Identification of psoriasis-associated hub genes by integrated transcriptomics and machine learning.

This study aims to systematically identify key hub genes in psoriasis pathogenesis through the integration of bioinformatics, machine learning and experimental validation methods. Four psoriasis microarray datasets were retrieved from the GEO database, with differentially expressed genes identified through comparative analysis. A dual machine learning approach screened core hub genes, whose expression levels and diagnostic efficacy were validated in an independent cohort. CIBERSORT was employed for immune cell infiltration analysis, whilst a multi-database approach constructed an mRNA-miRNA-ln

Machine-learning approach to dissect the clinical heterogeneity of IBD-associated fatigue.

OBJECTIVE: Extreme and persistent fatigue affects >50% of individuals with inflammatory bowel disease (IBD), with similar prevalence across many common immune-mediated inflammatory diseases (IMIDs). Despite its ubiquity, human scientific studies have yet to fully explain the mechanistic basis of this complex symptom. One fundamental reason is our inability to account for the clinical heterogeneity and multifactorial nature of fatigue. METHODS AND ANALYSIS: We present the conceptual machine-learning (ML) framework to dissect fatigue using one of the largest prospectively captured, real-world p

Impact of Sema3A Interference on Cerebellum-Dependent Motor Associative Learning and Memory.

Semaphorin 3A (Sema3A), a known axon chemorepulsive protein during development, is localised in perineuronal nets (PNNs) in the adult brain. PNNs are condensed aggregates of extracellular matrix molecules surrounding specific types of neurons, which regulate neuroplasticity and memory. However, the role of PNN-associated Sema3A in these processes remains unclear. To address this topic, we investigated the contribution of Sema3A to cerebellum-dependent learning and memory in adult mice using the eyeblink conditioning (EBC) paradigm. We interfered with Sema3A signalling by employing: (i) a molec

Machine learning-based risk scores are associated with conversion to dementia in Veterans.

BackgroundWe previously developed ancestry-specific risk scores for undiagnosed Alzheimer's disease and related dementias (ADRD) in Black and White American (BA and WA) Veterans by applying natural language processing and machine learning (ML) to Veterans Health Administration electronic health records. Using blinded manual chart reviews, we identified an association between ADRD risk scores and probable ADRD diagnosis at the time the scores were generated. However, it was unclear whether these scores were associated with future ADRD diagnoses and mortality.ObjectiveTo evaluate whether ADRD ri

Prospective contingency explains behavior and dopamine signals during associative learning.

Associative learning depends on contingency, the degree to which a stimulus predicts an outcome. Despite its importance, the neural mechanisms linking contingency to behavior remain elusive. In the present study, we examined the dopamine activity in the ventral striatum-a signal implicated in associative learning-in a Pavlovian contingency degradation task in mice. We show that both anticipatory licking and dopamine responses to a conditioned stimulus decreased when additional rewards were delivered uncued, but remained unchanged if additional rewards were cued. These results conflict with con

Integrating bioinformatics and machine learning to discover sumoylation associated signatures in sepsis.

Small Ubiquitin-like MOdifier-mediated modification (SUMOylation) is associated with sepsis; however, its molecular mechanism remains unclear. Herein, hub genes and regulatory mechanisms in sepsis was investigated. The GSE65682 and GSE95233 datasets were extracted from public databases. Differential analysis and Weighted Gene Co-expression Network Analysis (WGCNA) were conducted in GSE65682 to identify differentially expressed genes (DEGs) and key module genes. Candidate genes were derived by intersecting with SUMOylation-related genes (SUMO-RGs). The Least Absolute Shrinkage and Selection Ope

Deep learning-based prediction of rheumatoid arthritis-associated deformity on MRI.

INTRODUCTION: While the prevalence of surgery to correct atlantoaxial subluxation (AAS), subaxial subluxation (SAS) and vertical translocation (VT) in patients with rheumatoid arthritis (RA) had declined, cervical deformity is still observed regularly. RESEARCH QUESTION: The objective of this study is to develop a deep learning-based algorithm to predict RA-associated upper cervical spine deformity in patients before or close to RA diagnosis, with the purpose of early risk stratification. MATERIALS AND METHODS: Patients with RA in which follow-up cervical MRI studies (at least 3 years apart)

Vocal learning-associated convergent evolution in mammalian proteins and regulatory elements.

Vocal production learning ("vocal learning") is a convergently evolved trait in vertebrates. To identify brain genomic elements associated with mammalian vocal learning, we integrated genomic, anatomical, and neurophysiological data from the Egyptian fruit bat (Rousettus aegyptiacus) with analyses of the genomes of 215 placental mammals. First, we identified a set of proteins evolving more slowly in vocal learners. Then, we discovered a vocal motor cortical region in the Egyptian fruit bat, an emergent vocal learner, and leveraged that knowledge to identify active cis-regulatory elements in th

Prediction, perception, and psychosis: Application of associative learning theories to schizophrenia research.

