Autosomal recessive progressive myoclonus epilepsy due to impaired ceramide synthesis.

Autosomal recessive progressive myoclonus epilepsy due to impaired ceramide synthesis is an extremely rare condition, so far reported in a single family of Algerian origin presenting an unusual, severe form of progressive myoclonus epilepsy characterized by myoclonus, generalized tonic-clonic seizures and moderate to severe cognitive impairment, with probable autosomal recessive inheritance. Disease onset was between 6 and 16 years of age. Genetic study allowed to identify a homozygous nonsynonymous mutation in CERS1, the gene encoding ceramide synthase 1, a transmembrane protein of the endopl

Phosphatidic acid inhibits ceramide 1-phosphate-stimulated macrophage migration.

Ceramide 1-phosphate (C1P) was recently demonstrated to potently induce cell migration. This action could only be observed when C1P was applied exogenously to cells in culture, and was inhibited by pertussis toxin. However, the mechanisms involved in this process are poorly understood. In this work, we found that phosphatidic acid (PA), which is structurally related to C1P, displaced radiolabeled C1P from its membrane-binding site and inhibited C1P-stimulated macrophage migration. This effect was independent of the saturated fatty acid chain length or the presence of a double bond in each of t

Involvement of ceramide in ethanol-induced apoptotic neurodegeneration in the neonatal mouse brain.

Acute administration of ethanol to 7-day-old mice is known to cause robust apoptotic neurodegeneration in the brain. Our previous studies have shown that such ethanol-induced neurodegeneration is accompanied by increases in lipids, including ceramide, triglyceride (TG), cholesterol ester (ChE), and N-acylphosphatidylethanolamine (NAPE) in the brain. In this study, the effects of ethanol on lipid profiles as well as caspase 3 activation were examined in the cortex, hippocampus, cerebellum, and inferior colliculus of the postnatal day 7 mouse brain. We found that the cortex, hippocampus, and inf

Effect of sphingomyelinase-mediated generation of ceramide on aggregation of low-density lipoprotein.

This study addresses the response-to-retention hypothesis, which states that the subendothelial retention of atherogenic lipoproteins is the necessary and sufficient condition for the initiation of atherosclerosis. Here we focus on the relationship between the generation of ceramide in the low-density lipoprotein (LDL) phospholipid monolayer and the resulting aggregation of LDL particles. This study provides the first measurement of neutral, Mg (2+)-dependent Sphingomyelinase (Smase)-mediated ceramide formation from LDL-sphingomyelin and does so for a range of enzyme concentrations (0-0.22 uni

Safety studies of pseudo-ceramide SLE66: acute and short-term toxicity.

Topical application of ceramides is reported to improve the structure and texture of the skin. Synthetic pseudo-cermaide, SLE66 has been shown to reduce dryness/scaling/itching of human skin. Although efficacy of topically applied ceramides and their analogs has been investigated to some extent, safety information is scarce. The objective of the present investigation was to evaluate potential adverse effects of SLE66. The oral LD(50) of SLE66 in rats and mice was >5000 mg/kg, while dermal LD(50) in rats was >2000 mg/kg. In animal and human studies, SLE66 did not cause skin irritation or sensit

Enhancement of radiosensitivity by combined ceramide and dimethylsphingosine treatment in lung cancer cells.

The expression of inducible nitric oxide synthase (iNOS) is a critical factor in both normal physiological functions and the pathogenesis of disease. This study was undertaken to determine the molecular mechanism by which nitric oxide (NO) exerts negative feedback regulation on iNOS gene expression. Isolated rat hepatocytes stimulated with cytokines exhibited a marked increase in NO production as well as iNOS mRNA and protein levels, which were significantly reduced by pretreatment of the NO donors S-nitroso-N-acetyl-D,L-penicillamine (SNAP) and V-PYRRO/NO. This effect of SNAP was inhibited wh

Significant role of ceramide pathway in experimental gastric ulcer formation in rats.

Ceramides have emerged as key participants in the signaling pathway of cytokines and apoptosis. We previously revealed that phorbol 12-myristate 13-acetate (PMA) induced experimental ulcers in rat gastric mucosa. In this study, we investigated the role of ceramide in ulcer formation and its relation to the activation of transcription factors and apoptosis. PMA was subserosally injected to rat glandular stomach. Fumonisin B1 (FB1), an inhibitor of ceramide synthase, was administered together with the PMA. The time course of ceramide content was quantified using thin layer chromatography and the

Ceramide inhibits L-type calcium channel currents in rat pinealocytes.

In rat pinealocytes, ceramide can inhibit the KCl- and BayK 8644-mediated potentiation of cAMP and cGMP accumulation, suggesting that the L-type Ca2+ channel is a target of ceramide action. This was examined in the present study using intracellular Ca2+ measurement and patch-clamp studies. In fura-2-loaded pinealocytes, C2- and C6-ceramide inhibited the Ca2+ increase caused by BayK 8644 and KCl, but not that caused by norepinephrine, suggesting an inhibitory effect of ceramide on the L-type Ca2+ channels. Patch-clamp analysis confirmed that C2- and C6-ceramide, but not C2-dihydroceramide (the

Diet-Induced Ceramide Remodeling as a Mechanistic Link to Cardiac Metabolic Dysfunction.

