Endocrine disruptors affect larval zebrafish behavior: Testing potential mechanisms and comparisons of behavioral sensitivity to alternative biomarkers.

Larval zebrafish (Danio rerio) are a tool for assessing endocrine disruption during early development. Here, we investigated the extent to which a simple light/dark behavioral test at five days post fertilization could compliment current methods within the field. We exposed fertilized embryos to hormones (17β-estradiol, testosterone, dihydrotestosterone, 11-ketotestosterone, thyroxine, triiodothyronine, progesterone, and hydrocortisone) and other relevant compounds (17α ethinylestradiol, bisphenol A, bisphenol S, nonylphenol, flutamide, nilutamide, linuron, drospirenone, potassium perchlorate,

Surface-functionalized CdS clusters with recognition sites near the interface: selective luminescence response to lipophilic phenols.

A series of water-soluble cadmium sulfide clusters bearing an alkyl-chain layer between the inorganic core and the outer PEG layer were synthesized by the ligand-exchange reaction of Cd(10)S(4)(SPh)(12) with thiols functionalized by an N-(ω-PEGylated alkyl) amide moiety. The photoluminescence titration experiments in aqueous media revealed that clusters with a sufficiently hydrophobic inner environment exhibit definite emission enhancements upon the addition of bisphenol A or 4-nonylphenol. The dramatic effect of the alkyl chain length on the emission responses demonstrated that the hydrophobi

A simple Cr(VI)-S(IV)-O2 system for rapid and simultaneous reduction of Cr(VI) and oxidative degradation of organic pollutants.

Hexavalent chromium (Cr(VI)), a heavy-metal contaminant, can be easily reduced to less toxic trivalent chromium (Cr(III)) by sulfite ions (S(IV)). However, S(IV) has not drawn as much attention as the ferrous ion has. We report herein a novel Cr(VI)-S(IV)-O2 system containing sulfite ions that rapidly and simultaneously reduces Cr(VI) and oxidize organic pollutants in the presence of oxygen in aqueous solutions. This Cr(VI)-S(IV)-O2 system contains the initiator Cr(VI), the reductant S(IV), and the oxidant O2, which produce oxysulfur radicals (mainly SO4(-) and SO5(-)) and hydroxyl radicals (O

Influence of Bisphenol Compounds at Nanomolar Concentrations on Chromosome Damage Induced by Metabolically Activated Carcinogens in HepG2 Cells.

Bisphenol (BP) compounds are endocrine-disrupting organic pollutants. BPs may increase the messenger RNA (mRNA) transcripts of nuclear receptors (NRs) regulating the expression of xenobiotic-metabolizing cytochrome P450 (CYP) enzymes. Their impact on the genotoxicity of metabolically activated carcinogens, however, remains unknown. In this study, effects of the bisphenols A, F, S, and AF on the expression of the aryl hydrocarbon receptor (AhR), the pregnane X receptor (PXR), the constitutive androstane receptor, and individual xenobiotic-metabolizing CYP enzymes in a human hepatoma (HepG2) cel

Combined chemical exposure using exposure loads on human biomonitoring data of the 4th Flemish Environment and Health Study (FLEHS-4).

To improve our understanding of internal exposure to multiple chemicals, the concept exposure load (EL) was used on human biomonitoring (HBM) data of the 4th FLEHS (Flemish Environment and Health Study; 2016-2020). The investigated chemicals were per- and polyfluoroalkyl substances (PFASs), bisphenols, phthalates and alternative plasticizers, flame retardants, pesticides, toxic metals, organochlorine compounds and polycyclic aromatic hydrocarbons (PAHs). The EL calculates "the number of chemicals to which individuals are internally exposed above a predefined threshold". In this study, the 50th

Bisphenol S reduces locomotor performance and modifies muscle protein levels but not mitochondrial bioenergetics in adult zebrafish.

