Study of the cytolethal distending toxin (CDT)-activated cell cycle checkpoint. Involvement of the CHK2 kinase.

The bacterial cytolethal distending toxin (CDT) triggers a G2/M cell cycle arrest in eukaryotic cells by inhibiting the CDC25C phosphatase-dependent CDK1 dephosphorylation and activation. We report that upon CDT treatment CDC25C is fully sequestered in the cytoplasmic compartment, an effect that is reminiscent of DNA damage-dependent checkpoint activation. We show that the checkpoint kinase CHK2, an upstream regulator of CDC25C, is phosphorylated and activated after CDT treatment. In contrast to what is observed with other DNA damaging agents, we demonstrate that the activation of CHK2 can onl

Dose response characteristics of methylphenidate on different indices of rats' locomotor activity at the beginning of the dark cycle.

Using a computerized infrared activity analysis system, the dose-response relationship, timing, and duration for stimulation of motor activity after a single dose of methylphenidate was studied in Sprague-Dawley rats. After 5 days of acclimation and 2 days of monitored baseline activity, rats received a single subcutaneous injection of vehicle or of 0.6, 2.5, 10 or 40 mg/kg methylphenidate 1 h into the dark cycle. Recording was then resumed for an additional 36 h. Five locomotor indices were analyzed. Each locomotor parameter monitored different aspects of motor activity. The doses of 2.5, 10

The basal Mg2(+)-dependent ATPase activity is not part of the (H(+)+K+)-transporting ATPase reaction cycle.

Purified gastric (H(+)+K+)-transporting ATPase [(H(+)+K+)-ATPase] from the parietal cells always contains a certain amount of basal Mg2(+)-dependent ATPase (Mg2(+)-ATPase) activity. lin-Benzo-ATP (the prefix lin refers to the linear disposition of the pyrimidine, benzene and imidazole rings in the 'stretched-out' version of the adenine nucleus), an ATP analogue with a benzene ring formally inserted between the two rings composing the adenosine moiety, is an interesting substrate not only because of its fluorescent behaviour, but also because of its geometric properties. lin-Benzo-ATP was used

Impact of menstrual cycle and hormonal contraception on whole-body protein turnover in recreationally active females.

Protein turnover may be influenced by menstrual cycle hormones and hormones from oral contraceptives (OCs) and hormonal intrauterine devices (H-IUDs), yet data are limited. This study evaluated whole-body protein turnover in 40 healthy, active females (eumenorrheic = 15; OC = 13; H-IUD = 12) tested once in low- and high-hormone phases. Using a [15N]alanine tracer, net protein balance, synthesis, breakdown, and nitrogen flux were measured. No group differences were observed. Protein breakdown and nitrogen flux were higher in the low-hormone phase in the total sample. These small hormone cycle p

Physical activity of moderate-intensity optimizes myocardial citrate cycle in a murine model of heart failure.

INTRODUCTION: There is growing body of evidence that an enhanced concentration of branched-chain amino acids (BCAAs), as a consequence of an impaired myocardial oxidative metabolism, is involved in the occurrence and progression of heart failure (HF). The purpose of this study was to examine the effect of 8 weeks of spontaneous wheel running (8-sWR) (reflecting low-to-moderate intensity physical activity) on the myocardial [BCAAs] and mitochondrial oxidative metabolism markers, such as tricarboxylic acid (TCA) cycle intermediates (TCAi), mitochondrial electron transport chain (ETC) proteins an

Antifungal activity of cinnamaldehyde against Aspergillus fumigatus involves disruption of the TCA cycle and protein metabolism.

Aspergillus fumigatus is an environmental opportunistic fungal pathogen, which can lead to invasive aspergillosis in immunocompromised individuals, and resistant to conventional antifungual agents has become a growing concern. This study investigated the antifungal activity and the molecular antifungal mechanisms of Cinnamaldehyde (CA) against A. fumigatus, specifically its impact on metabolic pathways and protein metabolism. In susceptibility tests, CA was found to exhibit promising antifungal activity against A. fumigatus in both solid and liquid culture (biomass) systems, with the minimum i

Ubiquitin-activating enzyme1 (TgUAE1) acts as a key regulator of Toxoplasma gondii lytic cycle and homeostasis.

Ubiquitylation, regulated by the ubiquitin-proteasome system (UPS), is crucial for cell division and cycle transitions in Toxoplasma gondii. However, the primary E1 ubiquitin-activating enzyme (UAE1) in this process has been unclear. This study identified and characterized TGGT1_290290 (TgUAE1) as the canonical E1 enzyme in T. gondii. Through a combination of bioinformatics, biochemical, pharmacological, and genetic approaches, TgUAE1 was shown to exhibit typical E1 activity, particularly in forming K48- and K63-linked polyubiquitin chains. TAK-243, a UAE1 inhibitor, can effectively inhibit th

CCND2 Modified mRNA Activates Cell Cycle of Cardiomyocytes in Hearts With Myocardial Infarction in Mice and Pigs.

