Effects of topical Catalase application on dental pulp tissue: a histopathological evaluation.

OBJECTIVES: recently, the role of antioxidant enzymes in wound healing and their therapeutic potential has been a subject of interest in medical sciences. However there is restricted knowledge on the results of topical application, optimal therapeutic dosage and carrier types. The purpose of this study was to investigate the wound healing response of dental pulp after direct pulp capping with an antioxidant enzyme; Catalase (C-40). METHODS: three mongrel dogs were used in this study. A total of 32 perforated Cl V preparations were treated with either 0.8mg (n=16) or 1.6mg (n=16) Catalase (C-4

Catalase-negative Actinomyces neuii subsp. neuii isolated from an infected mammary prosthesis.

In this case report a catalase-negative strain of Actinomyces neuii subsp. neuii is described as the possible causative agent of an infected mammary prosthesis. DNA hybridization studies and 16S rRNA analysis confirmed that the strain belongs to the species Actinomyces neuii subsp. neuii. Since this strain is the first A. neuii subsp. neuii strain reported to be catalase negative, the catalase reaction should no longer be considered a key reaction for the diagnosis of this species but must be interpreted in conjunction with other characteristics.

Infection-associated decline of cape buffalo blood catalase augments serum trypanocidal activity.

Clearance of trypanosomes from the blood of infected Cape buffalo was associated with the development of two responses: (i) complement-dependent and clone-specific lytic activity and (ii) complement-independent trypanocidal activity that was not restricted by trypanosome clone or species. This latter activity was mediated by H2O2 and required the presence of xanthine oxidase in serum but not the addition of purine substrates. Expression of the xanthine oxidase-dependent trypanocidal activity in Cape buffalo serum was coincident with, and required, a decline in its H2O2 catabolic activity. The

Catalase mediates acetaldehyde formation from ethanol in fetal and neonatal rat brain.

Fetal ethanol (E) exposure has well documented deleterious effects on brain development, yet it is uncertain if the neurotoxicity of maternal E consumption is generated by E itself, by its primary metabolite acetaldehyde (AcHO), or both. The current studies present evidence that homogenates of immature rat brains can generate AcHO via a catalase (CAT)-mediated reaction and that AcHO may be produced in vivo by this system. Homogenates of day 19 fetal rat brain were incubated with E (50 mM). When incubated with CAT inhibitors (sodium azide or 3-aminotriazole), AcHO formation was blocked, whereas

beta-D(+) glucose-glucose oxidase-catalase for use as an antioxidant system.

The purpose of this study was to investigate the use of beta-D(+) glucose-glucose oxidase (EC 1.1.3.4)-catalase (EC 1.11.1.6) (BDG-GO-CAT) as a new antioxidant system in solutions. A novel method for estimation of activity of the system was developed using a dissolved oxygen (DO) meter and an oxygen probe. The method can be used to determine the enzymatic activity of the system at GO concentrations of 0.005 to 0.030 unit/ml, with an r2 of 0.995 for the linearity of the standard curve, and can be adapted for analysis in any solution. At room temperature of 23.0 degrees +/- 2.0 degrees C, the ma

Catalase: A Potential Pharmacologic Target for Hydrogen Peroxide in the Treatment of COVID-19.

BACKGROUND: Acute respiratory distress syndrome in the elderly with COVID-19 complicated by airway obstruction with sputum and mucus, and cases of asphyxia with blood, serous fluid, pus, or meconium in newborns and people of different ages can sometimes cause hypoxemia and death from hypoxic damage to brain cells, because the medical standard does not include intrapulmonary injections of oxygen-producing solutions. The physical-chemical repurposing of hydrogen peroxide from an antiseptic to an oxygen-producing antihypoxant offers hope for the development of new drugs. METHODS: This manuscript

Enzyme activity and expression of catalases in response to oxidative stress in Sporothrix schenckii.

Sporothrix schenckii is a dimorphic fungus, pathogenic to humans and animals, which is usually infective in the yeast form. Reactive oxygen species (ROS) play an important role in the host's defense, damaging the pathogen's DNA, proteins, and lipids. To prevent oxidative damage, the ROS are detoxified by pathogen-derived antioxidant enzymes such as catalases (CATs). In this work, we analyzed the activity and expression level of three S. schenckii genes, designated as CAT1, CAT2, and CAT3, that putatively encoded for three isoforms of monofunctional CAT with a predicted molecular weight of 57.6

Novel insights in mammalian catalase heme maturation: effect of NO and thioredoxin-1.