In recent years, there have been significant advances in our understanding of the positive symptoms of schizophrenia, such as hallucinations and delusions. This progress has been significantly aided by the use of associative learning-based approaches in human subjects and preclinical animal models. Here, we first review experimental research focusing on the abnormal processing of absent stimuli using three different conditioning phenomena: conditioned hallucinations, mediated conditioning, and trace conditioning. We then review studies investigating the ability to reduce focal processing of ph

Agranulocytosis Associated With Psychiatric Polypharmacy: Lessons Learned From a Clinical Case.

Psychiatric polypharmacy involves the use of two or more psychotropic medications to manage a mental and emotional condition. The prevalence of psychotropic polypharmacy has been increasing since the 1990s and has been attributed to the rise in multiple psychiatric conditions presenting in one patient. However, as the prevalence of polypharmacy increases to maximize therapeutic advantages, so does the adverse effect profile of those drugs used in combination, leading to very life-threatening effects such as agranulocytosis. Thus, we report a case of agranulocytosis secondary to polypharmacy in

The Nature of Paired Associate Learning Deficits in Chinese Children with Developmental Dyslexia.

Previous studies have found that individuals with dyslexia perform poorly in paired associate learning (PAL) tasks, which were explained by a deficit in cross-modal association or verbal demand in alphabetic language. However, the nature of PAL deficits in non-alphabetic languages remains unclear. In this study, we conducted PAL and priming tasks in visual-visual, visual-verbal, verbal-visual, and verbal-verbal conditions to dissociate the cross-modal and verbal demands in Chinese children with dyslexia. In Experiment 1, children with dyslexia performed worse in verbal-involved PAL (visual-ver

Deep Learning Algorithms to Detect Murmurs Associated With Structural Heart Disease.

Background The success of cardiac auscultation varies widely among medical professionals, which can lead to missed treatments for structural heart disease. Applying machine learning to cardiac auscultation could address this problem, but despite recent interest, few algorithms have been brought to clinical practice. We evaluated a novel suite of Food and Drug Administration-cleared algorithms trained via deep learning on >15 000 heart sound recordings. Methods and Results We validated the algorithms on a data set of 2375 recordings from 615 unique subjects. This data set was collected in real

Predicting genes associated with RNA methylation pathways using machine learning.

RNA methylation plays an important role in functional regulation of RNAs, and has thus attracted an increasing interest in biology and drug discovery. Here, we collected and collated transcriptomic, proteomic, structural and physical interaction data from the Harmonizome database, and applied supervised machine learning to predict novel genes associated with RNA methylation pathways in human. We selected five types of classifiers, which we trained and evaluated using cross-validation on multiple training sets. The best models reached 88% accuracy based on cross-validation, and an average 91% a

Cell-type-specific responses to associative learning in the primary motor cortex.

The primary motor cortex (M1) is known to be a critical site for movement initiation and motor learning. Surprisingly, it has also been shown to possess reward-related activity, presumably to facilitate reward-based learning of new movements. However, whether reward-related signals are represented among different cell types in M1, and whether their response properties change after cue-reward conditioning remains unclear. Here, we performed longitudinal in vivo two-photon Ca2+ imaging to monitor the activity of different neuronal cell types in M1 while mice engaged in a classical conditioning t

Associated factors of white matter hyperintensity volume: a machine-learning approach.

To identify the most important parameters associated with cerebral white matter hyperintensities (WMH), in consideration of potential collinearity, we used a data-driven machine-learning approach. We analysed two independent cohorts (KORA and SHIP). WMH volumes were derived from cMRI-images (FLAIR). 90 (KORA) and 34 (SHIP) potential determinants of WMH including measures of diabetes, blood-pressure, medication-intake, sociodemographics, life-style factors, somatic/depressive-symptoms and sleep were collected. Visually guided associative learning in pediatric and adult migraine without aura.

INTRODUCTION: The Rutgers Acquired Equivalence Test is a visually guided equivalence learning paradigm that involves rule acquisition and generalization. Earlier we found impaired performance in this paradigm among adult migraine patients without aura. The aim of the study was to investigate if similar impairments can be found already in the pediatric form of the disease and to compare the performance of the pediatric study population with that of an adult study population. We hypothesized that the deficits observed in adults would be observable already in the pediatric population. METHODS: T

A machine learning-based diagnostic model associated with knee osteoarthritis severity.

Knee osteoarthritis (KOA) is characterized by pain and decreased gait function. We aimed to find KOA-related gait features based on patient reported outcome measures (PROMs) and develop regression models using machine learning algorithms to estimate KOA severity. The study included 375 volunteers with variable KOA grades. The severity of KOA was determined using the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC). WOMAC scores were used to classify disease severity into three groups. A total of 1087 features were extracted from the gait data. An ANOVA and student's t-tes
