Soil microbial strategies for climate mitigation-report from a climate action workshop in Las Vegas, Nevada, February 2024.

Life on Earth faces an existential crisis due to the enduring repercussions of unsustainable human activities since the beginning of the Industrial Revolution. Among the most pressing issues are greenhouse gas emissions, primarily from fossil fuel combustion, unsustainable agricultural practices, as well as the global erosion of the world's topsoil. While agrochemicals have temporarily increased land productivity, their frequent use has adversely impacted the environment and microbial biodiversity. With half of the global soils already degraded by erosion and a projected 90% at risk by 2050, h

Socio-economic influences from egg to exit: Emerging biology.

Socio-economic status (SES) impacts life-long health from early development to old age. We review SES differences in biological factors that begin before fertilization and extend into later life, 'egg to exit'. SES gradients are shown in onset and prevalence of chronic conditions including cardiovascular disease, diabetes and dementia, leading up to a 15-year difference in life expectancy in some countries. Our analysis is limited to high-income countries and does not include detailed discussion of gender or ethnic differences. We develop a comprehensive multi-disciplinary framework for SES gr

Postmodern personhood: a matter of consciousness.

The concept of person is integral to bioethical discourse because persons are the proper subject of the moral domain. Nevertheless, the concept of person has played no role in the prevailing formulation of human death because of a purported lack of consensus concerning the essential attributes of a person. Beginning with John Locke's fundamental proposition that person is a 'forensic term', I argue that in Western society we do have a consensus on at least one necessary condition for personhood, and that is the capacity for conscious experience. When we consider the whole brain formulation of

Promoting sustainable pharmacy for tackling environmental pharmaceutical pollution.

Pharmaceuticals are essential for health but are increasingly impacting the environment, affecting ecosystems and human health, and contributing to biodiversity loss, antimicrobial resistance, and climate change. Active pharmaceutical ingredients designed for stability and biological activity, along with excipients and packaging materials, contaminate air, soil, and water globally. To address the issue of pharmaceutical pollution, the establishment of a sustainable pharmacy framework is required, beginning with drug development and the education of future professionals. In this article, we pre

Muscle biopsies show that FES of denervated muscles reverses human muscle degeneration from permanent spinal motoneuron lesion.

This paper presents biopsy analyses in support of the clinical evidence of muscle recovery induced by a new system of life-long functional-electrical-stimulation (FES) training in permanent spinal-motoneuron-denervated human muscle. Not earlier than 1 year after subjects experienced complete conus cauda lesion, their thigh muscles were electrically stimulated at home for several years with large skin surface electrodes and an expressly designed stimulator that delivered much longer impulses than those presently available for clinical use. The poor excitability of long-term denervated muscles w

Intravenous AAV9-ATP7A plus subcutaneous copper histidinate optimizes outcomes in a lethal Menkes disease mouse model.

Menkes disease is an X-linked recessive condition characterized by seizures, failure to thrive, and rapid, progressive neurodegeneration beginning within weeks after birth. Death usually occurs by 3 years of age. The disorder is caused by genetic variants in ATP7A, an evolutionarily conserved copper transporter that is crucial for normal brain development. The mottled-brindled mouse recapitulates salient features of the human illness. Affected male mice typically die by 14 postnatal days. We evaluated a single-dose intravenous adeno-associated virus gene therapy approach to deliver working cop

Why, How, and When to Integrate Narrative Identity Within Dimensional Approaches to Personality Disorders.

Researchers and clinicians are beginning to adopt dimensional approaches in the study and treatment of personality disorders (PD). Although dimensional approaches in the DSM-5 and ICD-11 hold considerable benefit, they need to better incorporate an appreciation of individuals' life stories, or narrative identities. Doing so will be necessary to flesh out the emphasis that both frameworks place on the role of identity in personality pathology. In this article, the authors review why, how, and when narrative identity theory and research can be integrated within dimensional approaches to PD. The

Brainstem mechanisms underlying the sudden infant death syndrome: evidence from human pathologic studies.

The brainstem hypothesis is one of the leading hypotheses concerning the sudden infant death syndrome (SIDS). It states that SIDS, or an important subset of SIDS, is due to abnormal brainstem mechanisms in the control of respiration, chemosensitivity, autonomic regulation, and/or arousal which impairs the infant's response to life-threatening, but often occurring, stressors during sleep (e.g., hypoxia, hypercarbia, asphyxia, hyperthermia) and leads to sudden death in a vulnerable developmental period. In this review, we summarize neuropathologic evidence from SIDS cases that support this hypot

FDA-Approved Radiopharmaceuticals in Oncologic PET Imaging: Current Status and Clinical Impact.

Cancer persists as a formidable adversary to human health, with its early-stage detection posing a persistent and intricate challenge. While radiologic imaging serves as a cornerstone in the diagnostic arsenal for oncological conditions, the incipient manifestations of malignancies frequently present with such subtlety that they elude recognition, resulting in missed opportunities for timely and potentially life-saving interventions. Several radiodiagnostic agents have received regulatory approval from the US FDA for clinical application in the oncological domain, specifically to facilitate th

A promising mouse model for Friedreich Ataxia progressing like human patients.

