Dental pulp stem cells (DPSCs) have excellent proliferative properties, mineralization potential and can be easily obtained from third molar teeth. Recently, many studies have focused on isolation and differentiation of DPSCs. In our study, we focused on biological properties of non-differentiated DPSCs in comparison with osteogenic differentiated cells from DPSCs. We analyzed morphology as well as mineralization potential using three varied osteogenic differentiation media. After fifteen days of differentiation, calcium deposit production was observed in all three osteogenic differentiation m

Treatment-induced neuroendocrine transdifferentiation (NEtD) complicates therapies for metastatic prostate cancer (PCa). Based on evidence that PCa cells can transdifferentiate to other neuroectodermally-derived cell lineages in vitro, we proposed that NEtD requires first an intermediary reprogramming to metastable cancer stem-like cells (CSCs) of a neural class and we demonstrate that several different AR+/PSA+ PCa cell lines were efficiently reprogrammed to, maintained and propagated as CSCs by growth in androgen-free neural/neural crest (N/NC) stem medium. Such reprogrammed cells lost featu

Since the first mouse clone was produced by somatic cell nuclear transfer, the success rate of cloning in mice has been extremely low. Some histone deacetylase inhibitors, such as trichostatin A and scriptaid, have improved the full-term development of mouse clones significantly, but the mechanisms allowing for this are unclear. Here, we found that two other specific inhibitors, suberoylanilide hydroxamic acid and oxamflatin, could also reduce the rate of apoptosis in blastocysts, improve the full-term development of cloned mice, and increase establishment of nuclear transfer-generated embryon

Huntington's disease (HD) is caused by Huntingtin (Htt) gene mutation resulting in the loss of striatal GABAergic neurons and motor functional deficits. We report here an in vivo cell conversion technology to reprogram striatal astrocytes into GABAergic neurons in both R6/2 and YAC128 HD mouse models through AAV-mediated ectopic expression of NeuroD1 and Dlx2 transcription factors. We found that the astrocyte-to-neuron (AtN) conversion rate reached 80% in the striatum and >50% of the converted neurons were DARPP32+ medium spiny neurons. The striatal astrocyte-converted neurons showed action po

Pituitary deficiencies, or hypopituitarisms, are defined as insufficient production of one or more adenohypophyseal hormones (growth hormone, TSH, ACTH, LH-FSH and prolactin). Congenital hypopituitarism, a rare disease with severely disabling consequences, often takes the form of isolated hormone deficiencies, such as isolated growth hormone deficiency (GHD) or isolated ACTH deficiency (ACTHD), or combined pituitary hormone deficiencies (CPHD), when several pituitary hormones are affected. They are mainly the result of genetic mutations, developmental malformations or the harmful action of env

Granulomas are specialized biologic defence mechanisms that form in response to infections by pathogens, foreign bodies or specific stimuli such as antimicrobials or fungi. These structures function to isolate foreign materials and pathogens that cannot be eliminated by immune cells, primarily through macrophage activity. In the skin, granulomas are a hallmark of several conditions, including sarcoidosis, granuloma annulare, tuberculosis and leprosy, each exhibiting distinct pathological and immunological features. Granulomas can also arise from lipid accumulation, as observed in xanthogranulo

Developing reproducible human disease models is a central goal of biomedical research and personalized medicine. Induced pluripotent stem (iPS) cell-derived models enable investigation of underlying cellular and molecular mechanisms in humans. However, obtaining reprogrammable somatic cells is a challenge, especially when patients have limited access to hospital and research facilities. Here we provide a step-by-step protocol for noninvasive isolation of human keratinocytes via trypsinization from scalp hair with intact follicles. The samples can be collected remotely by the donor, a family me

N6-methyladenosine (m6A) is one of the abundant post-transcription modification in cellular RNA. It regulates different biological processes, such as, protein synthesis, X-chromosome inactivation, cell stability, cell-reprogramming and miRNA regulation etc. Most recently, various studies claimed that mutations in m6A sites are linked with various diseases, such as, brain-tumor, heart attack, obesity and cancer. The correct identification of m6A sites is essential to overcome these diseases. However, the state-of-the-art predictors face many challenges for precise detection of m6A sites. Even f

BACKGROUND AND OBJECTIVES: Lipid metabolism reprogramming regulates cellular inflammatory and immune functions in macrophages. The effects of macrophage-specific PPARγ on lipid metabolism and oxidative stress remain unclear. This study aimed to elucidate the impact of the modulation of macrophage PPARγ expression on lipid metabolism, oxidative stress, inflammation, and the progression of nonalcoholic fatty liver disease.
METHODS: RAW264.7 cells, Kupffer cells, and bone marrow-derived macrophages were exposed to saturated fatty acids to establish a NAFLD macrophage model. Techniques, including

The pathogenic mechanisms of osteoporosis (OP), a systemic metabolic bone disease marked by an imbalance between bone growth and resorption, are still not fully understood.Through the specific binding of copper ions to mitochondrial the tricarboxylic acid (TCA) cycle acyl-CoA deoxygenases (like DLAT and PDHA1), cuproptosis-a novel copper-dependent form of programmed cell death-causes protein toxicity stress, protein aggregation, and iron-sulfur cluster (Fe-S cluster) depletion. This finding provides a fresh viewpoint on how to explain the molecular processes underlying oxidative stress. This st

