Berberine Synergistically Enhances the Antibacterial Activity of Ampicillin against Ampicillin-Resistant Staphylococcus epidermidis.

Although S. epidermidis is a skin commensal bacterium, it is thought to be a causative pathogen of skin infections. Prolonged and extensive use of antibiotics for skin infections, including ampicillin (AMP), has contributed to the increasing emergence of antibiotic-resistant bacteria and the shaping of skin commensal S. epidermidis into opportunistic pathogens. This highlights the urgent necessity for alternative therapeutic strategies to enhance antimicrobial efficiency and minimize toxic effects on host cells. The present study investigated the potential of berberine (BBR), with specific att

Integrative Multi-Omics Mendelian Randomization Identifies BLMH as a Keratinocyte-Enriched Protective Candidate Gene and Potential Therapeutic Target in Psoriasis.

BACKGROUND: Psoriasis is a chronic immune-mediated inflammatory skin disease driven by complex interactions between genetic susceptibility, immune dysregulation, and keratinocyte dysfunction. Although genome-wide association studies have identified numerous risk loci, most studies have focused on genetic associations rather than causal inference, limiting the identification of effective therapeutic targets. We employed an integrative multi-omics strategy to identify causal genes and potential therapeutic candidates for psoriasis. METHODS: We integrated psoriasis genome-wide association study

Nanometric Faujasite (FAU) Zeolite Ion-Exchanged With Metal Ions for Hemostatic and Antimicrobial Applications: A Thromboelastographic and Microbiological Study.

Integrating antimicrobial functionality with the intrinsic clot-promoting properties of zeolites represents a promising strategy for next-generation wound-care materials, where rapid hemorrhage control must be coupled with infection prevention. Here, we investigate nanocrystalline Faujasite (NanoFAU) and its ion-exchanged derivatives to elucidate how extra-framework cation identity governs the balance between hemostatic and antimicrobial performance. NanoFAU samples exchanged with Ag+, Ba2 +, Ca2 +, and Mg2 + were comprehensively characterized by XRD, SEM/HRTEM/EDS, AFM, BET surface area analy

Physicochemical and Biological Properties of Menthol and Thymol-Based Natural Deep Eutectic Solvents.

Seven hydrophobic deep eutectic solvents (hDESs) were characterised to evaluate their potential applicability in different industries and their environmental impact. Standard physicochemical properties were determined, yielding polarity and density values that were slightly higher for thymol-based hDESs than menthol-based ones, whereas for viscosity, the trend was opposite. Regarding biologically relevant activities, the antioxidative capacity and antimicrobial activity of hDESs were determined. Thymol-based hDESs are more potent as potential antioxidants, especially the one with coumarin as a

Functional characterization of the SDR42E1 reveals its role in vitamin D biosynthesis.

Vitamin D deficiency poses a widespread health challenge, shaped by environmental and genetic determinants. A recent discovery identified a genetic regulator, rs11542462, in the SDR42E1 gene, though its biological implications remain largely unexplored. Our bioinformatic assessments revealed pronounced SDR42E1 expression in skin keratinocytes and the analogous HaCaT human keratinocyte cell lines, prompting us to select the latter as an experimental model. Employing CRISPR/Cas9 gene-editing technology and multi-omics approach, we discovered that depleting SDR42E1 showed a 1.6-fold disruption in

Nanoplastic Size and Surface Chemistry Dictate Decoration by Human Saliva Proteins.

Environmental pollution with plastic polymers has become a global problem, leaving no continent and habitat unaffected. Plastic waste is broken down into smaller parts by environmental factors, which generate micro- and nanoplastic particles (MNPPs), ultimately ending up in the human food chain. Before entering the human body, MNPPs make their first contact with saliva in the human mouth. However, it is unknown what proteins attach to plastic particles and whether such protein corona formation is affected by the particle's biophysical properties. To this end, we employed polystyrene MNPPs of t

Anti-methicillin resistance Staphylococcus aureus and in vitro toxicology evaluation of corilagin-loaded gelatin/agar microspheres with potential biotextile applications.

Methicillin-resistant Staphylococcus aureus (MRSA) has emerged since the early 1960s. The increasing resistance of pathogens to currently used antibiotics requires the urgent discovery of new antimicrobials effective in combating drug-resistant bacteria. From past to present, medicinal plants are useful to cure human diseases. Corilagin (β-1-O-galloyl-3,6-(R)-hexahydroxydiphenoyl-d-glucose), commonly found in Phyllanthus species, exerts potentiating effect on β-lactams against MRSA. However, its biological effect may not be fully utilized. Therefore, incorporating microencapsulation technology

TNF-α induced extracellular release of keratinocyte high-mobility group box 1 in Stevens-Johnson syndrome/toxic epidermal necrolysis: Biomarker and putative mechanism of pathogenesis.

