The effects of caffeine on the contraction of the frog heart.

1. The force of contraction and the membrane potentials have been measured from preparations of frog heart, with methods that also allow the rapid exchange of the extracellular fluid.2. In regularly beating preparations, caffeine induces only weak contractures at temperatures above 15 degrees C, but it does cause a marked potentiation of the twitch responses; longer exposure results in a depression of contraction. The build up and decline of the twitch strength, on addition and on removal of caffeine approximates to a single exponential, time constant 26-45 sec, and this time constant is not a

Refractory ventricular fibrillation caused by caffeine intoxication.

A 30-year-old female with no history of cardiac or systemic disease presented with incessant ventricular fibrillation (VF) after an intake of 12 g of over-the-counter caffeine tablets for a suicidal purpose. Her VF was refractory, and repeated defibrillations were needed to attain a recovery of sinus rhythms. We then performed percutaneous cardiopulmonary support and therapeutic hypothermia to stabilize her circulation and prevent anoxic brain damage, respectively. A blood examination revealed an extremely high concentration of caffeine (172 mg/L). She fully recovered 16 days after her admissi

Caffeine preferentially protects against oxygen-induced retinopathy.

Retinopathy of prematurity (ROP) is the leading cause of childhood blindness, but current anti-VEGF therapy is concerned with delayed retinal vasculature, eye, and brain development of preterm infants. The clinical observation of reduced ROP severity in premature infants after caffeine treatment for apnea suggests that caffeine may protect against ROP. Here, we demonstrate that caffeine did not interfere with normal retinal vascularization development but selectively protected against oxygen-induced retinopathy (OIR) in mice. Moreover, caffeine attenuated not only hypoxia-induced pathologic an

Caffeine metabolism before and after liver transplantation.

AIM: To study drug metabolism in patients before and after liver transplantation using caffeine as a probe drug. Forty-five patients undergoing liver transplantation for various liver diseases and who had well documented dossiers were selected for the study. Before the liver transplantation and 1 month, 1 year, and 6 years after liver transplantation, they were given 200 mg of caffeine by the oral route in the morning after voiding their bladder. Twenty-four-hour urine samples were collected and caffeine and metabolites were determined by HPLC: 1-methylurate (1U), 1-methylxanthine (1X), 1.7-di

Caffeine-induced diuresis and atrial natriuretic peptides.

After a single-blind, randomized, cross-over protocol using decaffeinated coffee in a control experiment, the effect of an oral 250-mg caffeine dose on plasma immunoreactive atrial natriuretic peptide (ANF) was assessed in eight healthy students who had been on a methylxanthine-free diet for 1 week. One to 2 h after caffeine ingestion, both systolic blood pressure (SBP) and diastolic BP (DBP) increased by 12 mm Hg while heart rate (HR) also tended to increase. An increase in diuresis and in urinary sodium, potassium, and osmol excretion was observed within 1 h. Decaffeinated coffee induced no

Biodegradation of caffeine by whole cells of tea-derived fungi Aspergillus sydowii, Aspergillus niger and optimization for caffeine degradation.

BACKGROUND: Pu-erh tea is a traditional Chinese tea and produced by natural solid-state fermentation. Several studies show that the natural microbiota influence caffeine level in pu-erh tea. Our previous research also found that the caffeine declined significantly (p < 0.05) in the fermentation, which suggested that the caffeine level could be influenced by specific strains. The purpose of this study was to isolate and identify microorganisms for caffeine degradation, and this research explored the degradation products from caffeine and optimal condition for caffeine degradation. RESULTS: 11

Postnatal period of caffeine treatment and time of testing modulate the effect of acute caffeine on cortical epileptic afterdischarges in rats.

Recently, we have found that postnatal caffeine treatment results in a dose-dependent pro- or anticonvulsant effect during brain maturation which effect is transient and disappears in adult rats. The aim of the present study was to determine whether the developmental period of chronic caffeine treatment and the age of testing are crucial for the effects of acute caffeine on cortical epileptic afterdischarges (ADs) in rats. Rhythmic electrical stimulation of somatosensory cortex was applied to elicit EEG and motor phenomena in 18- and 25-day-old rats, respectively. Acute injection of caffeine (

Homologous recombination as a potential target for caffeine radiosensitization in mammalian cells: reduced caffeine radiosensitization in XRCC2 and XRCC3 mutants.

The radiosensitizing effect of caffeine has been associated with the disruption of multiple DNA damage-responsive cell cycle checkpoints, but several lines of evidence also implicate inhibition of DNA repair. The role of DNA repair inhibition in caffeine radiosensitization remains uncharacterized, and it is unknown which repair process, or lesion, is affected. We show that a radiosensitive cell line, mutant for the RAD51 homolog XRCC2 and defective in homologous recombination repair (HRR), displays significantly diminished caffeine radiosensitization that can be restored by expression of XRCC2

A schedule induction probe technique for evaluating abuse potential: comparison of ethanol, nicotine and caffeine, and caffeine-midazolam interaction.

