Genomic characterisation and antimicrobial resistance of Klebsiella species isolated from small stock and environmental sources in South Africa.

Klebsiella species pose a significant public health concern due to their association with various infections and the rising levels of antibiotic resistance. This study examined the antibiotic resistance profiles of nine Klebsiella species obtained from the culture collection at North-West University's Potchefstroom campus in South Africa using whole genome sequencing. The average nucleotide identity (ANI), plasmid types, and the identification of antibiotic resistance and virulence genes were performed on Klebsiella species. The nine sequenced Klebsiella spp. isolates were identified as K. pne

Antimicrobial Resistance: The Answers.

Antimicrobial resistance (AMR) has caused a global public health crisis, contributing to approximately five million deaths in 2019 and predicted deaths of approximately ten million annually by 2050. This equates to approximately 1.4-fold more deaths annually from AMR in 2050 than the entire COVID-19 pandemic to date. To tackle this AMR pandemic, regulatory and policy frameworks have been prepared at local, national and international levels with multi-faceted proposals and advances encompassing surveillance, diagnostics, infection prevention, antibiotic prescribing and variation of existing and

The Effects of Baicalin in Combination with Cefotaxime on the Biofilm and Metabolic Reprogramming of Multidrug-Resistant Pseudomonas aeruginosa.

Baicalin, a natural plant-derived compound, holds promise in addressing clinical bacterial resistance when combined with antibiotics. This study evaluated the antibacterial activity of the combination of baicalin and cefotaxime and explored its mechanism of action on the cell wall and biofilm of multidrug-resistant Pseudomonas aeruginosa (MRPA). The results showed that the combination of baicalin and cefotaxime exerted a synergistic inhibitory effect on the growth of MRPA, with a fractional inhibitory concentration index (FICI) of 0.28. Mechanistically, compared with cefotaxime alone, the comb

Hydrogel-Based Platforms for Wound Care: Integrated Strategies for Antimicrobial Delivery and Biofilm Management.

Chronic wounds, diabetic foot ulcers, venous leg ulcers, and pressure injuries affect millions of patients worldwide and cost healthcare systems in the order of $150 billion annually, yet treatment options have changed less than the scale of the problem would suggest. Biofilm formation, documented in up to 78% of chronic wounds, is a central cause: bacteria embedded in extracellular polymeric matrices tolerate antimicrobial concentrations up to 1000-fold higher than planktonic cells and sustain a chronic inflammatory state that actively prevents tissue repair. Hydrogels, crosslinked polymer ne

Comparison of Morphological Characteristics, Histological Tissue Structures, and Intestinal Function Among Eight Ornamental Fish Species Under Identical Aquaculture Conditions.

The intestine, particularly the gut microbiota, and the liver play key roles in digestion, nutrient transformation, and immune regulation in fish. However, limited information is available regarding how different ornamental fish species regulate these systems under identical aquaculture conditions. Therefore, this study systematically compared gut microbiota diversity, structural variation, and predicted ecological functions among eight ornamental fish species reared in the same environment, using 16S rRNA high-throughput sequencing combined with digestive enzyme indices and histological analy

Metabolic Competition-Driven Antitumor Immunity of glpX-Deficient Engineered Salmonella via Glucose Deprivation and STING Activation in Bladder Cancer.

Background: Bacteria-mediated cancer therapy is emerging as a promising approach due to its low cost, scalability, and sustained efficacy. While bacterial immunomodulation has been extensively studied, the role of bacterial metabolic competition, particularly for glucose, in tumor suppression remains unclear. We aimed to investigate how engineered Salmonella strains exploit metabolic competition to trigger antitumor immunity and improve therapeutic outcomes in bladder cancer models. Methods: Using genetic screening, we generated a gluconeogenesis-deficient Salmonella strain, the AISIH strain w

A conserved virulence backbone and heterogeneous type III secretion effector profiles in historical canine otitis-associated Pseudomonas aeruginosa from Hungary.

BACKGROUND: Canine otitis externa provides a chronic, inflamed and frequently antimicrobial-exposed niche in which Pseudomonas aeruginosa can persist through a coordinated repertoire of adhesion, biofilm, iron-acquisition, secretion and toxin systems. METHODS: We characterized the virulence-associated genomic architecture of a historical Hungarian collection obtained from canine otitis cases in 2010 and 2017. Of the 70 isolate identifiers in the nominal range 70-139, 60 had corresponding long-read assemblies and linked VFDB screening records. One assembly was excluded after genome-based taxon

Fungal-Bacterial Interactions in Polymicrobial Infections: Hidden Threats.

Polymicrobial infections involving fungi and bacteria represent a major and increasingly recognized clinical challenge, in which interkingdom interactions significantly amplify disease severity, antimicrobial resistance, and treatment failure. Rather than passive co-existence, fungal-bacterial communities form highly coordinated systems driven by physical adhesion, quorum sensing, metabolic interdependence, and biofilm-mediated structural reinforcement. These cooperative interactions, exemplified by pairs such as Candida albicans-Staphylococcus aureus and Pseudomonas aeruginosa-Aspergillus fum

Role of nanoclusters in the design and development of novel strategies for diagnosis and treatment of infectious diseases.

