Acid-resistant Bacillus velezensis effectively controls pathogenic Colletotrichum capsici and improves plant health through metabolic interactions.

UNLABELLED: Colletotrichum capsici is the etiological agent of Capsicum anthracnose. Bacillus velezensis has traditionally been recognized as an effective biocontrol agent; however, its efficacy decreases due to soil acidification. In this study, we domesticated Bacillus velezensis XY40-1 along an acid resistance gradient, resulting in a strain capable of growth at pH 4, and might adapt to acidic environments by regulating genes related to spore formation. Notably, the domesticated Bacillus velezensis XY40-1 exhibits significant antagonistic activity against Colletotrichum capsici in acidic du

Spatial Engineering of Yarrowia lipolytica for the Biosynthesis of Capsanthin.

Capsanthin is a high-value tetraterpenoid widely utilized in the food, cosmetic, and pharmaceutical industries. However, its traditional production via plant extraction and chemical synthesis faces severe sustainability and cost constraints. The oleaginous yeast Yarrowia lipolytica (Y. lipolytica) offers a superior chassis due to its robust mevalonate pathway and abundant lipid droplets for pigment sequestration. In this study, we systematically engineered Y. lipolytica for the de novo biosynthesis of capsanthin. By heterologously co-expressing of Capsicum annuum capsanthin/capsorubin synthase

Impact of capsaicin on aroma release: in vitro and in vivo analysis.

Capsaicin is the main bioactive compound in chili pepper that leads to the perception of "spiciness". However, the effect of capsaicin on aroma release in the nose remains unexplained. This is the first study designed to measure capsaicin's impact on aroma release during consumption. In vitro studies, using static headspace analysis by atmospheric pressure chemical ionization-mass spectrometry (APCI-MS), showed no impact of capsaicin (5 ppm) on the gas-liquid partitioning equilibria of a range of aroma compounds. However, a significant reduction in aroma release was observed in vivo, during or

The NLR protein CbAR9 and the hydrolase CbSAHH form a module driving systemic acquired resistance against Colletotrichum in pepper.

Anthracnose, which is caused by fungal pathogens of the genus Colletotrichum, poses a substantial threat to global pepper (Capsicum spp.) production. However, the principal regulators and signaling pathways that mediate anthracnose resistance remain largely unknown. In this study, we demonstrate the roles of ANTHRACNOSE RESISTANCE 9 (CbAR9) and S-ADENOSYLHOMOCYSTEINE HYDROLASE (CbSAHH) in the methyl salicylate (MeSA) mobile signal activating systemic acquired resistance (SAR) in Capsicum baccatum. Phenotypic and molecular analyses showed that the nucleotide-binding domain and leucine-rich repe

Pepper Mild Mottle Virus Coat Protein as a Novel Target to Screen Antiviral Drugs.

Pepper mild mottle virus (PMMoV) has caused serious economic losses to crop production in many countries. The coat protein (CP) of PMMoV is a multifunctional protein proved to be a determining factor in the assignment of virulence type. Therefore, we studied the interaction between drugs and PMMoV CP as a method to screen anti-PMMoV agents. In this study, vanisulfane (6f) exhibited good inactivation activity (68.5%) by biological activity screening. Meanwhile, the green fluorescent protein and PMMoV CP expression changes of vanisulfane against PMMoV were verified by western blot and qRT-PCR ex

Mutation increasing β-carotene concentrations does not adversely affect concentrations of essential mineral elements in pepper fruit.

Vitamin and mineral deficiencies are prevalent in human populations throughout the world. Vitamin A deficiency affects hundreds of millions of pre-school age children in low income countries. Fruits of pepper (Capsicum annuum L.) can be a major dietary source of precursors to Vitamin A biosynthesis, such as β-carotene. Recently, pepper breeding programs have introduced the orange-fruited (of) trait of the mutant variety Oranzheva kapiya, which is associated with high fruit β-carotene concentrations, to the mutant variety Albena. In this manuscript, concentrations of β-carotene and mineral elem

Temperature and water activity interactively regulate the growth and aflatoxin biosynthesis of Aspergillus flavus in dried red chili peppers: A predictive modeling and multi-omics study.

Dried red chili pepper is widely consumed yet prone to contamination by Aspergillus flavus and its carcinogenic aflatoxins, though how this fungus grows and produces toxins on this distinctive substrate remains poorly understood. This study examined the effects of temperature and water activity (aw) on A. flavus growth and aflatoxin (AF) production in dried red chili pepper, and explored the associated molecular and metabolic responses through multi-omics analyses. Four isolates tested at 15-42 °C and aw 0.86-0.98 showed optimal mycelial growth at 30-37 °C and aw 0.98, whereas AF production pe

Pectin-modified Nano-selenium inhibits Phytophthora capsici infection and enhances the nutritional quality of pepper fruits.

Phytophthora blight caused by Phytophthora capsici severely limits pepper production, and chemical fungicides induce pathogen resistance and reduce fruit quality. Nano‑selenium (nano-Se) shows biocontrol potential but suffers from poor stability. Here, metabolomic analyses were performed to explore the mechanism by which pectin-modified nano-Se (nano-Se@PT) acts in pepper. Results showed that nano-Se@PT enhanced plant resistance by modulating the phenylpropanoid pathway (elevating 6-nitrocoumarin by 30%) and the tricarboxylic acid cycle. Nano-Se@PT recruited beneficial rhizosphere microorganis

Pepper feeding derivative Brucella sp. in the gut of Spodoptera litura (Lepidoptera: Noctuidae) affect the growth and development of host larvae.

BLTY is a pepper cultivar that has resistance to young larvae of Spodoptera litura Fabricius, whereas FXBX is a pepper cultivar suitable for the growth of S. litura larvae. The obtained microbiomics data suggest that the BLTY leaves fed S. litura larval guts inhabited more Brucella. In this study, two Brucella spp. (2FC-4 and 2FC-15) and one Microbacterium sp. (2FC-22) were isolated from the guts of BLTY and FXBX leaf-fed S. litura, respectively. The loading of 2FC-4 and 2FC-15 reduced the survival rate, average larval weight, diet consumption, pupation rate, and emergence rate of S. litura, w