Previous research has shown that under daily 3h sessions of food schedule-induced drug solution polydipsia (on a fixed-time 1min (FT 1min) schedule, session probes at greater FT values induced comparatively more ml/pellet drinking of solutions of drugs possessing abuse potential (ethanol, cocaine, midazolam) than of vehicle or other drug solutions. The present study extended these findings to nicotine and caffeine, drugs that usually do not function as reinforcers in i.v. self-administration studies with animals, but are known to have reinforcing effects for humans. In vitro diagnosis of susceptibility to malignant hyperthermia: evaluation of tests with halothane-caffeine, potassium chloride, suxamethonium and caffeine-suxamethonium.

Susceptibility to MH was determined in 77 patients according to the protocol of the European MH Group. Additional muscle bundles were used for diagnostic in vitro tests with caffeine in the presence of 1% halothane, potassium chloride, suxamethonium, and caffeine-suxamethonium. Significant differences between MH-susceptible (MHS) and MH-negative (MHN) patients were obtained with the halothane-caffeine test and the potassium chloride test, whereas no differences were found with the suxamethonium test. In the test with caffeine-suxamethonium, a different response to caffeine but not to suxametho

Biosynthesis of caffeine by tea-leaf extracts. Enzymic formation of theobromine from 7-methylxanthine and of caffeine from theobromine.

1. Extracts prepared from tea leaves with Polyclar AT (insoluble polyvinylpyrrolidine) contained two methyltransferase activities catalysing the transfer of methyl groups from S-adenosylmethionine to 7-methylxanthine, producing theobromine, and to theobromine, producing caffeine. 2. The methyltransferases exhibited the same pH optimum (8.4) and a similar pattern of effects by metal ions, thiol inhibitors and metal-chelating reagents, both for theobromine and caffeine synthesis. Mg2+, Mn2+ and Ca2+ slightly stimulated enzyme activity but they were not essential. Paraxanthine was shown to be mos

Childhood respiratory outcomes after neonatal caffeine therapy.

Caffeine to prevent or treat apnea of prematurity in the newborn period is now standard of care for infants born very preterm. It has both short- and longer-term effects on respiratory health. In the short-term it reduces the duration of assisted ventilation and of oxygen therapy. It also reduces the rate of treatment for a patent ductus arteriosus, and of bronchopulmonary dysplasia. In the longer-term it improves expiratory airflow in childhood, and may have some benefits on respiratory health. Because it has not been used as a neonatal treatment for long enough, it is unknown if neonatal caf

Caffeine preserves quiet sleep in preterm neonates.

Caffeine is widely used in preterm neonates suffering from apnea of prematurity (AOP), and it has become one of the most frequently prescribed medications in neonatal intensive care units. Goal of this study is to investigate how caffeine citrate treatment affects sleep-wake behavior in preterm neonates. The observational study consists of 64 preterm neonates during their first 5 days of life with gestational age (GA) <32 weeks or very low birthweight of < 1500 g. A total of 52 patients treated with caffeine citrate and 12 patients without caffeine citrate were included. Sleep-wake behavior wa

Caffeine Sodium Benzoate for Electroconvulsive Therapy Augmentation.

OBJECTIVES: Because of an ongoing manufacturer shortage of injectable caffeine sodium benzoate (CSB), patients at our health system were given CSB compounded in-house to increase seizure response during electroconvulsive therapy (ECT). Therefore, we aimed to evaluate its effectiveness and safety as an ECT augmentation agent. METHODS: Medical records of patients who received compounded CSB at Virginia Commonwealth University Health System were reviewed to identify adults receiving it as part of an index ECT treatment course between June 2012 and December 2016. The primary outcome was change in

Bidirectional ventricular tachycardia induced by caffeine poisoning.

Bidirectional ventricular tachycardia (BVT) is a tachyarrhythmia characterized by 180-degree beat-to-beat alteration in the QRS axis. BVT is traditionally known as an electrocardiography (ECG) finding pathognomonic of digitalis poisoning and a hallmark of catecholamine-induced ventricular tachycardia. Apart from digitalis poisoning, aconitine poisoning is the only reported cause of poisoning-related BVT, and no report of caffeine-poisoning-related BVT is as yet available. A-27-year-old woman was transported to hospital with cardiac arrest from ventricular fibrillation after taking a massive do

Caffeine Prevents Memory Impairment Induced by Hyperhomocysteinemia.

L-Methionine chronic administration leads to impairment of memory. This impairment is due to the increase in the body oxidative stress, which damages neurons and prevents their firing. On the other hand, caffeine has antioxidant and neuroprotective effects that could prevent impairment of memory induced by L-methionine chronic administration. In the current study, this hypothesis was evaluated. L-methionine (1.7 g/kg/day) was orally administered to animals for 4 weeks and caffeine (0.3 g/L) treatment was added to the drinking water. The radial arm water maze (RAWM) was used to test spatial lea
