[Mechanisms of retinal photoreceptor cell fate determination].

PURPOSE: Understanding the molecular mechanisms by which distinct cell fate is determined during organogenesis is a central issue in cell development and disease. Using conditional gene ablation, we show that in mice the transcription factor Otx2 is essential for retinal photoreceptor cell fate determination and the development of the pineal gland. METHODS AND RESULTS: In the Otx2 knockout mice, differentiating photoreceptor cells were converted to amacrine-like neurons, and the pinealocytes in the pineal gland were totally absent. In addition, promoter assays revealed that Otx2 transactivate

A pathoconnectome of early neurodegeneration: Network changes in retinal degeneration.

Connectomics has demonstrated that synaptic networks and their topologies are precise and directly correlate with physiology and behavior. The next extension of connectomics is pathoconnectomics: to map neural network synaptology and circuit topologies corrupted by neurological disease in order to identify robust targets for therapeutics. In this report, we characterize a pathoconnectome of early retinal degeneration. This pathoconnectome was generated using serial section transmission electron microscopy to achieve an ultrastructural connectome with 2.18nm/px resolution for accurate identific

[A Potential Mechanism for Spontaneous Oscillation in the Abnormal Retina].

Rhythmic neural activities are observed in many brain regions, and these are considered to play an important role in neural information processing. On the other hand, distinct rhythmic neural activities emerge under several pathological conditions, suggesting that rhythmic neural activity has a close relation to brain function and dysfunction. In many pathological cases, the intrinsic property of unusual rhythm generation in a neuron or a neuronal network is prevented under normal conditions, but released by the pathological condition. Therefore, it may be useful to explore which conditions de

Mammalian Retinal Bipolar Cells: Morphological Identification and Systematic Classification in Rabbit Retina With a Comparative Perspective.

Retinal bipolar cells (BCs) convey visual signals from photoreceptors to more than 50 types of rabbit retinal ganglion cells (Famiglietti, 2020). More than 40 years ago, 10-11 types of bipolar cells were recognized in rabbit and cat retinas (Famiglietti, 1981). Twenty years later, 10 were identified in mouse, rat, and monkey, while recent molecular genetic studies indicate that there are 15 types of bipolar cell in mouse retina (Shekhar et al., 2016). The present detailed study of more than 800 bipolar cells in 11 Golgi-impregnated rabbit retinas indicates that there are 14-16 types of cone bi

Involvement of retinal neurons and pigment epithelial cells in a murine model of sandhoff disease.

BACKGROUND/AIMS: To investigate the effects of deficient degradation of glycolipids on the morphological appearance of all retinal cells in a murine model of G(M2) gangliosidosis (Sandhoff disease). METHODS: The morphological appearance of the retina in Sandhoff mice at symptomatic stages (3 and 4 months of age) was examined by immunohistochemistry, lectin histochemistry and electron microscopy. RESULTS: Under a light microscope, intense immunoreactivity for G(M2) ganglioside was observed in the ganglion cell, inner plexiform, and inner nuclear layers in the Sandhoff mice. The ganglion cell

LIM-Homeodomain Transcription Factor LHX4 Is Required for the Differentiation of Retinal Rod Bipolar Cells and OFF-Cone Bipolar Subtypes.

Retinal bipolar cells (BCs) connect with photoreceptors and relay visual information to retinal ganglion cells (RGCs). Retina-specific deletion of Lhx4 in mice results in a visual defect resembling human congenital stationary night blindness. This visual dysfunction results from the absence of rod bipolar cells (RBCs) and the loss of selective rod-connecting cone bipolar cell (CBC) subtypes and AII amacrine cells (ACs). Inactivation of Lhx4 causes the apoptosis of BCs and cell fate switch from some BCs to ACs, whereas Lhx4 overexpression promotes BC genesis. Moreover, Lhx4 positively regulates

Spatiotemporal coordination of Slit-Robo repulsion and neurturin-Gfrα attraction guides multipolar migration during retinal lamination.

