Phenotypic Variation in Two Siblings Affected with Shwachman-Diamond Syndrome: The Use of Expert Variant Interpreter (eVai) Suggests Clinical Relevance of a Variant in the KMT2A Gene.

Introduction. Shwachman-Diamond Syndrome (SDS) is an autosomal-recessive disorder characterized by neutropenia, pancreatic exocrine insufficiency, skeletal dysplasia, and an increased risk for leukemic transformation. Biallelic mutations in the SBDS gene have been found in about 90% of patients. The clinical spectrum of SDS in patients is wide, and variability has been noticed between different patients, siblings, and even within the same patient over time. Herein, we present two SDS siblings (UPN42 and UPN43) carrying the same SBDS mutations and showing relevant differences in their phenotypi

Spermatogenic Activity and Sperm Traits in Post-Pubertal and Adult Tomcats (Felis catus): Implication of Intra-Male Variation in Sperm Size.

Tomcats are considered to be adults at 1 year of age, although many reach sexual maturity at an earlier age. Nevertheless, we still know little about whether the spermatogenic activity and sperm quality of mature under one-year-old tomcats differ from those of tomcats that are over one-year-old. This study aims to evaluate the spermatogenic activity, sperm traits, and their relationships in mature tomcats at two different ages. Sixteen tomcats showing complete spermatogenesis and spermatozoa in their epididymal caudae were used and classified according to their age as post-pubertal (<1 year ol

Clinical pharmacology and dosing regimen optimization of neonatal opioid withdrawal syndrome treatments.

In this paper, we review the management of neonatal opioid withdrawal syndrome (NOWS) and clinical pharmacology of primary treatment agents in NOWS, including morphine, methadone, buprenorphine, clonidine, and phenobarbital. Pharmacologic treatment strategies in NOWS have been mostly empirical, and heterogeneity in dosing regimens adds to the difficulty of extrapolating study results to broader patient populations. As population pharmacokinetics (PKs) of pharmacologic agents in NOWS become more well-defined and knowledge of patient-specific factors affecting treatment outcomes continue to accu

Intratumor heterogeneity in human parathyroid tumors.

Parathyroid tumors are the second most common endocrine neoplasia after thyroid neoplasia. They are mostly associated with impaired parathormone (PTH) synthesis and release determining the metabolic and clinical condition of primary hyperparathyroidism (PHPT). PHPT is the third most prevalent endocrine disorder, mainly affecting postmenopausal women. Parathyroid benign tumors, both adenomas of a single gland or hyperplasia involving all the glands, are the main histotypes, occurring in more than 95% of PHPT cases. The differential diagnosis between benign and malignant parathyroid lesions is a

Integrating electric field modeling and neuroimaging to explain inter-individual variability of tACS effects.

Transcranial electrical stimulation (tES) of the brain can have variable effects, plausibly driven by individual differences in neuroanatomy and resulting differences of the electric fields inside the brain. Here, we integrated individual simulations of electric fields during tES with source localization to predict variability of transcranial alternating current stimulation (tACS) aftereffects on α-oscillations. In two experiments, participants received 20-min of either α-tACS (1 mA) or sham stimulation. Magnetoencephalogram (MEG) was recorded for 10-min before and after stimulation. tACS caus

CellDiffusion: Enhancing scRNA-seq data integration with accurate inference of cell-cell association.

UNLABELLED: Integrating single-cell transcriptomic datasets across laboratories, conditions, and technologies remains challenging due to cell-type-specific batch effects and heterogeneous biological compositions. Current methods often fail to separate genuine biological variation from technical noise, misaligning distinct cell populations or leaving batch effects uncorrected. To address these challenges, we present CellDiffusion, an unsupervised graph-based model that propagates cellular features through a diffusion process to strengthen meaningful cell-cell associations in data integration. Heritability of body size matches trait evolution in the range expansion of a biological control agent.

Adaptive evolution requires both natural selection and genetic variation. In introduced species, the selective dynamics of range expansion are predicted by theory to lead to differences between the core and the leading edge, with edge individuals evolving to be more fecund (under r-selection) and have greater dispersal ability than core individuals. In arthropods, both fecundity and dispersal ability are often positively correlated with body size. Here, we quantify genetic variation available for evolution of body size in a beetle (Diorhabda carinulata) introduced into North America as a biolo

Genetic variation among Drosophila melanogaster isofemale lines influences encapsulation ability of the parasitoid Asobara tabida.

One of the most important factors which affect the outcome of the host-parasite interactions is the genetic variation in particular those of the host. The aim of this study was to test the effect of variation within Drosophila melanogaster population on the encapsulation ability against the parasitoid Asobara tabida. To this end, nine isofemale lines of D. melanogaster which represent different levels of encapsulation ability were used. The encapsulation ability and its related parameters such as infestation, avoidance, and parasitism success and super-parasitism rates were assessed following

Ethnic variation in respiratory morbidity and lung function in childhood.

In a population of 5689 primary schoolchildren there were few important differences between children of European (n = 5287), African (n = 198), and Indian origin (n = 204) in the prevalence of a history of past respiratory illnesses or current respiratory symptoms. The reported 12 month period prevalence of the symptom "ever wheezy" was 15%, 18%, and 17% respectively in the three ethnic groups (differences not significant). In a subsample of 973 European, 47 African, and 40 Indian children forced expiratory volume in one second (FEV1) and forced vital capacity (FVC) were significantly lower by

Intraspecific variation of Bothrops pictus venom reveals geographic and phylogenetic patterns of functional diversity.

