All major life forms from bacteria to humans have internal clocks that regulate essential biological processes in a roughly 24-h cycle. In mammals, the central clock in the suprachiasmatic nucleus (SCN) is historically considered the top of a hierarchical organisation that dominates subordinate clocks in peripheral tissues and dictates the circadian behaviours of an organism. Recent studies, however, underscore the importance of the local circadian oscillators, such as the skeletal muscle clock, in regulating local metabolism and physiology. Studies in animal models show that the muscle periph

BACKGROUND: Epigenetic age acceleration (EAA) and epigenetic gestational age acceleration (EGAA) are biomarkers of physiological development and may be affected by the perinatal environment. The aim of this study was to evaluate performance of epigenetic clocks and to identify biological and sociodemographic correlates of EGAA and EAA at birth and in childhood. In the Project Viva pre-birth cohort, DNA methylation was measured in nucleated cells in cord blood (leukocytes and nucleated red blood cells, N = 485) and leukocytes in early (N = 120, median age = 3.2 years) and mid-childhood (N = 460

The biological clock of the suprachiasmatic nuclei drives numerous physiological and behavioural circadian rhythms. In this study, we addressed the question as to whether different components of the clock may control separately various circadian functions. Using the rat transpineal microdialysis tool, we analysed the effect of clock perturbation by exogenous melatonin injection on two hormonal clock outputs: pineal melatonin and adrenal corticosterone secretions. As already reported, a single melatonin injection at the light/dark transition induces a marked increase in the endogenous pineal me

Lack of sleep time is a menace to modern people, and it leads to chronic diseases and mental illnesses. Circadian processes control sleep, but little is known about how sleep affects the circadian system. Therefore, we performed a 28-day sleep restriction (SR) treatment in mice. Sleep restriction disrupted the clock genes' circadian rhythm. The circadian rhythms of the Cry1 and Per1/2/3 genes disappeared. The acrophase of the clock genes (Bmal1, Clock, Rev-erbα, and Rorβ) that still had a circadian rhythm was advanced, while the acrophase of negative clock gene Cry2 was delayed. Clock genes' u

BACKGROUND AND AIM: Low-grade inflammation (LGI) and metabolic dysfunction are tightly linked processes central to the onset and progression of complex diseases and aging. LGI acts both as a driver and a consequence of metabolic disturbances and represents a hallmark of "inflammaging", the chronic, systemic inflammatory state arising from sustained immune activation that characterizes aging and underlies many age-related pathologies. Targeting LGI through lifestyle changes, particularly diet, offers a promising approach for healthy aging. Epigenetic clocks have recently emerged as biomarkers o

The nerve center responsible for controlling our circadian rhythm is located in a cluster of cells known as the suprachiasmatic nucleus in the hypothalamus. Various physiological functions such as sleep, arousal, blood pressure, body temperature, and hormone secretion are regulated in a 24-hour rhythm by this circuit. Somatic cells of other organs have a peripheral clock gene and by synchronizing the rhythm of the central and peripheral clocks, it is possible to live a healthy life. Due to aging and degenerative disease, circadian rhythm gradually collapses. Factors that can contribute to this

Migraine is among the most dynamic and disabling neurological disorders, affecting over one billion people globally. Attacks demonstrate rhythmic patterns of onset across the day and seasonally, highlighting that attack onset is influenced by an individual's biological rhythms. Indeed, an individual's chronotype (their endogenous circadian clock rhythm) predicts when they are most likely to have an attack. These biological rhythms are regulated by an endogenous master biological clock in the hypothalamus that coordinates the function of peripheral clocks, aligning biological processes and beha

BACKGROUND: Cancer and its treatments are associated with accelerated biological aging and cognitive impairment among breast cancer survivors. DNA methylation-based epigenetic clocks provide measures of biological aging that predict morbidity and mortality, yet little is known about whether these aging trajectories can be modified by lifestyle intervention. This secondary analysis of a subset of participants in a randomized controlled trial examined the impact of a 12-month exercise intervention on markers of epigenetic aging among breast cancer survivors.
METHODS: Sedentary breast cancer sur

The mammalian circadian system is a complex hierarchical temporal network which is organized around an ensemble of uniquely coupled cells comprising the principal circadian pacemaker in the suprachiasmatic nucleus of the hypothalamus. This central pacemaker is entrained each day by the environmental light/dark cycle and transmits synchronizing cues to cell-autonomous oscillators in tissues throughout the body. Within cells of the central pacemaker and the peripheral tissues, the underlying molecular mechanism by which oscillations in gene expression occur involves interconnected feedback loops