Background/Objectives: Dietary patterns characterized by excess saturated fat intake contribute to obesity, type 2 diabetes, and cardiac metabolic dysfunction. Ceramides, bioactive sphingolipids synthesized in response to nutrient overload, have emerged as key molecular mediators linking dietary lipid composition to alterations in cardiac metabolic signaling. This review aims to integrate current evidence on diet-induced ceramide remodeling and its impact on intracellular pathways regulating cardiac metabolism. Methods: We analyzed experimental and clinical studies investigating the effects of

A Reproducible Ceramide Phenotype of Sepsis Across Aetiologies - A Monocenter Cohort Study.

PURPOSE: Sepsis due to SARS-CoV-2 shares features with non-COVID-19 sepsis. Ceramides-bioactive sphingolipids-exist as long-chain (LC) and very-long-chain (VLC) species with opposing signaling effects. Whether LC and VLC ceramides are altered similarly in COVID-19 vs non-COVID-19 sepsis, and the therapeutic relevance of sphingomyelinase inhibition, remains uncertain. We characterised circulating ceramides in patients with systemic inflammatory response syndrome (SIRS) or sepsis with and without SARS-CoV-2. PATIENTS AND METHODS: We performed targeted lipidomics on serum/plasma in two cohorts:

Glial ceramide orchestrates lipid homeostasis and age-dependent motor function.

Glial cells have emerged as equal partners to neurons in the development and maintenance of the nervous system. In this study, we examine the consequences of modulating glial ceramide metabolism on adult motor function. Glial reduction of ER-localised ceramide synthesis, as exemplified by knockdown (KD) of schlank or ifc or lace, leads to age-dependent motor defects, as does KD of ceramide transfer protein (CERT). Motor defects in CERT-null animals can be significantly rescued by glial CERT expression, further highlighting the importance of glial ceramide homeostasis. Expectedly, glial schlank

Targeted inhibition of hepatic de novo ceramide synthesis ameliorates MASH.

Increasing evidence implicates ceramides in the pathogenesis of metabolic dysfunction-associated steatohepatitis (MASH). However, the therapeutic potential of liver-targeted ceramide lowering remains unclear. In this study, we demonstrate that elevated ceramide levels in MASH patients and mouse models are closely associated with the activation of hepatic de novo ceramide synthesis. The analysis of human hepatic single-nucleus RNA sequencing (snRNA-seq) data revealed predominant up-regulation of SPTLC2, which encodes a subunit of the rate-limiting enzyme in the de novo ceramide synthesis pathwa

Therapeutic potential of targeting ceramide for early cardiometabolic lipotoxicity in vivo study.

Ceramides (Cer) drive cardiometabolic disease (CMD) by promoting lipotoxicity and oxidative stress, yet their therapeutic potential remains underexplored. We investigated Cer dysregulation in high-fat diet (HFD) fed rats and evaluated three antidiabetic drugs (Sitagliptin, Liraglutide, Saxagliptin) for Cer-modulating effects. HFD elevated CerC16:0 and CerC18:0 across different tissues (serum, heart and liver), with pronounced cardiac CerC18:0 accumulation (1.37 ± 0.103 nmol/g, p < 0.001). Cer changes preceded alterations in traditional metabolic markers (glucose, cholesterol and triglycerides)

Plasma ceramide mediates the association of peripheral T cells with Alzheimer's disease.

INTRODUCTION: Neuroinflammation involving peripheral immune cells, particularly T lymphocytes, has been implicated in Alzheimer's disease (AD) progression. Metabolic dysregulation in sphingolipid pathways may influence neuroinflammatory processes in AD pathogenesis. However, the specific roles of T cell subsets and their interactions with metabolic pathways in AD pathogenesis remain unclear. OBJECTIVES: To determine the causal contributions of peripheral T cell subsets to AD risk and investigate whether blood metabolites mediate this association. METHODS: We integrated single-cell RNA sequen

Ceramide synthase 4 interferes with replication of influenza virus but is downregulated by infection.

Influenza continues to pose a serious threat to humans. Influenza-host interaction is incompletely understood, requiring identification of host factors that regulate viral pathogenicity. Ceramide synthases (CerSs) are responsible for producing and controlling ceramide levels within cells. Ceramides are structural and signaling sphingolipid components that mediate various biological functions and affect the infectivity of multiple viruses. However, the role of CerSs during virus infections remains unclear. In this study, we investigated the possible function of CerSs in host defense against inf

Ceramide enhanced the hepatic glucagon response through regulation of CREB activity.

BACKGROUND & AIMS: Hyperglycemia is associated with lipid disorders in patients with diabetes. Ceramides are metabolites involved in sphingolipid metabolism that accumulate during lipid disorders and exert deleterious effects on glucose and lipid metabolism. However, the effects of ceramide on glucagon-mediated hepatic gluconeogenesis remain largely unknown. This study was designed to investigate the impact of ceramides on gluconeogenesis in the context of the hepatic glucagon response, with the aim of finding new pharmacological interventions for hyperglycemia in diabetes. METHODS: Liquid ch
