Effects of lesions of the perirhinal cortex on amygdala kindling in rats.

The perirhinal cortex (PRC), the region of temporal cortex adjacent to the rhinal sulcus, has been suggested as a critical substrate for the development and expression of generalized motor seizures in the late stages of kindling development. For further investigation of the role of the PRC in limbic kindling, excitotoxic lesions centered on PRC by microinjection of ibotenate were performed in rats 2 weeks before onset of amygdala kindling. Rats with large bilateral or unilateral PRC lesions showed the same rate and pattern of kindling development as sham-lesioned controls. The only significant

Laterality of dopamine function and neuroleptic action in the amygdala in the rat.

Rats selected according to turn preference in an open field were categorised as showing left or right hemispheric dominance (turning to right or left respectively). Dopamine was persistently infused into the central area of the left or right amygdala of animals showing left or right hemispheric dominance. Infusion was effected for 9 days by Alzet osmotic minipumps, implanted subcutaneously, which delivered 25 micrograms/24 hr of dopamine via chronically-indwelling, stereotaxically-located injection units. Dopamine caused marked and consistent hyperactivity only when infused into the left amygd

Control of innate olfactory valence by segregated cortical amygdala circuits.

Animals exhibit innate behaviors that are stereotyped responses to specific evolutionarily relevant stimuli in the absence of prior learning or experience. The posterolateral cortical amygdala (plCoA) mediates innate attraction and aversion to odor. Here, we sought to define the circuit features of plCoA that give rise to innate attraction and aversion to odor in mice. First, we examined odor-evoked responses in these areas and found sparse encoding of odor identity, but not valence. We next considered a topographic organization and found that optogenetic stimulation of the anterior and poster

Basolateral amygdala neurons await oscillatory recruitment into valence-relevant ensembles.

Impaired valence processing is a core feature of psychiatric illnesses. The ability to rapidly evaluate situations and stimuli and determine whether they have positive or negative implications, termed valence processing, is a highly adaptive brain function that is essential for survival. Despite its importance, we still lack a complete understanding of the neural computations involved. The basolateral amygdala (BLA) plays a critical role in valence processing and valence ensembles have been identified by their anatomical location within the BLA, their projection targets, their genetic identity

Maternal Prenatal Depressive Symptoms and Fetal Amygdala-Cerebellum Functional Connectivity.

OBJECTIVE: Prenatal maternal depression (PMD) is a common public health concern, affecting up to 20% of pregnancies worldwide. Children exposed to PMD exhibit early self-regulatory and emotional difficulties, although its impact on the developing fetal brain remains largely unexplored. The amygdala and cerebellum are both implicated in emotion regulation and psychiatric vulnerability, are densely interconnected, and undergo rapid maturation in utero. Characterizing the relationship of PMD symptoms to amygdala-cerebellar connectivity before birth offers a unique opportunity to isolate in utero

Amygdala Function, Blood Flow, and Functional Connectivity in Nonclinical Schizotypy.

BACKGROUND AND HYPOTHESIS: Schizotypy can be utilized as a phenotypic risk marker for schizophrenia and its spectrum and might relate to putative dimensional biological markers of the psychosis spectrum. Among these are amygdala function and structure, which are impaired in schizophrenia, but possibly also correlated with subclinical expression of schizotypy in nonclinical samples. We tested whether different parameters relating to amygdala function would be different in healthy subjects with relatively higher vs lower schizotypy traits. STUDY DESIGN: Sixty-three psychiatrically healthy subje

Central amygdala astrocyte plasticity underlies GABAergic dysregulation in ethanol dependence.

Dependence is a hallmark of alcohol use disorder characterized by excessive alcohol intake and withdrawal symptoms. The central nucleus of the amygdala (CeA) is a key brain structure underlying the synaptic and behavioral consequences of ethanol dependence. While accumulating evidence suggests that astrocytes regulate synaptic transmission and behavior, there is a limited understanding of the role astrocytes play in ethanol dependence. The present study used a combination of viral labeling, super resolution confocal microscopy, 3D image analysis, and slice electrophysiology to determine the ef

Amygdala-centered fusional connections characterized nonmotor symptoms in Parkinson's disease.

