Cystic adventitial disease.

Cystic adventitial disease represents an unusual cause of peripheral vascular insufficiency. A mucinous cyst in the outer media or adventitia progressively compromises the arterial lumen. The therapy can be either aspiration of the cyst or resection with graft interposition. We present two cases involving the popliteal artery that were treated by resection. This lesion may be recognized by its angiographic appearance associated with a lack of generalized atherosclerotic disease.

[Isolation, culture and validation of CD34+ vascular wall-resident stem cells from mice].

Vascular wall-resident stem cells (VW-SCs) play a critical role in maintaining normal vascular function and regulating vascular repair. Understanding the basic functional characteristics of the VW-SCs will facilitate the study of their regulation and potential therapeutic applications. The aim of this study was to establish a stable method for the isolation, culture, and validation of the CD34+ VW-SCs from mice, and to provide abundant and reliable cell sources for further study of the mechanisms involved in proliferation, migration and differentiation of the VW-SCs under various physiological

Regional delamination strength in the human aorta underlies the anatomical localization of the dissection channel.

Aortic dissection is a life-threatening event, during which a primary tear propagates along the aorta causing catastrophic delamination of the inner (intima with most of the media) from the outer layers (leftover media with adventitia). Our understanding of mode-I fracture resistance at different aortic regions is incomplete, although the anatomical localization of the dissection channel may be assigned to this factor. To determine whether the susceptibility to dissection propagation varied with aortic region, the average and standard deviation of peel tension (indices of adhesive strength bet

Behçet's disease with ruptured anterior communicating artery aneurysm following ruptured thoracic aortic aneurysm.

A 48-year-old Japanese woman with Behçet's disease suffered successive rupture of thoracic aortic and cerebral aneurysms within one year. The patient underwent successful surgical treatment for both aneurysms. Histological examination of the aneurysm walls revealed lymphocyte infiltration into the adventitia with smooth muscle cell hyperplasia in the thickened intima. This is an extremely unusual presentation of Behçet's disease associated with both cerebral and aortic aneurysms.

Diffuse pulmonary angiomatosis.

A six year old girl with diffuse pulmonary angiomatosis presented with haemoptysis and diffuse interstitial changes with bilateral pleural effusions on the chest radiograph. The lung lesion as seen on biopsy specimens (and confirmed at necropsy) consisted of bloodless, thin walled, endothelium lined channels, affecting the interstitial septae, pleura, bronchi, and adventitia of large vessels. There was no response to oral corticosteroids or a trial of cyclosphamide. This lesion may be an example of an angiogenic disease.

Vascular adventitial cells and vascular remodeling: when a harp of thousand strings does not keep in tune so long.

The tunica adventitia of blood vessels is a highly heterogeneous microenvironment comprising diverse cell types. Beyond providing structural scaffolding, adventitial fibroblasts act as dynamic, responsive cells that contribute to vascular homeostasis mainly through regulation of collagen, fibronectin and elastin expression and paracrine effects that promote vascular inflammation. Their dysregulation leads to the onset and progression of several diseases. Signaling pathways in adventitial fibroblasts, particularly DDR2, hypoxia-ERK1/2 MAPK and PI3K/Akt signaling, impact cell proliferation, pro-

Three-Dimensional Imaging of Aortic Tissues in Atherosclerosis.

Recent research has advanced the understanding of atherosclerosis as a transmural chronic inflammatory disease involving all three layers of the arterial wall, including the intima plaque, the media, and the adventitia, which forms the outer connective tissue coat of arteries. Our recent studies have suggested that the adventitia is used by the peripheral nervous system as a conduit for reaching all tissue cells. We also found that the peripheral nervous system, that is, the sensory and sympathetic nervous system, undergoes major remodeling processes involving the neogenesis of axon networks a

Pathological Study on the Expression of Vasohibins in Peripheral Artery Disease.

Vasohibin-2 (VASH2) is a gene that promotes local angiogenesis. The tubulin carboxypeptidase activity of vasohibin causes detyrosination of alpha-tubulin and may play an important role in the regulation of various phenomena. Pathological and therapeutic angiogenesis are involved in atherosclerotic lesions. This study aimed to investigate whether the expression of VASH2 is associated with peripheral artery disease (PAD) in relation to angiogenesis, tubulin detyrosination, and severity of atherosclerotic lesions. An analysis of femoral and tibial arteries obtained from 86 patients with PAD or ab

A stereological study of 3D printed tissues engineered from rat vaginas.

BACKGROUND: This study aims to retrieve the stereological data from rat vaginas for 3D printing tissue-engineered vaginas. METHODS: In this study, five female Sprague-Dawley rats, aged 8-12 weeks, were used to obtaining the vagina tissues. Each vagina was divided into eight segments fixed in 4% paraformaldehyde and embedded in paraffin, whose two consecutive sections of each block were stained using hematoxylin and eosin (H&E) and anti-α-actin antibody with immunohistochemistry staining, respectively. The thickness of the epithelium, lamina propria, the smooth muscle layer, and the adventitia

Dipeptidyl peptidase-4 is increased in the abdominal aortic aneurysm vessel wall and is associated with aneurysm disease processes.

BACKGROUND: Abdominal aortic aneurysm (AAA) is a potentially life-threatening disease, and until today there is no other treatment available than surgical intervention. Dipeptidyl peptidase-4 (DPP4)-inhibitors, used clinically to treat type 2 diabetes, have in murine models been shown to attenuate aneurysm formation and decrease aortic wall matrix degradation, inflammation and apoptosis. Our aim was to investigate if DPP4 is present, active and differentially expressed in human AAA. METHODS AND RESULTS: DPP4 gene expression was elevated in both media and adventitia of AAA tissue compared with

Upregulation of Plasminogen Activator Inhibitor-1 in Irradiated Recipient Arteries and Veins from Free Tissue Transfer Reconstruction in Cancer Patients.

