Hydroxamic acid-containing hydrogels for nonabsorbed iron chelation therapy: synthesis, characterization, and biological evaluation.

Iron overload is a severe clinical condition and can be largely prevented by the use of iron-specific chelating agents. A successful iron chelator needs to be orally active, nontoxic, and selective. In this study, hydrogels containing pendant hydroxamic acid groups have been synthesized as potential nonabsorbed chelators for iron in the gastrointestinal tract. The synthetic method employed to introduce hydroxamic acid groups to polymer chains involved reaction of polymer gels based on N-acryloxysuccinimide, acryloyl chloride, and (2-hydroxyethyl)acrylate monomers with hydroxylamine. These hydr

Factors affecting the adherence energy of experimental resin cements bonded to a nickel-chromium alloy.

Reliable adherence of resin-based cements is of prime importance for the longevity of cemented restorations. The present study investigated whether a relationship exists between adherence energy to a metal substrate and the degree of cross-linking and wetting characteristics of resin-based luting agents. The adherence energies between a sand-blasted metal surface and a series of experimental resin cements were measured by means of the wedge test. The degree of cross-linking was calculated from the monomer composition of the resin cements. The measured wetting characteristics were work of adhes

Discovery of Highly Selective AKR1C3 Inhibitors to Overcome EGFR C797S-Mediated Osimertinib Resistance in Non-Small Cell Lung Cancer.

Osimertinib resistance driven by the cis-C797S/T790M EGFR triplet mutation remains clinically intractable. We identify aldo-keto reductase 1C3 (AKR1C3) as a metabolic vulnerability that sustains glutathione-reactive oxygen species (GSH-ROS) homeostasis in resistant non-small cell lung cancer (NSCLC). Starting from the selective inhibitor S07-2001, six rounds of structure-guided optimization delivered 55 analogues. The most advanced, SG-55, is a noncompetitive AKR1C3 inhibitor with nanomolar potency, exhibiting a half-maximal inhibitory concentration (IC50) of 5 ± 1 nM, whereas the IC50 values

MicroRNA Mimics Based on the miR-15/107 Consensus Sequence Sensitise NSCLC Cells to Targeted Therapy.

Non-small cell lung cancer (NSCLC) is the leading cause of lung cancer deaths, with resistance to targeted therapies posing a major clinical challenge. Drug-tolerant persister (DTP) cells are key contributors to resistance, and targeting them offers new strategies to enhance existing treatments. MicroRNAs (miRNAs), particularly the tumour-suppressive miR-15/107 family, offer promise due to their ability to target multiple oncogenic pathways. This study evaluated a synthetic consensus miRNA mimic, conmiR-15/107, in NSCLC cell line models. Dose-response assays showed robust, dose-dependent growt

Role of VEGFA/VEGFR2 signaling in predicting clinical outcomes of EGFR-TKI treatment in EGFR-mutant non-small cell lung cancer.

BACKGROUND: Dual inhibition of epidermal growth factor receptor (EGFR) and vascular endothelial growth factor (VEGF) pathways may improve outcomes in metastatic EGFR-mutant NSCLC, but VEGF inhibitors are not universally effective. We evaluated the impact of VEGFA and VEGFR2 expressions on EGFR-TKI outcomes. METHODS: EGFR-mutant NSCLC patients from the National Cancer Center Hospital, were retrospectively analysed. The early-stage cohort comprised stage I-IIIA patients (1997-2019). The advanced-stage cohort included metastatic patients (2018-2022). VEGFA/VEGFR2 expressions were dichotomised by

Clinical Outcomes and Prognostic Factors of ABCP Therapy in EGFR-mutated NSCLC in the Osimertinib Era.

BACKGROUND/AIM: Atezolizumab, bevacizumab, carboplatin, and paclitaxel (ABCP) therapy has demonstrated efficacy in epidermal growth factor receptor (EGFR)-mutated non-small cell lung cancer (NSCLC) after EGFR-tyrosine kinase inhibitor (TKI) failure. However, real-world evidence in the osimertinib era remains limited. This study aimed to evaluate the efficacy, safety, and prognostic factors of ABCP therapy in patients with EGFR-mutated NSCLC. PATIENTS AND METHODS: This retrospective study evaluated patients with EGFR-mutated NSCLC who received ABCP therapy after EGFR-TKI failure at two institu

EGFR-TKI plus chemotherapy versus TKI monotherapy in advanced EGFR-mutant NSCLC with high PD-L1 expression: a real-world retrospective study.

BACKGROUND: Patients with advanced EGFR-mutant NSCLC and high PD-L1 expression (TPS ≥50%) typically respond poorly to EGFR-TKI monotherapy. We compared EGFR-TKI plus chemotherapy versus TKI alone in this population. RESEARCH DESIGN AND METHODS: This retrospective study included 212 patients receiving first-line osimertinib with chemotherapy (n = 98) or as monotherapy (n = 114). Primary endpoint was progression-free survival (PFS). RESULTS: Combination therapy significantly improved median PFS overall (22.5 vs 13.8 months; HR = 0.513, 95% CI:0.372-0.803; p < 0.001) and in PD-L1-high patients

Targeted degradation of estrogen receptor β to overcome Osimertinib resistance in non-small cell lung carcinoma.

