With the increase in numbers of joint replacements, spinal surgeries, and dental implantations, there is an urgent need to combat implant-associated infection. In addition to stringent sterile techniques, an efficacious way to prevent this destructive complication is to create new implants with antimicrobial properties. Specifically, these implants must be active in the dental implant environment where the implant is bathed in the glycoprotein-rich salivary fluids that enhance bacterial adhesion, and propagation, and biofilm formation. However, in designing an antimicrobial surface, a balance

Mycoplasma pneumoniae is a leading cause of community-acquired pneumonia (CAP) and upper respiratory tract infections, particularly in children and immunocompromised individuals. The growing global prevalence of macrolide-resistant M. pneumoniae (MRMP) further emphasizes the urgent need to elucidate its pathogenic mechanisms. Among these, adhesion plays a central role, serving as a prerequisite for colonization and disease progression, and thus warrants detailed investigation. The terminal organelle of M. pneumoniae mediates both adhesion and gliding motility, facilitating colonization, tissue

D-mannose is widely used as non-antibiotic treatment for bacterial urinary tract infections. This application is based on a well-studied mechanism of binding to the type 1 bacterial pili and, therefore, blocking bacteria adhesion to the uroepithelial cells. To implement D-mannose into carrier systems, the mechanism of action of the sugar in the bladder environment is also relevant and requires investigation. Herein, two different MANNosylation strategies using mesoporous silica nanoparticles (MSNs) are described. The impact of different chemical linkers on bacterial adhesion and bladder cell r

Classical antimicrobial drugs target proliferation and therefore place microbes under extreme selective pressure to evolve resistance. Alternative drugs that target bacterial virulence without impacting survival directly offer an attractive solution to this problem, but to date few such molecules have been discovered. We previously discovered a widespread group of bacterial adhesins, termed Multivalent Adhesion Molecules (MAMs) that are essential for initial binding of bacteria to host tissues and virulence. Thus, targeting MAM-based adherence is a promising strategy for displacing pathogens f

Cronobacter sakazakii (C. sakazakii) is a foodborne pathogen whose initial process involves intestinal cell adhesion mediated by numerous virulence factors encoded in various genes. The key metabolic enzyme, glyceraldehyde-3-phosphate dehydrogenase (GAPDH, also known as GapA), is encoded by gapA in the glycolysis pathway and acts as an adhesin in some bacteria. In C. sakazakii, there is also a key enzyme in the gluconeogenesis pathway, Gap, with type I GAPDH and erythrose-4-phosphate dehydrogenase activities and encoded by gap. This study aimed to investigate the virulence properties of GapA a

Prosthetic devices are frequently used for temporary or permanent drainage of cerebrospinal fluid (CSF), i.e., ventricular catheters with or without external monitoring devices and shunts. Infections constitute a serious complication in the use of biomaterials in contact with CSF; coagulase-negative staphylococci (CNS) are the most common aetiological agents. In the present study, polyvinylchloride (PVC) and PVC with endpoint-attached heparin were exposed to human CSF under perfusion to mimic conditions in vivo. Adhesion of strains of CNS isolated from patients with or without biomaterial-asso

In cattle, bovine respiratory syncytial virus (BRSV) is associated with secondary bacterial infections; however, the mechanisms of the interaction between BRSV and bacteria are unclear. Trueperella pyogenes (T. pyogenes) causes pneumonia in cattle and is involved in secondary infections following viral infections. In this study, we evaluated the effect of BRSV infection on the adhesion of T. pyogenes to BRSV-infected cells. BRSV infection significantly enhanced the adhesion of T. pyogenes to cells in a multiplicity of infection- and time-dependent manner. The BRSV-mediated change in the adhesi

Early caries is initiated by bacterial colonization and biofilm maturation, which drive localized demineralization of enamel. This study developed a borneol-grafted poly(amidoamine) dendrimer to inhibit bacterial adhesion, suppress biofilm formation, and enhance enamel remineralization. Borneol-grafted poly(amidoamine) dendrimer was synthesized via imine linkage and evaluated for antibacterial activity against Streptococcus mutans and Streptococcus sanguinis assessed by adhesion inhibition, and biofilm biomass/viability assays. Mechanisms were explored by zeta potential analysis and scanning e

