Astrocytic Gq pathway activation enhances the efficacy of rehabilitation after stroke.

Stroke is a leading cause of functional impairment. As rehabilitation has a limited effect, it is essential to advance future treatments and enhance the efficacy of rehabilitation. This study focused on the use of astrocytes to augment the efficacy of rehabilitation for strokes. It utilized chemogenetics to specifically activate astrocytes in the peri-infarct region of a mouse photothrombosis model. The mice were subjected to immunohistochemical analysis and behavioral tests to evaluate the effects of astrocytic Gq pathway activation after an induced stroke. The timing of astrocytic activation

Oxytocin and Nociceptive Processing: Mechanisms and Therapeutic Potential.

BACKGROUND: Oxytocin (OXT), a neuropeptide synthesized in the hypothalamic paraventricular and supraoptic nuclei, is traditionally known for its roles in parturition and lactation. However, accumulating evidence indicates that OXT also functions as an endogenous analgesic modulator acting through central and peripheral pathways. SUMMARY: In this review, we summarize recent advances in the understanding of the role of OXT for pain modulation, focusing on descending hypothalamo-spinal projections, peripheral neuroendocrine mechanisms, and novel experimental approaches using transgenic and chemo

Glutamatergic Projection from the Ventrolateral Periaqueductal Gray to the Parafascicular Nucleus of the Thalamus Contributes to the Modulation of Neuropathic Pain in Mice.

Chronic pain is frequently accompanied by increased pain sensitivity and affective disturbances. The ventrolateral periaqueductal gray (vlPAG) and the parafascicular nucleus of the thalamus (PF) are key nodes in pain modulation. However, the functional role of the vlPAG-PF neural pathway remains poorly understood. In this study, we employed an integrative approach combining neuroanatomical tracing and chemogenetic techniques to investigate the role of the vlPAGvGluT2-PF neural pathway in pain modulation. Our results demonstrate that vGluT2-positive neurons in the vlPAG project to the PF. Chemo

Medial Prefrontal Cortex Modulation of the Peripheral Immune Response in Sepsis Via the Autonomic Nervous System.

BACKGROUND: Sepsis is a life-threatening organ dysfunction syndrome caused by a dysregulated host response to infection, in which neuroimmune dysfunction plays a central role. The medial prefrontal cortex (mPFC) is involved in autonomic and immune regulation. However, its role in modulating peripheral immune responses during sepsis remains unclear. This study investigated whether glutamatergic neurons in the mPFC regulate systemic inflammation and organ injury during sepsis via autonomic pathways and adrenergic signaling. METHODS: Glutamatergic neurons in the mPFC were selectively manipulated

Adolescent alcohol exposure disrupts astrocyte-synaptic structural and functional coupling in the male dorsal hippocampus.

Adolescence is a window of heightened vulnerability to the neurotoxic effects of binge ethanol exposure. Adolescent intermittent ethanol (AIE) exposure has been shown to induce long-lasting cognitive and behavioral impairments in patients and rodent models that increase the risk of developing alcohol use disorder (AUD). Our previous work shows that these behavioral deficits coincide with persistent astrocyte dysfunction. Here, we aim to understand how astrocyte-synaptic structural and functional crosstalk are disrupted following AIE to provide better mechanistic understanding of why behavioral

A Common Neuronal Ensemble in the Lateral Habenula Regulates Ciprofol Anesthesia in Mice.

General anesthetics were first used over 170 years ago; however, the mechanisms of how general anesthetics induce loss of consciousness (LOC) remain unclear. Ciprofol, a novel intravenous anesthetic, has been developed by incorporating cyclopropyl into the chemical structure of propofol. This modification offers the benefits of rapid onset and minimal injection pain. Recent studies have revealed that the glutamatergic neurons of the lateral habenula (LHb) play a crucial role in modulating the LOC induced by propofol and sevoflurane. Nevertheless, the specific involvement of LHb in the anesthet

Principal neurons in the olfactory cortex mediate bidirectional modulation of seizures.

