Development and Application of a Desirability of Outcome Ranking (DOOR) Endpoint for Acute Bacterial Skin and Skin Structure Infections.

BACKGROUND: Desirability of outcome ranking (DOOR), a paradigm for the design, analysis, and interpretation of clinical trials based on patient-centered benefit-risk evaluation, could help inform drug development for acute bacterial skin and skin structure infections (ABSSSIs). METHOD: A multidisciplinary committee created an infectious diseases DOOR endpoint, incorporating clinical response, infectious complications, serious adverse events, and mortality, and adapted this for ABSSSI. We retrospectively applied DOOR to 2 previous ABSSSI randomized controlled registrational trials (OASIS-1 and

Omadacycline for the treatment of acute bacterial skin and skin structure infections: a systematic review and network meta-analysis.

BACKGROUND: Few studies have compared the efficacy and safety of omadacycline with other treatments for acute bacterial skin and skin structure infections (ABSSSI). Therefore, updated meta-analyses on this topic is necessary. METHODS: We conducted a network meta-analysis (NMA) of randomized controlled trials (RCTs) to compare omadacycline with other anti-MRSA antibiotics. A systematic search of seven databases was undertaken to identify eligible RCTs enrolling adults with ABSSSI. Two reviewers independently assessed the quality of the included studies and three reviewers independently extract

Cerebral arteritis in bacterial meningitis: Structural adaptations of the vascular wall in response to an infectious nidus - A narrative review.

Despite dramatic improvements in diagnosis and antimicrobial treatment of bacterial meningitis over the last few decades, adverse postinfective sequelae and mortality remain exceedingly high in adults. Of note, the unfavorable clinical outcome is usually attributable to the presence of intracranial complications during the acute phase of infection, such as cerebral edema and increases in intracranial pressure and cerebral blood flow disturbances. Involvement of the cerebral vasculature during bacterial meningitis is overwhelmingly clear from clinical and laboratory evidence highlighting cerebr

The fermentation of Cordyceps militaris polysaccharide influenced gut bacterial LPS structure formation and changed its antigenicity.

Gut bacterial lipopolysaccharide (LPS) could be released into the circulatory system via the gut-liver axis and cause inflammatory immune response, while Cordyceps militaris polysaccharide (CMP40) has been reported to be effective in alleviating this inflammatory response. In this study, the effects of CMP40 gut fermentation on internal LPS structure formation and the subsequent immune response were explored. Results showed that CMP40 could change antigenicity of LPS of Vibrio parahaemolyticus, Salmonella enterica, and enterotoxigenic Escherichia coli, indicated by a reduced level of NO, IL-1β

Dalbavancin for the treatment of acute bacterial skin and skin structure infections (ABSSSI) in pediatric patients: a case series.

Acute bacterial skin and skin-structure infections (ABSSSI) are a significant cause of morbidity in pediatric patients, requiring timely and effective treatment. Dalbavancin, a long-acting lipoglycopeptide antibiotic recently approved for pediatric use, offers advantages such as excellent bactericidal activity against Gram-positive bacteria (including multidrug-resistant pathogens) and high tissue penetration. We present a case series of pediatric patients with ABSSSI treated with dalbavancin. Five cases were described demonstrating the efficacy of dalbavancin in different clinical scenarios.

Seasonal dynamics of bacterial community structure and function in the surf zone seawater of a recreational beach in Ostend, Belgium.

Despite the importance of bacteria in surf zone water quality, detailed insights into their community composition, functions, and seasonal dynamics at recreational beaches are scarce. This study conducted year-long, weekly monitoring of bacterial communities and environmental factors at a recreational beach in Ostend, Belgium. Using full-length 16S rRNA gene sequencing, we correlated bacterial composition and predicted functions with environmental factors to identify potential drivers. Bacterial communities were significantly affected by seasonal variations in chlorophyll a (Chl a), net primar

Structural analysis of neomycin B and kanamycin A binding Aminoglycosides Modifying Enzymes (AME) and bacterial ribosomal RNA.

