Catechol-grafted chitosan-based antioxidant hydrogel with rapid self-healing property for wound healing.

Chronic wounds, particularly those with non-healing ulcers, show high levels of oxidant stress, such as excessive reactive oxygen species (ROS), which delays the healing process. Bacterial infections in wounds increase the need for antibiotics, which brings up serious concerns regarding antibiotic resistance. The innovative antibiotic-free strategies to endow the hydrogels with antibacterial and antioxidant features including the grafting of ROS-scavenging moieties onto the hydrogel structure are in high demand for wound management. Herein, an antibacterial and antioxidant hydrogel with rapid

Aquatic toxicity of hydroquinone and catechol following metal oxide treatment to Ceriodaphnia dubia and Pimephales promelas.

Metal oxides comprise a large group of chemicals used in water treatment to adsorb organic pollutants. The ability of titanium dioxide (TiO2) and iron (III) oxide (Fe2O3) to reduce the chronic toxicity of (phenolic) C6H6(OH)2 isomers, namely hydroquinone (HQ) and catechol (CAT) to Ceriodaphnia dubia and Pimephales promelas (less than 24 h-old) were investigated. The toxic endpoints following metal oxide treatment were compared to endpoints of untreated CAT and HQ. In chronic toxicity testing, HQ resulted in greater toxicity than CAT for both test organisms; the median lethal concentrations (LC

Poly-Catecholic Functionalization of Biomolecules for Rapid Gelation, Robust Injectable Bioadhesion, and Near-Infrared Responsiveness.

Mussel-inspired catechol-functionalization of degradable natural biomaterials has garnered significant interest as an approach to achieve bioadhesion for sutureless wound closure. However, conjugation capacity in standard coupling reactions, such as carbodiimide chemistry, is limited by low yield and lack of abundant conjugation sites. Here, a simple oxidative polymerization step before conjugation of catechol-carrying molecules (i.e., 3,4-dihydroxy-l-phenylalanine, l-DOPA) as a potential approach to amplify catechol function in bioadhesion of natural gelatin biomaterials is proposed. Solution

Dinuclear copper(II) complexes with derivative triazine ligands as biomimetic models for catechol oxidases and nucleases.

The research reported herein focuses on the synthesis of two new Cu(II) complexes {[Cu2(2-X-4,6-bis(di-2-picolylamino)-1,3,5-triazine], with X = butane-1,4-diamine (2) or N-methylpyrenylbutane-1,4-diamine (3)}, the latter with a pyrene group as a possible DNA intercalating agent. The structure of complex (3) was determined by X-ray crystallography and shows the dinuclear {CuII(μ-OCH3)2CuII} unit in which the CuII···CuII distance of 3.040 Å is similar to that of 2.97 Å previously found for 1, which contains a {CuII(μ-OH)2CuII} structural unit. Complexes (2) and (3) were also characterized in sp

Catechol-O-methyltransferase (COMT) polymorphisms modulate working memory in individuals with schizophrenia and healthy controls.

OBJECTIVE: Cognitive impairment is a core feature of schizophrenia, related to dopaminergic dysfunction in the prefrontal cortex (PFC). It is hypothesized that functional single nucleotide polymorphism (SNP) rs4680 of the catechol-O-methyltransferase (COMT) gene could mediate the relationship between cognition and dopamine activity in the PFC. Other COMT SNPs could also play a role. METHODS: We evaluated the role of three COMT SNPs (rs737865, rs165599, and rs4680) in schizophrenia and their impact on three working memory tasks. For genetic association analyses, 212 individuals with schizophre

Preliminary Evidence for an Association Between Variants of the Catechol-O-Methyltransferase (COMT) Gene and Premature Ejaculation.

BACKGROUND: Studies have suggested that dopamine plays a role in the neurobiological mechanism that triggers ejaculation, leading scientists to hypothesize that dopamine-related genetic polymorphisms could contribute to symptoms of premature ejaculation (PE). AIM: To investigate associations between dopamine receptor and catechol-O-methyltransferase (COMT; an enzyme involved in the catabolism of dopamine) gene-linked polymorphisms and PE. METHODS: PE status in patient groups was determined by clinical diagnosis performed by a physician specializing in sexual medicine. Self-reported PE sympto

Genetic variation in catechol-O-methyltransferase modifies effects of clonidine treatment in chronic fatigue syndrome.

Clonidine, an α2-adrenergic receptor agonist, decreases circulating norepinephrine and epinephrine, attenuating sympathetic activity. Although catechol-O-methyltransferase (COMT) metabolizes catecholamines, main effectors of sympathetic function, COMT genetic variation effects on clonidine treatment are unknown. Chronic fatigue syndrome (CFS) is hypothesized to result in part from dysregulated sympathetic function. A candidate gene analysis of COMT rs4680 effects on clinical outcomes in the Norwegian Study of Chronic Fatigue Syndrome in Adolescents: Pathophysiology and Intervention Trial (NorC

Catechol-O-methyltransferase (COMT) genotype moderates the effects of childhood trauma on cognition and symptoms in schizophrenia.