Friedreich Ataxia (FRDA) is a genetic disease caused by an expended GAA repeat in the FXN gene leading to a reduction in frataxin protein production. Frataxin is an essential protein involved in mitochondrial iron-sulfur-cluster formation, its absence affecting numerous cellular rections. In patients, the disease leads to a progressive neuromuscular degeneration and, most of the time, death from heart failure. In order to determine if a treatment is effective or not, it is essential to have the mouse model, which best reflects all of the characteristics of this disease. Many groups were workin

Genome-resolved metaproteomic characterization of preterm infant gut microbiota development reveals species-specific metabolic shifts and variabilities during early life.

BACKGROUND: Establishment of the human gut microbiota begins at birth. This early-life microbiota development can impact host physiology during infancy and even across an entire life span. However, the functional stability and population structure of the gut microbiota during initial colonization remain poorly understood. Metaproteomics is an emerging technology for the large-scale characterization of metabolic functions in complex microbial communities (gut microbiota). RESULTS: We applied a metagenome-informed metaproteomic approach to study the temporal and inter-individual differences of

Brain death in the pediatric patient: historical, sociological, medical, religious, cultural, legal, and ethical considerations.

OBJECTIVE: To detail the origins of the definition of death, the development of the criterion of whole brain death as fulfilling the definition of death, and the tests used to fulfill that criterion. DATA SOURCES: A review of the literature was performed. No Institutional Review Board approval was necessary. DATA EXTRACTION: In 1959, patients were described as being in "coma dépassé" or beyond coma. In 1967, the first successful heart transplantation took place, with the organ coming from a brain-dead, beating-heart donor. However, anxiety over the definitions of death did not begin with the

The Anatomy and Clinical Significance of Sensory Disturbance in Parkinson's Disease.

The clinical features of Parkinson's disease (PD) include tremors and rigidity. However, paresthesia has not drawn clinical attention. PD involves the whole body and begins with gastrointestinal lesions, which do not start in the midbrain substantia nigra, but from the beginning of the medulla oblongata of the glossopharyngeal nerve nuclei, to the motor nerve dorsal nucleus of the vagus nerve, to the pons and midbrain, and finally to the neocortex. The human eye, ear, nose, tongue, and body perceive the external world. (1) Visual impairment in patients with PD can be easily confused with senil

Delayed Induction of Human NTE (PNPLA6) Rescues Neurodegeneration and Mobility Defects of Drosophila swiss cheese (sws) Mutants.

Human PNPLA6 gene encodes Neuropathy Target Esterase protein (NTE). PNPLA6 gene mutations cause hereditary spastic paraplegia (SPG39 HSP), Gordon-Holmes syndrome, Boucher-Neuhäuser syndromes, Laurence-Moon syndrome, and Oliver-McFarlane syndrome. Mutations in the Drosophila NTE homolog swiss cheese (sws) cause early-onset, progressive behavioral defects and neurodegeneration characterized by vacuole formation. We investigated sws5 flies and show for the first time that this allele causes progressive vacuolar formation in the brain and progressive deterioration of negative geotaxis speed and en

Association Between Prenatal Exposure to Maternal Infection and Offspring Mood Disorders: A Review of the Literature.

The purpose of this article is to provide a systematic review of studies that have examined the association between prenatal exposure to maternal infection and development of mood disorders across the life course. Drawing from both human- and animal-based studies, we give an overview of hypothesized biological mechanisms by which exposure to maternal infection during critical periods of gestation may contribute to fetal programming of mood disorders in offspring. We discuss studies examining the association between prenatal exposure to maternal infection with pathogens including influenza as w

Single-cell atlas of the small intestine throughout the human lifespan demonstrates unique features of fetal immune cells.

Proper development of mucosal immunity is critical for human health. Over the past decade, it has become evident that in humans, this process begins in utero. However, there are limited data on the unique features and functions of fetal mucosal immune cells. To address this gap, we integrated several single-cell ribonucleic acid sequencing datasets of the human small intestine (SI) to create an SI transcriptional atlas throughout the human life span, ranging from the first trimester to adulthood, with a focus on immune cells. Fetal SI displayed a complex immune landscape comprising innate and

A Stoichioproteomic Analysis of Samples from the Human Microbiome Project.

Ecological stoichiometry (ES) uses organism-specific elemental content to explain differences in species life histories, species interactions, community organization, environmental constraints and even ecosystem function. Although ES has been successfully applied to a range of different organisms, most emphasis on microbial ecological stoichiometry focuses on lake, ocean, and soil communities. With the recent advances in human microbiome research, however, large amounts of data are being generated that describe differences in community composition across body sites and individuals. We suggest

Mouse Models of Pain in Sickle Cell Disease.

Sickle cell disease (SCD) is a genetic blood disorder that impacts millions of individuals worldwide. SCD is characterized by debilitating pain that can begin during infancy and may continue to increase throughout life. This pain can be both acute and chronic. A characteristic feature specific to acute pain in SCD occurs during vaso-occlusive crisis (VOC) due to the blockade of capillaries with sickle red blood cells. The acute pain of VOC is intense, unpredictable, and requires hospitalization. Chronic pain occurs in a significant population with SCD. Treatment options for sickle pain are lim