Osimertinib resistance remains a major challenge in advanced non-small cell lung cancer (NSCLC) patients. Estrogen receptor β (ERβ) has been identified as a vital promoter in the emergence of Osimertinib resistance in NSCLC cells, however, the mechanism of action of ERβ underlining the development of acquired resistance to Osimertinib for NSCLC treatment remain unclear. To explore its therapeutic potential, we designed and synthesized a series of novel ERβ targeting PROteolysis Targeting Chimera (PROTAC) degraders for the first time to overcome Osimertinib resistance in NSCLC. Targeting the SYVN1-EGFR axis: a breakthrough strategy for TKI-resistant NSCLC.

Non-small cell lung cancer (NSCLC) is the leading cause of cancer-related death. Currently, molecular targeted therapy remains a crucial approach to the treatment of NSCLC. However, the development of acquired drug resistance poses significant challenges for subsequent treatment. Identifying new therapeutic targets is of great significance for improving the prognosis of patients with NSCLC. Here, we verify synoviolin-1 (SYVN1) as a potential new therapeutic target for NSCLC. SYVN1 is highly expressed in NSCLC, and its upregulation is associated with poor prognosis. We show that the N-terminus

Inhibition of histone deacetylases 3 attenuates imiquimod-induced psoriatic dermatitis via targeting cGAS-STING signaling in keratinocytes.

BACKGROUND: Psoriasis is a common chronic inflammatory skin disease characterized by epidermal keratinocyte hyperproliferation and persistent immune activation. Histone deacetylase 3 (HDAC3), a member of the class I HDAC family, plays critical roles in regulating immunity and inflammation. However, its precise expression profile and functional contribution to psoriasis pathogenesis remain poorly defined. METHODS: We first performed bioinformatics analysis of HDAC3 expression using the Gene Expression Omnibus (GEO) database. Subsequently, we employed a combination of cellular and molecular tec

Osimertinib after definitive chemoradiotherapy in unresectable stage III epidermal growth factor receptor-mutated non-small-cell lung cancer: analyses of central nervous system efficacy and distant progression from the phase III LAURA study.

BACKGROUND: Distant metastases in non-small-cell lung cancer (NSCLC) are a poor prognostic factor that negatively impact quality of life. The central nervous system (CNS) is a common site of distant progression in epidermal growth factor receptor-mutated (EGFRm) NSCLC. Osimertinib is a third-generation EGFR-tyrosine kinase inhibitor recommended for advanced EGFRm NSCLC and as adjuvant treatment for resected EGFRm NSCLC. In LAURA (NCT03521154), osimertinib demonstrated statistically significant improvement in progression-free survival (PFS) versus placebo in unresectable stage III EGFRm NSCLC w

Taxus chinensis var. mairei (Lemée et Lévl) Cheng et L.K. Fu overcomes the resistance to osimertinib in EGFR-mutant non-small-cell lung cancer via suppression of ERK1/2-related cholesterol biosynthesis.

ETHNOPHARMACOLOGICAL RELEVANCE: Acquired resistance to osimertinib limits its clinical efficacy in non-small cell lung cancer (NSCLC) with EGFR mutations. The widespread recognition of Taxus chinensis var. Mairei (Lemée et Lévl) Cheng et L.K. Fu (Chinese yew) as a natural anti-cancer medication is well-established. However, the specific contribution of Taxus chinensis var. Mairei (Lemée et Lévl) Cheng et L.K. Fu in addressing resistance to osimertinib is still uncertain. AIM OF THE STUDY: Based on the biological behaviors and lipid metabolism, we investigated whether aqueous extract of Taxus

Risk factors of metachronous brain metastasis in patients with EGFR-mutated advanced non-small cell lung cancer.

BACKGROUND: NSCLC patients with EGFR mutation were at a higher incidence of developing brain metastasis (BM). Patients with BM are associated with high mortality. Reducing BM incidence becomes increasingly significant for NSCLC patients to achieve prolonged survival. The aim of the study was to explore the possible risk factors of developing metachronous BM during EGFR-TKIs treatment, and to identify the potential candidates for prophylactic cranial irradiation (PCI) or the first-line Osimertinib treatment. METHODS: A total of 157 consecutive EGFR-mutated advanced NSCLC patients without BM at

Safety Profile of Epidermal Growth Factor Receptor Tyrosine Kinase Inhibitors: A Disproportionality Analysis of FDA Adverse Event Reporting System.

Adverse event reports submitted to the US Food and Drug Administration (FDA) were analyzed to map the safety profile of epidermal growth factor receptor-tyrosine kinase inhibitors (EGFR-TKIs). We conducted a disproportionality analysis of the adverse events (AEs) of EGFR-TKIs (gefitinib, erlotinib, afatinib, osimertinib) by data mining using the FDA adverse event reporting system (AERS) database, and by calculating the reporting odds ratios (ROR) with 95% confidence intervals. The FDA AERS database contained 27,123 EGFR-TKI-associated AERs within the reporting period from January 1, 2004 to Ma
