Naftifine gel in the treatment of tinea pedis: two double-blind, multicenter studies. Naftifine Gel Study Group.

Three hundred and sixteen patients with a diagnosis of tinea pedis were evaluated in two multicenter, double-blind randomized studies. One hundred and fifty-nine patients applied naftifine gel 1 percent and 157 applied gel vehicle to affected areas twice daily for four weeks. At the end of treatment, the naftifine-treated groups had higher mycologic cure rates (66 percent and 63 percent) than the respective vehicle-treated groups (34 percent and 27 percent, p is less than 0.01 and p is less than 0.001). The rate of global improvement in patients' conditions and the reduction in the incidence a

Naftifine cream 1% versus econazole cream 1% in the treatment of tinea cruris and tinea corporis.

Data from 104 subjects with tinea cruris or tinea corporis were evaluated in this double-blind, randomized study. The subjects applied naftifine cream 1% or econazole nitrate cream 1% to affected areas twice daily for 4 weeks. After 1 week of treatment naftifine had an overall cure rate of 19% compared with 4% for econazole (p = 0.03). A difference in favor of naftifine, although not statistically significant after the first week, persisted throughout treatment. Two weeks after the end of treatment both medications had overall cure rates of approximately 80%. Three percent of the naftifine-tre

Topical antifungal treatments for tinea cruris and tinea corporis.

BACKGROUND: Tinea infections are fungal infections of the skin caused by dermatophytes. It is estimated that 10% to 20% of the world population is affected by fungal skin infections. Sites of infection vary according to geographical location, the organism involved, and environmental and cultural differences. Both tinea corporis, also referred to as 'ringworm' and tinea cruris or 'jock itch' are conditions frequently seen by primary care doctors and dermatologists. The diagnosis can be made on clinical appearance and can be confirmed by microscopy or culture. A wide range of topical antifungal

Endothelium-inspired coating for hemodialysis needle: Heparin-modulated sustained NO release for synergistic anti-thrombotic and anti-infective effects.

Thrombosis and infection remain the principal barriers to durable vascular access in hemodialysis, while clinical needles themselves are key drivers of these complications. Inspired by the vascular endothelium, surface modification leveraging the synergistic effect of nitric oxide (NO) and heparin has been widely adopted to address this issue. While N-diazeniumdiolate (NONOate) donors are ideal for the short duration of dialysis, their NO release often suffers from an initial burst. Here, a unique surface design leverages heparin not only to functionally complement NO, but more importantly, to

Polyelectrolyte Multilayer Coatings As a Tunable Tool to Control Inflammatory Response to Biomaterials in Vitro.

The immune response to implanted medical devices can impact their performance and longevity in the body. Instead of designing implants to produce the desired immune responses, this might more easily be achieved with the use of surface modification technology, which can alter the biological profile of the implant without compromising function or performance. In the present study, we generated two polyelectrolyte (PE) multilayer (PEM) coatings from polystyrenesulfonate (PSS) and poly(allylamine hydrochloride) (PAH). These two coatings had identical chemical compositions, but the wettability and

Complexation of novel thiomers and insulin to protect against in vitro enzymatic degradation - towards oral insulin delivery.

A significant barrier to oral insulin delivery is its enzymatic degradation in the gut. Nano-sized polymer-insulin polyelectrolyte complexes (PECS) have been developed to protect insulin against enzymatic degradation. Poly(allylamine) (Paa) was trimethylated to yield QPaa. Thiolation of Paa and QPaa was achieved by attaching either N-acetylcysteine (NAC) or thiobutylamidine (TBA) ligands (Paa-NAC/QPaa-NAC and Paa-TBA/QPaa-TBA thiomers). PEC formulations were prepared in Tris buffer (pH 7.4) at various polymer: insulin mass ratios (0.2:1-2:1). PECS were characterized by %transmittance of light

High terbinafine resistance in Trichophyton interdigitale isolates in Delhi, India harbouring mutations in the squalene epoxidase gene.