Although the piriform cortex (PC) has been previously implicated as a critical node for seizure generation and propagation, the underlying neural mechanism has remained unclear. Here, we found increased excitability in PC neurons during amygdala kindling acquisition. Optogenetic or chemogenetic activation of PC pyramidal neurons promoted kindling progression, whereas inhibition of these neurons retarded seizure activities induced by electrical kindling in the amygdala. Furthermore, chemogenetic inhibition of PC pyramidal neurons alleviated the severity of kainic acid-induced acute seizures. Th

Protocol for targeting the magnocellular neuroendocrine cell ensemble via retrograde tracing from the posterior pituitary.

The hypothalamic magnocellular neuroendocrine cells (MNCs) project to the posterior pituitary (PPi), regulating reproduction and fluid homeostasis. It has been challenging to selectively label and manipulate MNCs, as they are intermingled with parvocellular neuroendocrine cells projecting to the median eminence. Here, we provide a step-by-step protocol for specifically targeting the MNCs by infusing retrograde viral tracers into the PPi. When combined with optogenetics, chemogenetics, and transgenic animals, this approach allows cell-type-specific manipulation of MNCs in multiple sites for fun

GABA Cells in the Central Nucleus of the Amygdala Promote Cataplexy.

Cataplexy is a hallmark of narcolepsy characterized by the sudden uncontrollable onset of muscle weakness or paralysis during wakefulness. It can occur spontaneously, but is typically triggered by positive emotions such as laughter. Although cataplexy was identified >130 years ago, its neural mechanism remains unclear. Here, we show that a newly identified GABA circuit within the central nucleus of the amygdala (CeA) promotes cataplexy. We used behavioral, electrophysiological, immunohistochemical, and chemogenetic strategies to target and manipulate CeA activity selectively in narcoleptic (or

Acute Stress Attenuates Hepatic Ischemia-Reperfusion Injury via Hypothalamic CRH Neuron-Induced HPA Axis Activation.

BACKGROUND: Hepatic ischemia-reperfusion injury (HIRI) is a pathologic process commonly encountered during liver surgery, which seriously threatens patient prognosis. Currently, effective interventions or preventive measures are still lacking. Notably, patients with liver disease commonly experience brief acute stress prior to surgery; however, the impact of acute stress on HIRI remains unclear. METHODS: A 30-min restraint stress was used to simulate acute restraint stress (ARS). Hematoxylin-eosin staining and ELISA were employed to assess HIRI. Immunofluorescence staining and electrophysiolo

Activation of a locus coeruleus to ventral tegmental area noradrenergic pathway blunts binge-like ethanol intake and promotes aversive unconditioned responses in TH-ires-Cre mice.

Ethanol comprises innately aversive properties that may act as a deterrent to overconsumption. We have previously found that chemogenetic activation or inhibition of a noradrenergic locus coeruleus (LC) to rostromedial tegmental nucleus (RMTg) pathway bidirectionally modulates binge-like ethanol intake and aversive reactivity in male and female TH-ires-Cre mice. We previously hypothesized that noradrenergic RMTg circuitry may modulate ethanol intake and aversion through inhibitory inputs to the ventral tegmental area (VTA), a region that is both densely innervated by the RMTg and proposed to c

Effects of inhibiting the orexin system on nicotine reward memory and fear memory.

The orexin system is associated with various brain functions, including reward processing and the stress response. Previous studies have suggested that orexin receptors (OXRs) antagonists could be beneficial in treating several mental disorders, such as drug addiction and post-traumatic stress disorder. However, the specific role of the orexin system in both appetitive (reward-related) and aversive (fear-related) memories remains unclear. In this study, we used nicotine-induced conditioned place preference (CPP) and fear conditioning models to evaluate the effects of suvorexant, a dual OXRs an

Electroacupuncture Regulates Sympathetic Nerve Through the NTSGlu-RVLM Circuit to Relieve Spontaneous Pain in SNI Rats.