Aminoglycosides are crucial antibiotics facing challenges from bacterial resistance. This study addresses the importance of aminoglycoside modifying enzymes in the context of escalating resistance. Drawing upon over two decades of structural data in the Protein Data Bank, we focused on two key antibiotics, neomycin B and kanamycin A, to explore how the aminoglycoside structure is exploited by this family of enzymes. A systematic comparison across diverse enzymes and the RNA A-site target identified common characteristics in the recognition mode, while assessing the adaptability of neomycin B a

Modulation of ultrathin nanosheet structure and nitrogen defects in graphitic carbon nitride for efficient photocatalytic bacterial inactivation.

The efficient generation and utilization of ROSs is a key step in determining the achievement of safe drinking water by photocatalytic bacterial inactivation technology. Although graphitic carbon nitride (g-C3N4) serves as a green and promising photocatalyst for water disinfection, insufficient bacterial capturing capacity and serious charge recombination of pristine g-C3N4 extremely restrict its bactericidal activity. Herein, we develop a facile thermal exfoliation and thermal polymerization method to prepare the nitrogen-defective ultrathin g-C3N4 nanosheets (DUCN-500). Our results showed th

Effects of continuous straw returning on bacterial community structure and enzyme activities in rape-rice soil aggregates.

Straw returning is an effective management measure to improve or maintain soil fertility in agricultural ecosystems. This study investigated the effects of straw returning combined with compound fertilizer on the bacterial community, enzyme activities, and soil nutrients' contents in a rape-rice rotation soil aggregates. To do so, a 5-year field trial (November 2016 to October 2021) was carried out in a paddy soil with three treatments: no straw + no fertilization (CK), compound fertilizer (F), and straw returning + compound fertilizer (SF). Soil aggregates were classified into mega-aggregates

Effect of g-C3N4 on biodiversity and structure of bacterial community in sediment of Xiangjiang River under tetracycline pressure.

Photocatalysts have been widely prepared and used in wastewater treatment. Although the influence of photocatalyst application on survival and activity of organisms has been examined, its impact on composition and diversity of microbial community is not fully understood. In this study, the impact of photocatalyst g-C3N4 (Graphitic carbon nitride) on microbial communities in riverbed sediments polluted by antibiotic tetracycline (TC) was investigated. The sediment samples collected from the Xiangjiang River of China were exposed to different concentrations of TC, g-C3N4 and TC/g-C3N4 and the ba

Sulfadiazine destruction by chlorination in a pilot-scale water distribution system: Kinetics, pathway, and bacterial community structure.

Sulfadiazine (SDZ) has been frequently detected in surface waters in recent years. We evaluated the kinetics, mechanisms, intermediate products and bacterial community structure that result from the reaction of SDZ with free chlorine (HOCl/OCl-). We examined this in a pilot-scale water distribution system. Neutral pH had the fastest rate of destruction of SDZ. A second-order reaction constant for the destruction of SDZ by chlorine increased with increasing concentration of free chlorine (FC). For different pipe materials, the rate of SDZ degradation decreased as follows: stainless steel (SS) p

Risk factors associated with bacteremia correlated with mortality in patients with acute bacterial skin and skin structure infection.

Acute bacterial skin and skin structure infections (ABSSSI) is a common cause of acute admissions worldwide, but the disease is not well understood epidemiologically with respect to factors that determine positive blood cultures or patient mortality. To understand the utility of blood cultures and the association between bacteremia and mortality in patients with ABSSSI, we conducted a retrospective study to investigate factors associated with positive blood cultures and mortality in patients with ABSSSI. A retrospective cohort study of hospitalized adult patients with ABSSSI was conducted in a

Management of acute bacterial skin and skin structure infections with a focus on patients at high risk of treatment failure.

Over the last 25 years, the terminology of skin and soft tissue infections, as well as their classification for optimal management of patients, has changed. The so-called and recently introduced term 'acute bacterial skin and skin structure infections' (ABSSSIs), a cluster of fairly common types of infection, including abscesses, cellulitis, and wound infections, require an immediate effective antibacterial treatment as part of a timely and cautious management. The extreme level of resistance globally to many antibiotic drugs in the prevalent causative pathogens, the presence of risk factors o

Structural and biochemical analyses indicate that a bacterial persulfide dioxygenase-rhodanese fusion protein functions in sulfur assimilation.