Human activity has now introduced novel chemicals into most aquatic ecosystems. Endocrine-disrupting compounds originating from plastic pollution and manufacture can have pronounced biological effects by disrupting hormone-mediated processes. Bisphenol A (BPA) is one of the most commonly produced endocrine-disrupting compounds, which interferes with signalling by a broad range of hormones. In recognition of its potentially harmful effects, BPA is being replaced by substitutes such as bisphenol S (BPS). However, toxicological studies revealed that BPS too can bind to hormone receptors and disru

Transgenerational thyroid endocrine disruption induced by bisphenol S affects the early development of zebrafish offspring.

Maternal thyroid hormones (THs) play an essential role in the embryonic and larval development of fish. Previous studies in fish have reported that parental exposure to thyroid disrupting chemicals (TDCs) changed maternal TH levels in the offspring; however, whether this transgenerational thyroid endocrine disruption can further disturb the early development of the offspring still remains largely unknown. Bisphenol S (BPS), a substitute of bisphenol A, has been reported to be a potential TDC. In this study, zebrafish (F0) were exposed to environmentally relevant concentrations (1, 10, and 100

Ultrasensitive electrochemical detection of bisphenol A using copper(ii) metal-organic framework ternary quantum dot (Cu-MOF/TQD) composite-modified gold electrodes.

A copper(ii) metal-organic framework (MOF) formulated as [Cu4(NITA)4(H2O)2(DMF)4]·(DMF)4 was prepared and characterized by single-crystal X-ray crystallography. The compound crystallized as a paddled wheel binuclear complex with each copper(ii) ion in a distorted square pyramidal geometry. The MOF was reacted with silver indium sulphide (AgInS2) ternary quantum dots (TQDs), prepared using a hydrothermal technique, to construct a composite electrochemical sensor, formulated as {AgInS2@[Cu4(NITA)4(H2O)2(DMF)4]·(DMF)4} with improved electrochemical performance. The composite was characterized by

Exploring the chromatographic performance of octadecanethiol modified spherical covalent organic frameworks stationary phase.

In recent years, covalent organic frameworks (COFs) have attracted considerable attentions with the advantages of high specific surface area, ordered pores, and excellent stability. However, the nanoscale size and irregular morphology of COFs have led to high back pressure, which hindered their application and development in the field of separation. Herein, we synthesized uniform spherical COFs via Schiff-base condensation by regulating the amount of acetic acid. Then, octadecanethiol was modified on the COFs via a simple click chemistry reaction to construct a novel stationary phase material

Transplacental transfer efficiency of environmental chemicals in association with gestational diabetes mellitus.

BACKGROUND: Many environmental chemicals have been shown to cross the placental barrier, threatening the health of offspring. Additionally gestational diabetes mellitus (GDM) may affect the transplacental transfer ratios (TTRs) of environmental chemicals, but the rate of transfer of a wide variety of environmental chemicals have not been widely studied. OBJECTIVE: We examined the TTRs of per- and poly- fluoroalkyl substances (PFAS), phthalates, parabens, bisphenols, and benzophenones in pregnant women, and if GDM increases transplacental transfer of these chemicals. METHODS: We examined 576

Prenatal exposure to environmental phenolic compounds and their association with childhood atopic dermatitis, asthma, and allergic rhinitis in the ECHO cohort.

Phenolic compounds may be harmful to the developing fetus, but many have not been studied in-depth for adverse childhood allergic and respiratory health effects. We hypothesized that higher levels of phenolic compounds in prenatal spot urine would be associated with greater odds of childhood atopic dermatitis, allergic rhinitis, and asthma, and that child sex may modify these associations. 3198 mother-child paired cases were enrolled from 16 cohorts in the U.S. ECHO consortium. Fifteen phenols (e.g. benzophenones, parabens, bisphenols, triclosans) were measured from mother's urine during pregn

Bisphenol S exposure before sexual maturity impairs follicular development via Cathepsin L-mediated granulosa cell death.