BACKGROUND: Clinical studies have shown that radiotherapy can induce vascular disease at the site of exposure but is usually not clinically evident until years after treatment. We have studied irradiated human arteries and veins to better understand the underlying biology in search of future treatments. The aim was to investigate whether radiotherapy contributed to a sustained expression of plasminogen activator inhibitor-1 (PAI-1) in human arteries and veins. METHODS: Irradiated arteries and veins were harvested, together with unirradiated control vessels, from patients undergoing free tissu

Methodological Approach to Use Fresh and Cryopreserved Vessels as Tools to Analyze Pharmacological Modulation of the Angiogenic Growth.

The sprouting of new vessels is greatly influenced by the procedure chosen. We sought to optimize the experimental conditions of the angiogenic growth of fresh and cryopreserved vessels cultured in Matrigel with the aim to use this system to analyze the pharmacological modulation of the process. Segments of second-order branches of rat mesenteric resistance arteries, thoracic aorta of rat or mouse, and cryopreserved rat aorta and human femoral arteries were cultured in Matrigel for 7-21 days in different mediums, as well as in the absence of endothelial or adventitia layer. Adventitial transduction of lentivirus-shRNA-VEGF-A in arteriovenous fistula reduces venous stenosis formation.

Venous neointimal hyperplasia (VNH) causes hemodialysis vascular access failure. Here we tested whether VNH formation occurs in part due to local vessel hypoxia caused by surgical trauma to the vasa vasorum of the outflow vein at the time of arteriovenous fistula placement. Selective targeting of the adventitia of the outflow vein at the time of fistula creation was performed using a lentivirus-delivered small-hairpin RNA that inhibits VEGF-A expression. This resulted in significant increase in mean lumen vessel area, decreased media/adventitia area, and decreased constrictive remodeling with

Changes in tissue substance P levels in patients with carpal tunnel syndrome.

BACKGROUND: Although carpal tunnel syndrome (CTS) is the most common entrapment neuropathy in adults, its etiology is not completely known. Chronic inflammation, fibrosis of the transverse carpal ligament (TCL), and altered sensory response contribute to the symptoms. OBJECTIVE: Because substance P (SP) is known to be involved in neuropathic pain, chronic inflammation, and fibrosis, the present study evaluated changes in SP levels in patients with CTS. METHODS: TCL, median nerve adventitia, and synovial connective tissue of the middle flexor digitorum superficialis tendon samples from patien

A study of the course of the internal carotid artery in the parapharyngeal space and its clinical importance.

The differences in the course and shape of the internal carotid artery (ICA) in the parapharyngeal space were investigated to determine the possible risks for serious hemorrhage during tonsillectomy, drainage of peritonsillar abscess, soft palate injuries, adenoidectomy and velopharyngeoplasty. The course of the ICA was studied in the parapharyngeal spaces of 50 adult cadavers. From each specimen, circumferential sections were obtained and they stained with hematoxylin-eosin and Verhoeff's elastic staining. The cervical course of the ICA showed no curvature in 70 cases; but in 25 cases it had

Identification of fibrillins as a major component of coronary atherosclerotic plaques.

Fibrillins are two distinct 350 kD RGD-containing glycoproteins found in microfibrils in the extracellular matrix. Fibrillin-containing microfibrils are required for elastic fiber formation and serve as an anchor for protein binding via β1 and/or αvβ3 receptors. Fibrillin-1 is causally linked to Marfan's syndrome, which is in part characterized by disruption of the aortic media. Fibrillin-2 is causally linked to congenital contractural arachnodactyly, which is rarely characterized by cardiovascular symptoms. We hypothesized that fibrillins might be present in coronary atherosclerotic plaques a

Microanatomy and morphometry of the hydronephrotic "obstructed" renal pelvis in asymptomatic infants.

There has been limited histological study of the obstructed ureteropelvic junction in patients less than 1 year old. We present our prospective studies on the histomorphometry of the ureteropelvic junction complexes of 35 infants, of whom 23 underwent pyeloplasty for obstruction and 12 were age matched autopsy normals. Qualitatively, the major abnormalities of the pelvic microanatomy in the hydronephrotic obstructed kidneys included a lamina muscularis that was significantly thicker than normal and the presence of collagen fibers between muscle fascicles. Additionally, variable amounts of elas

Prolonged prostaglandin E1 infusion: histologic effects on the patent ductus arteriosus.

An infant with Ebstein's malformation of the tricuspid valve and severe pulmonic stenosis underwent a 39-day course of prostaglandin E1 infusion, and a histologic study of the ductus arteriosus was undertaken after autopsy. There were marked alterations in the ductal and juxtaductal structures following this prolonged infusion of prostaglandin E1. The internal elastic lamella of the ductus was disrupted in many areas. The media showed widespread areas of disruption with cavity formation. The adventitia adjacent to the junction of the ductus with the pulmonary artery was thickened and infiltrat

Enhanced imaging of porcine thoracic aorta vasa vasorum with contrast micro-computed tomography.

The vasa vasorum (VV) have gathered considerable interest over the last decade due to its role in vascular wall biology and pathology; however, while the coronary VV are relatively well studied, the anatomy of peripheral VV, such as those of the aorta, remains poorly described, hampering knowledge of their role in peripheral vascular diseases. Through careful retrieval of the porcine thoracic aorta and successful microthrombi removal, the thoracic aorta was perfused with BriteVu™ followed by micro-computed tomography (micro-CT) scanning to image the VV and obtain a 3D reconstruction. We used c