Decreased epidermal high-mobility group box 1 (HMGB1) expression is an early marker of epidermal injury in Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN). Etanercept, an anti-tumor necrosis factor therapeutic, is effective in the treatment of SJS/TEN. The objective was to characterize antitumor necrosis factor-alpha (TNF-α)-mediated HMGB1 keratinocyte/epidermal release and etanercept modulation. HMGB1 release from TNF-α treated (± etanercept), or doxycycline-inducible RIPK3 or Bak-expressing human keratinocyte cells (HaCaTs) was determined by western blot/ELISA. Healthy skin exp

In vitro and in vivo acute toxicity of a novel citrate-coated magnetite nanoparticle.

Magnetic nanoparticles (MNps) have become powerful tools for multiple biomedical applications such as hyperthermia drivers, magnetic resonance imaging (MRI) vectors, as well as drug-delivery systems. However, their toxic effects on human health have not yet been fully elucidated, especially in view of their great diversity of surface modifications and functionalizations. Citrate-coating of MNps often results in increased hydrophilicity, which may positively impact their performance as drug-delivery systems. Nonetheless, the consequences on the intrinsic toxicity of such MNps are unpredictable.

Low extracellular pH inhibits nucleotide excision repair.

Nucleotide excision repair (NER) is the main pathway to repair bulky DNA damages including pyrimidine dimers, and the genetic dysregulation of NER associated proteins is well known to cause diseases such as cancer and neurological disorder. Other than the genetic defects, 'external factors' such as oxidative stress and environmental chemicals also affect NER. In this study, we examined the impact of extracellular pH on NER. We prepared the culture media, whose pH values are 8.4 (normal condition), 7.6, 6.6 and 6.2 under atmospheric CO2 conditions. Human keratinocytes, HaCaT, slightly died afte

2-Hydroxy-4-methoxybenzophenone Enhances the Suppression of Superoxide Anion Radicals Generated via UVA-induced Photosensitizing by t-Butyl Methoxydibenzoylmethane.

4-tert-Butyl-4'-methoxydibenzoylmethane (BMDM) is widely used throughout the world as a highly effective UVA absorber that can prevent the progression of photoaging in skin. However, due to its low photostability, BMDM is also known for the disadvantage of having a reduced capability to absorb UVA during prolonged exposure to sunlight. Although many studies have been carried out to overcome this disadvantage of BMDM, little attention has been paid to how the radicals generated from BMDM during UV exposure influence the skin. Therefore, the purpose of this study was twofold: One goal was to cla

The Tryptophan-Derived Endogenous Aryl Hydrocarbon Receptor Ligand 6-Formylindolo[3,2-b]Carbazole Is a Nanomolar UVA Photosensitizer in Epidermal Keratinocytes.

Endogenous UVA chromophores may act as sensitizers of oxidative stress underlying cutaneous photoaging and photocarcinogenesis, but the molecular identity of non-DNA key chromophores displaying UVA-driven photodyamic activity in human skin remains largely undefined. Here we report that 6-formylindolo[3,2-b]carbazole (FICZ), a tryptophan photoproduct and endogenous high-affinity aryl hydrocarbon receptor (AhR) agonist, acts as a nanomolar photosensitizer potentiating UVA-induced oxidative stress irrespective of AhR ligand activity. In human HaCaT and primary epidermal keratinocytes, photodynami

A time-lapse approach to examine chromium and nickel effects on wound healing in vitro.

Chromium and nickel cause allergic contact dermatitis, a common biological skin response to sensitizing agents. This study used a conventional in vitro wounding model to study the impact of sensitizing agents on the innate immune response of human keratinocytes. Experiments were designed to evaluate the involvement of specific Toll-like receptors and metalloproteinases as effectors molecules downstream, at a molecular level. Further, keratinocytes were co-cultured with monocytes (THP-1 cells) to reproduce an inductive stimulus on monocytes made by metals. Human keratinocytes (HaCat) were grown

R3ACT: An Oxygen-Controlled Biosensor-Based NAM to Support Animal-Free Chemical Safety Assessment.

Most in vitro chemical safety workflows are performed in ambient air, although many human tissues experience substantially lower physiological oxygen levels. This mismatch can alter redox tone, stress-response signaling, and interpretation of chemical effects. Here, we present R3ACT (Redox & Antioxidant Capacity Tracker), an animal-free, oxygen-controlled live-cell NAM for mechanistic profiling of redox perturbation and antioxidant-response activation. In human HaCaT keratinocytes adapted to 5 kPa O₂, R3ACT combines viability-gated concentration selection, oxygen-controlled plate-reader measur

In vitro efficacy of different PEGylation designs on cathelicidin-like peptide with high antibacterial and antifungal activity.

Recent reports on antibiotic resistance have highlighted the need to reduce the impact of this global health issue through urgent prevention and control. The World Health Organization currently considers antibiotic resistance as one of the most dangerous threats to global health. Therefore, Antimicrobial peptides (AMPs) are promising for the development of novel antibiotic molecules due to their high antimicrobial effects, non-inducing antimicrobial resistance (AMR) properties, and broad spectrum. Hence, in this study, we developed novel antimicrobial peptide/polymer conjugates to reduce the a
