OBJECTIVE: To determine, for the average older adult, the odds of demonstrating an auditory processing abnormality for each of seven speech-based measures of auditory processing and how these odds vary based on a number of independent subject variables.
DESIGN: Using a cross-sectional design, 1576 adults aged 55 years and older were assessed with speech measures of central auditory processing and questionnaires pertaining to health status, cognitive and perceived auditory function. The speech-based measures from which abnormal/normal outcomes were derived were (a) right ear Macquarie Syntheti

AIM: This study aimed to investigate the association between the superoxide dismutase 2 (SOD2)-rs4880 polymorphism and age-related hearing loss (ARHL) in an Iranian population, assessing its impact on age, hearing loss severity, and SOD activity, alongside bioinformatics and molecular docking analyses with TOM20.
MATERIALS AND METHODS: This case-control study included 145 ARHL patients and 155 healthy controls. Serum SOD activity was measured using a commercial kit and rs4880 genotyping was done via PCR-RFLP. Bioinformatics tools focusing on HADDOCK-2.4 were used to assess the interaction bet

Age-related hearing loss may increase listening difficulties in challenging listening conditions (e.g., speech-in-noise), limiting cognitive resources available to perform common, complex multitasking behaviours like listening while driving. Older adults with hearing loss may compensate by increasing prefrontal cortex (PFC) activation in response to multitasking demands. Few realistic, controlled studies have examined how competing attentional demands of listening while driving affect performance and brain activation, and how these patterns may differ between older adults with and without audi

A/J mouse is a model of age-related hearing loss (AHL). Mutation in the citrate synthase (Cs) gene of the mouse plays an important role in the hearing loss and degeneration of cochlear cells. To investigate the pathogenesis of cochlear cell damage in A/J mice resulted from Cs mutation, we downregulated the expression level of CS in HEI-OC1, a cell line of mouse cochlea, by shRNA. The results showed that low CS expression led to low ability of cell proliferation. Further study revealed an increase level of reactive oxygen species (ROS), activation of ATF6 mediated endoplasmic reticulum stress (

C57BL/6J mice have long been studied as a model of age-related hearing loss (ARHL). In C57BL/6J mice, ARHL begins in the high-frequency range at 3 months of age and spreads toward low frequencies by 10 months of age. We previously confirmed that c.753A>G genome editing of an ahl allele (c.753A) in the cadherin 23 gene (Cdh23) suppressed the onset of ARHL until 12 months of age. We further investigated the hearing phenotypes of the original and genome-edited C57BL/6J-Cdh23+/+ (c.753G/G) mice until 24 months of age. The hearing tests revealed that most of the C57BL/6J mice maintained good hearin

Distinct forms of age-related hearing loss are hypothesized based on evidence from animal models of aging, which are identifiable in human audiograms. The Sensory phenotype results from damage (e.g., excessive noise or ototoxic drugs) to outer hair cells and sometimes inner hair cells, producing large threshold increases predominately at high frequencies. The Metabolic phenotype results from a decline in endocochlear potential that can reduce outer hair cell motility throughout the cochlea, producing gradually sloping thresholds from lower to higher frequencies. Finally, the combined Metabolic

OBJECTIVES/HYPOTHESIS: To obtain actual status of age-related hearing loss in a general unscreened population of older Dutch adults and to investigate whether the prevalence or degree has changed over time.
STUDY DESIGN: To investigate the prevalence and degree of hearing loss, we conducted a large prospective cohort study of older adults between February 2011 and July 2015.
METHODS: Pure-tone air- and bone-conduction thresholds were measured for 4,743 participants. Results were compared to previous cohort studies.
RESULTS: As expected, hearing loss increased with age. We found a correlatio

Auditory nerve (AN) fibers that innervate inner hair cells in the cochlea degenerate with advancing age. It has been proposed that age-related reductions in brainstem frequency-following responses (FFR) to the carrier of low-frequency, high-intensity pure tones may partially reflect this neural loss in the cochlea (Märcher-Rørsted et al., 2022). If the loss of AN fibers is the primary factor contributing to age-related changes in the brainstem FFR, then the FFR could serve as an indicator of cochlear neural degeneration. In this study, we employed electrocochleography (ECochG) to investigate t

The Klotho mouse is a recently developed model that exhibits phenotypes resembling human aging. We used this model to investigate sensorineural hearing loss from the point of view that it may be considered an issue of aging. Using reverse transcription-polymerase chain reaction, Western blotting and immunohistochemical staining, we were able to confirm klotho gene transcription and protein synthesis in the kidney and inner ear. Klotho protein was mainly expressed in the stria vascularis and spiral ligament of the inner ear and in the distal convoluted tubule of the kidney, likely serving a com

OBJECTIVES: Both age-related peripheral or central hearing loss, and cardiometabolic multimorbidity (CMM), which are independent association with global and domain-specific cognitive impairment, are common among older adults. Cardiometabolic diseases also are independent risk factors of age-related hearing loss. The first aim of the study was to investigate the independent and joint influence of CMM and low- and high-frequency hearing loss or central auditory processing dysfunction (CAPD) on global and domain-specific cognitive impairment. The second aim was to investigate whether CMM mediate

Age-related hearing loss (ARHL) is a common neurodegenerative disorder in the elderly, but its molecular mechanisms and diagnostic biomarkers remain unclear. Here, by using the GSE49543 dataset (n = 40, Affymetrix microarray) from the Gene Expression Omnibus (GEO) with limma, 17 ARHL-associated differentially expressed genes were identified, which were enriched in immune effector functions and the IL-17 signaling pathway. Weighted gene co-expression network analysis (WGCNA) further revealed that 168 genes were associated with hearing loss (module significance > 0.3). Intersecting DEGs with thi

Mitochondrial dysfunction is associated with aging and age-related hearing loss (AHL). However, the precise mechanisms underlying the pathophysiology of hearing loss remain unclear. Cdk5 regulatory subunit-associated protein 1 (CDK5RAP1) enables efficient intramitochondrial translation by catalyzing the deposition of 2-methylthio modifications on mitochondrial tRNAs. Here we investigated the effect of defective mitochondrial protein translation on hearing and AHL in a Cdk5rap1 deficiency C57BL/6 mouse model. Compared to control C57BL/6 mice, Cdk5rap1-knockout female mice displayed hearing loss

Wolfram syndrome (WS) is caused by recessive mutations in the Wolfram syndrome 1 (WFS1) gene. Sensorineural hearing impairment (HI) is a frequent feature in WS and, furthermore, certain mutations in WFS1 cause nonsyndromic dominantly inherited low-frequency sensorineural HI. These two phenotypes are clinically distinct indicating that WFS1 is a reasonable candidate for genetic studies in patients with other phenotypes of HI. Here we have investigated, whether the variation in WFS1 has a pathogenic role in age-related hearing impairment (ARHI). WFS1 gene was investigated in a population sample

Speech perception difficulties are common among elderlies; yet the underlying neural mechanisms are still poorly understood. New empirical evidence suggesting that brain senescence may be an important contributor to these difficulties has challenged the traditional view that peripheral hearing loss was the main factor in the etiology of these difficulties. Here, we investigated the relationship between structural and functional brain senescence and speech perception skills in aging. Following audiometric evaluations, participants underwent MRI while performing a speech perception task at diffe
