Transcriptomic and Phenotypic Analyses Reveal the Molecular Mechanism of Dwarfing in Tetraploid Robinia pseudoacacia L.

Polyploid breeding techniques aid in the cultivation of new forestry cultivars, thus expanding the suite of strategies for the improvement of arboreal traits and innovation within the field of forestry. Compared to diploid Robinia pseudoacacia L. (black locust) 'D26-5①' (2×), its dwarfed homologous tetraploid 'D26-5②' (4×) variety has better application prospects in garden vegetation guardrails and urban landscape. However, the molecular mechanism of the generation and growth of this dwarf variety is still unclear. Here, plant growth and development as well as histological differences between

Biological control of potato common scab and growth promotion of potato by Bacillus velezensis Y6.

Potato common scab, caused mainly by Streptomyces scabies, causes surface necrosis and reduces the economic value of potato tubers, but effective chemical control is still lacking. In this study, an attempt was made to control potato common scab by inoculating potatoes with Bacillus velezensis (B. velezensis) and to further investigate the mechanism of biological control. The results showed that B. velezensis Y6 could reduce the disease severity of potato common scab from 49.92 ± 25.74% [inoculated with Streptomyces scabies (S. scabies) only] to 5.56 ± 1.89% (inoculated with S. scabies and Y6

Making the 'Green Revolution' Truly Green: Improving Crop Nitrogen Use Efficiency.

Traditional breeding for high-yielding crops has mainly relied on the widespread cultivation of gibberellin (GA)-deficient semi-dwarf varieties, as dwarfism increases lodging resistance and allows for high nitrogen use, resulting in high grain yield. Although the adoption of semi-dwarf varieties in rice and wheat breeding brought big success to the 'Green Revolution' in the 20th century, it consequently increased the demand for nitrogen-based fertilizer, which causes severe threat to ecosystems and sustainable agriculture. To make the 'Green Revolution' truly green, it is necessary to develop

Cotton GhMKK5-GhMPK13 cascade regulates resistance to verticillium wilt by phosphorylating GhBES1-GhPEL75-mediated molecular module.

Verticillium wilt, caused by Verticillium dahliae, is a soil-borne vascular disease that harms many plants, including cotton. The mitogen-activated protein kinase cascade is a highly conserved signal transduction pathway that transmits various extracellular stimuli to downstream targets during plant defense responses. Although there have been many studies on the key role of the mitogen-activated protein kinase cascade in plant disease resistance, mitogen-activated protein kinase signal transduction in cotton remains to be elucidated. Here, we found that GhMKK5 interacted with GhMPK13 and direc

The pathogen-induced peptide CEP14 is perceived by the receptor-like kinase CEPR2 to promote systemic disease resistance in Arabidopsis.

Secreted plant peptides that trigger cellular signaling are crucial for plant growth, development, and adaptive responses to environmental stresses. In Arabidopsis (Arabidopsis thaliana), the C-TERMINALLY ENCODED PEPTIDE (CEP) family is a class of secreted signaling peptides that is phylogenetically divided into 2 groups: group I (CEP1-CEP12) and group II (CEP13-CEP15). Several group I CEP peptides regulate root architecture and nitrogen starvation responses, whereas the biological activity and roles of group II CEPs remain unknown. Here, we report that a group II CEP peptide, CEP14, functions

Unraveling hydrogen sulfide-promoted lateral root development and growth in mangrove plant Kandelia obovata: insight into regulatory mechanism by TMT-based quantitative proteomic approaches.

Mangroves are the main intertidal ecosystems with varieties of root types along the tropical and subtropical coastlines around the world. The typical characteristics of mangrove habitats, including the abundant organic matter and nutrients, as well as the strong reductive environment, are favor for the production of hydrogen sulfide (H2S). H2S, as a pivotal signaling molecule, has been evidenced in a wide variety of plant physiological and developmental processes. However, whether H2S functions in the mangrove root system establishment is not clear yet. Here, we reported the possible role of H

Brassinolide-mediated physiological and biochemical adaptation in rye (Secale cereale L.) under aluminum and freeze-thaw stress.

