The use of oil adjuvants in therapeutic vaccines.

Water in oil emulsions represents one of the new promising generations of adjuvants for immunotherapy. Fifty years ago, incomplete freund adjuvant (IFA) has been used in clinical trials for prophylactic vaccines like poliomyelitis or flu vaccine because of its strong potency. However, even if the quality of the raw materials has been improved in order to avoid secondary reactions, the risk benefit ratio was not favorable to its use for prophylactic vaccines. Moreover, emulsions were highly viscous with a weak stability. The development of new adjuvants concepts like liposomes, oil in water emu

Adenovirus vector-based vaccine for infectious diseases.

Replication-incompetent adenovirus (Ad) vectors have been widely used as gene delivery vehicles in both gene therapy studies and basic studies for gene function analysis due to their highly advantageous properties, which include high transduction efficiencies, relatively large capacities for transgenes, and high titer production. In addition, Ad vectors induce moderate levels of innate immunity and have relatively high thermostability, making them very attractive as potential vaccine vectors. Accordingly, it is anticipated that Ad vectors will be used in vaccines for the prevention of infectio

Live attenuated, nef-deleted SIV is pathogenic in most adult macaques after prolonged observation.

OBJECTIVE: A live attenuated SIV vaccine strain, termed SIVmac239Delta3 and containing large deletions in, and the negative regulatory element, was previously shown to cause AIDS mostly in monkeys vaccinated as infants. In the present study, we demonstrate that SIVmac239Delta3 is pathogenic in most vaccinated adult monkeys, given enough time. METHODS: Eleven rhesus macaques vaccinated as adults with SIVmac239Delta3 were followed for extended periods (up to 6.8 years). RESULTS: We found signs of immune dysregulation in all 11 adult vaccinees. All animals developed persistently inverted CD4 :

SHIV transmission and susceptibility to re-exposure through social contact following vaccination with an HIV synthetic peptide-cocktail: a case study.

An effective vaccine against human immunodeficiency virus (HIV) should not only protect from infection and development of acquired immunodeficiency syndrome (AIDS), but also prevent potential transmission to naïve partners. We recently reported protection of rhesus macaques from chronic simian-human immunodeficiency virus (SHIV) infection and AIDS by an HIV envelope peptide-cocktail vaccine. In the present case study, we observed that one of the vaccinated females, with undetectable circulating virus, when housed in a pair with a naïve male, did not transmit the infection over a 35-week period

Developing Antiviral Vaccines Based on Attenuated Salmonella.

Attenuated Salmonella strains are a promising gene delivery vector in the development of vaccines targeting various human diseases. For example, the Ty21a vaccine is a Salmonella-based vaccine that elicits a robust local, cellular, and systemic immune response against Salmonella typhi infection. Human immunodeficiency virus (HIV) is responsible for the AIDS pandemic and requires lifelong treatment to manage. Human papillomavirus (HPV) is the cause of the vast majority of cervical cancer cases in the global population. Influenza is a seasonal, pandemic virus that can lead to high death tolls in

Advances in nanomaterial vaccine strategies to address infectious diseases impacting global health.

Despite the overwhelming success of vaccines in preventing infectious diseases, there remain numerous globally devastating diseases without fully protective vaccines, particularly human immunodeficiency virus (HIV), malaria and tuberculosis. Nanotechnology approaches are being developed both to design new vaccines against these diseases as well as to facilitate their global implementation. The reasons why a given pathogen may present difficulties for vaccine design are unique and tied to the co-evolutionary history of the pathogen and humans, but there are common challenges that nanotechnology

Pediatric rheumatic diseases and vaccinations: a promising alliance.

BACKGROUND: Immunosuppressive drugs, disease-modifying antirheumatic drugs, as methotrexate, glucocorticoids and biological agents can limit the immune response to vaccines and, in some cases, contraindicate their administration. Non-live vaccines are safe also for immunosuppressed paediatric patients. Seroprotection is maintained in children with rheumatic diseases or autoinflammatory diseases (AID) undergoing vaccinations on immunosuppression, except for those treated with high-dose glucocorticoids and B-cell depleting drugs. METHODS: We analyzed in a retrospective observational study 107 p

Induction of circulating T follicular helper cells and regulatory T cells correlating with HIV-1 gp120 variable loop antibodies by a subtype C prophylactic vaccine tested in a Phase I trial in India.

A Phase I HIV-1 vaccine trial sponsored by the International AIDS Vaccine Initiative (IAVI) was conducted in India in 2009 to test a subtype C prophylactic vaccine in a prime-boost regimen comprising of a DNA prime (ADVAX) and MVA (TBC-M4) boost. The trial demonstrated that the regimen was safe and well tolerated and resulted in enhancement of HIV-specific immune responses. Preliminary observations on vaccine-induced immune responses were limited to analysis of neutralizing antibodies and IFN-γ ELISPOT response. The present study involves a more detailed analysis of the nature of the vaccine-i

Edible Vaccines: A Patent-Driven Exploration of Immunization Technologies.

