[Excitatory effects of glucocorticoids on neuronal activity in the medial vestibular nucleus--mediation by glucocorticoid receptor on the membrane].

Although glucocorticoids are sometimes used for the treatment of vertigo in certain disorders such as Meniere's disease, the mechanism underlying anti-vertigo effect remains unknown. The present study was performed to examine the effects of a glucocorticoid, dexamethasone, on neuronal activity in the medial vestibular nucleus (MVN) to determine whether or not the drug acts directly on the MVN neuron using alpha-chloralose-anesthetized cats which were fixed in a stereotaxic instrument placed on a turn-table. Single neuronal activities in the MVN were extracellularly recorded with a glass-insula

Effect of cortical spreading depression on activity of trigeminovascular sensory neurons.

The effect of cortical spreading depression, a proposed initiating event for migraine pain, on cortical blood flow (laser Doppler method) and on the spontaneous firing rate and stimulus-evoked responses of trigemino-cervical neurons with craniovascular input was studied in 17 neurons in 8 cats anesthetized with chloralose. Cortical spreading depression, induced via cortical pinprick injury, produced an initial wave of cortical hyperemia (243+/-57% of control) and a later and smaller phase of oligemia (96+/-4% of control). Neither the basal discharge rate (6.7+/-1.7 sec(-1)) nor the evoked resp

Response of the goat carotid body to acute and prolonged hypercapnia.

The effect of prolonged hypercapnia on carotid chemoreceptor discharge frequency has not been elucidated. In addition, the effect of acute hypercapnia on chemoreceptor discharge has not been determined in the goat, a species commonly used for ventilatory control studies. Therefore, we determined the effects of acute and prolonged normoxic-hypercapnia on single fiber output of the carotid body of chloralose anesthetized goats. The animals were paralyzed and artificially ventilated. The average acute response curve for 12 single fibers was linear over the range of 30-80 Torr PaCO2 with a mean sl

The action of intravenous anaesthetic and central depressant drugs on the contractures elicited by tetraethylammonium in the chick biventer cervicis muscle.

A group of intravenous anaesthetic drugs was compared with methohexitone sodium for their ability to potentiate tetraethylammonium induced contractures of the chick biventer cervicis muscle (TEA test). The equipotent concentrations in the TEA test were: methohexitone 8.8 X 10(-5) M, propanidid 1.78 X 10(-4) M, althesin 3.58 X 10(-5) M (in terms of alphaxalone), etomidate 1.6 X 10(-4) M, thiopentone 2.13 X 10(-5) M and valium 4.95 X 10(-5) M. There was no relation between activity in the TEA test and excitatory muscular activity reported clinically. The central depressant drugs ethyl alcohol an

Drug interactions and vasoconstrictors used in local anesthetic solutions.

This study examined widely advertised interactions between sympathomimetic amine vasoconstrictors currently used in dental local anesthetic solutions and MAO inhibitors (phenelzine, 5 mg/kg), phenothiazines (chlorpromazine, 2 mg/kg), and tricyclic antidepressants (desipramine, 2 mg/kg). Twelve greyhound dogs premedicated with morphine and anesthetized with urethane and alpha-chloralose were prepared for physiologic recordings. During a control period, the dogs received bolus injections of epinephrine, norepinephrine, and levonordefrin sufficient to construct log-linear dose-response curves for

Mechanism of cardiac output response to hypertonic sodium chloride infusion in dogs.

Hypertonic sodium chloride and tris(hydroxymethyl)amino-methane (Tris) hydrochloride (0.3 mmol/kg.min) were infused intravenously into chloralose-anesthetized dogs over a 20-min period. Cardiac output, maximum left ventricular dP/dt, and (dP/dt)/P increased by 55, 33, and 23%, respectively, during NaCl infusion, but Tris HCl infusion had no effects. NaCl infusion did not change heart rate or left ventricular end-diastolic pressure. Mean systemic and pulmonry arterial blood pressures increased, whereas total peripheral and pulmonary vascular resistances decreased. Responsiveness of cardiac beta

The effect of sympathetic nerve stimulation on serum gastrin, gastric acid secretion and mucosal blood flow responses to meat extract stimulation in anaesthetized cats.

Chloralose anaesthetized cats were prepared with fundic and antral pouches. Fundic mucosal blood flow was measured by the amidopyrine technique and serum gastrin was measured by radioimmunoassay. 2. Meat extract suspension in the pyloric antrum produced a highly significant sixfold increase in arterial serum gastrin concentration (P less than 0-001). 3. The mean ratio of the fundic mucosal blood flow to acid secretory responses (deltaMBF/deltaH+ ratio) of 0-142 +/- 0-026 (25) ml./muequiv H+, is very similar to the values previously published for exogenous gastrin stimulation. 4. Effects of sugammadex on heart rate variability in anesthetized male rats.