BACKGROUND: Environmental stresses such as extreme temperatures, salinity, drought, metal toxicity and freeze-thaw (FT) cycles severely limit crop productivity. The increasing frequency of FT events combined with aluminum (Al) toxicity, exacerbated by global warming, poses a serious threat to seedling survival, especially in early spring. Brassinolide (BL), a phytohormone, enhances plant resilience under abiotic stress, yet its role under combined FT and Al stress in rye (Secale cereale L.) remains unexplored. RESULTS: Al stress increased malondialdehyde content, indicating membrane lipid per

Integrated ATAC-Seq and RNA-Seq Data Analysis to Reveal OsbZIP14 Function in Rice in Response to Heat Stress.

Transcription factors (TFs) play critical roles in mediating the plant response to various abiotic stresses, particularly heat stress. Plants respond to elevated temperatures by modulating the expression of genes involved in diverse metabolic pathways, a regulatory process primarily governed by multiple TFs in a networked configuration. Many TFs, such as WRKY, MYB, NAC, bZIP, zinc finger protein, AP2/ERF, DREB, ERF, bHLH, and brassinosteroids, are associated with heat shock factor (Hsf) families, and are involved in heat stress tolerance. These TFs hold the potential to control multiple genes,

Rhizosphere microbiome-mediated aluminum detoxification in acidic soils: From microbial mechanisms to microecology-based management.

Aluminum (Al) toxicity in acidic soils severely constrains global crop production. Classical mechanisms of plant Al tolerance, established primarily in sterile laboratory systems, have centered on organic acid secretion. However, such mechanisms do not capture the critical contributions of rhizosphere microorganisms under field conditions. This review synthesizes current advances in microbe-mediated Al detoxification, aiming to reinterpret classical tolerance mechanisms from a microbial perspective and explore strategies for translating these insights into sustainable field applications. Leaf development is associated with coupled metabolome-fungi trajectories in the phyllosphere of Panax ginseng.

BACKGROUND: Leaf development underpins physiological performance and secondary metabolite accumulation, yet its influence on phyllosphere fungal communities in perennial medicinal plants such as ginseng (Panax ginseng C.A. Meyer) remains poorly understood. Understanding how developmental stage affects leaf chemistry and microbial assembly is important for linking plant metabolism with aboveground microbial ecology. RESULTS: We analyzed ginseng leaves across four developmental stages (S1-S4) using physiological measurements, ginsenoside profiling, untargeted metabolomics, ITS amplicon sequenci

Integrated transcriptome-metabolome analysis reveals a GmMYB093-GmCYP90A1-BR module supporting soybean adaptation to iron deficiency.

Iron deficiency is a major abiotic constraint that limits soybean growth, nodulation, symbiotic nitrogen fixation, and yield, yet objective criteria for evaluating low-Fe tolerance and the regulatory mechanisms linking root-nodule responses with shoot adaptation remain insufficiently defined. Here, we established an entropy-weight-based evaluation system using 62 soybean accessions and identified Wanhuang506 (Wh506) as a highly tolerant cultivar and Flyer as a highly sensitive cultivar. Physiological validation showed that Wh506 maintained higher Fe accumulation, chlorophyll retention, antioxi

GASA/snakin proteins in plants: structural conservation, phylogenomic expansion, and functional diversity.

Plant GASA/Snakin proteins (GASA: Gibberellic Acid-Stimulated Arabidopsis) constitute a family of secreted cysteine-rich peptides. The family is defined by 12 highly conserved cysteine residues forming up to six disulfide bonds within a thermostable helical scaffold. Phylogenetically restricted to vascular plants, the family ranges from 10 members in rice to approximately 40 in polyploid species such as peanut (Arachis hypogaea), with paralogous expansion driven by whole-genome, segmental, and tandem duplication events, while the 12-cysteine spacing pattern is maintained under purifying select

Plant immune co-receptors: bridging gaps toward next-generation crop defense.

Plant innate immunity is a crucial, multi-layered defense system that protects crops from a wide range of bacterial, fungal, and viral pathogens, all of which pose a significant threat to global food security. Co-receptors function as central molecular modulators that amplify and fine-tune immune signaling. This signaling is initiated by cell-surface pattern-recognition receptors (PRRs) and intracellular nucleotide-binding leucine-rich repeat (NLR) proteins. Critical co-receptors for strong immune signal propagation include the SOMATIC EMBRYOGENESIS RECEPTOR KINASE (SERK) protein family, repre

The loss of COP1 delays senescence and inhibits root growth through modulation of sucrose allocation and availability.

CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1) participates in diverse signaling pathways that regulate plant growth and development, but its role in sucrose-mediated signaling remains unclear. Here, we show that COP1 contributes to leaf senescence and root growth by modulating sucrose allocation and availability in Arabidopsis. Under sucrose-free conditions, cop1 mutants developed chlorotic leaves with reduced chlorophyll content, impaired primary root growth, and decreased starch granule formation in root columella cells. cop1 mutants also showed reduced expression of sucrose transport- and cataboli

Comparative transcriptome analysis provides insights into the dwarfing mechanism of pear trees.

BACKGROUND: Dwarfism is crucial for intensive cultivation and labor-saving management in modern orchards. However, the molecular mechanisms underlying pear tree dwarfing remain largely unclear. Here, we performed comparative transcriptome analysis between dwarf pear germplasm and pear cultivar 'Cuiguan'. RESULTS: The dwarf pear germplasm exhibited significantly shorter internode and branch length compared to 'Cuiguan'. Histological analysis revealed that cortical cells in the dwarf pear germplasm were disordered and irregular, and longer than those of 'Cuiguan'. Comparative transcriptome anal

MicroRNA-mediated regulation of cucumber (Cucumis sativus L.) fruit curvature.

Curved cucumber fruits are classified as a malformation, significantly compromising their commercial value. However, research on cucumber curvature remains limited, and there are no existing reports addressing post-transcriptional regulation mediated by microRNA. In this study, histological and endogenous hormone analyses revealed different distributions of auxin, brassinosteroid, and cytokinin between the concave and convex sides of curved fruits. This asymmetry in hormone distribution led to uneven growth. The growth was driven by parenchyma cells division on the second day post anthesis, an

Combination treatment with epibrassinolide overcomes tamoxifen resistance in breast cancer cells via endoplasmic reticulum stress induction.

BACKGROUND: Breast cancer (BC) is the most frequently diagnosed cancer among women and the leading cause of cancer-related mortality globally. One of the current systemic therapies against estrogen receptor-positive BC is hormonal therapy with tamoxifen (TAM), a selective estrogen receptor modulator. Recent studies showed that TAM resistance may occur often due to enhanced drug efflux mediated by ATP-binding cassette (ABC) transporters and ultimately can affect treatment efficacy. Epibrassinolide (EBR) is an apoptotic agent by inducing endoplasmic reticulum (ER) stress and the unfolded protein

Environmental stress responses and climate vulnerability of Abies beshanzuensis: combined physiological and transcriptomic analysis.

Abies beshanzuensis M. H. Wu is a critically endangered conifer endemic to cool, high-elevation forests; its narrow range renders it vulnerable to climate warming and associated heat and water stress. We simulated climate warming by growing seedlings along an elevational gradient (500-1200 m) to assess physiological and molecular stress responses. Seedlings at mid-high elevation (1000-1200 m) grew optimally, whereas low-elevation (500 m) seedlings showed stress symptoms. At 500 m, Yield and ETR declined sharply; stomatal conductance also dropped, and NPQ collapsed entirely, dropping below the

Drought-activated BZR1 reprograms flavonoid metabolism via transcriptional cascades to amplify baicalin biosynthesis in Scutellaria baicalensis.

Environmental stress triggers a dual response in medicinal plants by inhibiting growth while enhancing secondary metabolite production. However, the regulatory mechanism of this trade-off remains unclear. Here, we identified SbBZR1 as a brassinosteroid (BR)-responsive transcription factor that orchestrates drought resilience and baicalin biosynthesis in Scutellaria baicalensis through integrated multi-omics and metabolic flux analysis. In general, drought stress upregulated BR signaling and SbBZR1 expression. Transgenic Arabidopsis expressing SbBZR1 and its hyperactive mutant (Sbbzr1-1D) exhib

Thirty years of plant farnesylation research: Few substrates, many gaps, and a clear path forward.

Protein farnesylation - the covalent attachment of a 15-carbon farnesyl isoprenoid to a C-terminal CaaX motif by protein farnesyltransferase (PFT, FTase) - is one of the most functionally important post-translational modifications in eukaryotic biology. In mammals, the biomedical significance of farnesylation is well established. The FTase inhibitor (FTI) lonafarnib treats Hutchinson-Gilford progeria syndrome. FTase remains an oncology target, and chemical proteomic strategies have mapped more than 100 farnesylated substrates at single-protein resolution. In plants, genetic evidence establishe
