce maize (Zea mays L.) performance via negative plant-soil feedbacks relative to the m…