The administration of sugammadex has been associated with bradycardia and rarely asystole; however, the underlying mechanisms remain unclear. We examined heart rate variability (HRV) as an index of autonomic nervous system activity and investigated the effects of sugammadex on HRV in α-chloralose and urethane anesthetized male rats. In protocol 1, we examined the effect of intravenous administration of sugammadex (10 mg/kg) alone. In protocol 2, sugammadex was administered 3 min after the administration of rocuronium bromide. In protocol 3, the effect of sugammadex was assessed after vagotomy.

Functional MRI of the rat somatosensory cortex: effects of hyperventilation.

Functional mapping of the rat somatosensory cortex was performed with T2*-sensitized MRI using a forepaw electrical stimulation model in alpha-chloralose-anesthetized rats at 7 T under both normocapnia and mild hyperventilation-induced hypocapnia. A highly localized activation area, consistent with the known somatosensory cortical region, was detected in all seven animals studied during hypocapnia and in five of the same animals during normocapnia. Quantitatively, hypocapnia was found to significantly increase both the size of the fMRI activation area (3.4 +/- 0.6 mm2 versus 1.5 +/- 0.6 mm2 in

[Role of the lateroventral periolivary nucleus in the central chemoreception of respiration in rats].

Using the methods of microinjection, microiontophoresis, electrolytic destruction and electric stimulation, the present work was undertaken to investigate whether the lateroventral periolivary nucleus (LVPO) might play a role in the central mechanism of chemoreception on respiration. Experiments were performed on 38 chloralose-urethane anesthetized Sprague-Dawley rats. The results were as follows: (1) Bilateral microinjection of acidified mock CSF into the LVPO region, evoked an enhancement in respiratory activities. (2) Microiontophoretic application of H+ caused mainly excitatory effects on

Reflex carotid body chemoreceptor control of phrenic sympathetic neurons.

The reflex reaction of phrenic sympathetic neurons to stimulation of carotid body chemoreceptors was tested in chloralose-anesthetized and paralyzed cats with both vago-aortic nerves cut. During systemic hypoxia (animals ventilated with 10% O2 in N2) the sympathetic phrenic nerve activity increased from 100% in the control to 269%. This increase was markedly attenuated after cutting both sinus nerves. Reflex excitatory response in phrenic sympathetic neurons with the latency of 150 msec was evoked by electrical stimulation of the right carotid sinus nerve (3 pulses of 0.2 msec, 333 Hz). The ce

Vago-vagal activation of naloxone-sensitive non-adrenergic, non-cholinergic jejunal contractions in the anaesthetized cat.

Experiments were performed on chloralose-anaesthetized cats, pretreated with guanethidine and with ligated adrenals and sectioned splanchnic nerves. The animals were paralysed and artificially ventilated. Jejunal motility was recorded by an intraluminal balloon, as volume changes at a constant pressure. Cervical or abdominal vagal afferent stimulation was performed before and after the administration of atropine. In both circumstances jejunal contractions were elicited. The latency of onset and the duration of the responses were significantly shorter before than after atropine. Strength-durati

Mediation of the fastigial pressor response and a somatosympathetic reflex by ventral medullary neurones in the cat.

In chloralose-anaesthetized, artificially ventilated cats, the effects have been studied of inactivating neurones of the 'glycine-sensitive area' of the rostral ventrolateral medulla on evoked renal sympathetic and vasomotor responses. Glycine applied topically and bilaterally to the 'glycine-sensitive area' besides markedly lowering blood pressure and heart rate, reversibly abolished or severely attenuated both the pressor response and the renal nerve volley produced by electrical stimulation of the fastigial nucleus as well as the tibial to renal nerve somatosympathetic reflex. Responses of T2-T4 spinal neurons to stimulation of the greater splanchnic nerves of the cat.

Effects of electrical stimulation of the greater splanchnic nerves on T2-T4 spinal neurons were determined in 16 cats anesthetized with alpha-chloralose. Of 77 neurons responding to somatic stimuli, 65 (84%) were excited, inhibited, or both excited and inhibited by splanchnic input. Each of the splanchnic responsive cells also was responsive to electrical stimulation of cardiopulmonary sympathetic afferent fibers. All but one neuron with left splanchnic input also received input from the right splanchnic nerve. Short- and long-latency excitatory responses were observed after splanchnic stimula