The interaction of genetic and environmental factors may affect the course and development of psychotic disorders. We examined whether the effects of childhood trauma on cognition and symptoms in schizophrenia were moderated by the Catechol-O-methyltransferase (COMT) Val(158)Met polymorphism, a common genetic variant known to affect cognition and prefrontal dopamine levels. Participants were 429 schizophrenia/schizoaffective cases from the Australian Schizophrenia Research Bank (ASRB). Cognitive performance was assessed using the Repeatable Battery for Assessment of Neuropsychological Status (

Catechol O-methyltransferase and monoamine oxidase A genotypes, and plasma catecholamine metabolites in bipolar and schizophrenic patients.

Metabolites of dopamine and norepinephrine measured in the plasma have long been associated with symptomatic severity and response to treatment in schizophrenic, bipolar and other psychiatric patients. Plasma concentrations of catecholamine metabolites are genetically regulated. The genes encoding enzymes that are involved in the synthesis and degradation of these monoamines are candidate targets for this genetic regulation. We have studied the relationship between the Val158Met polymorphism in catechol O-methyltransferase gene, variable tandem repeat polymorphisms in the monoamine oxidase A g

Catechol-O-methyltransferase (COMT) gene variants predict response to bupropion therapy for tobacco dependence.

BACKGROUND: Although bupropion is efficacious for smoking cessation, only a minority of smokers are able to quit. Pharmacogenetic research may improve treatment outcomes through discovery of DNA sequences predictive of successful pharmacotherapy for subgroups of smokers. We investigated variants in the catechol-O-methyltransferase (COMT) gene in a smoking cessation trial of bupropion. METHODS: A double-blind, placebo-controlled, 10-week trial of bupropion and counseling (with a 6-month follow-up period) was conducted at two university-based smoking cessation research programs. Polymorphisms of the estrogen-metabolizing genes CYP17 and catechol-O-methyltransferase and risk of epithelial ovarian cancer.

Because some studies have linked polymorphic variants of the estrogen-metabolizing genes CYP17 and catechol O-methyl transferase (COMT) with risk for hormonally related cancers, we sought to determine whether selected polymorphisms of these genes differed between women with and without ovarian cancer. From a population-based study of ovarian cancer, we analyzed DNA from a total of 480 cases and controls. PCR amplification was performed using primers that amplify restriction sites for MspAI (A2 polymorphism-CYP17) and NlaIII (Val/Met polymorphism-COMT). Digestion of the PCR products was perform

Identification and functional analysis of 2,3-dihydroxybenzoic acid and catechol catabolic pathway genes in Aspergillus niger.

Microorganisms catabolize many plant-derived aromatic compounds through the β-ketoadipate pathway. This pathway has two branches, beginning with protocatechuic acid and catechol, which converge on β-ketoadipate before further catalysis to produce tricarboxylic acid cycle intermediates. The protocatechuic acid branch of the pathway has been characterized in fungi. Here, we characterized the genes of the catechol branch of the β-ketoadipate pathway in the filamentous fungus Aspergillus niger. We predicted the genes involved using comparative transcriptomics and homology to characterized genes. We

Visible-Light-Curable Catechol-Reinforced Gelatin Bioglue for Rapid Hemostasis and Engineered Tissue Assembly.

Hydrogel-based tissue adhesives hold promise for diverse biomedical applications, yet achieving strong, durable adhesion to wet, dynamic biological surfaces remains challenging. The catechol-containing amino acid 3,4-dihydroxy-l-phenylalanine (DOPA) is a crucial functional motif in mussel-inspired adhesives and has been extensively investigated for incorporation into various biomaterials. However, conventional approaches of DOPA incorporation and subsequent crosslinking have shown limited potential for clinical translation. Discovery of Catechol-Benzothiazole Conjugates as Antibacterial Synergists against Pseudomonas aeruginosa by Inhibiting Biofilm Formation.

The formation of Pseudomonas aeruginosa (P. aeruginosa) biofilm hinders the efficacy of antibiotics, making clinical treatment challenging. Given that disrupting iron homeostasis represents a promising strategy for treating biofilm infections, a series of catechol-conjugated benzothiazole derivatives with iron-chelating properties was designed and synthesized. Among them, 4p was identified as the hit compound, demonstrating potent biofilm inhibition (IC50 = 0.27 μM). Mechanistic studies demonstrated that 4p attenuates biofilm formation by inhibiting heme oxygenase (HemO), impairing iron homeos