Multipolar migration is a conserved neuronal migration mode in the developing brain, enabling emerging neurons to navigate in crowded environments and reach precise laminar positions. Yet, how these cells interpret external cues to guide their migration is not fully understood. We investigate this question using retinal horizontal cells as a model. Combining transcriptomics, targeted CRISPR screening, and live imaging, we reveal the spatiotemporal guidance system underlying horizontal cell lamination: repulsive Slit1b/2-Robo2 signaling in the amacrine cell layer initiates apical horizontal cel

Ultrastructural study on centrifugal fibers in the feline retina.

Morphological studies were carried out to elucidate whether centrifugal fibers are present in the cat retina. Unilateral optic nerve transection with the minimum vascular damage was performed at 6 mm behind the eyeball via the soft palate approach in adult specific pathogen free cats. Light and electron microscopic observations were performed at 40 and 96 hours after the optic nerve transection. At 40 hours, when there was no evidence of degenerative change in the retinal ganglion cells, degenerative changes were observed in the neuronal processes in the outer part of the inner plexiform layer

The M5 Cell: A Color-Opponent Intrinsically Photosensitive Retinal Ganglion Cell.

Intrinsically photosensitive retinal ganglion cells (ipRGCs) combine direct photosensitivity through melanopsin with synaptically mediated drive from classical photoreceptors through bipolar-cell input. Here, we sought to provide a fuller description of the least understood ipRGC type, the M5 cell, and discovered a distinctive functional characteristic-chromatic opponency (ultraviolet excitatory, green inhibitory). Serial electron microscopic reconstructions revealed that M5 cells receive selective UV-opsin drive from Type 9 cone bipolar cells but also mixed cone signals from bipolar Types 6,

Intravitreal MPTP drives retinal ganglion cell loss with oral nicotinamide treatment providing robust neuroprotection.

Neurodegenerative diseases have common underlying pathological mechanisms including progressive neuronal dysfunction, axonal and dendritic retraction, and mitochondrial dysfunction resulting in neuronal death. The retina is often affected in common neurodegenerative diseases such as Parkinson's and Alzheimer's disease. Studies have demonstrated that the retina in patients with Parkinson's disease undergoes changes that parallel the dysfunction in the brain. These changes classically include decreased levels of dopamine, accumulation of alpha-synuclein in the brain and retina, and death of dopa

Phenotypic expression of photoreceptor and endocrine cell properties by cultured pineal cells of the newborn rat.

Pineal glands of newborn rats were dissociated and maintained under cell culture conditions. The phenotypic expression of both photoreceptor and endocrine cell properties was investigated using immunohistochemical techniques (specific antibodies against opsin or serotonin). After one week in culture, a number of small round cells appeared on top of a sheet of flat epithelium. Among those cells, opsin-like immunoreactive cells were observed. These cells showed a neuron-like morphology with neuritic processes and often formed rosettes. Immunoreactivity was found on the plasma membrane of both th

Characterization of the cell death promoter, Bad, in the developing rat retina and forebrain.

Neuronal programmed cell death, or apoptosis, occurs during development, following injury or in certain disease processes, and is regulated by members of the B-cell leukemia-2 (Bcl-2) protein family. These molecules include both positive and negative regulators of cell death and act by selective dimerization that results in permissive or inhibitory effects on a cascade of cellular events, including mitochondrial release of cytochrome c, stimulation of cysteine protease activity and subsequent cellular deterioration. Here, we have characterized the expression of the cell death agonist, Bad, in

Elevated PRDM13 Disrupts Photoreceptor Function and Survival in the Mammalian Retina.