In the last few years, infections caused by dermatophytes along with a concomitant increase in the number of difficult to treat cases have increasingly been recognised, indicating that dermatophytosis remains a challenging public health problem. The majority of infections are caused by Trichophyton rubrum and Trichophyton mentagrophytes complex. Terbinafine, an allylamine antifungal used orally and topically is considered to be a first-line drug in the therapy of dermatophyte infections. Terbinafine resistance has been predominately attributed to point mutations in the squalene epoxidase (SQLE

Confined Flocculation of Ionic Pollutants by Poly(L-dopa)-Based Polyelectrolyte Complexes in Hydrogel Beads for Three-Dimensional, Quantitative, Efficient Water Decontamination.

The development of simple and recyclable adsorbents with high adsorption capacity is a technical imperative for water treatment. In this work, we have successfully developed new adsorbents for the removal of ionic pollutants from water via encapsulation of polyelectrolyte complexes (PECs) made from positively charged poly(allylamine hydrochloride) (PAH) and negatively charged poly(l-3,4-dihydroxyphenylalanine) (PDopa), obtained via the self-polymerization of l-3,4-dihydroxyphenylalanine (l-Dopa). Given the outstanding mass transport through the hydrogel host matrixes, the PDopa-PAH PEC guests

Development of a new medium useful for the recovery of dermatophytes from clinical specimens by minimizing the carryover effect of antifungal agents.

Two surface-active compounds, egg lecithin and polysorbate 80, usually used as the deactivators of various preservatives were tested whether they also counteract either or all of the three major topical antifungal drugs, bifonazole (BFZ), lanoconazole (LCZ) and terbinafine (TBF). Both egg lecithin and polysorbate 80, when added to culture media up to final concentrations of 1.0 and 0.7%, respectively, antagonized the anti-dermatophytic activity of the three drugs in a concentration-dependent manner. A greater extent of antagonistic action was exerted when the two deactivators combined at their

Once daily treatment with terbinafine 1% cream (Lamisil) for one week is effective in the treatment of tinea corporis and cruris. A placebo-controlled study.

Duration of therapy is an important factor in determining patients' compliance in dermatomycosis. Terbinafine (Lamisil) is an allylamine antifungal agent. Its fungicidal properties against dermatophytes should allow physicians to reduce treatment duration without affecting the cure rate. This study was carried out to determine the efficacy and tolerability of terbinafine 1% cream, applied once daily for 7 days, in adult patients with tinea corporis/cruris. In a multicentre, randomized, double-blind, parallel-group study, patients with a clinical diagnosis of tinea corporis/cruris confirmed by

The antimycotic drug terbinafine in contrast to ketoconazole lacks acute effects on the pituitary-testicular function of healthy men: a placebo-controlled double-blind trial.

Among the side-effects of the antimycotic drug ketoconazole, inhibition of testosterone biosynthesis is highly pronounced. The decrease in testosterone may cause impotence and gynecomastia, and this side-effect has been exploited in drug tests for the treatment of androgen-dependent tumours. Terbinafine, an allylamine derivate, from a new group of antifungal substances, did not show similar inhibiting effects on steroid synthesis in vitro and in vivo in animal experiments. In a double-blind, placebo-controlled study the influence of terbinafine and ketoconazole on the pituitary-testicular axis

Persistent and recurrent tinea corporis in children treated with combination antifungal/ corticosteroid agents.

BACKGROUND: Combination antifungal/corticosteroid preparations are widely used by nondermatologists in the treatment of superficial fungal infections in patients of all ages. Over half of the prescriptions written for the most commonly used combination agent clotrimazole 1%/betamethasone diproprionate 0.05% cream (Lotrisone, Schering, Kenilworth, NJ) were prescribed for children younger than 4 years old. Our pediatric dermatology division has recently encountered a series of children with recurrent or persistent tinea corporis, especially tinea faciei, treated initially with combination antifu

Genetic confirmation of terbinafine resistance in Trichophyton rubrum mediated by the squalene epoxidase Leu393Phe mutation via targeted gene replacement.

