Antigenic Multi-Epitope Vaccine Against Ross River Virus Using Capsid and Envelope Proteins via Immunoinformatics and Reverse Vaccinology Approaches.

Ross River virus (RRV), a mosquito-borne alphavirus of the Togaviridae family, is the causative agent of Ross River fever a debilitating infectious disease characterized by fever, rash, and persistent polyarthritis. The virus remains endemic across Australia and the Pacific regions, with sporadic outbreaks reported in new geographic areas due to climate change and increased mosquito activity. Despite its growing public health impact, no licensed vaccine or antiviral therapy currently exists, underscoring the urgent need for an effective preventive strategy. In this study, an immunoinformatics-

A conserved capsid-based multi-epitope vaccine targeting dengue virus serotypes (DENV1-4): an integrated computational and in vivo study.

BACKGROUND: Dengue virus (DENV), comprising four antigenically distinct serotypes (DENV1-4), continues to pose a major global health burden. The lack of a universally protective vaccine capable of inducing balanced immunity against all serotypes remains a critical challenge, largely due to antigenic diversity and the risk of antibody-dependent enhancement (ADE). Conserved capsid proteins represent promising targets for epitope-based vaccine development because of their structural stability and immunogenic potential. METHODS: An integrated reverse vaccinology and immunoinformatics strategy was

Current Concepts in Periprosthetic Joint Infection: Modern Biomarkers, Microbiome Insights, Molecular Diagnostics, and Emerging Prevention Strategies.

AIM: Periprosthetic joint infection (PJI) remains one of the leading causes of revision arthroplasty and is associated with substantial morbidity, mortality, and healthcare costs. Diagnosis remains challenging because of heterogeneous clinical presentations, the limited sensitivity of conventional inflammatory markers in chronic and low-grade infections, culture-negative cases, and biofilm-mediated pathogen persistence. This narrative review summarizes recent advances in the diagnosis and prevention of PJI, with a particular focus on synovial and serum biomarkers, microbiome-related susceptibi

Periprosthetic Joint Infection: Modern Diagnostic Markers and Their Limitations.

Joint arthroplasty (JA) is a common surgical procedure that provides significant relief from degenerative joint pain, notably increases quality of life for patients, and markedly improves functional status. However, despite its many benefits, JA is not without risk, and one of the greatest risks patients who undergo JA face is periprosthetic joint infections (PJIs). PJIs are defined as infections occurring after the implantation of an artificial joint and have been shown to result in substantial morbidity, high treatment costs, and increased long-term mortality. Despite improvements in diagnos

Protective effect and mechanism of nanoantimicrobial peptide ND-C14 against Streptococcus pneumoniae infection.

BACKGROUND: Streptococcus pneumoniae (S. pneumoniae) is a common pathogen that causes bacterial pneumonia. However, with increasing bacterial resistance, there is an urgent need to develop new drugs to treat S. pneumoniae infections. Nanodefensin with a 14-carbon saturated fatty acid (ND-C14) is a novel nanoantimicrobial peptide designed by modifying myristic acid at the C-terminus of human α-defensin 5 (HD5) via an amide bond. However, it is unclear whether ND-C14 is effective against lung infections caused by S. pneumoniae. METHODS: In vitro, three groups were established, including the con

Dietary sanguinarine supplementation improves the growth performance and intestinal immunity of broilers.

Dietary sanguinarine (SAN) can enhance the growth performance of poultry and livestock, but the regulatory mechanism of the SAN monomer on intestinal homeostasis and how it promotes growth performance has not yet been clarified. In this study, 200 chickens were divided into four groups and fed different doses of SAN (0, 0.225, 0.75, 2.25 mg/kg) for transcriptome and microbiota analysis. The data showed that different doses of SAN supplementation increased the feed conversion rate (FCR) of 22 to 42 d old and 1 to 42 d old broilers (P < 0.01), and 0.225 mg/kg SAN reduced the contents of alanine

Impact of Citrus Pulp or Inulin on Intestinal Microbiota and Metabolites, Barrier, and Immune Function of Weaned Piglets.

We investigated the use of citrus pulp (CP) as a novel prebiotic capable of exerting microbiota and immunomodulating capacities to alleviate weaning stress. Inulin (IN), a well-known prebiotic, was used for comparison. Hundred and 28 male weaned piglets of 21 days old were assigned to 32 pens of 4 piglets each. Piglets were assigned to one of the four treatments, i.e., control, IN supplemented at 0.2% (IN0.2%), and CP supplemented either at 0.2% (CP0.2%) or at 2% (CP2%). On d10-11 and d31-32 post-weaning, one pig per pen was euthanized for intestinal sampling to evaluate the growth performance

Prior exposure of Arabidopsis seedlings to mechanical stress heightens jasmonic acid-mediated defense against necrotrophic pathogens.