PURPOSE: Retinal degeneration is a common cause of blindness, but there is a gap in our understanding of the molecular mechanisms causing degeneration. Dysregulated PRDM13 has been linked to retinal dystrophy, indicating a role for PRDM13 in the retina. PRDM13 knockout studies have shown that PRDM13 specifies amacrine cell fates, but no studies have shown the phenotypic and mechanistic outcomes of its elevated activity in the retina. METHODS: We developed a mouse model to induce aberrant PRDM13 expression in a controlled, time-dependent manner. ERG and histological analyses were performed to

Global Ca2+ signaling drives ribbon-independent synaptic transmission at rod bipolar cell synapses.

Ribbon-type presynaptic active zones are a hallmark of excitatory retinal synapses, and the ribbon organelle is thought to serve as the organizing point of the presynaptic active zone. Imaging of exocytosis from isolated retinal neurons, however, has revealed ectopic release (i.e., release away from ribbons) in significant quantities. Here, we demonstrate in an in vitro mouse retinal slice preparation that ribbon-independent release from rod bipolar cells activates postsynaptic AMPARs on AII amacrine cells. This form of release appears to draw on a unique, ribbon-independent, vesicle pool. Exp

Cellular Characterization and Interspecies Evolution of the Tree Shrew Retina across Postnatal Lifespan.

Tree shrews (TSs) possess a highly developed visual system. Here, we establish an age-related single-cell RNA sequencing atlas of retina cells from 15 TSs, covering 6 major retina cell classes and 3 glial cell types. An age effect is observed on the cell subset composition and gene expression pattern. We then verify the cell subtypes and identify specific markers in the TS retina including CA10 for bipolar cells, MEGF11 for H1 horizontal cells, and SLIT2, RUNX1, FOXP2, and SPP1 for retinal ganglion cell subpopulations. The cross-species analysis elucidates the cell type-specific transcriptiona

Variation in GABA mini amplitude is the consequence of variation in transmitter concentration.

Miniature postsynaptic currents (minis) in cultured retinal amacrine cells, as in other central neurons, show large variations in amplitude. To understand the origin of this variability, we have exploited a novel form of synapse in which pre- and postsynaptic receptors sample the same quantum of transmitter. At these synapses, mini amplitudes measured simultaneously in the 2 cells show a strong correlation, accounting for, on average, more than half of the variance in amplitude. Two pieces of evidence support the conclusion that variations in the amount of transmitter in different quanta under

Dicer is essential for proper maturation, composition, and function in the postnatal retina.

The impact of Dicer/miRNAs during postnatal retinogenesis is unknown. This study aimed to deplete Dicer in postnatal retinal progenitor/precursor cells (RPCs/PCs) and to evaluate the structure and function of the adult Dicer-cKO retina using optical coherence tomography (OCT), electroretinography (ERG), and histology. We found impaired rod function, a significantly reduced number of rod bipolar cells and their associated functions, a decreased Müller glia population, and an increased population of HuC/D + Pax6/+ amacrine cells in the adult Dicer-cKO retina. Transcriptional analyses conducted i

Metabolic regulation of visual acuity.

Photoreceptors signal ON and OFF pathways via a synapse with bipolar cells that are transmitted to retinal ganglion cells (RGCs) for luminance and contrast detection. Retinal neurons metabolize glucose whose transport is mediated by photoreceptor contact with the adjacent retinal pigment epithelium (RPE). Rod loss in retinitis pigmentosa (RP) reduces RPE contact, diminishing glucose transport. We show diminished glucose leads to light hyperresponsiveness driven by deregulated ON cone bipolar signaling. Transmission of this constitutive signal to RGCs causes ON > OFF signaling imbalance and fai

[Endocrine-cell stomach tumors].

Screening of the endocrine cell participation in the stomach carcinoma has been performed. Endocrine cells are found in all stomach tumors and those in which these cells occupy more than 75% of the surface are distinguished as endocrine cell carcinomas (ECC). They are subdivided into well (WD), moderately (MD) and poorly differentiated (PD). ECC are more frequently observed in males, their predominant location is cardia and fundus. The growth in the deep parts of mucosa and submucosa (this determines late clinical symptoms) is characteristic for these tumors. Alveolar, trabecular and glandular
