Solitary fibrous tumor of the spinal cord.

We report a case of primary solitary fibrous tumor occurring in the intramedullary thoracic spinal cord in a 47-year-old man. The tumor predominately consisted of spindle cells separated by abundant collagen; a few areas of hemangiopericytomatous morphology were also present. The diagnosis was confirmed by immunohistochemistry and electron microscopy. The tumor was reactive to vimentin and CD34 but was negative for glial fibrillary acid protein (GFAP), S-100, smooth muscle actin, epithelial membrane antigen, HMB-45, myelin basic protein, and keratin; ultrastructural examination showed fairly u

Histological and histochemical investigations of achondroplastic mice: a possible model of human achondroplasia.

Histological and histochemical investigations of tibial epiphyses, costo-chondral junctions and caudal vertebrae of achondroplastic (cn) mice have shown that, in spite of conspicuous reduction of bone length, endochondrial ossification occurs in much the same way as in controls. Moreover, the histological structure of seriated cartilage, and the distribution of proteoglycans in resting cartilage are the same in cn/cn mice and in controls. The only difference betweeen the two types of animals is represented by the occurence of early aging-like changes of chondrocytes and cartilage matrix in ach

Inducing In Situ Functional Maturation of Transplanted Human Induced Pluripotent Stem Cell-Derived Cardiomyocytes: Establishing Strategies for Treating Myocardial Injury.

Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) have demonstrated significant potential for the treatment of heart diseases. However, hiPSC-CMs generated by the current methods still exhibit structural and electrophysiological immaturity, resembling fetal cardiomyocytes. Although various strategies have been developed to promote in vitro maturation, cell loss and death remain persistent challenges during transplantation. Therefore, a multipronged approach is developed to induce in situ hiPSC-CMs functional maturation and enhance the cell transplantation rate, not only al

Healed Perforated Corneal Ulcers in Human.

This study investigates the pathophysiological process of healed perforated corneal ulcers (HPCUs) in humans. All subjects underwent keratoplasty due to opacities or leakage from HPCUs. Half of each specimen was fixed with 4% glutaraldehyde for transmission electron microscope (TEM) examination. The other half was fixed in 10% formaldehyde for immunofluorescence (IF) examination. TEM identified layered structures with two cell types (polygonal and elongated) connected by gap or adherent junctions during early stage of healing. Both apoptotic and mitotic changes were found in both types of cell

TGFβ signaling networks in ovarian cancer progression and plasticity.

Epithelial ovarian cancer (EOC) is a leading cause of cancer-related death in women. Late-stage diagnosis with significant tumor burden, accompanied by recurrence and chemotherapy resistance, contributes to this poor prognosis. These morbidities are known to be tied to events associated with epithelial-mesenchymal transition (EMT) in cancer. During EMT, localized tumor cells alter their polarity, cell-cell junctions, cell-matrix interactions, acquire motility and invasiveness and an exaggerated potential for metastatic spread. Key triggers for EMT include the Transforming Growth Factor-β (TGFβ

The disturbed blood-brain barrier in human glioblastoma.

The aim of this article is to describe alterations of the blood-brain barrier (BBB) in gliomas. The main clinical problem of human gliomas is the edematous swelling and the dramatic increase of intracerebral pressure, also compromising healthy areas of the brain. According to our concept, one of the main reasons on the cellular level for these clinical problems is the loss or reduction of astroglial polarity. Astroglial polarity means the specific accumulation of potassium and water channels in the superficial and perivascular astroglial endfeet membranes. The most important water channel in t

Genome-Wide Association Study to Identify Novel Genetic Variants Associated With Primary Budd-Chiari Syndrome.

BACKGROUND: Primary Budd-Chiari syndrome (BCS) is a rare hepatic vascular disorder characterized by hepatic venous outflow obstruction involving the hepatic veins and/or inferior vena cava. Marked geographic variation in vascular anatomy and prothrombotic profile suggests an underlying genetic predisposition. This study aimed to identify genetic variants associated with primary BCS using genome-wide association study (GWAS) and to characterize implicated biological pathways. METHODS: In a case-control study (June 2020-May 2022), 310 adults with primary BCS and 312 age- and sex-matched healthy

Modification of the composition of polycystin-1 multiprotein complexes by calcium and tyrosine phosphorylation.