The importance of nonmotor symptoms in understanding the pathogenesis of the heterogeneity of Parkinson's disease has been highlighted. However, the validation of specific brain network biomarkers in nonmotor symptom subtypes is currently lacking. By performing a new approach to compute functional connectivity with structural prior using magnetic resonance imaging, the present study computed both functional connectivity and fusional connectivity features in the nonmotor symptom subtypes of Parkinson's disease, one characterized by cognitive impairment with late onset and the other depression w

Activation of Centromedial Amygdala GABAergic Neurons Produces Hypotension in Mice.

The central amygdala (CeA) is a crucial modulator of emotional, behavioral, and autonomic functions, including cardiovascular responses. Despite its importance, the specific circuit by which the CeA modulates blood pressure remains insufficiently explored. Our investigations demonstrate that photostimulation of GABAergic neurons in the centromedial amygdala (CeMGABA), as opposed to those in the centrolateral amygdala (CeL), produces a depressor response in both anesthetized and freely-moving mice. In addition, activation of CeMGABA axonal terminals projecting to the nucleus tractus solitarius

Anterograde amnesia recurrence in temporal lobe epilepsy with amygdala-enlargement.

Temporal lobe epilepsy (TLE) can cause different types of memory impairments. Here, we report a case of immediate improvement of memory impairment following antiepileptic drug (AED) treatment in a patient with TLE with amygdala enlargement (TLE-AE), who rapidly developed recurrence. The patient was a man in his 60s whose family members complained of his amnesia. Neuropsychological investigations detected obvious recent and remote memory loss and executive function impairments. Our examinations revealed evidence of TLE and bilateral amygdala enlargement without any results suggesting organic di

Amygdala lesions are associated with improved mood after epilepsy surgery.

Neuroimaging studies in healthy and clinical populations strongly associate the amygdala with emotion, especially negative emotions. The consequences of surgical resection of the amygdala on mood are not well characterized. We tested the hypothesis that amygdala resection would result in mood improvement. In this study, we evaluated a cohort of 52 individuals with medial temporal lobectomy for intractable epilepsy who had resections variably involving the amygdala. All individuals achieved good post-surgical seizure control and had pre- and post-surgery mood assessment with the Beck Depression

Sleep deprivation altered encoding of basolateral amygdala on fear acquisition.

Sleep deprivation (SD) may lead to the development of fear- and anxiety-related emotional disorders. However, the neural mechanisms underlying the effects of SD on fear acquisition are unclear. Here, we tested whether and how SD influences the behavioral and neural manifestations of fear acquisition. We found that subjective fear ratings and objective fear indices (skin conductance response [SCR]) in the SD group were greater than those in the control group during fear acquisition, suggesting that SD facilitated fear acquisition (nSD = 18 and ncontrol = 23 for self-reported rating analysis; nS

Cue- versus reward-encoding basolateral amygdala projections to nucleus accumbens.

In substance use disorders, drug use as unconditioned stimulus (US) reinforces drug taking. Meanwhile, drug-associated cues (conditioned stimulus [CS]) also gain incentive salience to promote drug seeking. The basolateral amygdala (BLA) is implicated in both US- and CS-mediated responses. Here, we show that two genetically distinct BLA neuronal types, expressing Rspo2 versus Ppp1r1b, respectively, project to the nucleus accumbens (NAc) and form monosynaptic connections with both dopamine D1 and D2 receptor-expressing neurons. While intra-NAc stimulation of Rspo2 or Ppp1r1b presynaptic terminal

Abnormal amygdala functional connectivity in MDD patients with insomnia complaints.