Terbinafine (TBF) is a first-line allylamine antifungal used to treat dermatophytosis caused by Trichophyton species, targeting squalene epoxidase (SQLE). The recent emergence of TBF-resistant Trichophyton rubrum (T. rubrum) strains represents a significant global clinical challenge. Although the SQLE Leu393Phe substitution is a commonly reported resistance mechanism in Japan, direct genetic evidence demonstrating its effect when introduced into a T. rubrum recipient strain has been lacking. In this study, we established the causal relationship between the SQLE Leu393Phe substitution and TBF r

Antimicrobial efficiency of pH-responsive PAA/PAH-coated and flucloxacillin loaded titanium implants in the Galleria mellonella model.

Implant-associated infections are a leading cause of implant failure in orthopedics and dentistry, often necessitating revision surgery. These infections arise primarily from biofilm formation on implant surfaces, which shields pathogenic microorganisms from host immunity and systemic antibiotics. To address this issue, we used a pH-responsive poly(acrylic acid)/poly(allylamine hydrochloride) (PAA/PAH) coating applied to nanostructured titanium rods with controlled nanotube pore sizes and loaded with flucloxacillin (FCX) to address Staphylococcus aureus infections. The release behavior of FCX

Surface Chemistry-Dependent Binding Interactions between Kraft Lignin and Polyelectrolyte-Encapsulated Gold Nanoparticles.

This study investigated the relationship between gold nanoparticle (AuNP) surface chemistry and the binding affinity of unfractionated kraft lignin to polyelectrolyte-coated AuNPs. Specifically, fluorescence quenching titrations were employed to determine lignin's binding affinity (K a) to different charged polymer surfaces displayed on 90 nm citrate-stabilized AuNPs, (poly-(allylamine hydrochloride)) (PAH), polyacrylate (PAA), and poly-(diallyldimethylammonium chloride) (PDADMAC). AuNP-lignin conjugates were characterized by UV-vis absorbance spectroscopy, ζ-potential analysis, and dynamic li

Reagentless Sensing of Vancomycin Using an Indium Tin Oxide Electrode Grafted with Molecularly Imprinted Polymer including Ferrocenyl Group.

Vancomycin (VCM) is a first-line antimicrobial agent against methicillin-resistant Staphylococcus aureus, a cause of nosocomial infections. Therapeutic drug monitoring is strongly recommended for VCM-based chemotherapy. The authors attempted to develop a simple VCM sensor based on molecularly imprinted polymer (MIP), which can be used with simple operations. Methacrylic acid (MAA), acrylamide, methylenebisacrylamide, and allylamine carboxypropionate-3-ferrocene (ACPF) were copolymerized in the presence of VCM and grafted from the surface of indium-tin oxide (ITO) to obtain MIP-coated electrode

Antifungal agents for onychomycosis: new treatment strategies to improve safety.

Onychomycosis is a common and difficult-to-treat fungal infection of the nail unit that gradually leads to dystrophic changes of the nail plate and nail bed. If untreated, infection progresses and may lead to discomfort, reduced quality of life, and risk of complications in patients with comorbid conditions (eg, diabetes, human immunodeficiency virus, peripheral vascular disease). Onychomycosis treatments are designed to eradicate causative pathogens (most commonly Trichophyton rubrum and Trichophyton mentagrophytes), restore healthy nails, and prevent recurrence or spread of infection. Given

Terbinafine Hydrochloride Trans-ungual Delivery via Nanovesicular Systems: In Vitro Characterization and Ex Vivo Evaluation.

Treating a nail infection like onychomycosis is challenging as the human nail plate acts as a formidable barrier against all drug permeation. Available oral and topical treatments have several setbacks. Terbinafine hydrochloride (TBH), belonging to the allylamine class, is mainly used for treatment of onychomycosis. This study aims to formulate TBH in a nanobased spanlastic vesicular carrier that enables and enhances the drug delivery through the nail. The nanovesicles were formulated by ethanol injection method, using either Span® 60 or Span® 65, together with Tween 80 or sodium deoxycholate
