Environmental discontinuity hypothesis: Buffer dysfunctions as a source of human disease.

Adaptive physiologic buffers enable organisms to respond to environmental variation with appropriate plasticity. Modern humans have substantially remodeled their environment such that many interactions with the environment have become relatively discontinuous functions compared to the past. Examples include sunlight, temperature, and altitude. We propose that environmental discontinuity represents a Darwinian maladaptation and may promote disease by inducing buffer dysfunctions. Skin pigmentation is an adaptive, dynamic buffer that normalizes sunlight exposure to balance the potential harm of

Buffering effect of antacids in the mouth--a new treatment of dental erosion?

Antacids are drugs of choice in the treatment of reflux esophagitis and peptic ulcer disease, where acid regurgitation may cause dental erosion. Since there are no specific preventive measures or effective treatment for erosions, the present study was made to assess whether the keeping of antacids in the mouth before swallowing would be beneficial with regard to their effect on mouth pH, after acidic challenge. Five healthy subjects formed the test panel. After measuring the acid binding capacity of all the antacid preparations in the Finnish market, the two most effective were selected for te

Influence of food on aspirin absorption from tablets and buffered solutions.

After a standard meal, 12 normal volunteers received three aspirin dosage forms in a single-dose, complete crossover study. The three dosage forms were an unbuffered tablet, an effervescent solution with 16 meq of buffer, and an effervescent solution with 34 meq of buffer. Plasma and urine aspirin, salicylic acid, and salicyluric acid were measured for 10 hr. Significant differences in the absorption kinetics of aspirin were observed, with aspirin from the two solutions being absorbed faster than from the tablet. Urine pH and renal clearance for all three acid compounds were influenced by the

Venus: chemical weathering of igneous rocks and buffering of atmospheric composition.

Data from the Pioneer Venus radar mapper, combined with measurements of wind velocity and atmospheric composition, suggest that surface erosion on Venus varies with altitude. Calcium- and magnesium-rich weathering products are produced at high altitudes by gas-solid reactions with igneous minerals, then removed into the hotter lowlands by surface winds. These fine-grained weathering products may then rereact with the lower atmosphere and buffer the composition of the observed gases carbon dioxide, water vapor, sulfur dioxide, and hydrogen fluoride in some regions of the surface. This process i

[Barbital buffer in tablets as a solvent for complement fixation test (CFT)].

The purpose of this paper was to develop a tableting technology of barbital buffer as a standard diluent easy in use for complement fixation test (CFT). As starting point, barbital (veronal) buffer after Alton et al. [1] was assumed. Among 4 solubilizers polyvinyl pyrrolidone (PVP 10 000) was chosen, which provided a good solubility of the barbital in distilled water at 20 degrees C and it affected favourably the complement activity (C'). PVP 10 000 added to the components of barbital buffer at an amount of 0.275 per 10 tablets, in the form of solution for granulation, provided the required ph

The denaturation of human oxyhaemoglobin A, A2 and S by isopropanol/buffer method.

Human oxyhaemoglobin A and A2 from normal individuals and oxyhaemoglobin S from patients with sickle cell anaemia and sickle cell trait were studied using Isopropanol/buffer method at 37 degrees C and 40 degrees C. Hb S was less stable than Hb A, whereas Hb A2 was considerably more stable than either. Denaturation of Hb S was dependent on temperature and its concentration. Between the patients with sickle cell trait it was not possible to verify the influence of the concentration probably due to the small range used (from 38% to 44%).

Physical exercise after induced alkalosis (bicarbonate or tris-buffer).

The influence of bicarbonate and Tris-buffer infusions on the performance capacity for maximal, brief exercise (400 m run) was studied using 10 normal males in their twenties. Run time, maximal lactate concentration and heart rate remained unchanged after the buffer infusions. As a result of the induced elevated buffering capacity, the average pH after exercise was about 0.1 unit higher. Corresponding values for base excess and standard bicarbonate were found. The arterial pCO2 was higher after infusion as a result of the active respiratory compensation. Since the reduction in the work-related

In-office compounding of buffered lidocaine and epinephrine - A stability and safety review.

