Artificial visual environments to enhance mating success in colonised Anopheles funestus.

BACKGROUND: Establishing and maintaining laboratory colonies of the malaria vector, Anopheles funestus using newly collected material has proven challenging, in part because of their low propensity to mate in captivity. In this study we assessed how cage conditions influence the mating success of two An. funestus strains originating from different geographic areas, Angola (FANG) and Mozambique (FUMOZ). METHODS: The visual environment in adult mosquito-rearing cages was manipulated either by covering the cages in different planes with black opaque cloth (referred to as black horizons) or by pl

Gene drive-based population suppression in the malaria vector Anopheles stephensi.

Gene drives are alleles that can bias the inheritance of specific traits in target populations for the purpose of modification or suppression. Here, we construct a homing suppression drive in the major urban malaria vector Anopheles stephensi targeting the female-specific exon of doublesex, incorporating two gRNAs and a nanos-Cas9 to reduce functional resistance and improve female heterozygote fitness. Our results show that the drive was recessive sterile in both females and males, with various intersex phenotypes in drive homozygotes. Both male and female drive heterozygotes show only moderat

Anopheles hyrcanus (Diptera: Culicidae): yet another invasive mosquito species in Germany.

From August to October 2024, 62 specimens of the non-native mosquito species Anopheles hyrcanus were trapped in the federal state of Brandenburg, northeastern Germany. At one site, 59 specimens, and at a second site, 3 specimens were collected, with both sites located in floodplain areas, approximately 25 km apart. The records represent the northernmost collection sites of this species worldwide. Anopheles hyrcanus is considered a potential vector of malaria parasites, dirofilarial worms, and various viruses, although vector capacity appears to be generally low. The findings in Germany follow

Odor repellency and neuronal responses of commercial products towards Anopheles mosquitoes.

BACKGROUND: The bites from infected female Anopheles mosquitoes lead to hundreds of thousands of deaths annually. To find a human, mosquitoes rely on their keen sense of smell. Repellents are chemicals that can target a mosquito's chemosensory senses. Therefore, targeting olfaction using repellents can be an effective way to prevent their attraction to humans and subsequent biting. However, the precise mechanisms underlying the mode of action for most commonly used repellents is surprisingly not well known. Recent evidence suggests that commonly used insect repellents target Anopheles mosquito

Selection for insecticide resistance can promote Plasmodium falciparum infection in Anopheles.

Insecticide resistance is under strong selective pressure in Anopheles mosquitoes due to widespread usage of insecticides in vector control strategies. Resistance mechanisms likely cause changes that profoundly affect mosquito physiology, yet it remains poorly understood how selective pressures imposed by insecticides may alter the ability of the mosquito to host and transmit a Plasmodium infection. From pyrethroid-resistant field-derived Anopheles gambiae s.l. mosquitoes, we established resistant (RES) and susceptible (SUS) colonies by either selection for, or loss of insecticide resistance.

The sex-specific factor SOA controls dosage compensation in Anopheles mosquitoes.

The Anopheles mosquito is one of thousands of species in which sex differences play a central part in their biology, as only females need a blood meal to produce eggs. Sex differentiation is regulated by sex chromosomes, but their presence creates a dosage imbalance between males (XY) and females (XX). Dosage compensation (DC) can re-equilibrate the expression of sex chromosomal genes. However, because DC mechanisms have only been fully characterized in a few model organisms, key questions about its evolutionary diversity and functional necessity remain unresolved1. Here we report the discover

Effective Oral RNA Interference (RNAi) Administration to Adult Anopheles gambiae Mosquitoes.

RNA interference has been a heavily utilized tool for reverse genetic analysis for two decades. In adult mosquitoes, double-stranded RNA (dsRNA) administration has been accomplished primarily via injection, which requires significant time and is not suitable for field applications. To overcome these limitations, here we present a more efficient method for robust activation of RNAi by oral delivery of dsRNA to adult Anopheles gambiae. Long dsRNAs were produced in Escherichia coli strain HT115 (DE3), and a concentrated suspension of heat-killed dsRNA-containing bacteria in 10% sucrose was offere

The ecdysone receptor regulates several key physiological factors in Anopheles funestus.

BACKGROUND: Malaria is a devastating disease, transmitted by female Anopheles mosquitoes infected with Plasmodium parasites. Current insecticide-based strategies exist to control the spread of malaria by targeting vectors. However, the increase in insecticide resistance in vector populations hinder the efficacy of these methods. It is, therefore, essential to develop novel vector control methods that efficiently target transmission reducing factors such as vector density and competence. A possible vector control candidate gene, the ecdysone receptor, regulates longevity, reproduction, immunity

Mating-regulated atrial proteases control reinsemination rates in Anopheles gambiae females.

Anopheles gambiae mosquitoes are the most important vectors of human malaria. The reproductive success of these mosquitoes relies on a single copulation event after which the majority of females become permanently refractory to further mating. This refractory behavior is at least partially mediated by the male-synthetized steroid hormone 20-hydroxyecdysone (20E), which is packaged together with other seminal secretions into a gelatinous mating plug and transferred to the female atrium during mating. In this study, we show that two 20E-regulated chymotrypsin-like serine proteases specifically e

Evaluation of Pistia stratiotes fractions as effective larvicide against Anopheles mosquitoes.

