Physical activity, body mass index and bone mineral density-associations in a prospective population-based cohort of women and men: the Canadian Multicentre Osteoporosis Study (CaMos).

BACKGROUND: Physical activity (PA) is an important modifiable risk factor for both bone mineral density (BMD) and body mass index (BMI). However, BMI is itself strongly predictive of BMD. Our aim was to determine the association between PA and BMD, with consideration of BMI as a potential mediating factor. METHODS: The Canadian Multicentre Osteoporosis Study (CaMos) is a population-based prospective cohort study of Canadian women and men. PA was determined from interviewer-administered questionnaires at baseline and Year 5 and summarized as daily energy expenditure in total metabolic equivale

Predictors of aortic stenosis in homozygous familial hypercholesterolemia: A study from the Canadian HoFH registry.

BACKGROUND: Homozygous familial hypercholesterolemia (HoFH) is a rare genetic disease of low-density lipoprotein cholesterol (LDL-C) metabolism. This disease is associated with a major risk of both atherosclerotic cardiovascular disease and aortic stenosis (AS). The predictors of AS in this population are not well established. OBJECTIVE: To investigate the univariable and multivariable predictors of AS in patients from the Canadian HoFH registry. METHODS: Individuals from the Canadian HoFH registry were included in this retrospective longitudinal study. Clinical data were obtained from the t

Evaluation of late-onset Alzheimer disease genetic susceptibility risks in a Canadian population.

We performed case-control studies using 2 Canadian cohorts to examine the role of 10 promising Alzheimer's disease (AD) loci identified in recent genomewide association studies. Patients age 65 years and older diagnosed with AD at baseline (prevalent cases) or who developed AD during follow-up assessments (incident cases) were compared with control subjects with no cognitive impairment. Our prevalent case study comparing prevalent AD cases (n = 428) with participants with no cognitive impairment (n = 524) revealed a significant association of rs6656401 and rs3818361 (CR1), rs2075650 (TOMM40),

Childhood behavioral inhibition and attachment: Links to generalized anxiety disorder in young adulthood.

Generalized anxiety disorder (GAD) is under-treated yet prevalent among young adults. Identifying early risk factors for GAD would contribute to its etiological model and identify potential targets for intervention. Insecure attachment patterns, specifically ambivalent and disorganized, have long been proposed as childhood risk factors for GAD. Similarly, childhood behavioral inhibition has been consistently associated with anxiety disorders in adulthood, including GAD. Intolerance of uncertainty (IU), the tendency to react negatively to uncertain situations, has also been shown to be a crucia

Carotid intima media thickness in multiple sclerosis: A CLSA study.

BACKGROUND: People with multiple sclerosis (MS) have an increased incidence of atherosclerotic disease, including ischemic heart disease and stroke, compared to people without MS even after accounting for risk factors such as hypertension, dyslipidemia, diabetes and smoking. We compared carotid intima media thickness (CIMT), a surrogate of atherosclerosis, in people with MS and in two groups of people without MS (rheumatoid arthritis [RA]; all other participants). METHODS: We used data from participants in the Canadian Longitudinal Study on Aging (CLSA) who did not have known vascular disease

Active Child, Accomplished Youth: Middle Childhood Active Leisure Fuels Academic Success by Emerging Adulthood.

Background/Objectives: Physical activity is an important protective factor throughout life. However, little research has observed the associations between the practice of physical activity and academic success longitudinally, and none have done so with a pan-Canadian sample. This article aims to examine the prospective associations between active leisure in middle childhood and academic achievement in emerging adulthood, for both boys and girls, beyond several family factors. Methods: Participants are 2775 children from the National Longitudinal Study on Children and Youth (NLSCY) aged between

Prognostic factors associated with minimal improvement following acute whiplash-associated disorders.