Mutations in the PKD1 gene are responsible for >85% of autosomal dominant polycystic kidney disease (ADPKD). The protein product of PKD1, polycystin-1, is a large, modular membrane protein, with putative ligand-binding motifs in the extracelluar N-terminal portion, 9-11 transmembrane domains and an intracellular C-terminal portion with phosphorylation sites. A role for polycystin-1 as a cell surface receptor involved in cell-matrix and cell-cell interactions has been proposed. In this study, we have analyzed polycystin-1 and associated protein distribution in normal human epithelial cells and

Spontaneous malignant myoepithelioma in the parotid gland of an aged female Crl:CD (SD) rat.

An aged female Crl:CD (SD) rat developed a subcutaneous tumor in the left occipital region. Histologically, the tumor was predominantly composed of spindle-shaped cells proliferating in a loosely cohesive fascicular arrangement in a basophilic myxoid matrix with a minor population of epithelioid cells. Metastases were detected in the cervical lymph nodes, kidneys, lungs, and the pleural cavity. Duct-like structures, considered to be entrapped normal salivary ducts, were scattered within the tumor. Immunohistochemically, the tumor cells were diffusely positive for vimentin, focally positive for

Endothelial LRP1 protects against neurodegeneration by blocking cyclophilin A.

The low-density lipoprotein receptor-related protein 1 (LRP1) is an endocytic and cell signaling transmembrane protein. Endothelial LRP1 clears proteinaceous toxins at the blood-brain barrier (BBB), regulates angiogenesis, and is increasingly reduced in Alzheimer's disease associated with BBB breakdown and neurodegeneration. Whether loss of endothelial LRP1 plays a direct causative role in BBB breakdown and neurodegenerative changes remains elusive. Here, we show that LRP1 inactivation from the mouse endothelium results in progressive BBB breakdown, followed by neuron loss and cognitive defici

Ultrastructurally "invasive" microvilli in an aggressively metastasizing biphasic malignant mesothelioma.

The presence of long, slender, often branching microvilli on cell surfaces is a characteristic feature of malignant and benign mesothelial cells. However, these typical microvilli are seen only in better-differentiated lumens within epithelial areas of malignant mesotheliomas. Presented here are the clinical and ultrastructural findings in a biphasic malignant mesothelioma that lacked lumens, but possessed very long microvilli. These invaginated deeply into the cytoplasm of neighboring tumor cells, as well as into the surrounding stromal matrix. The tumor cells had well-formed intercellular de

Chordomalike soft tissue sarcoma in the leg: a light and electron microscopic and immunohistochemical study.

A soft tissue tumor in the leg of a 67-year-old woman is described. This large tumor below the knee area infiltrated extensively the deep and superficial soft tissues but did not involve the bones. The tumor cells formed nodules resembling the architecture seen in chondroid tumors and chordoma. The tumor cells were often vacuolized, and there was extracellular myxoid matrix similar to that in chordoma or myxoid chondrosarcoma. Immunohistochemistry showed keratins 8 and 19, epithelial membrane antigen, and vimentin in most tumor cells, and there was also S-100 protein positivity in a number of

The Impaired Wound Healing Process Is a Major Factor in Remodeling of the Corneal Epithelium in Adult and Adolescent Patients With Keratoconus.

PURPOSE: Keratoconus (KTCN) is the most common corneal ectasia, characterized by pathological cone formation. Here, to provide an insight into the remodeling of the corneal epithelium (CE) during the course of the disease, we evaluated topographic regions of the CE of adult and adolescent patients with KTCN. METHODS: The CE samples from 17 adult and 6 adolescent patients with KTCN, and 5 control CE samples were obtained during the CXL and PRK procedures, respectively. Three topographic regions, central, middle, and peripheral, were separated toward RNA sequencing and MALDI-TOF/TOF Tandem Mass

Supramolecular Assemblies of Glycopeptides Enhance Gap Junction Maturation of Human Induced Pluripotent Stem Cell-Derived Cardiomyocytes via Inducing Spheroids Formation to Optimize Cardiac Repair.