AIM: Patients suffering from neuropathic pain often experience sympathetic dysfunction. Acupuncture has shown promise in alleviating pain and modulating the activity of the autonomic nervous system. This study aims to explore the potential mechanism through which electroacupuncture (EA) modulates sympathetic nerves to alleviate neuropathic pain. METHODS: Spared nerve injury (SNI) was utilized to induce neuropathic pain. EA was administered at acupoints Huantiao and Yanglingquan for 30 min every other day after SNI. Pain behavior was evaluated using paw withdrawal thresholds (PWTs) and spontan

Methylone regulates fear memory and amygdala activity: A potential treatment for posttraumatic stress disorder?

Methylone (3,4-methylenedioxy-N-methylcathinone) is a rapid-acting entactogen that has demonstrated significant benefits for patients with post-traumatic stress disorder (PTSD) and exhibits good tolerability in phase 1 clinical trials. Despite these promising results, its preclinical effects on fear memory regulation and the underlying mechanisms remain largely unexplored. This study aims to investigate the impact of methylone on auditory fear extinction and its influence on neuronal and synaptic activity in the basolateral amygdala (BLA). Using C57BL/6 mice, we employed an auditory fear condi

GluCl.Cre ON enables selective inhibition of molecularly defined pain circuits.

Insight into nociceptive circuits will ultimately build our understanding of pain processing and aid the development of analgesic strategies. Neural circuit analysis has been advanced greatly by the development of optogenetic and chemogenetic tools, which have allowed function to be ascribed to discrete neuronal populations. Neurons of the dorsal root ganglion, which include nociceptors, have proved challenging targets for chemogenetic manipulation given specific confounds with commonly used DREADD technology. We have developed a cre/lox dependant version of the engineered glutamate-gated chlo

Lateral hypothalamic galanin neurons are activated by stress and blunt anxiety-like behavior in mice.

Despite the prevalence of anxiety disorders, the molecular identity of neural circuits underlying anxiety remains unclear. The lateral hypothalamus (LH) is one brain region implicated in the regulation of anxiety, and our recent data found that chemogenetic activation of LH galanin neurons attenuated the stress response to a novel environment as measured by the marble burying test. Thus, we hypothesize that LH galanin neurons may contribute to anxiety-related behavior. We used chemogenetics and fiber photometry to test the ability of LH galanin neurons to influence anxiety and stress-related b

A Subset of Nucleus Accumbens Neurons Receiving Dense and Functional Prelimbic Cortical Input Are Required for Cocaine Seeking.

BACKGROUND: Prelimbic cortical projections to the nucleus accumbens core are critical for cue-induced cocaine seeking, but the identity of the accumbens neuron(s) targeted by this projection, and the transient neuroadaptations contributing to relapse within these cells, remain unknown. METHODS: Male Sprague-Dawley rats underwent cocaine or sucrose self-administration, extinction, and cue-induced reinstatement. Pro-opiomelanocortin neurons in the nucleus of the solitary tract mediate endorphinergic endogenous analgesia in mice.

The nucleus of the solitary tract (NTS) contains pro-opiomelanocortin (POMC) neurons that are 1 of the 2 major sources of β-endorphin in the brain. The functional role of these NTS POMC neurons in nociceptive and cardiorespiratory function is debated. We have shown that NTS POMC optogenetic activation produces bradycardia and transient apnoea in a working heart-brainstem preparation and chemogenetic activation with an engineered ion channel (PSAM) produced opioidergic analgesia in vivo. To better define the role of the NTS POMC neurons in behaving animals, we adopted in vivo optogenetics (Chri

Pancreatic β-Cells Communicate With Vagal Sensory Neurons.

BACKGROUND AND AIMS: Destroying visceral sensory nerves impacts pancreatic islet function, glucose metabolism, and diabetes onset, but how islet endocrine cells interact with sensory neurons has not been studied. METHODS: We characterized the anatomical pattern of pancreatic sensory innervation by combining viral tracing, immunohistochemistry, and reporter mouse models. To assess the functional interactions of β-cells with vagal sensory neurons, we recorded Ca2+ responses in individual nodose neurons in vivo while selectively stimulating β-cells with chemogenetic and pharmacologic approaches.