STUDY DESIGN: Retrospective clinical cohort study. OBJECTIVE: To identify the prognostic factors associated with a poor response to treatment in the early stages of a whiplash-associated disorder (WAD). SUMMARY OF BACKGROUND DATA: Several demographic and clinical factors related to recovery following acute WADs have been identified. However, few longitudinal studies have investigated a multivariable model of recovery that includes socio-demographic, treatment, clinical, and nonclinical factors. METHODS: A study cohort of 2,185 patients with acute or subacute WADs presenting to 48 rehabilita

Characteristics Associated with Variation in Corticosteroid Exposure in Children with Steroid-Sensitive Nephrotic Syndrome: Results from a Canadian Longitudinal Study.

BACKGROUND: Variation in dose and duration of corticosteroids for childhood-onset steroid-sensitive nephrotic syndrome occurs worldwide, likely reflecting the evolving evidence on optimal dosing and variable severity of the disease observed between patients. We conducted a study to determine the associations between site, physician, and patient factors, and average daily corticosteroid dose and duration of therapy. METHODS: Data were derived from the Canadian Childhood Nephrotic Syndrome (CHILDNEPH) Project, an observational longitudinal study from 2013 to 2019 of children with nephrotic synd

Sociodemographic characteristics of SARS-CoV-2 serosurveillance studies with diverse recruitment strategies, Canada, 2020 to 2023.

BACKGROUND: Serological testing was a key component of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) surveillance. Social distancing interventions, resource limitations, and the need for timely data led to serosurveillance studies using a range of recruitment strategies, which likely influenced study representativeness. Characterizing representativeness in surveillance is crucial to identify gaps in sampling coverage and to assess health inequities. METHODS: We retrospectively analyzed three pre-existing longitudinal cohorts, two convenience samples using residual blood, and on

Rationale and Design of the Canadian Lipoprotein(a) Registry.

Lipoprotein(a) [Lp(a)] is an independent, heritable risk factor for atherosclerotic cardiovascular disease. Guidelines recommend Lp(a) testing once in a lifetime and recognize it as a risk-enhancing factor. However, management of elevated Lp(a) in real-world clinical practice is not well described. Here we describe the rationale, design, and preliminary baseline characteristics of the Canadian Lp(a) Registry, a prospective, longitudinal observational study of patients with Lp(a) ≥ 100 nmol/L (≥ 50 mg/dL). Patient demographics, cardiovascular risk factors, laboratory results, and clinical outco

Genetics versus clinical risk scores for melanoma prediction.

BACKGROUND: Cutaneous melanoma is a common cancer, for which risk stratification has been proposed to aid early detection. OBJECTIVES: To assess the performance of a polygenic risk score (PRS) in predicting the risk of invasive cutaneous melanoma, alone and combined with clinical risk factors. METHODS: The PRS was derived from the most recent genome-wide association study meta-analysis of cutaneous melanoma, which included 28 849 patients with melanoma and 78 922 control participants from 20 studies from the UK, the USA, Australia and Europe. Then, it was tuned in the Canadian Longitudinal S

Maternal concentrations of perfluoroalkyl sulfonates and alterations in white matter microstructure in the developing brains of young children.

BACKGROUND: Maternal exposure to per- and polyfluoroalkyl substances (PFAS) has been linked to child neurodevelopmental difficulties. Neuroimaging research has linked these neurodevelopmental difficulties to white matter microstructure alterations, but the effects of PFAS on children's white matter microstructure remains unclear. We investigated associations between maternal blood concentrations of six common perfluoroalkyl sulfonates and white matter alterations in young children using longitudinal neuroimaging data. METHODS: This study included 84 maternal-child pairs from a Canadian pregna

Social Determinants of Health and Symptomatic Recovery from Pediatric Concussion.

Social determinants of health (SDoH) are the non-medical factors that influence an individual's health and well-being. This study aimed to examine the relationships between SDoH and post-concussion symptom severity in Canadian children. We conducted a secondary analysis of data collected through Advancing Concussion Assessment in Pediatrics (A-CAP), a prospective longitudinal cohort study. Children aged 8-16 years with concussion (n = 633) or orthopedic injury (OI; n = 334) were recruited from five Canadian pediatric emergency departments. Post-concussion symptoms were rated by children and pa