Stem cell-based therapies have demonstrated significant potential for use in heart regeneration. An effective paradigm for heart repair in rodent and large animal models is the transplantation of human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs). Despite this, the functional and phenotypical immaturity of 2D-cultured hiPSC-CMs, particularly their low electrical integration, poses a caveat for clinical translation. In this study, a supramolecular assembly of a glycopeptide containing a cell adhesion motif-RGD, and saccharide-glucose (Bio-Gluc-RGD) is designed to enable the

The Kallikrein-Kinin System: Proteolytic Orchestrators of Tissue Barrier Disruption in Inflammation and Cancer.

The kallikrein-kinin system (KKS) and the kallikrein-related peptidase (KLK) family are interconnected proteolytic networks that regulate inflammatory signaling, vascular permeability, extracellular matrix remodeling, and tissue barrier dynamics. Beyond their classical vasoactive and inflammatory functions, accumulating evidence indicates that kinin peptides, including bradykinin, Lys-bradykinin, and their des-Arg9 metabolites, together with selected KLKs, modulate cell-cell and cell-extracellular matrix adhesion. Through B1 and B2 kinin receptor activation, the KKS influences endothelial adhe

[Regenerative medicine: stem cells, cellular and matricial interactions in the reconstruction of skin and cornea by tissue engineering].

Considering that there is a shortage of organ donor, the aim of tissue engineering is to develop substitutes for the replacement of wounded or diseased tissues. Autologous tissue is evidently a preferable transplant material for long-term graft persistence because of the unavoidable rejection reaction occuring against allogeneic transplant. For the production of such substitutes, it is essential to control the culture conditions for post-natal human stem cells. Furthermore, histological organization and functionality of reconstructed tissues must approach those of native organs. For self-renew

Integration of Stromal Cells and Hydrogel Below Epithelium Results in Optimal Barrier Properties of Small Intestine Organoid Models.

Background/Objectives: The barrier properties of the human small intestine play a crucial role in regulating digestion, nutrient absorption and drug metabolism. Current in vitro organotypic models consist only of an epithelium, which does not take into account the possible role of stromal cells such as fibroblasts or the extracellular matrix (ECM) which could contribute to epithelial barrier properties. Therefore, the aim of this study was to determine whether these stromal cells or ECM were beneficial or detrimental to barrier function when incorporated into an organotypic human small intesti

Nfe2l2-knockout mouse model exhibits oxidative stress-associated stromal degeneration and inflammatory activation resembling keratoconus pathology.

Oxidative stress is central to keratoconus (KC) pathogenesis, and NRF2 is a key antioxidant regulator. However, an in vivo model linking NRF2 loss to KC has been lacking. We generated CRISPR/Cas9 Nfe2l2-knockout (KO) mice and assessed 4-month-old corneas by slit-lamp fluorescein staining, optical coherence tomography (OCT), histology/immunofluorescence and transmission electron microscopy (TEM), reactive oxygen species (ROS) assays, βIII-tubulin whole-mounts, and whole-cornea single-cell RNA-seq with Seurat/CellChat. Nfe2l2 loss caused central corneal thinning, increased fluorescein uptake, an

Human organ chip models recapitulate FRC-driven high endothelial venule differentiation, immune recruitment, and cancer-induced disruption.

High endothelial venules (HEVs) are specialized post-capillary venules that gate lymphocyte entry into lymph nodes, yet human in vitro models that capture their specialization remain limited. Here, we develop a perfusable human HEV-on-chip in which blood endothelial cells (BECs) form an engineered microvessel within a collagen-fibrin matrix populated by human fibroblastic reticular cells (FRCs). The presence of FRCs, without exogenous LIGHT, drives HEV-associated marker expression (PNAd/MECA-79 and VCAM-1) and a plump endothelial wall enwrapped by a thickened laminin+ basement membrane, recapi

Collagen XVII/BP180: a collagenous transmembrane protein and component of the dermoepidermal anchoring complex.

Collagen XVII, or BP180, is a collagenous transmembrane protein and a structural component of the dermoepidermal anchoring complex. Molecular studies reveal that it has a globular cytosolic amino-terminal domain and flexible-rod extracellular carboxyterminal domain. The extracellular portion of collagen XVII is constitutively shed from the cell surface by ADAMs (proteinases that contain adhesive and metalloprotease domains). Cell biological analyses suggest that collagen XVII functions as a cell-matrix adhesion molecule through stabilization of the hemidesmosome complex. This concept is suppor