Acute ischaemic stroke care has long been guided by the maxim "time is brain," yet outcome variability suggests that clock time alone is insufficient. We propose the Time, Tissue, and Systems (TTS) framework, a quantitative model integrating temporal progression, tissue vulnerability, and health-system performance. Biological injury is conceptualized as accelerated neuronal loss scaled by tissue phenotype and systems friction, and clinical decision-making is reframed as net utility optimization. Illustrative modeling across four archetypal stroke profiles demonstrates how identical clock times

The sensitivity of the human circadian system to light has been the subject of considerable debate. Using computer simulations of a recent quantitative model for the effects of light on the human circadian system, we investigated these effects of light during different experimental protocols. The results of the simulations indicate that the nonuniform distribution over the circadian cycle of exposure to ordinary room light seen in classical free-run studies, in which subjects select their exposure to light and darkness, can result in an observed period of approximately 25 h, even when the intr

Understanding how respiratory oscillations are generated and controlled in the mammalian brainstem requires synthesis of neural function at cellular, synaptic, and network levels. Modeling the respiratory network has entered a new era where simulations incorporating complex cellular properties and network interactions exhibited by respiratory neurons are possible. Given the complexity and unknowns of the real system, there are obvious limitations of the modeling in its present form. Nevertheless, if we are to produce neurobiologically satisfying, mechanistic explanations that synthesize cellul

ObjectivePsychosis is a significant neuropsychiatric symptom in Alzheimer's Disease (AD) and Mild Cognitive Impairment (MCI). While female APOE4 homozygotes are at the highest risk of psychosis, mechanisms underlying this association remain unclear. We investigated whether epigenetic age acceleration (EAA) is associated with psychosis in individuals with AD and MCI, and whether this association varies by sex and APOE4 status.MethodsParticipants with clinical MCI or AD were drawn from the Alzheimer's Disease Neuroimaging Initiative database. EAA was calculated from DNA methylation data using th

Adverse birth outcomes, such as early gestational age and low birth weight, can have lasting effects on morbidity and mortality, with impacts that persist into adulthood. Identifying the maternal factors that contribute to adverse birth outcomes in the next generation is thus a priority. Epigenetic clocks, which have emerged as powerful tools for quantifying biological aging and various dimensions of physiological dysregulation, hold promise for clarifying relationships between maternal biology and infant health, including the maternal factors or states that predict birth outcomes. The circadian clock is a generator of self-sustaining physiological and behavioral rhythms, which can be guided by external environmental factors, so as to synchronize biological behaviors with external environmental changes. The modern lifestyles make the human body incapable of synchronization to the external time with the circadian rhythm, and thus form a social jet lag. Non-alcoholic fatty liver disease (NAFLD) is a disorder closely related to metabolic abnormalities. The circadian clock is closely related to metabolic abnormalities and NAFLD and changes among them may be involved with fee

BACKGROUND: Aging-associated changes are major contributors to the onset and progression of chronic diseases. Different aging clocks have been developed to assess biological aging, demonstrating their utility in predicting mortality, diagnosing disease, and evaluating the efficacy of antiaging interventions. However, the protein profile underlying the accelerating or decelerating rates of aging, hidden behind aging clocks, remains poorly understood.
METHOD: Based on the UK Biobank (n = 53, 013; age range 39-71 years; 53.9% men and 46.1% women), we built a proteomic-based aging clock, ProteAge

Biological clocks are evolved time-keeping systems by which organisms rhythmically coordinate physiology within the body, and align it with rhythms in their environment. Clocks are highly sensitive to light and are at the interface of several major endocrine pathways. Worryingly, exposure to artificial-light-at-night (ALAN) is rapidly increasing in ever more extensive parts of the world, with likely impact on wild organisms mediated by endocrine-circadian pathways. In this overview, we first give a broad-brush introduction to biological rhythms. Then, we outline interactions between the avian

Many aspects of life in plants, animals and humans are controlled by light. Endogenous, so-called circadian rhythms in the body deviate from the exact 24-hour day and have typically a period of around 25.5 hours in man. Normally these rhythms adapt to the external 24-hour day-and night changes but under constant conditions the rhythms can free run. Many studies show how important the interplay between light and the circadian rhythms are for man as well as for other organisms. The control of these rhythms by light is mediated via the retina and the melatonin system in man. The adaptation of the

Physiological processes and behaviors in many mammals are rhythmic. Recently there has been increasing interest in the role of circadian rhythmicity in the control of reproductive function. The circadian rhythm of the pineal hormone melatonin plays a role in synchronizing the reproductive responses of animals to environmental light conditions. There is some evidence that melatonin may have a role in the biological regulation of circadian rhythms and reproduction in humans. Moreover, circadian rhythms and clock genes appear to be involved in optimal reproductive performance. These rhythms are c
