AOP-based framework for predicting the joint action mode of di-(2-ethylhexyl) phthalate and bisphenol A co-exposure on autism spectrum disorder.

Autism spectrum disorder (ASD), also known as autism, is a common, highly hereditary and heterogeneous neurodevelopmental disorder. The global prevalence of ASD among children continues to rise significantly, which is partially attributed to environmental pollution. It has been reported that pre- or post-natal exposure to di-(2-ethylhexyl) phthalate (DEHP) or bisphenol A (BPA), two prevalent environmental endocrine disruptors, increases the risk of ASD in offspring. Yet, the joint action mode linking DEHP and BPA with ASD is incompletely understood. This study aims to unravel the joint action

Evaluation of the endocrine disrupting potential of Di-isononyl phthalate.

Low molecular weight ortho-phthalate compounds have been implicated in disruption of androgen pathways when exposure occurs during the masculinization programming window. Di-isononyl phthalate (DINP) is a high molecular weight phthalate and a high production volume chemical. To understand the potential for DINP and its metabolites to disrupt endocrine pathways, a weight of evidence assessment was conducted according to the European Chemicals Agency (ECHA)/ European Food Safety Authority (EFSA) Endocrine Disruptor Guidance (2018). Toxicological data related to estrogen (E), androgen (A), thyroi

Neurodevelopmental and neurofunctional outcomes in children with congenital diaphragmatic hernia.

The objective of this review was to provide a critical overview of our current understanding on the neurocognitive, neuromotor, and neurobehavioral development in congenital diaphragmatic hernia (CDH) patients, focusing on three interrelated clinical issues: (1) comprehensive outcome studies, (2) characterization of important predictors of adverse outcome, and (3) the pathophysiological mechanism contributing to neurodevelopmental disabilities in infants with CDH. Improved survival for CDH has led to an increasing focus on longer-term outcomes. Neurodevelopmental dysfunction has been recognize

Development of a decision flow-based read-across approach for assessing skin sensitization using publicly available tools.

Animal tests like the guinea pig maximization test (GPMT) and murine local lymph node assay (LLNA) were historically used to assess skin sensitization in cosmetics. The EU banned animal testing for cosmetics in 2013, prompting the development of alternative in vitro methods and new approach methodologies (NAMs) based on adverse outcome pathways (AOPs). These NAMs are integrated into Defined Approaches (DAs) to evaluate skin sensitization hazards and potency, which challenge is incorporating them into next-generation risk assessment (NGRA) frameworks. To address this challenge, we aim to achiev

Proteomic analysis identifies pathways related to immune dysregulation in patients with hematologic malignancies after COVID-19 infection.

Patients with hematologic malignancies (HMs) are particularly vulnerable to coronavirus disease 2019 (COVID-19) because of underlying immune dysfunction and treatment-related immunosuppression. However, proteomic features associated with different clinical trajectories in this population remain insufficiently characterized. We performed serum proteomic analysis in 40 HM patients with COVID-19 and 15 healthy controls. Compared with controls, HM patients showed impaired immune-related responses during the acute phase of COVID-19. Acute-phase proteomic patterns differed across outcome groups; how

Functional perturbation reveals context-dependent contributions of nuclear receptors to drug-induced hepatic steatosis.

Drug-induced hepatic steatosis is mediated by diverse molecular mechanisms, yet several nuclear receptors have been proposed as molecular initiating events or early key events within adverse outcome pathways for hepatic steatosis. However, direct functional evidence supporting these mechanistic roles in human-relevant experimental systems remains limited. The present study evaluated the contribution of selected nuclear receptors to drug-induced hepatic steatosis using complementary human hepatic in vitro models. Stable short hairpin RNA-mediated knockdown of individual nuclear receptors was es

The evolutionary landscape of chronic lymphocytic leukemia treated with ibrutinib targeted therapy.

Treatment of chronic lymphocytic leukemia (CLL) has shifted from chemo-immunotherapy to targeted agents. To define the evolutionary dynamics induced by targeted therapy in CLL, we perform serial exome and transcriptome sequencing for 61 ibrutinib-treated CLLs. Here, we report clonal shifts (change >0.1 in clonal cancer cell fraction, Q < 0.1) in 31% of patients during the first year of therapy, associated with adverse outcome. We also observe transcriptional downregulation of pathways mediating energy metabolism, cell cycle, and B cell receptor signaling. Known and previously undescribed mutat

Predicting the need for home gavage or g-tube feeds in asphyxiated neonates treated with therapeutic hypothermia.

