Methamphetamine (METH) abuse is a global health concern with serious repercussions, including potential disruption of the male reproductive system. Testicular integrity is particularly vulnerable to the toxic effects of METH. This review aims to comprehensively examine the current literature to elucidate the key mechanisms by which METH induces testicular damage, focusing on the cellular and molecular pathways involved. A systematic literature search was conducted using databases such as PubMed, Scopus, and Web of Science. Keywords included "methamphetamine," "testicular toxicity," "reproducti

Protein phosphatase, Mg2+/Mn2+ dependent, 1G (PPM1G) regulates protein function via dephosphorylation. PPM1G participates in the assembly of adherens junctions by dephosphorylating α-catenin. Here, we demonstrated through siRNA transfection and intratesticular injection that PPM1G is critical for maintaining blood-testis barrier function and regulating Sertoli cell apoptosis. We observed that upon knocking down Ppm1g in rat testes, the function of the blood testis barrier was compromised, and the localization of α-catenin and β-catenin became aberrant. Further investigation in rat Sertoli cell

The testis is known as an immunologically privileged organ. In particular, the blood-testis barrier formed by Sertoli cells protects auto-immunogenic spermatids from attack by the self-immune system. We review here the micro-status of testicular tissues in mice from the viewpoint of induction of inflammatory cell responses. Many studies have demonstrated that the testis is the most resistant to various forms of non-autoimmune inflammation among the male reproductive organs. However, it was found that testicular inflammation of autoimmune origin is inducible by immunization with testis antigens

Three aspects of the control of movements of fluids and substances into, out of and inside the testis are discussed: the tubular barrier, the interstitial extracellular fluid and the testicular blood vessels. The functional basis for the tubular barrier is twofold; there are significant differences in the concentration of many substances inside and outside the tubules and marker substances enter or leave the tubular fluid at widely different rates, depending on lipid solubility and the presence of specific carrier systems. The anatomical basis for this barrier appears to be the specialized jun

In this study, we have explored the role of the KATNB1 gene, a microtubule-severing protein, in the seminiferous epithelium of the rat testis. Our data have shown that KATNB1 expressed in rat brain, testes, and Sertoli cells. KATNB1 was found to co-localize with α-tubulin showing a unique stage-specific distribution across the seminiferous epithelium. Knockdown of KATNB1 by RNAi led to significant disruption of the tight junction (TJ) permeability barrier function in primary Sertoli cells cultured in vitro with an established functional TJ-barrier, as well as perturbations in the microtubule a

ABCC1 gene is expressed in various tissues and organs of the human body, and can transport substrates including drugs, heavy metals, toxic substances and organic anions. Previous research on ABCC1 gene has mostly focused on tumor multidrug resistance. Recently, ABCC1 has been proposed as a candidate gene for hereditary hearing impairment, which has attracted much attention. ABCC1-associated deafness may be related to its role in biological barriers. This article has summarized recent progress in the study of the role of ABCC1 in the blood-testis barrier, placental barrier, blood-brain barrier,

Eight-week-old male Kunming mice were administered either melamine (MA, 30, 140, or 700 mg/kg/day), a melamine and cyanuric acid mixture (MC, each at 15, 70, or 350 mg/kg/day), or vehicle (control) for 3 consecutive days. Testicular toxicity was evaluated on days 1 and 5 after the final exposure. The testicular and epididymal weights and serum testosterone level were significantly decreased in the highest MC group (350 mg/kg/day). Histopathologically, both MA and MC caused obvious lesions in the testis and epididymis, with significant increases in sperm abnormalities. By TEM, the blood-testis

Tight junctions (TJ) in inter-Sertoli junctional areas and epididymal epithelia are important for the formation of blood-testis barrier (BTB) and blood-epididymal barrier (BEB). In this study, the expression of occludin, an integral member of the TJ, was verified in canine testis and epididymis. Both low molecular weight (MW) (25-28 kDa) forms as well as high MW (68-72 kDa) forms of occludin were detected in the testis and epididymis using Western blot. The relative amount of the high MW forms of occludin vs low MW forms was higher in the testis than in the epididymis. Some difference in the c