BACKGROUND: Insomnia is one of the major symptom relevant factors in major depressive disorder (MDD), but the neurological mechanisms underlying the multiple effect between insomnia and depression have not been well interpreted. This study aimed at exploring the potential mechanisms between insomnia and depression based on amygdala-based resting-state functional connectivity (RSFC). METHODS: In total 56 MDD patients with low insomnia (MDD-LI) patients, 46 MDD patients with high insomnia (MDD-HI) patients, and 57 healthy controls (HCs) were employed and underwent a resting-state functional mag

Regulation of learned fear expression through the MgN-amygdala pathway.

Heightened fear responding is characteristic of fear- and anxiety-related disorders, including post-traumatic stress disorder. Neural plasticity in the amygdala is essential for both initial fear learning and fear expression, and strengthening of synaptic connections between the medial geniculate nucleus (MgN) and amygdala is critical for auditory fear learning. However, very little is known about what happens in the MgN-amygdala pathway during fear recall and extinction, in which conditional fear decreases with repeated presentations of the auditory stimulus alone. In the present study, we fo

Associations between amygdala nuclei volumes, psychosis, psychopathy, and violent offending.

The amygdala is involved in fear perception and aggression regulation, and smaller volumes have been associated with psychotic and non-psychotic violence. We explored the relationship between amygdala nuclei volumes in violent offenders with and without psychosis, and the association to psychopathy traits. 3T MRI scans (n = 204, males, 18-66 years) were obtained from psychotic violent offenders (PSY-V, n = 29), non-psychotic violent offenders (NPV, n = 19), non-violent psychosis patients (PSY-NV, n = 67), and healthy controls (HC, n = 89). Total amygdala and 9 amygdala nuclei volumes were obta

Ethanol modulation of cortico-basolateral amygdala circuits: Neurophysiology and behavior.

This review highlights literature relating the anatomy, physiology, and behavioral contributions by projections between rodent prefrontal cortical areas and the basolateral amygdala. These projections are robustly modulated by both environmental experience and exposure to drugs of abuse including ethanol. Recent literature relating optogenetic and chemogenetic dissection of these circuits within behavior both compliments and occasionally challenges roles defined by more traditional pharmacological or lesion-based approaches. In particular, cortico-amygdala circuits help control both aversive a

Amygdala pathology in amyotrophic lateral sclerosis and primary lateral sclerosis.

Temporal lobe studies in motor neuron disease overwhelmingly focus on white matter alterations and cortical grey matter atrophy. Reports on amygdala involvement are conflicting and the amygdala is typically evaluated as single structure despite consisting of several functionally and cytologically distinct nuclei. A prospective, single-centre, neuroimaging study was undertaken to comprehensively characterise amygdala pathology in 100 genetically-stratified ALS patients, 33 patients with PLS and 117 healthy controls. The amygdala was segmented into groups of nuclei using a Bayesian parcellation

Reward-related dynamical coupling between basolateral amygdala and nucleus accumbens.

Recognizing reward-related stimuli is crucial for survival. Neuronal projections from the basolateral amygdala (BLA) to the nucleus accumbens (NAc) play an important role in processing reward-related cues. Previous studies revealed synchronization between distant brain regions in reward-sensitive neurocircuits; however, whether the NAc synchronizes with the BLA is unknown. Here, we recorded local field potentials simultaneously from the BLA and NAc of rats during social preference tests and an appetitive conditioning test in which explicit stimuli were associated with food. BLA-NAc coherence i

Amygdala rapid kindling impairs breathing in response to chemoreflex activation.

Temporal lobe epilepsy is often accompanied by behavioral, electroencephalographic and autonomic abnormalities. Amygdala kindling has been used as an experimental model to study epileptogenesis. Although amygdala kindling has been extensively investigated in the context of its clinical relevance to the epilepsies, potential associated respiratory alterations are not well known. Here, our main objective was to better investigate the mechanisms involved in respiratory physiology impairment in the amygdala rapid kindling (ARK) model of epileptogenesis. Male Wistar rats with electrodes implanted i