Snake venom composition is shaped by ecological, biological, and evolutionary pressures, with intraspecific variability playing an important role in venom evolution and toxin diversity. Bothrops pictus, an endemic viper of the Peruvian coastal desert, has been the subject of biochemical and proteomic studies; however, interindividual venom variation among geographically distinct coastal populations remains insufficiently characterized. Here, we evaluated toxinological variation using an integrative approach combining enzymatic assays, electrophoretic and chromatographic profiling, immunologica

Short-Term Precision Error of Body Composition Assessment Methods in Resistance-Trained Male Athletes.

Athletic populations require high-precision body composition assessments to identify true change. Least significant change determines technical error via same-day consecutive tests but does not integrate biological variation, which is more relevant for longitudinal monitoring. The aim of this study was to assess biological variation using least significant change measures from body composition methods used on athletes, including surface anthropometry (SA), air displacement plethysmography (BOD POD), dual-energy X-ray absorptiometry (DXA), and bioelectrical impedance spectroscopy (BIS). Tangled Evolutionary History: Genetically Divergent Taxa and Hybrids Characterise Lantana Invasions in Australia.

This study investigated population genomic diversity and structure in the taxonomically complicated and highly invasive Lantana camara L. (Verbenaceae) species complex in Australia. We considered taxonomy, spatial distribution and patterns of morphological and genetic variation in populations across the main invaded area (i.e., coastal and subcoastal eastern Australia across 22° latitude), and reconstructed their relationships with native-range plants sampled from the Americas. We used DArTseq to generate genome-wide single nucleotide polymorphism (SNP) data for > 600 individuals and analysed

Quantitative ecology of ticks as a basis for transmission models of tick-borne pathogens.

Tick population ecology is the basis of spatiotemporal variation in the risk of infection by tick-borne pathogens. The continental distribution of tick-borne encephalitis (TBE) in Europe is statistically associated with a specific pattern of the seasonal dynamics of Ixodes ricinus, and a particular characteristic of the seasonal land surface temperature profile. A tick population model would allow us to move from this statistical analysis to a biological, process-based model for TBE virus and the many other pathogens vectored by Ixodes spp. ticks in Europe and the United States. Long-term fiel

The interplay between genetic variation and gene expression of the glucocorticoid receptor gene NR3C1 and blood cortisol levels on verbal memory and hippocampal volumes.

The hypothalamus-pituitary-adrenal axis is the main physiological stress response system and regulating the release of cortisol. The two corticoid receptors encoded by the genes NR3C1 and NR3C2 are the main players in regulating the physiological response to cortisol. This biological system has been linked to neurocognitive processes and memory, yet the mechanisms remain largely unclear. In two independent general population studies (SHIP, total sample size > 5500), we aim to diseantangle the effects of genetic variation, gene expression and cortisol on verbal memory and memory associated brai

Biologics in T2 Severe Asthma: Unveiling Different Effectiveness by Real-World Indirect Comparison.

Background: Indirect comparison among biologics in severe asthma (SA) is a challenging but desirable goal for clinicians in real life. The aim of the study is to define characteristics of a biologic-treated T2-driven-SA population and to evaluate the effectiveness of biologic treatments in a real-world setting by variation in intra/inter-biologic parameters in an up to 4-year follow-up. Methods: Demographic, clinical, functional, and biological characteristics were evaluated retrospectively in 104 patients recruited until July 2022 at baseline (T0) and over a maximum of 4 years (T4) of biologi

Population Pharmacokinetics of Candesartan in Patients with Chronic Heart Failure.

Heart failure (HF) causes pathological changes in multiple organs, thus affecting the pharmacokinetics (PK) of drugs. The aim of this study was to investigate the PK of candesartan in patients with HF while examining significant covariates and their related impact on estimated clearance using a population PK (Pop-PK) modeling approach. Data from a prospective, multicenter study were used. Modeling and simulations were conducted using Nonlinear Mixed-Effects Modeling (NONMEM) and R software. A total of 281 white patients were included to develop the Pop-PK model. The final model developed for a

Non-Hodgkin lymphoma and gluten-sensitive enteropathy: estimate of risk using meta-analyses.

OBJECTIVES: Gluten-sensitive enteropathy, including coeliac disease and dermatitis herpetiformis, is associated with non-Hodgkin lymphoma (NHL), and particularly enteropathy-associated T-cell lymphoma (EATCL). We conducted a meta-analysis to quantify the association. METHODS: Fifty-four risk estimates (range 0.28-300) were pooled using random-effects meta-analysis. Potential sources of variation were examined using sensitivity analyses and meta-regression. RESULTS: Thirty-one estimates with gluten-sensitive enteropathy diagnosed by serology then biopsy, serology alone, or recorded in medical

Population Model of Serum Creatinine as Time-Dependent Covariate in Neonates.

Serum creatinine (sCr) is a commonly measured biomarker to estimate glomerular filtration rate (GFR) and therefore widely used as a covariate in population pharmacokinetic models of renally excreted drugs. In neonates, sCr dynamically changes during the first few weeks after birth. Missing covariates are a common problem in pharmacokinetic modeling of neonates due to the limited availability of blood sampling in number and volume. The objective of this work is to develop a parsimonious population model describing time courses of sCr in neonates with the intent to be incorporated into pharmacok