BACKGROUNDS: The Sertoli cell that plays a vital role during spermatogenesis is a known target of physiological and pathological factors affecting testicular development. Tumor necrosis factor alpha (TNFα) participates in the blood-testis barrier reconstruction, cell apoptosis, and inflammatory response by recognizing receptors on Sertoli cell. TNFα has also been shown to induce the proliferation of immature Sertoli cell in vitro, yet the mechanism still remains unclarified.
OBJECTIVES: This study was designed to investigate the effect of TNFα on blood-testis barrier development during pubert

BACKGROUND: Male immunological infertility (MII), driven by anti-sperm antibody (AsAb), is a complex autoimmune disorder. The herbal formulation TuominJian (TMJ) has shown clinical efficacy against MII, but its immunomodulatory mechanisms involving Tfh cells, B cells, and cytokines remain unclear.
PURPOSE: This study aimed to elucidate the immunoregulatory effects and underlying mechanism of TMJ in MII mice.
METHODS: Active compounds in TMJ were identified using UPLC-Q/TOF-MS. An MII mouse model was established using sperm immunization. After verifying the model, testicular and epididymal ti

Blood-tissue barriers are specialized interfaces that safeguard organ homeostasis by restricting pathogen dissemination. Zika virus (ZIKV), an emerging flavivirus of global concern, exhibits an exceptional ability to breach multiple barriers-including the blood-brain, blood-placental, blood-testis, and blood-retinal barriers-enabling neuroinvasion, vertical and sexual transmission, and ocular disease. ZIKV employs diverse strategies to cross these barriers: receptor-mediated entry, disruption of tight junctions, and hijacking immune cells or extracellular vesicles as viral carriers. Adaptive m

Manganese (Mn2+ ) is an environmental pollutant, and testis is one of the main target organs. Coridius chinensis (C. chinensis), a traditional Chinese medicine, has been shown widely used in treating various kinds of pain, nephropathy and erectile dysfunction. In our recent study, we found that Mn2+ exposure caused testicular injury could be rescued in part by the antioxidant activity of C. chinensis extracts (CcE). However, there is dearth of extensive knowledge on the therapeutic effects of C. chinensis on manganese-induced reproductive toxicity. In the present study, Sprague-Dawley (SD) rat

The testis is an immune-privileged organ that balances protection of developing germ cells with the need to respond to pathogens. This review summarizes the dual nature of testicular immunity. A tolerogenic microenvironment is maintained through the blood-testis barrier (BTB) and immunomodulatory factors from Sertoli and Leydig cells, which suppress immune activation and preserve spermatogenesis. When infection, inflammation, or environmental stress disrupts this balance, immune responses shift toward pathology, inducing inflammatory cascades, apoptosis, and impaired fertility. We highlight th

The testis is a unique organ that simultaneously supports spermatogenesis and steroidogenesis, and it must maintain host defense against infection while suppressing excessive immune responses in an environment that includes germ cell antigens that emerge after sexual maturation. Testicular macrophages reside in the testicular interstitium starting in fetal life and localize near Leydig cells and around seminiferous tubules. Through cytokine and lipid mediator production, phagocytosis of dying cells, and antigen presentation, testicular macrophages can influence both steroidogenesis and spermat

The male reproductive system, particularly the male gamete, offers a unique barrier to the immune system. The growing germ cells in the testis need to be shielded from autoimmune damage. Hence the testis has to establish and sustain an immune-privileged milieu. Sertoli cells create this safe space, protected by the blood-testis barrier. Cytokines are a type of immune reaction that can positively and negatively affect male reproductive health. Inflammation, disease, and obesity are just a few physiological conditions for which cytokines mediate signals. They interact with steroidogenesis, shapi

Infertility impacts roughly 1 in 6 adults globally, where 50% of infertility cases are because of male factors. It is estimated that up to 15% of male infertility cases are because of disruption of testicular immune regulation. Thus, understanding mechanisms of immune privilege dysregulation resulting in blood-testis barrier disruption and germ cell loss could bring us one step closer to identifying ways to address infertility issues. In this views and reviews article, we introduce how testicular immune privilege is established at the blood-testis barrier through junction proteins between adja