Following pilus-mediated adhesion to human brain endothelial cells, meningococcus (N. meningitidis), the bacterium causing cerebrospinal meningitis, initiates signaling cascades, which eventually result in the opening of intercellular junctions, allowing meningeal colonization. The signaling receptor activated by the pathogen remained unknown. We report that N. meningitidis specifically stimulates a biased β2-adrenoceptor/β-arrestin signaling pathway in endothelial cells, which ultimately traps β-arrestin-interacting partners, such as the Src tyrosine kinase and junctional proteins, under bact

Biomaterials including catheters, artificial joints, dental implants, and contact lenses are highly susceptible to bacterial adhesion, colonization, and biofilm formation, leading to biomaterials-associated infections (BAIs). BAIs pose a significant challenge to the healthcare sector, contributing to increased morbidity, mortality, and economic burden. Preventing bacterial adhesion and biofilm formation is crucial to eliminate BAIs. This special issue, dedicated to Prof. Henk J. Busscher (The University of Groningen, The Netherlands) on the occasion of his retirement, presents original researc

The attachment of Escherichia coli O157:H7 to intestinal epithelial cells is indispensable for its pathogenesis. Besides translocated-intimin receptor (Tir), E. coli O157:H7 interacts with host cell surface receptors to promote intimate adhesion. This study showed that integrin β1 was increased in Caco-2 cells upon E. coli O157:H7 infection, while Caco-2 cells subjected to integrin β1 antibody blocking or CRISPR/Cas9 knockout had reduced bacterial attachment. Infection of E. coli O157:H7 inactivated focal adhesion kinase (FAK) and paxillin, increased focal adhesion (FA) and actin polymerizatio

Starch-based wood adhesives are widely used as environmentally friendly and formaldehyde-free alternatives in the manufacturing of wood products. However, their poor water resistance and inability to inhibit bacterial/fungal growth limit their practicality. To address this issue, a physicochemical double-crosslinked network structure between citric acid, starch, and wood was prepared in this study by physically entangling the wood structure and chemically crosslinking the starch molecules, with citric acid preventing the activity of various enzymes and denatured proteins within microbial cells

INTRODUCTION: IbeA is a critical virulence factor that is present in the GimA island in Escherichia coli (E. coli) K1, the most common cause of Gram-negative bacterial meningitis in newborns. IbeA has multiple virulence functions, including adhesion, invasion, and biofilm formation in the pathogenesis of E. coli K1 meningitis. However, it is unclear how IbeA coordinates its role in bacterial adhesion, invasion, and biofilm formation during crossing the blood-brain barrier (BBB).
METHODS: An isogenic ibeA deletion mutant (ZD1) was generated from the neonatal meningitis-associated E. coli K1 st

UNLABELLED: Essentials Staphylococcus aureus (S. aureus) binds to endothelium via von Willebrand factor (VWF). Secreted VWF-binding protein (vWbp) mediates S. aureus adhesion to VWF under shear stress. vWbp interacts with VWF and the Sortase A-dependent surface protein Clumping factor A (ClfA). VWF-vWbp-ClfA anchor S. aureus to vascular endothelium under shear stress.
SUMMARY: Objective When establishing endovascular infections, Staphylococcus aureus (S. aureus) overcomes shear forces of flowing blood by binding to von Willebrand factor (VWF). Staphylococcal VWF-binding protein (vWbp) interac

BACKGROUND: Streptococcus suis serotype 2 (SS2) is an important zoonotic pathogen that causes severe infections in pigs and humans. Bacterial autolysins play essential roles in cell-wall remodelling and virulence; however, the structural and functional characteristics of the SS2 autolysin ATL remain poorly understood. With the increasing emergence of antimicrobial resistance, anti-virulence strategies that attenuate bacterial pathogenicity without directly inhibiting growth have attracted increasing attention.
METHODS: ATL domain organization was analysed bioinformatically. The catalytic doma

Since Marshall's discovery before 20 years, Helicobacter pylori (H. pylori) infection is reportedly to be associated with a variety of clinical outcomes including peptic ulcer disease and gastric cancer. The first step of the H. pylori colonization might be its adhesion to the surface epithelial cells, which evokes gastric inflammatory events initiated by neutrophil recruitment from the microcirculation. Mongolian gerbil is one of the suitable animal models for H. pylori infection, which exerts gastric ulcer and cancer with its bacterial infection. In H. pylori-colonized gerbils, extensive lev