Vaccines are biological preparations that improve immunity to particular diseases. Particularly for poor developing nations, edible vaccines show significant potential as a financially advantageous, simple to administer, straightforward to store, fail-safe, and socially and culturally acceptable vaccine delivery system. A vaccine incorporates the gene-encoding bacterial or viral disease-causing agent in plants without losing its immunogenic property. Potatoes, tomatoes, rice, soybeans, and bananas are the primary plants for edible vaccines. It activates the systemic and mucosal immunity respon

Vector-based genetically modified vaccines: Exploiting Jenner's legacy.

The global vaccine market is diverse while facing a plethora of novel developments. Genetic modification (GM) techniques facilitate the design of 'smarter' vaccines. For many of the major infectious diseases of humans, like AIDS and malaria, but also for most human neoplastic disorders, still no vaccines are available. It may be speculated that novel GM technologies will significantly contribute to their development. While a promising number of studies is conducted on GM vaccines and GM vaccine technologies, the contribution of GM technology to newly introduced vaccines on the market is disapp

Innate gamma/delta T-cells during HIV infection: Terra relatively Incognita in novel vaccination strategies?

Despite a long-lasting global effort, the Holy Grail quest for a protective vaccine, able to confer prevention to HIV infection, did not reach the hoped for results, nor seems able to do so in the near future. Since mucosal surfaces of the host serve as the main entry point for HIV, it seems now logical to switch from a systemic to a localized view of events, in order to reveal critical steps useful in designing new and different vaccination strategies. In this context, the recent description of the very early phases of infection, from the eclipse to the viremia peak phase, seems to define a p

Brief report: Influenza vaccination and health care workers in the United States.

OBJECTIVE: To determine influenza vaccination rates among U.S. health care workers (HCWs) by demographic and occupational categories. DESIGN AND PARTICIPANTS: We analyzed data from the 2000 National Health Interview Survey (NHIS). Weighted multivariable analyses were used to evaluate the association between HCW occupation and other variables potentially related to receipt of influenza vaccination. HCWs were categorized based on standard occupational classifications as health-diagnosing professions, health-assessing professions, health aides, health technicians; or health administrators. MAIN

[Microorganisms strike back--infectious diseases during the last 50 years].

In the first half of the 20th century, improved living conditions, preventive measures, vaccines and antibiotics led to a marked reduction in morbidity and mortality from infectious diseases. It was predicted that the conquest of all infectious diseases was imminent. However, 50 years later, in 1999, they were still the major cause of disease worldwide, and caused nearly one third of all deaths (a total of 55.9 million). The eradication of smallpox in the 1970s and the approaching eradication of poliomyelitis represent major achievements. The prevalence of measles, pertussis and tetanus neonat

The Current State and Progress of Mpox Vaccine Research.

On July 23, 2022, the World Health Organization (WHO) declared the monkeypox (mpox) outbreak a "Public Health Emergency of International Concern." Since 2022, outbreaks of mpox in many countries around the world have primarily resulted in fatalities among immunocompromised individuals, such as untreated HIV/AIDS patients. Since the eradication of smallpox was declared by the WHO in 1980, the global vaccination against smallpox has been gradually discontinued. China also stopped routine smallpox vaccination in 1981. The protective effect of the smallpox vaccine has decreased over time due to ag

Kaposi's Sarcoma Lesion Progression in BKV-Tat Transgenic Mice Is Increased by Inflammatory Cytokines and Blocked by Treatment with Anti-Tat Antibodies.

Kaposi's sarcoma (KS) is an angioproliferative tumor showing an increased frequency and aggressiveness in HIV-infected subjects (AIDS-KS), due to the combined effects of inflammatory cytokines (IC), angiogenic factors, and the HIV-1 Tat protein. While the introduction of effective combined antiretroviral regimens greatly improved AIDS-KS incidence and course, it continues to be an incurable disease and the development of new rational targeted therapies is warranted. We used the BKV/Tat transgenic mouse model to evaluate the effects of IC and anti-Tat antibodies (Abs) treatment on KS-like lesio

Effect of inactivated COVID-19 vaccination on intrauterine insemination cycle success: A retrospective cohort study.

BACKGROUND: Vaccine hesitancy was found in couples seeking artificial reproductive technology (ART) services. As the main vaccine used in China, investigations into the influence of inactivated coronavirus disease 2019 (COVID-19) vaccines on human fertility is needed. METHODS: This retrospective cohort study included data on COVID-19 vaccination, clinical characteristics, and reproductive outcome of 1,000 intrauterine insemination (IUI) cycles in 653 couples from March 2021 to March 2022 in a single university hospital-based center for reproductive medicine. The IUI cycles were divided into t

New Perspectives of HIV/AIDS Therapy Study.

HIV/AIDS (acquired immune deficient syndrome), a human infectious disease was once listed as the No. 1 disease killer in US (1993). After the invention of antiviral drug cocktails-high active anti-retroviral therapy (HAART), most HIV-infected patients can survive much longer than with single antiviral drugs or vaccines alone. However, it turns out to be a chronic disease owing to being incapable to eradicate HIV from infectious patients. Furthermore, potential newly outbreak of HIV epidemics caused by widespread drug-resistance or viral mutations is still looming over the globe. In order to co