Hydrogen sulfide (H2S) is a signaling molecule that is toxic at elevated concentrations. In eukaryotes, it is cleared via a mitochondrial sulfide oxidation pathway, which comprises sulfide quinone oxidoreductase, persulfide dioxygenase (PDO), rhodanese, and sulfite oxidase and converts H2S to thiosulfate and sulfate. Natural fusions between the non-heme iron containing PDO and rhodanese, a thiol sulfurtransferase, exist in some bacteria. However, little is known about the role of the PDO-rhodanese fusion (PRF) proteins in sulfur metabolism. Herein, we report the kinetic properties and the crys

Increasing the Size of the Microbial Biomass Altered Bacterial Community Structure which Enhances Plant Phosphorus Uptake.

Agricultural production can be limited by low phosphorus (P) availability, with soil P being constrained by sorption and precipitation reactions making it less available for plant uptake. There are strong links between carbon (C) and nitrogen (N) availability and P cycling within soil P pools, with microorganisms being an integral component of soil P cycling mediating the availability of P to plants. Here we tested a conceptual model that proposes (i) the addition of readily-available organic substrates would increase the size of the microbial biomass thus exhausting the pool of easily-availab

Continuous desulfurization and bacterial community structure of an integrated bioreactor developed to treat SO2 from a gas stream.

Sulfide dioxide (SO2) is often released during the combustion processes of fossil fuels. An integrated bioreactor with two sections, namely, a suspended zone (SZ) and immobilized zone (IZ), was applied to treat SO2 for 6months. Sampling ports were set in both sections to investigate the performance and microbial characteristics of the integrated bioreactor. SO2 was effectively removed by the synergistic effect of the SZ and IZ, and more than 85% removal efficiency was achieved at steady state. The average elimination capacity of SO2 in the bioreactor was 2.80g/(m(3)·hr) for the SZ and 1.50g/(m

Structural details of human tuba recruitment by InlC of Listeria monocytogenes elucidate bacterial cell-cell spreading.

The human pathogen Listeria monocytogenes is able to directly spread to neighboring cells of host tissues, a process recently linked to the virulence factor InlC. InlC targets the sixth SH3 domain (SH3-6) of human Tuba, disrupting its physiological interaction with the cytoskeletal protein N-WASP. The resulting loss of cortical actin tension may slacken the junctional membrane, allowing protrusion formation by motile Listeria. Complexes of Tuba SH3-6 with physiological partners N-WASP and Mena reveal equivalent binding modes but distinct affinities. The interaction surface of the infection com

Diversity and Structural Composition of Bacterial Communities in Mineral Springs of the Andean-Amazonian Transition Zone in Northern Peru.

Mineral springs in the Andean-Amazonian transition represent poorly explored aquatic ecosystems whose hydrogeochemical heterogeneity may favor the establishment of specialized microbial communities. In this study, bacterial communities in sediments from 19 mineral springs in northern Peru were characterized using 16S rRNA gene sequencing, and their relationships with hydrogeochemical conditions and predicted functional potential were evaluated. The springs exhibited temperatures ranging from 15.1 to 38.2 °C and total dissolved solids (TDS) concentrations ranging from 0.022 to 162.5 mg L-1, ref

Species-specific structuring of gut bacterial and fungal communities in honey bees Apis cerana and Apis mellifera.

Honey bee gut microbiome studies have primarily emphasized bacteria, leaving fungal communities comparatively overlooked despite their ecological and functional importance. Whole-genome shotgun metagenomics of Apis cerana and Apis mellifera revealed fungal assemblages dominated by Ascomycota, with Basidiomycota and Microsporidia in minor proportions, alongside gut bacterial communities composed mainly of Pseudomonadota, Bacillota, and Actinomycetota. The bacterial diversity was markedly higher in A. mellifera (Shannon = 5.90; Simpson = 0.98) than in A. cerana (Shannon = 4.01; Simpson = 0.94; p

AIE-Active Antibacterial Photosensitizer Disrupting Bacterial Structure: Multicenter Validation against Drug-Resistant Pathogens.

Antimicrobial resistance (AMR) has emerged as a global challenge in treating bacterial infections, creating an urgent need for broad-spectrum antimicrobial agents that can effectively combat multidrug-resistant (MDR) bacteria. Despite advancements in novel antimicrobial agents, many fail to comprehensively cover common resistant bacterial strains or undergo rigorous multi-center validation. Herein, a cationic AIE-active photosensitizers are developed, ITPM, derived from a triphenylamine-pyridine backbone to address the MDR challenge. Rigorous validation demonstrates that ITPM possesses superio