Causative mutations and variants associated with cardiac diseases have been found in genes encoding cardiac ion channels, accessory proteins, cytoskeletal components, junctional proteins, and signaling molecules. In most cases the functional evaluation of the genetic alteration has been carried out by expressing the mutated proteins in in-vitro heterologous systems. While these studies have provided a wealth of functional details that have greatly enhanced the understanding of the pathological mechanisms, it has always been clear that heterologous expression of the mutant protein bears the int

The application of conditional reprogramming culture (CRC) methods to nasal airway epithelial cells would allow more wide-spread incorporation of primary airway epithelial culture models into complex lung disease research. In this study, we adapted the CRC method to nasal airway epithelial cells, investigated the growth advantages afforded by this technique over standard culture methods, and determined the cellular and molecular basis of CRC cell culture effects. We found that the CRC method allowed the production of 7.1 × 10(10) cells after 4 passages, approximately 379 times more cells than

Cancer-initiating/sustaining stem cell subsets (CSCs) have the potential to regenerate cancer cell populations and are resistant to routine therapeutic strategies, thus attracting much attention in anticancer research. In this study, an innovative framework of endogenous microenvironment-renewal for addressing such a dilemma has been just developed. CSCs in three-dimensional multipotent spheroid-engineered biologics were prepared with 150 Gy radiation and inoculated into 15-mo-old BALB/c and C57BL/6 mice bearing diverse advanced tumors covering Mammary 4T1, liver Hepa, lung LL/2, and colon C26

Several studies have reported inducing adult cells into sweat gland-like cells; however, slow transition and low efficiency limit the potential for cell-based treatment. Here, we show that overexpression of the transcription factor FoxC1 was sufficient to reprogram epidermal cells to induced functional sweat gland-like cells (iSGCs). The iSGCs expressing secreting-related genes, had a global gene expression profile between fetal SGCs (P5) and adult SGCs (P28). Moreover, iSGCs transplanted into the burn mice model facilitated wound repair and sweat gland regeneration. We further demonstrated th

Nuclear reprogramming mediated by somatic cell nuclear transfer (SCNT) has many applications in medicine. However, animal clones show increased rates of abortion and reduced neonatal viability. Herein, we used exosomal-miRNA profiles as a non-invasive biomarker to identify pathological pregnancies. MiRNAs play important roles in cellular proliferation and differentiation during early mammalian development. Thus, the aim of this study was to identify exosomal-miRNAs in maternal blood at 21 days of gestation that could be used for diagnosis and prognosis during early clone pregnancies in cattle.

Eggs and oocytes have a remarkable ability to induce transcription of sperm after normal fertilization and in somatic nuclei after somatic cell nuclear transfer. This ability of eggs and oocytes is essential for normal development. Nuclear actin and actin-binding proteins have been shown to contribute to transcription, although their mode of action is elusive. Here, we find that Xenopus Wave1, previously characterized as a protein involved in actin cytoskeleton organization, is present in the oocyte nucleus and is required for efficient transcriptional reprogramming. Moreover, Wave1 knockdown

OBJECTIVE: CD20+CD3+ T cells, a distinct immune cell population implicated in immune responses, contribute to the development of multiple diseases. However, their role in RA progression, particularly their potential to drive the reprogramming of monocyte-derived macrophages toward a pathogenic state akin to synovial tissue macrophages (STMs), remains unclear.
METHODS: Single-cell RNA sequencing and flow cytometry were conducted on RA patients and healthy controls. CD14+ monocytes isolated from the peripheral blood of RA patients were co-cultured with CD20+CD3+ T cells to study their effect on

This review synthesizes the molecular mechanisms of cold tolerance in pepper, integrating multi-omics data, genome editing, and precision breeding strategies to accelerate the development of cold-resilient cultivars. Cold stress is a significant environmental factor that affects the growth, productivity, and fruit quality of Capsicum annuum by impairing membrane integrity photosynthesis and cellular redox homeostasis. Although pepper has several endogenous cold-responsive regulators such as CaNAC035 and CabHLH035, along with antioxidant defense systems, its cold tolerance remains limited due to

Tertiary lymphoid structures (TLSs) are immune cell aggregates that emerge in nonlymphoid tissues during various disease states, including chronic inflammation, autoimmunity, and cancer. TLSs are structurally and functionally analogous to secondary lymphoid organs, and exhibit a maturation continuum (progressing from initial aggregation to mature structures with germinal centers). TLS formation is synergistically regulated by local chemokine networks (e.g. CXCL13, CCL19, and CCL21), lymphotoxin signaling axes, stromal cells, metabolic reprogramming, and the microbiome. This review comprehensiv

BACKGROUND: Despite the relevance of induced pluripotent stem cells (iPSCs) technology for biotechnological applications in cattle, the generation of bona fide bovine iPSCs (biPSCs) remains challenging. This study aimed to evaluate the use of a non-integrating Sendai virus system carrying polycistronic human KLF4-OCT3/4-SOX2, c-MYC, and KLF4 to reprogram bovine fetal mesenchymal stem/stromal cells (MSCs) derived from adipose tissue (AT-MSCs) and bone marrow (BM-MSCs) into biPSCs.
METHODS AND RESULTS: AT-MSCs, BM-MSCs, and control fibroblasts (FBs) were isolated from bovine fetuses (n = 9; 8 m