BACKGROUND: Prolonged mechanical stress (MS) causes thigmomorphogenesis, a stress acclimation response associated with increased disease resistance. What remains unclear is if; 1) plants pre-exposed to a short period of repetitive MS can prime defence responses upon subsequent challenge with necrotrophic pathogens, 2) MS mediates plant immunity via jasmonic acid (JA) signalling, and 3) a short period of repetitive MS can cause long-term changes in gene expression resembling a stress-induced memory. To address these points, 10-days old juvenile Arabidopsis seedlings were mechanically stressed f

Genetics of rotator cuff tears: no association of col5a1 gene in a case-control study.

BACKGROUND: The incidence of RC tears increases with aging, affecting approximately 30 to 50% of individuals older than 50 years, and more than 50% of individuals older than 80 years. Intrinsic factors (age or gender), extrinsic factors (sports activity or occupation), and biological factors were identified in the onset and progression of RC tears. The attention in the study of aetiology of RC tendinopathy has shifted to the identification of gene variants. A multi-epitope pan-betacoronavirus vaccine construct predicted to induce broad-spectrum and durable immune responses: an immunoinformatics approach.

INTRODUCTION: Recurrent zoonotic spillovers and continuous antigenic evolution among betacoronaviruses, including SARS-CoV, MERS-CoV, and SARS-CoV-2, highlight the urgent need for a broad-spectrum vaccine capable of eliciting cross-protective immunity. Conventional vaccines, although effective against specific strains, may be limited by antigenic mismatch and waning immunity. This study aimed to design a multi-epitope pan-betacoronavirus vaccine targeting conserved regions within the receptor-binding domain (RBD) using an integrated immunoinformatics and reverse vaccinology framework. Exploring the interplay of vitamin D, salivary antimicrobial peptides, and cytokines in oral immunity and disease prevention: an insight for implications in oral health policy.

INTRODUCTION: Oral diseases such as caries and periodontitis are complex, multifactorial diseases and remain the most prevalent worldwide. Both diseases are associated with vitamin D deficiency (VDD) and its pathophysiological process. The prevalence of metabolic syndrome (MetS) is rising in developing countries, and studies have shown that oral diseases are more prevalent among the metabolic group. This study aims to assess and correlate the relationship between serum vitamin D levels, salivary antimicrobial peptides (AMPs), and their role in oral health. MATERIALS AND METHODS: A total of 80

Pan-genomic insights into resistance, virulence, and stress adaptation in Clostridium perfringens from the Tibetan Plateau.

Clostridium perfringens is a ubiquitous foodborne and zoonotic pathogen responsible for necrotizing enteritis, food poisoning, and gas gangrene in humans and livestock, with mortality reaching 100% in specific subtypes. The Qinghai-Tibetan Plateau, with its high altitude, hypoxia, large temperature fluctuations, and strong UV radiation, imposes a strong selective regime that drives distinctive, plateau-specific microbial evolution. We isolated and sequenced eight C. perfringens isolates from yaks and Tibetan pigs and analyzed them with 129 public genomes. Multilocus sequence typing (MLST) reve

Novel integrated approach modeling proanthocyanidins and bacteriophages to combat multidrug Salmonella Typhimurium in challenged broilers.

The emergence of multidrug bacterial isolates, including Salmonella (S.) Typhimurium, which primarily spreads to humans through chicken products, is correlated with a rising prevalence of antimicrobial therapy failure. Thus, we performed a comprehensive analysis of the combined impact of Salmonella bacteriophage (BP) and grape seed oligomeric pro-anthocyanidins (GSOPs) on growth performance, immune functions, antioxidant capacity, cecal microbiota, gut integrity, and S. Typhimurium resistance in challenged broilers. A total of 250 Ross-308 male broiler chicks were offered either a control diet

Effect of progestin-based contraceptives on HIV-associated vaginal immune biomarkers and microbiome in adolescent girls.

Adolescent girls bear a disproportionate burden of both the HIV epidemic and unintended pregnancies; yet important questions remain unanswered regarding the effects of hormonal contraceptives on the vaginal immune microenvironment, which can impact HIV susceptibility in this group. Multiple studies report genital immune alterations associated with the progestin-based contraceptive Depot medroxyprogesterone acetate (DMPA) in adult women, but there is little available data in adolescents. The objective of this longitudinal cohort study was to evaluate the effects of short-term use of three proge