Despite major advances in sanitation, vaccination, and antimicrobial chemotherapy, infectious diseases continue to pose a huge threat to global health. The ever-increasing occurrences of antimicrobial resistance, emerging pathogens, and persistent diagnostic delays further exacerbate this situation. Over the years, nanotechnology has generated a lot of material platforms to address these failures, and in this study, we critically examine nanoclusters-ultrasmall, atomically precise assemblies (<2 nm cores) with discrete electronic states, as an under-recognized but potentially transformative cl

Dietary AiiO-AIO6 mitigated Aeromonas hydrophila-induced intestinal inflammation in juvenile grass carp (Ctenopharyngodon idella) involving in NF-κB signaling and pyroptosis.

AiiO-AIO6 is a quorum-sensing quenching enzyme that could decrease the virulence of pathogenic bacteria, and improve animal immunity. However, the precise regulatory mechanism of AiiO-AIO6 on intestinal immunity remains unknown. Thus, this study aimed to reduce this knowledge gap. After feeding with graded levels of AIO-AIO6 (0.0 (un-supplemented group), 2.5, 5.0, 7.5, 10.0, and 12.5 U/g) for 70 days, juvenile grass carp was selected from each group for a 6-day Aeromonas hydrophila challenge test. Meanwhile, some other fish selected from un-supplemented control group were injected with saline

Fractional inhibitory concentration of bio-actives from agricultural waste disassembles biofilms and quenches virulence of nosocomial pathogens.

Introduction. The contact surfaces in hospitals serve as reservoirs for pathogens and account for 20-40% of hospital-acquired infections. This resistance is mainly attributed to the biofilm-forming ability of the microbes. These biofilms restrict the entry of the antibiotics to penetrate them, thus giving rise to drug resistance. Hence, there is a renewed interest in formulating an environmentally friendly, non-allergic, quick mode of action, broad-spectrum disinfectant.Hypothesis. We hypothesize that the pure compounds present in the pyrolysis aqueous phase could act as an anti-infective and

Activation of CzcS/CzcR during zinc excess regulates copper tolerance and pyochelin biosynthesis of Pseudomonas aeruginosa.

UNLABELLED: Zinc is an important transition metal that is essential for numerous physiological processes while excessive zinc is cytotoxic. Pseudomonas aeruginosa is a ubiquitous opportunistic human pathogen equipped with an exquisite zinc homeostatic system, and the two-component system CzcS/CzcR plays a key role in zinc detoxification. Although an increasing number of studies have shown the versatility of CzcS/CzcR, its physiological functions are still not fully understood. In this study, transcriptome analysis was performed, which revealed that CzcS/CzcR is silenced in the absence of the z

Unraveling the ecological interactions between dairy strains Bacillus licheniformis and Bacillus cereus during the dual-species biofilm formation.

Bacillus cereus and Bacillus licheniformis are widely presented in dairy products. They can form thick biofilms on surfaces of dairy processing equipment, which may pose serious safety issues and spoilage of final dairy products. However, how ecological interactions between B. cereus and B. licheniformis affect the functions and stability of mixed-species biofilm remains uncovered. In this work, the altered profiles of a dual-species biofilm by dairy-derived B. cereus 121 and B. licheniformis 919 were investigated by RNA-sequencing analysis in combined with phenotype validation (bacterial grow

Two Permeases Associated with the Multifunctional CtaP Cysteine Transport System in Listeria monocytogenes Play Distinct Roles in Pathogenesis.

The soil-dwelling bacterium Listeria monocytogenes survives a multitude of conditions when residing in the outside environment and as a pathogen within host cells. Key to survival within the infected mammalian host is the expression of bacterial gene products necessary for nutrient acquisition. Similar to many bacteria, L. monocytogenes uses peptide import to acquire amino acids. Peptide transport systems play an important role in nutrient uptake as well as in additional functions that include bacterial quorum sensing and signal transduction, recycling of peptidoglycan fragments, adherence to

The Autoinducer N-Octanoyl-L-Homoserine Lactone (C8-HSL) as a Potential Adjuvant in Vaccine Formulations.

Autoinducers AI-1 and AI-2 play an important role in bacterial quorum sensing (QS), a form of chemical communication between bacteria. The autoinducer N-octanoyl-L-Homoserinehomoserine lactone (C8-HSL) serves as a major inter- and intraspecies communicator or 'signal', mainly for Gram-negative bacteria. C8-HSL is proposed to have immunogenic properties. The aim of this project is to evaluate C8-HSL as a potential vaccine adjuvant. For this purpose, a microparticulate formulation was developed. The C8-HSL microparticles (MPs) were formulated by a water/oil/water (W/O/W) double-emulsion solvent

Microbial isolation and characterization from two flex lines from the urine processor assembly onboard the international space station.

Urine, humidity condensate, and other sources of non-potable water are processed onboard the International Space Station (ISS) by the Water Recovery System (WRS) yielding potable water. While some means of microbial control are in place, including a phosphoric acid/hexavalent chromium urine pretreatment solution, many areas within the WRS are not available for routine microbial monitoring. Due to refurbishment needs, two flex lines from the Urine Processor Assembly (UPA) within the WRS were removed and returned to Earth. The water from within these lines, as well as flush water, was microbiall

Fungi-A Component of the Oral Microbiome Involved in Periodontal Diseases.

The human oral cavity is a diverse ecological niche favorable for colonization by hundreds of different species of microorganisms. They include not only bacteria but also numerous species of fungi, many of which are able to cause opportunistic infections when the host's immunity is impaired, predominantly by systemic and chronic diseases like diabetes, pulmonary diseases, renal disorders, or acquired immunodeficiency syndrome. Within the dental biofilm and subgingival sites, fungi of the genus Candida are often found, also in individuals affected with periodontitis. Moreover, fungal species of
