In the introduction to this paper an outline is given of dysostotic malformations of the jaw. Following this, the author reports observations made on sixty-five patients. Clinical, roentgenological, electromyographic, histological, and karyological, examinations made on this patient stock yielded numerous, hitherto undescribed results which, among other things, allow the previous classification of syndromes of the bronchial arch to be extended to include additional conditions.

Hamartomas are tumor-like abnormalities typified by the presence of cellular proliferation indigenous to the native site. However, hamartomas maintain growth cessation without the potential for further growth or malignant transformation. Hamartomas are commonly seen in the lungs, kidney, liver, and spleen and rarely occur in the orofacial region. Various hamartomatous oral lesions include hemangiomas, lymphangiomas, tori, exostosis, dens invaginatus, dens evaginatus, odontomas, nevi, and cherubism. Infantile hemangiomas are benign vascular tumors that emerge soon after birth and experience rap

BACKGROUND: Premature loss of primary teeth can occur as a consequence of dental trauma, neonatal tooth extraction, early childhood caries, or periodontal problems, or it can be a manifestation of systemic disease. This review aims to present systemic disorders that can lead to premature loss of deciduous teeth in children and to provide a comprehensive resource for clinical practice for both physicians and dentists.
METHODS: This study is a narrative review of original studies and case reports published in English and Polish between 1957 and 2021 that was conducted by searching electronic sc

PURPOSE OF REVIEW: This article addresses the prevalence and relationship between autoinflammatory diseases and vasculitis.
RECENT FINDINGS: Autoimmune diseases (AIDs) are a group of syndromes characterized by episodes of unprovoked inflammation due to dysregulation of the innate immune system. Despite the common occurrence of rashes and other skin lesions in these diseases, vasculitis is reported in only a few. On the other hand, neutrophilic dermatoses are more prevalent. Large vessel vasculitis is reported in patients with Behcet's and Blau's syndromes. Small and medium size vasculitides a

This chapter explores the intricate interplay between immune regulation and craniofacial development, emphasizing the critical role of the immune system in both normal and pathological conditions. The process of craniofacial development, from the formation of pharyngeal arches and facial prominences to palate formation, is outlined, highlighting the susceptibility of these structures to maternal immune disruptions.Key immune interactions at the maternal-fetal interface are discussed, focusing on immune tolerance mechanisms that safeguard the fetus from maternal immune rejection. Dysregulation

BACKGROUND: Systemic autoinflammatory diseases (SAIDs) are a group of disorders related to defective regulation of the innate immune system. Recurrence of inflammation can severely affect the patients' outcomes with a direct or indirect impact on their physical and mental health and/or global quality of life (QoL). We therefore sought to identify currently available QoL studies for these diseases as well as measurement tools at our disposal.
BASIC PROCEDURES: A systematic literature review was carried out with a focus on monogenic SAIDs. We inventoried the study designs developed in the selec

Fibrous-cystic osteitis is a bone metabolic disorder related to hyperparathyroidism. This pathological condition shows a bone catabolism enhancement, due to increased level of PTH. Brown tumour is a uni- or multi-focal bone lesion, which represents the terminal stage of the hyperparathyroidism-dependent bone pathology. This focal lesion is not a real neoplasm showing itself as a cellular reparative process, mainly interesting the jaws, specially the mandible. Because of the similar radiological features (cyst-like radiolucency) showed by other lesions, the diagnosis can be difficult. Histology

"Giant cell reparative granuloma" was introduced into medical literature by Jaffe in 1953. Prior to that time most authors considered this lesion to be a variant of the benign giant cell tumor of the long bones, or a giant cell variant of osteitis fibrosa. Bernier and Cahn established the subdivision between the rare central giant cell reparative granuloma and the common peripheral epulis. In the past, considerable emphasis has been placed on the importance of differentiating the true giant cell tumor from the giant cell reparative granuloma of the jaw bones. Most authors now believe the true

Autoinflammatory bone diseases (AIBDs) constitute a recently identified subset of autoinflammatory diseases. These conditions are characterized by an exaggerated inflammatory response in the bones without any apparent etiology. Inflammatory bone lesions associated with AIBDs exhibit chronic inflammation, are typically culture-negative, and do not exhibit discernible microorganisms on histopathological examination. The most common and representative AIBD is chronic non-bacterial osteomyelitis (CNO), which is also known as chronic recurrent multifocal osteomyelitis. Another variant of CNO, which

Impaired growth is common in patients with glycogen storage disease (GSD), who also may have "cherubic" facies similar to the "moon" facies of Cushing syndrome (CS). An infant presented with moon facies, growth failure, and obesity. Laboratory evaluation of the hypothalamic-pituitary-adrenal (HPA) axis was consistent with CS. He was subsequently found to have liver disease, hypoglycemia, and a pathogenic variant in PHKA2, leading to the diagnosis of GSD type IXa. The cushingoid appearance, poor linear growth and hypercortisolemia improved after treatment to prevent recurrent hypoglycemia. We s

Genetic disorders of bone constitute a large number of alterations approaching almost 300 types. This article focuses on the most common disorders in this group, which at the same time are the most clinically significant because of their incidence and degree of involvement of the craniofacial skeleton.

Within the framework of a broad definition of the extracellular matrix (ECM), this review discusses three genetic disorders in which major pathogenetic features have been traced back to alterations in the levels/activities of matrix components. In each case, disease-associated alterations are found both intra- and extracellularly. The nature of the ECM involvement is surprising, offers an exciting therapeutic opportunity, and deepens our understanding of ECM-cell interactions. The first of these disorders, cherubism, is a case of inflammatory bone loss in the jaws of children for reasons that

BACKGROUND/AIM: In patients afflicted with the autosomal dominant hereditary tumor predisposition syndrome neurofibromatosis type 1 (NF1), central giant cell granuloma of the jaw (CGCG) is observed in rare cases. In NF1, the lesion is caused by mutations in the NF1 gene. The report summarizes the diagnosis, treatment and follow-up of 2 NF1 patients and provides a brief evaluation of case reports on NF1-associated CGCG.
CASE REPORT: Both females with CGCG (age: 14 and 22 years) had either one or more jaw lesions. The lesions were removed with curettage to avoid dental injuries. In both cases,

The authors presents two cases of basocellular nevomatosis with multiple maxillary cysts. He thought it important to point out the presence of a cyst in the left styloid apophysis, as up to now, epidermoid cysts have never been observed in other facial bones apart from the maxillae in BCN. In conclusion, the author presents three casses, which although showing multiple cysts or pseudocysts of the maxillae, do not belong to the BCN syndrome.

Diseases of the jaw bones in children can be monostotic or polyostotic. Examples are fibrous dysplasia and Langerhans cell granulomatosis. Sarcomas of the jaw bones are rare, but do occur with some preference in children. The dentist should be aware of the clinical and even more so of the rather typical radiologic changes in such diseases.

Biallelic loss-of-function variants in TBC1D2B have been reported in five subjects with cognitive impairment and seizures with or without gingival overgrowth. TBC1D2B belongs to the family of Tre2-Bub2-Cdc16 (TBC)-domain containing RAB-specific GTPase activating proteins (TBC/RABGAPs). Here, we report five new subjects with biallelic TBC1D2B variants, including two siblings, and delineate the molecular and clinical features in the ten subjects known to date. One of the newly reported subjects was compound heterozygous for the TBC1D2B variants c.2584C>T; p.(Arg862Cys) and c.2758C>T; p.(Arg920*)