Bisphenol S (BPS) poses potential risks to the reproductive system, but its effects during pre-sexual maturity on follicle development and its underlying mechanisms remain unclear. This study aimed to investigate the association between pre-sexual BPS exposure and follicular dysplasia, as well as to decipher the molecular mechanisms involved. Urine samples from females aged 3-16 years were first analyzed, revealing a 95.7% detection rate of BPS, with a geometric mean concentration of 51.74 ng/L. Furthermore, the pre-sexual rat model revealed that BPS (0.05, 5, and 250 mg/kg body weight/day) re

Bisphenol S induces hepatic steatosis and insulin resistance through AMPK inhibition and endoplasmic reticulum stress.

Bisphenol S (BPS), a common substitute for bisphenol A, has emerged as a widespread environmental contaminant, yet its metabolic effects remain poorly understood. Increasing evidence links BPS exposure to metabolic dysfunction-associated steatotic liver disease (MASLD), although the underlying mechanisms are unclear. Here, we investigated the metabolic effects of chronic BPS exposure (4, 25, and 50 μg/kg/day for 12 weeks) in C57BL/6 mice and evaluated its impact on human Huh-7 hepatocytes. In vivo, exposure to 25 μg/kg/day exhibited obesogenic potential, while all doses induced insulin resista

Bisphenol S and high-fat diet drive structural and inflammatory remodeling of the colon in C57Bl/6 male mice.

Bisphenol S (BPS) is an emerging environmental contaminant with endocrine-disrupting properties capable of triggering early gastrointestinal alterations linked to intestinal remodeling. This study investigated the effects of BPS and a high-fat diet (HFD) on colon remodeling in male mice. Forty animals were divided into four groups: standard chow (SC), SC+BPS (SCB), HFD, and HFD+BPS (HFB). After 12 weeks, biometric parameters and colon morphology were analyzed. Both BPS and HFD induced weight gain, adiposity, and reductions in colon length, crypt depth, and mucus-positive area. Altered tight ju

Bisphenol S induces oxidative stress-mediated impairment of testosterone synthesis by inhibiting the Nrf2/HO-1 signaling pathway.

Bisphenol S (BPS) is an environmental endocrine disruptor widely used in industrial production. BPS induces oxidative stress and exhibits male reproductive toxicity in mice, but the mechanisms by which BPS impairs steroid hormone synthesis are not fully understood. Nuclear factor erythroid 2-related factor 2(Nrf2)/HO-1 signaling is a key pathway in improving cellular antioxidant defense capacities. Therefore, this study explored the effects of exposure to BPS on testosterone synthesis in adult male mice and its mechanisms with regard to the Nrf2/HO-1 signaling pathway. Adult male C57BL/6 mice

Transgenerational effects on intestinal inflammation status in mice perinatally exposed to bisphenol S.

Increasing evidence has highlighted the critical role of early life environment in shaping the future health outcomes of individuals in subsequent generations. Bisphenol S (BPS) has been widely used as a substitute for various plastic materials due to the limited application of Bisphenol A (BPA) which is an endocrine disruptor. However, the lack of efficient evaluation of BPS leaves doubts about the relevant substitute of BPA. Few studies of transgenerational inheritance have examined the effects of environmental exposures to endocrine disruptors on the immune system. In this study, we analyze

Transgenerational effects of bisphenol S exposure on the development of experimental asthma.

Early exposure to bisphenol A (BPA) is associated with increased asthma prevalence. To examine the effects of the most widely used BPA substitute, bisphenol S (BPS), on the development of childhood asthma, we conducted the study using the mouse model of early exposure. To simulate the burden from the human chronic exposure, we used a short-term exposure with 10 µg/ml BPS in the drinking water of female BALB/c mice (F0) from one week before pregnancy until the weaning of F1 pups. The pups were sensitized with low doses of ovalbumin (OVA) injection on postnatal day 4 and inhalation of OVA two we