BACKGROUND: Experiments in mammalian models of cardiac injury suggest that the cardiomyocyte-specific overexpression of CCND2 (cyclin D2, in humans) improves recovery from myocardial infarction (MI). The primary objective of this investigation was to demonstrate that our specific modified mRNA translation system (SMRTs) can induce CCND2 expression in cardiomyocytes and replicate the benefits observed in other studies of cardiomyocyte-specific CCND2 overexpression for myocardial repair. METHODS: The CCND2-cardiomyocyte-specific modified mRNA translation system (cardiomyocyte SMRTs) consists of

Aberrant activation of neuronal cell cycle caused by dysregulation of ubiquitin ligase Itch results in neurodegeneration.

It is critical for the neuronal cell cycle to remain suppressed in terminally differentiated neurons as its activation results in aberrant cell cycle re-entry that causes neuronal apoptosis (CRNA), which has been observed in several neurodegenerative disorders like Alzheimer's disease (AD). In the present study, we report that E3 ubiquitin ligase Itch is a major regulator of CRNA and elucidated the mechanism via which it is regulated in this process. Neurotoxic amyloid peptide Aβ42-treated neurons or neurons from an AD transgenic mouse model (TgAD) exhibited aberrant activation of the JNK path

Cryopreservation of Paramecium bursaria Chlorella Virus-1 during an active infection cycle of its host.

Best practices in laboratory culture management often include cryopreservation of microbiota, but this can be challenging with some virus particles. By preserving viral isolates researchers can mitigate genetic drift and laboratory-induced selection, thereby maintaining genetically consistent strains between experiments. To this end, we developed a method to cryopreserve the model, green-alga infecting virus, Paramecium bursaria Chlorella virus 1 (PBCV-1). We explored cryotolerance of the infectivity of this virus particle, whereby freezing without cryoprotectants was found to maintain the hig

Asynchronic steroid activity of Leydig and Sertoli cells related to spermatogenic and testosterone cycle in Phymaturus antofagastensis.

The severe environments where Phymaturus lizards inhabit in the Andes highlands and in Patagonia, Argentina, impose restrictions on their reproduction, offering a framework for the development of life history strategies to overcome hard weather conditions. Among them, prolonged female cycles, asynchrony between sexes in receptivity, and sperm storage in males, were described. Asynchrony in the reproductive timing between males and females is a consequence of different energy requirements for gametogenesis, and often imply the existence of cellular mechanisms to enhance fertilization, such as t

Inhibition of proteasome activity blocks cell cycle progression at specific phase boundaries in African trypanosomes.

Proteasomes are one of the cellular complexes controlling protein degradation from archaebacteria to mammalian cells. We recently purified and characterized the catalytic core of the proteasome, the 20S form, from Trypanosoma brucei, a flagellated protozoa which causes African trypanosomiasis. To identify the role of proteasomes in African trypanosomes, we used lactacystin, a specific inhibitor of proteasome activity. Lactacystin showed potent inhibition of the activity of 20S proteasomes purified from both bloodstream and procyclic (insect) forms of T. brucei (IC50 = 1 microM). It also inhibi

EMG-activity and muscular performance of lower leg during stretch-shortening cycle after cooling.

To test the effect of cooling on EMG-activity of muscles working as an agonist and antagonist in the lower leg, 12 men dressed in shorts and jogging shoes performed a drop-jump exercise after 60 min exposures to 27 degrees C and 10 degrees C. Cooling decreased mean skin temperature 5.6 +/- 0.4 degrees C (mean +/- SD, P < 0.001), whereas rectal temperature was unaffected. The muscle temperature measured from m. gastrocnemius medialis decreased 4.1 +/- 0.3 degrees C (P < 0.01) at the depth of 30 mm below skin surface. To find the optimal stretching velocity for potentiation of elastic energy, th

Human embryonic stem cell-derived cardiovascular progenitor cells stimulate cardiomyocyte cell cycle activity via activating the PI3K/Akt pathway.

Promoting endogenous cardiomyocyte proliferation is crucial for repairing infarcted hearts. Implantation of human pluripotent stem cell-derived cardiovascular progenitor cells (hCVPCs) promotes healing of infarcted hearts. However, little is known regarding their impact on host cardiomyocyte proliferation. Here, we revealed that hCVPC implantation into mouse infarcted hearts induced dedifferentiation and cell cycle re-entry of host cardiomyocytes, which was further confirmed in vitro by hCVPC-conditioned medium. Mechanistically, the PI3K/Akt signaling pathway mediated hCVPC-induced cardiomyocy

The clock gene cycle synchronizes circadian metabolic oscillations uniquely in reproductively active Culex pipiens females.

The circadian clock regulates changes in daily metabolite abundance, coordinating animal activity, feeding and rest. The Northern house mosquito, Culex pipiens, also displays seasonal changes between periods of reproductive activity and overwintering diapause, regulated by changes in daylength. However, differences in metabolic oscillations between diapause and non-diapause have yet to be characterized. We hypothesized that reproductively active, non-diapausing mosquitoes would show large metabolic oscillations compared to diapausing mosquitoes. Our previous work demonstrated that CYCLE regula