Buffered lidocaine is widely used in dermatologic and other in-office procedures to reduce injection pain and expedite onset of anesthesia. Current United States Pharmacopeia <797> standards limit immediate-use compounded sterile preparations to administration within 4 hours, creating significant barriers for dermatology practices where buffered lidocaine is routinely prepared in advance of patient visits. In collaboration with the U.S. Food and Drug Administration, Centers for Disease Control and Prevention, and United States Pharmacopeia, the American Academy of Dermatology Association, Amer

A Buffered LC-MS Method for Resolving and Quantifying Albiflorin and Paeoniflorin.

Albiflorin and paeoniflorin are bioactive isomers found in Paeoniae Radix Alba, a key component of Traditional Chinese Medicine. Their accurate and simultaneous quantification is essential for pharmacological research. Standard separation methods often rely on LC-MS using a mobile phase containing 0.1% formic acid. This study demonstrates that the use of 0.1% formic acid generates a secondary peak for albiflorin that exhibits an identical mass-to-charge ratio and similar fragmentation pattern as the main peak. To address this, an LC-MS method was developed using a Kinetex phenyl-hexyl column w

ADAR1 promotes cancer fitness by buffering endogenous RNA sensing.

Tumors exploit multifaceted strategies to survive and avoid destruction by the immune system. The RNA-editing enzyme adenosine deaminase acting on RNA 1 (ADAR1) regulates double-stranded (ds)RNA metabolism by catalyzing the adenosine-to-inosine (A-to-I) change, which is one of the most prevalent RNA modifications in mammalian cells. dsRNA binding and editing by ADAR1 skew the activation of cytosolic RNA sensors of innate immunity generally occurring during viral infections or upon accumulation of unedited endogenous RNAs. ADAR1 expression and global A-to-I RNA editing levels are elevated acros

Neutral Borate Buffer Activates Diazo for Rapid Protein Labeling.

Diazo, a carbene precursor, offers an effective chemistry for bioorthogonal labeling upon activation by acids, metals, and/or photo-illumination. Here, we report neutral borate buffer can trigger diazo compounds into carbene for protein labeling without the need for acid, metal, or light. We show that such boron-diazo-mediated covalent modification rapidly occurs within seconds under physiological conditions and preferentially targets carboxylate-containing amino acid residues. Towards applications of such boron-diazo protein chemistry, we exemplify the design of an affinity-based covalent inh

Influence of buffer distance on environmental geological hazard susceptibility assessment.

Previous researches seldom studied the selection of buffer distance between geological hazards (positive samples) and non-geological hazards (negative samples), and its reasonable selection plays a very important role in improving the accuracy of susceptibility zoning, protecting the environment and reducing the cost of hazard management. Based on GIS technology and random forest (RF) and frequency-ratio random forest (FR-RF) models, this study innovatively explored the influence of randomly selected non-geological hazard samples outside different buffer distances on the susceptibility evaluat

Pseudopod Tracking and Statistics During Cell Movement in Buffer and Chemotaxis.

Amoeboid cells such as the protist Dictyostelium, human neutrophils, and the fungus B.d. chytrid move by extending pseudopods. The trajectories of cell movement depend on the size, rhythm, and direction of long series of pseudopods. These pseudopod properties are regulated by internal factors such as memory of previous directions and by external factors such as gradients of chemoattractants or electric currents. Here a simple method is described that defines the X, Y time coordinates of a pseudopod at the start and the end of the extension phase. The connection between the start and end of an

Semitransparent Perovskite Solar Cells with Ultrathin Protective Buffer Layers.

Semitransparent perovskite solar cells (ST-PSCs) are increasingly important in a range of applications, including top cells in tandem devices and see-through photovoltaics. Transparent conductive oxides (TCOs) are commonly used as transparent electrodes, with sputtering being the preferred deposition method. However, this process can damage exposed layers, affecting the electrical performance of the devices. In this study, an indium tin oxide (ITO) deposition process that effectively suppresses sputtering damage was developed using a transition metal oxides (TMOs)-based buffer layer. An ultrat

Translational buffering by ribosome stalling in upstream open reading frames.

Upstream open reading frames (uORFs) are present in over half of all human mRNAs. uORFs can potently regulate the translation of downstream open reading frames through several mechanisms: siphoning away scanning ribosomes, regulating re-initiation, and allowing interactions between scanning and elongating ribosomes. However, the consequences of these different mechanisms for the regulation of protein expression remain incompletely understood. Here, we performed systematic measurements on the uORF-containing 5' UTR of the cytomegaloviral UL4 mRNA to test alternative models of uORF-mediated regu