BACKGROUND: Asphyxiated infants treated with therapeutic cooling can have persistent oral feeding difficulty because of involvement of neural pathways in the brainstem, cortex, and basal ganglia. The goal is to predict the composite adverse outcome of death or persistent oral feeding difficulty using precooling/cooling attributes, and the severity and distribution of hypoxic-ischemic lesions, especially brainstem lesions on post-cooling brain magnetic resonance imaging (MRI). METHODS: Retrospective review of 86 asphyxiated infants cooled from January 2006 to August 2014. Persistent feeding di

Outcomes following major dysvascular lower limb amputation: an updated systematic review.

BACKGROUND: Adults with major lower limb amputation (LLA) due to dysvascular etiology often experience diverse health outcomes. However, the absence of standardized outcome reporting limits the ability to synthesize evidence, evaluate care models, and develop consistent clinical pathways. This review updates a systematic review on outcome measures and indicators reported in this population. METHODS: A systematic review update was conducted across six databases (MEDLINE, EMBASE, EBM Reviews, PsycINFO, Web of Science, and CINAHL) from 20 April 2017 to 1 December 2022. All study designs were eli

Effects of hyperhomocysteinemia on placental morphology and function.

OBJECTIVE: Hyperhomocysteinemia (HHcy), characterized by elevated homocysteine levels, is associated with adverse pregnancy outcomes, though its mechanistic link to placental dysfunction remains unclear. This study aimed to explore how HHcy disrupts placental development to identify conserved molecular mechanisms related to adverse pregnancy outcomes. METHODS: We develop murine HHcy model using a high-methionine diet and observe placental and fetal developmental outcome at key pregnancy times (E7.5, E10.5, E13.5). Structural and functional anomalies were detected histopathologically and molec

Anti-inflammatory interventions in pregnancy: now and the future.

A growing body of evidence implicates inflammatory pathways in adverse reproductive outcomes. This expanding evidence suggests that anti-inflammatory interventions may hold promise in reducing the maternal and neonatal morbidities and mortalities associated with these obstetrical complications. Preterm birth, preeclampsia, pregnancy loss and adverse neonatal outcomes have all been associated with the activation of inflammatory pathways during pregnancy. Because of the number of observational human studies, as well as animal models of preterm birth, the mechanisms by which inflammation may prom

Adverse prognosis gene expression patterns in metastatic castration-resistant prostate cancer.

Metastatic castration-resistant prostate cancer (mCRPC) is a heterogeneous disease. Several studies have identified transcriptional subtypes of mCRPC, but comprehensive analysis of prognostic gene expression pathways has been limited. Therefore, we aggregated a cohort of 1012 mCRPC tissue samples from 769 patients and investigated the association of gene expression-based pathways with clinical outcomes and intrapatient and intratumor heterogeneity. Survival data were obtained for 272 patients. Pathway-level enrichment was evaluated using gene set variation analysis. scRNA-seq datasets from mCR

Horner's Syndrome Following Dural Puncture Epidural Analgesia for Labor.

Horner's syndrome has been identified as an adverse outcome associated with the administration of epidural analgesia during labor. This syndrome is attributed to the upward spread of the local anesthetic, which may extend toward the superior cervical sympathetic chain. This process could disrupt the sympathetic pathways supplying the facial and ocular areas. We describe a case of a 26-year-old primigravid female with transient isolated Horner's syndrome following dural puncture epidural analgesia during labor.

From etiology to outcome: investigating the link between placental pathology and clinical risk for preterm birth and neurodevelopment.

Understanding the etiology of preterm birth may provide insights into the risk for adverse neurodevelopmental outcome. This commentary contextualizes the study of El-Shahad et al alongside the complex multifactorial pathway to outcomes in the preterm infant, while noting some limitations within the study. Placental origins of preterm birth do not appear to provide major insights into risk for adverse neurodevelopmental outcome in the preterm infant.

Influence of perinatal distress on adverse birth outcomes: A prospective study in the Tigray region, northern Ethiopia.

BACKGROUND: In low-income countries, where socioeconomic adversities and perinatal distress are common, adverse birth outcomes are significant public health problems. In these settings, perinatal distress, i.e., high symptoms of anxiety, depression, and/or stress during pregnancy, may be linked with adverse birth outcomes. However, few prospective studies have investigated the impact of perinatal distress on adverse birth outcomes such as preterm birth (gestational age <37 weeks), low birth weight (<2.5 kg), and small for gestational age birth (birth weight below the 10th percentile for gestat