Catalase is a tetrameric heme-containing enzyme with essential antioxidant functions in biology. Multiple factors including nitric oxide (NO) have been shown to attenuate its activity. However, the possible impact of NO in relation to the maturation of active catalase, including its heme acquisition and tetramer formation, has not been investigated. We found that NO attenuates heme insertion into catalase in both short-term and long-term incubations. The NO inhibition in catalase heme incorporation was associated with defective oligomerization of catalase, such that inactive catalase monomers

Catalases play differentiated roles in the adaptation of a fungal entomopathogen to environmental stresses.

The catalase family of Beauveria bassiana (fungal entomopathogen) consists of catA (spore-specific), catB (secreted), catP (peroxisomal), catC (cytoplasmic) and catD (secreted peroxidase/catalase), which were distinguished in phylogeny and structure and functionally characterized by constructing single-gene disrupted and rescued mutants for enzymatic and multi-phenotypic analyses. Total catalase activity decreased 89% and 56% in ΔcatB and ΔcatP, corresponding to the losses of upper and lower active bands gel-profiled for all catalases respectively, but only 9-12% in other knockout mutants. The catalases of Paracoccidioides brasiliensis are differentially regulated: protein activity and transcript analysis.

Paracoccidioides brasiliensis is a fungal pathogen of humans. The P. brasiliensis response to oxidative stress is largely unexplored. We report the analysis of three catalases, PbCatA, PbCatP and PbCatC. The former are monofunctional catalases and the latter is a catalase-peroxidase. Differential expression of catalases as measured by activity and by quantitative analysis of transcripts was observed in the morphological conversion and in response to different stress conditions. PbCatA manifested higher activity in the mycelial phase, showed increased activity during transition from mycelium to

Cytotoxicity of lawsone and cytoprotective activity of antioxidants in catalase mutant Escherichia coli.

Lawsone is an active naphthoquinone derivative isolated from henna (Lawsonia inermis L.), a widely used hair dye. Previous study on the toxicity of lawsone remains unclear since the involvement of oxidative stress and the kind of ROS (reactive oxygen species) involved have not been fully resolved yet. This present study reports the cytotoxic effects of lawsone and henna. We carried out CAT assay (a zone of inhibition test of bacterial growth and colony-forming efficiency test of transformant Escherichia coli strains that express mammalian catalase gene derived from normal catalase mice (Cs(a))

Liver targeting of catalase by cationization for prevention of acute liver failure in mice.

To achieve hepatic delivery of CAT for the prevention of CCl4-induced acute liver failure in mice, two types of cationized CAT derivatives, HMD- and ED-conjugated CAT, were developed. Slight structural changes occurred during cationization and the number of increased free amino groups was 3.1 in HMD-CAT and 13.6 in ED-CAT. 111In-cationized CAT derivatives showed an increased binding to HepG2 cells, and were rapidly taken up by the liver. H2O2-induced cytotoxicity in HepG2 cells was significantly prevented by preincubation of the cells with cationized CAT derivatives. A bolus intravenous inject

Inhibition of catalase by sulfite and oxidation of sulfite by H2O2 cooperating with ascorbic acid.

An oxidative detoxification of sulfite, which originates from sulfur dioxide taken up into a leaf, has not yet been fully understood. In this study, we discuss that redox reactions between sulfite and H2O2 have an important role for the detoxification of sulfite. Sulfite was oxidized by H2O2 and during the redox reaction, oxygen consumption was observed. The oxygen consumption was partially inhibited by superoxide dismutase, indicating that O2- is generated during the redox reaction. Oxidation of sulfite by H2O2 was also observed in the presence of ascorbic acid, and during the oxidation, no s

Role of cytochrome P-450 IIE1 and catalase in the oxidation of acetonitrile to cyanide.

Acetonitrile is a common industrial solvent and laboratory agent, which can be toxic if ingested. The toxicity of nitriles appears to be due to the production of cyanide, and detailed studies by Freeman and Hayes [(1988) Biochem. Pharmacol. 37, 1153-1159; (1987) Fundam. Appl. Toxicol. 8, 263-271] have shown that microsomes oxidize acetonitrile to cyanide. Treatment of rats with inducers of cytochrome P-450 IIE1 such as pyrazole, 4-methylpyrazole, and ethanol resulted in a 4- to 5-fold increase in cyanide production from acetonitrile by isolated microsomes. Phenobarbital treatment had a small s

Catalase and sulfur in the rice rhizosphere: An ultrastructural histochemical demonstration of a symbiotic relationship.

An ultrastructural study has been made of a symbiotic association between a sulfur bacterium and the roots of the rice plant (Oryza saliva L.). This association is proposed to have useful economic consequences in ameliorating hydrogen sulfide toxicity and associated Akiochi or Straighthead disease in lowland rice cultivation. The presence of catalase (E.C. 1.11.1.6) in rice roots and in some rhizosphere bacteria has been demonstrated in ultrathin sections of field-grown rice using a catalasespecific medium. Catalase was detected in capsules surrounding the catalase-negative S-bacteria. These c