Mosquito are well-known vectors that cause diseases particularly malaria and filariasis which are detrimental to human health. These vectors occur mainly in tropical countries where more than 2 billion people live in endemic regions with about one million deaths been claimed yearly from malaria and filariasis. The study is aimed at evaluating the larvicidal activity of Pistia stratiotes fractions on Anopheles mosquitoes (Diptera: Culicidae). The ethyl acetate extract of P. stratiotes was obtained through percolation process and was chromatographed to yield nine fractions. The larvicidal activi

Pure early zygotic genes in the Asian malaria mosquito Anopheles stephensi.

BACKGROUND: The Asian malaria mosquito, Anopheles stephensi, is a major urban malaria vector in the Middle East and on the Indian subcontinent. Early zygotic transcription, which marks the maternal-to-zygotic transition, has not been systematically studied in An. stephensi or any other Anopheles mosquitoes. Improved understanding of early embryonic gene expression in An. stephensi will facilitate genetic and evolutionary studies and help with the development of novel control strategies for this important disease vector. RESULTS: We obtained RNA-seq data in biological triplicates from four ear

Genome-Wide Divergence in the West-African Malaria Vector Anopheles melas.

Anopheles melas is a member of the recently diverged An. gambiae species complex, a model for speciation studies, and is a locally important malaria vector along the West-African coast where it breeds in brackish water. A recent population genetic study of An. melas revealed species-level genetic differentiation between three population clusters. An. melas West extends from The Gambia to the village of Tiko, Cameroon. The other mainland cluster, An. melas South, extends from the southern Cameroonian village of Ipono to Angola. Bioko Island, Equatorial Guinea An. melas populations are genetical

Antennal-expressed ammonium transporters in the malaria vector mosquito Anopheles gambiae.

The principal Afrotropical malaria vector mosquito, Anopheles gambiae remains a significant threat to human health. In this anthropophagic species, females detect and respond to a range of human-derived volatile kairomones such as ammonia, lactic acid, and other carboxylic acids in their quest for blood meals. While the molecular underpinnings of mosquito olfaction and host seeking are becoming better understood, many questions remain unanswered. In this study, we have identified and characterized two candidate ammonium transporter genes, AgAmt and AgRh50 that are expressed in the mosquito ant

Distinct olfactory signaling mechanisms in the malaria vector mosquito Anopheles gambiae.

Anopheles gambiae is the principal Afrotropical vector for human malaria, in which olfaction mediates a wide range of both adult and larval behaviors. Indeed, mosquitoes depend on the ability to respond to chemical cues for feeding, host preference, and mate location/selection. Building upon previous work that has characterized a large family of An. gambiae odorant receptors (AgORs), we now use behavioral analyses and gene silencing to examine directly the role of AgORs, as well as a newly identified family of candidate chemosensory genes, the An. gambiae variant ionotropic receptors (AgIRs),

Isolation of a Plasmodium vivax refractory Anopheles culicifacies strain from India.

Anopheles culicifaciessensu lato comprises five sibling species. We report the isolation of an An. culicifacies species B strain which is completely refractory to Plasmodium vivax sporogonic development and partially refractory to P. falciparum. Parasite development in this strain is arrested by a melanotic encapsulation mechanism in the mid-gut. We compare the infectivity of this refractory strain and four other species B strains from different epidemiological zones of India with P. vivax in the laboratory.

Nitric oxide metabolites induced in Anopheles stephensi control malaria parasite infection.

Malaria parasite infection in anopheline mosquitoes is limited by inflammatory levels of nitric oxide metabolites. To assess the mechanisms of parasite stasis or toxicity, we investigated the biochemistry of these metabolites within the blood-filled mosquito midgut. Our data indicate that nitrates, but not nitrites, are elevated in the Plasmodium-infected midgut. Although levels of S-nitrosothiols do not change with infection, blood proteins are S-nitrosylated after ingestion by the mosquito. In addition, photolyzable nitric oxide, which can be attributed to metal nitrosyls, is elevated after

Salivary gland-specific gene expression in the malaria vector Anopheles gambiae.

Molecular studies on the tissue-specific gene expression in the salivary glands of Anopheles gambiae may provide useful tools for the development of new strategies for the control of the most efficient malaria vector in the sub-Saharan Africa. We summarize here the results of a recent investigation focused on the isolation of secreted factors and putative receptors from the salivary glands of An. gambiae. Using the Signal Sequence Trap technique we have identified the first cDNAs specifically expressed in the An. gambiae salivary glands. Among these, four are exclusively expressed in female gl

Field studies on inversion polymorphism in Anopheles superpictus from southern Italy.

Polytene chromosome studies were carried out on various population samples of Anopheles superpictus from different localities in Southern Italy. More than 7,000 female specimens, mostly obtained from daytime collections of indoor resting mosquitoes, were successfully scored for nurse cell polytene chromosomes. Night biting samples were also examined in some localities. Only one chromosomal polymorphism, due to a paracentric inversion involving the central third of the 2L arm, was recorded in all samples. In all localities, the inverted 2La arrangement showed remarkably stable frequency althoug
