The ABCA2 transporter: intracellular roles in trafficking and metabolism of LDL-derived cholesterol and sterol-related compounds.

ATP-binding cassette (ABC) transporters comprise a family of critical membrane bound proteins functioning in the translocation of molecules across cellular membranes. Substrates for transport include lipids, cholesterol and pharmacological agents. Mutations in ABC transporter genes cause a variety of human pathologies and elicit drug resistance phenotypes in cancer cells. ABCA2, the second member the A subfamily to be identified, was highly expressed in ovarian carcinoma cells resistant to the anti-cancer agent, estramustine, and more recently, in human vestibular schwannomas. Cells expressing

Combined effects of cholesterol reduction and apolipoprotein A-I expression on atherosclerosis in LDL receptor deficient mice.

Reduction of total and LDL cholesterol reduces atherosclerosis and clinical cardiovascular events. High density lipoprotein (HDL) cholesterol levels have a strong inverse association with atherosclerosis, and overexpression of apolipoprotein A-I (apoA-I), the major protein component of HDL, reduces atherosclerosis in hypercholesterolemic animals. However, little is known about the potential for additive or synergistic effects between cholesterol reduction and apoA-I overexpression on atherosclerosis. In the current study, we tested the hypothesis that significant reduction of plasma cholestero

Cholesterol efflux in vivo from a depot of cationized LDL injected into a thigh muscle of small rodents.

We have developed a model system to measure quantitatively removal of cholesterol from a well-defined depot in vivo. To that end, lipoproteins were injected into the rectus femoris muscle of small rodents, using a 25 microliters Hamilton syringe and a 27-gauge needle. In most experiments, the injected volume was 10 microliters containing 200 micrograms of cholesterol. The lipoproteins tested were native or modified LDL labeled with trace amounts of [3H]free cholesterol ([3H]FC). The amount of label or of cholesterol mass recovered at various time intervals after injection was normalized to tha

Diagnostic and therapeutic inertia for LDL cholesterol management of consecutively verified patients with acute myocardial infarction.

BACKGROUND: Actionable quality gaps persist in low-density lipoprotein cholesterol (LDL-C) management after acute myocardial infarction (AMI). We aimed to quantify diagnostic and therapeutic inertia in LDL-C management after discharge in a real-world AMI population with verified and quantified consecutive inclusion. METHODS: We conducted a quality improvement audit of patients discharged alive with a primary diagnosis of AMI. Consecutiveness was quantified using the consecutive index (e.g., the ratio of included patients with available 6-month follow-up information to the total AMI discharges

LDL-cholesterol reduction is not associated with deterioration of cellular integrity after myocardial infarction - a longitudinal analysis.

BACKGROUND: Intensive low-density lipoprotein cholesterol (LDL-C) lowering is a cornerstone of secondary prevention after myocardial infarction, but theoretical concerns persist regarding potential adverse effects of very low LDL-C on cellular membrane integrity. Phase angle at 50 kHz (PhA50), derived from bioelectrical impedance analysis (BIA), is a non-invasive marker reflecting cell membrane bioelectrical properties. This study assessed whether LDL-C reduction after myocardial infarction is associated with deterioration in PhA50. METHODS: This longitudinal analysis included 45 patients und

Sex differences in monitoring and achievement of blood pressure and LDL-cholesterol targets in chronic coronary syndrome.

BACKGROUND: The cornerstones of secondary prevention in chronic coronary syndrome (CCS) are blood pressure lowering and lipid-lowering therapy (LLT). The purpose of this study was to examine the control of systolic blood pressure (SBP) and low-density lipoprotein cholesterol (LDL-C) in patients with CCS followed in primary care. METHODS: This was a retrospective population-based cohort study of patients diagnosed with CCS at cardiology clinics in Uppsala Region, Sweden, between 2012 and 2020. Data from electronic health records were analyzed for SBP and LDL-C monitoring and secondary preventi

Analysis of 26 Studies of the Impact of Coconut Oil on Lipid Parameters: Beyond Total and LDL Cholesterol.

Coconut oil (CNO) is often characterized as an "artery-clogging fat" because it is a predominantly saturated fat that ostensibly raises total cholesterol (TChol) and LDL cholesterol (LDL-C). Whereas previous analyses assessed CNO based on the relative effects on lipid parameters against other fats and oils, this analysis focuses on the effects of CNO itself. Here, we review the literature on CNO and analyze 984 lipid profile data sets from 26 CNO studies conducted over the past 40 years. This analysis shows considerable heterogeneity among CNO studies regarding participant selection, the amoun

Intensive Lowering of LDL Cholesterol Levels With Evolocumab in Autoimmune or Inflammatory Diseases: An Analysis of the FOURIER Trial.

BACKGROUND: Patients with an autoimmune or inflammatory disease (AIID) are at increased cardiovascular risk and may benefit more from statin therapy. In the FOURIER trial (Further Cardiovascular Outcomes Research with PCSK9 Inhibition in Subjects with Elevated Risk), the PCSK9 (proprotein convertase subtilisin/kexin type 9) inhibitor evolocumab lowered low-density lipoprotein cholesterol levels, but not hsCRP (high-sensitivity C-reactive protein) levels, and reduced the risk of cardiovascular events. METHODS: FOURIER was a randomized trial of evolocumab versus placebo in 27 564 patients with

LDL-cholesterol control in the primary prevention of cardiovascular diseases: An expert opinion for clinicians and health professionals.

AIMS: Although adequate clinical management of patients with hypercholesterolemia without a history of known cardiovascular disease is essential for prevention, these subjects are often disregarded. Furthermore, the scientific literature on primary cardiovascular prevention is not as rich as that on secondary prevention; finally, physicians often lack adequate tools for the effective management of subjects in primary prevention and have to face some unsolved relevant issues. This document aims to discuss and review the evidence available on this topic and provide practical guidance. DATA SYNT

Epicardial fat thickness is significantly increased and related to LDL cholesterol level in patients with familial hypercholesterolemia.

PURPOSE: Increased epicardial fat thickness (EFT) is accepted as a risk factfcardiovascular diseases in some studies. There are conflicting results about the relation between plasma low-density lipoprotein cholesterol (LDL-C) level and EFT. As well, the relation between EFT and familial hypercholesterolemia is not clearly documented in the literature. Our aim was to investigate EFT in familial hypercholesterolemia patients and to determine which parameters were related to EFT increase. METHODS: A total of 150 subjects enrolled in our study. Subjects were separated into two groups: 75 hypercho

LDL-Cholesterol Is Not the Only Clinically Relevant Biomarker for Coronary Artery Disease or Acute Coronary Syndrome.

Coronary artery disease (CAD) is a progressive disorder and its risk is best determined using multiple biomarkers and risk factors. Many novel biomarkers have been shown to improve cardiovascular disease risk beyond that of the traditional risk factors, including polygenic risk scores, small very low-density lipoprotein (VLDL), ApoB, triglycerides (TG), non-HDL-c, lipoprotein (a) (Lp(a)), coronary calcium scores, C-reactive protein (CRP), B-type natriuretic peptide (BNP), troponins, and many other novel biomarkers that have been measured using high throughput assays.

LDL cholesterol and apolipoprotein B are associated with ascending aorta dilatation in bicuspid aortic valve patients.

BACKGROUND: The factors related to ascending aorta dilation (AAD) in patients with bicuspid aortic valve (BAV) are not completely understood. In addition, the role of cholesterol metabolism in AAD has not been studied. METHODS: We analyzed the relationship between different lipid parameters and the ascending aorta diameter/presence of aortic dilatation in 91 consecutive patients with BAV. RESULTS: We observed a positive linear correlation between the total cholesterol, low-density lipoprotein (LDL) cholesterol and apolipoprotein B (ApoB) levels and the ascending aorta diameter. Significant impact of chromosomal locus 1p13.3 on serum LDL cholesterol and on angiographically characterized coronary atherosclerosis.

OBJECTIVES: Recently, a significant impact of a new locus on chromosome 1p13.3 on serum LDL (low-density lipoprotein) cholesterol and clinical events of coronary artery disease (CAD) was described. Potential associations between variants on this locus and angiographically characterized coronary atherosclerosis are unknown. We therefore aimed at investigating the association of variants of locus 1p13.3 with coronary atherosclerosis. METHODS: We performed genotyping of variants rs599839, rs646776, and rs4970834 on chromosome 1p13.3 in a large cohort of 1610 consecutive Caucasian patients underg

Age-multiplied low density lipoprotein cholesterol (LDL-C) burden (AM-LDL) index stratifies bleeding and ischemic risk in acute coronary syndrome: implications for early therapeutic intensification.

BACKGROUND: Early identification of high-risk individuals after Acute Coronary Syndrome (ACS) is critical for tailoring intensive lipid-lowering therapy, a cornerstone of secondary prevention to alleviate the growing global burden of cardiovascular diseases, while practical tools integrating lipid burden and vascular aging remain lacking. This study proposes the age-multiplied low density lipoprotein cholesterol (LDL-C) burden (AM-LDL) index to quantify cumulative atherogenic exposure and stratify 1-year ischemic and bleeding risk. METHODS: This post-hoc analysis utilized data from the prospe

Low-density lipoprotein cholesterol (LDL-C) levels and LDL-C goal attainment among elderly patients treated with rosuvastatin compared with other statins in routine clinical practice.

BACKGROUND: Reducing low-density lipoprotein cholesterol (LDL-C) levels lowers the risk of consequences of cardiovascular disease. Research has confirmed these benefits in elderly patients. The 3-hydroxy-3-methylglutaryl coenzyme A inhibitors (ie, statins) have long-standing proven efficacy in reducing levels of LDL-C and total cholesterol. OBJECTIVE: The goal of this study was to compare change in LDL-C from baseline and National Cholesterol Education Program (NCEP) Adult Treatment Panel (ATP) III LDL-C goal attainment in a population of elderly patients (aged > or =65 years) treated with ro

Long-term LDL cholesterol burden and stroke: mechanistic insights and emerging strategies for personalized lipid management.

Low-density lipoprotein cholesterol (LDL-C) is a causal and modifiable driver of atherosclerotic cardiovascular disease and an important contributor to atherosclerotic ischemic stroke. Increasing evidence suggests that cumulative LDL-C exposure-the combined effect of LDL-C magnitude and duration over time-captures vascular injury more accurately than a single measurement obtained at one clinical encounter. This review uses cumulative LDL-C exposure as an organizing framework for stroke prevention. We summarize current approaches to quantifying long-term LDL-C burden, review epidemiological evi
