Highly contagious classical swine fever (CSF) remains a major trade and health problem in the pig industry, resulting in large economic losses worldwide. In CSF-endemic countries, attenuated CSF virus (CSFV) vaccines have been routinely used to control the disease. However, eradication of CSFV in a geographical area would require permanent reduction to zero presence of the virus. It is therefore of paramount importance to develop a safe, potent, and non-infectious CSF vaccine. We have previously reported on a cost-effective CSF E2 subunit vaccine, KNB-E2, which can protect against CSF symptoms

Control of infectious diseases in wildlife is often considered challenging due to the limited availability of information. Some infectious diseases in wildlife can also affect livestock, posing significant problems for the animal farming industry. In Japan, classical swine fever (CSF) reemerged in September 2018. Given the availability of commercial vaccines, control measures mainly involve the vaccination of domestic pigs and the distribution of oral vaccines to wild boars. Despite these efforts, the disease continues to spread, primarily due to wild boars. This transmission is further exacer

The outbreak of infectious diseases in swine, such as classical swine fever (CSF), has become a significant concern in the pig-farming industry. In Japan, after the re-emergence of CSF in 2018, farms are now exposed to the risk of transmission from infected wild boar and CSF-contaminated farms. This study aimed to identify biosecurity measures that were effective for the prevention of CSF introduction into farms during the period from the beginning of the CSF epidemic to the implementation of a vaccination campaign for domestic pigs at risk. The probability of virus introduction was assumed to

Classical swine fever (CSF) is a highly contagious, devastating, and transboundary viral disease that afflicts swine industries worldwide. Immunization with vaccines is one of the most effective strategies for controlling this disease. However, shifts in the antigenicity and pathogenicity of novel evolving viral strains have the potential to evade vaccination. In this study, 352 samples from swines exhibiting fever, hemorrhages, lethargy, and diarrhea in different pig farms located in 9 provinces of Vietnam were collected. CSFV was identified even within farms that had been vaccinated against

Control of classical swine fever virus (CSFV) in endemic countries relies on vaccination, mostly using vaccines that do not allow for differentiation of vaccinated from infected animals (DIVA). FlagT4G vaccine is a novel candidate that confers robust immunity and shows DIVA capabilities. The present study assessed the immune response elicited by FlagT4G and its capacity to protect pigs for a short time after vaccination. Five days after a single dose of FlagT4G vaccine, animals were challenged with a highly virulent CSFV strain. A strong, but regulated, interferon-α response was found after va

Classical swine fever (CSF) is an economically important infectious disease of pigs caused by Classical swine fever virus (CSFV). To facilitate the eradication of CSF in endemic areas, a marker vaccine enabling differentiation of infected from vaccinated animals (DIVA) is urgently needed. Previously, we have demonstrated that the DIVA vaccine rAdV-SFV-E2, an adenovirus-vectored Semliki Forest virus replicon expressing the E2 glycoprotein of CSFV, induces complete protection from lethal CSFV challenge. The aim of this study was to investigate whether maternally derived antibodies (MDAs) from so

Investigations were carried out to establish the clinical and morphological reactions in rabbits following the consecutive infection with the virulent Vratsa strain and the lapinized K strain of the swine fever virus. It was found that although the virulent strain did not cause a thermal response there were morphological lesions in the body of the rabbits, characterizing the presence of an immune response. The latter was shown to inhibit the thermal reaction in the animals after infecting with lapinized strain. It is stated that such inhibition of the thermal reaction which is typical for stra

A farm in North-West Germany experienced a high morbidity and mortality in their sow herd. Sows showed fever, lethargy, oedema, mucosal discharge and dyspnoea. Necropsy revealed a severe fibrinous and purulent polyserositis. Haematological and histological examinations confirmed septicaemia. Streptococcus equi subspecies zooepidemicus was isolated in high yields from major organs. Sequence typing of this isolate (21/455) revealed a new sequence type showing a significantly higher proliferation rate in comparison to two other isolates. Other infectious agents (influenza A virus, Porcine Reprodu

Given the importance of pork in the Bhutanese diet and to enhance food security, the Government of Bhutan has generated policies to promote development of the swine sector in the country. One of the main threats to swine farming is classical swine fever (CSF), a transboundary disease of wild and domestic swine thought to be enzootic in the country. In 2018, three outbreaks of CSF were notified in Bhutan. The type of animals, their vaccine status, and the aftermath of these outbreaks highlight the interplay of epidemiological and context-specific factors that may result in CSF undermining the e

Classical swine fever virus (CSFV) and Porcine reproductive and respiratory syndrome virus (PRRSV) are both important pathogens which seriously harm the economic swine industry worldwide. We have previously demonstrated that rPRRSV-E2 is a promising live, virus-vectored vaccine that provides 100% protection against highly pathogenic PRRSV (HP-PRRSV) and CSFV. Here, we evaluated the duration of immunity (DOI) of the vaccine strain, rPRRSV-E2. Vaccine or cell culture medium was administered to piglets at 4 weeks of age. All immunized piglets developed high levels of antibodies, which could maint

Classical swine fever is a disease that infects wild boars and pigs and had a significant negative economic impact on the swine industry. Oral vaccination is an effective method for controlling classical swine fever. However, information on oral vaccination program has been limited, and its efficiency has not been clarified in Japan. The purpose of this study was to determine the seasonal variation in factors affecting the ingestion of oral vaccines by wild boars. The Gifu Prefecture oral vaccination program was initiated in March 2019, and by February 2021, six seasonal programs had been cond

African swine fever (ASF) is caused by the African swine fever virus (ASFV) and is a highly contagious, acute, febrile disease that has high morbidity and mortality rates in domestic and wild swine. However, a safe and effective vaccine against ASF remains unavailable as single antigens fail to provide sufficient protection. Therefore, a combination of multiple antigens with an efficient delivery system might be an alternative strategy. Herein, a de novo-designed antigen with multiple T-cell epitopes (TEPs) of ASFV was conjugated for surface display on self-assembled nanoparticles (NPs) of Aqu

Classical swine fever (CSF), caused by the classical swine fever virus (CSFV), is an acute, febrile, and highly contagious disease that has led to significant economic losses in the global swine industry. Although the attenuated lapinized CSF vaccine (C-strain) has effectively controlled CSF outbreaks in China since the 1950s, it remains challenging to serologically differentiate infected from vaccinated animals (DIVA). Currently, the application of E2 subunit vaccines allows for DIVA by detecting antibodies against the Erns protein. Therefore, this study aimed to develop a blocking ELISA for

In several countries, classical swine fever (CSF) has not been detected in domestic pigs, but has been detected in wild boars, making the disease difficult to control. To overcome this problem, we inoculated pigs with a CSF live marker vaccine (Flc-LOM-BErns strain), which has "distinguish infection from vaccinated animals (DIVA)" function, to determine whether it is suitable as an oral vaccine specifically for wild boars. Pigs inoculated intramuscularly or orally with the Flc-LOM-BErns vaccine were challenged 2 or 4 weeks later, respectively, with virulent CSFV. Pigs administered the oral Flc

BACKGROUND: Classical Swine Fever (CSF) is still one of the most economically important viral diseases of pigs. In endemic countries, the disease is controlled mostly through vaccination; hence, the availability of safe and effective vaccines is of utmost importance. Vaccines intended for application in developing countries must also be thermally stable, since the infrastructure needed to maintain a cold chain in those countries is usually lacking. Porvac® is a second-generation subunit marker vaccine against CSF that has demonstrates to be safe and protective. Previous studies have also shown

We analyzed the pathogenicity of a recent Japanese classical swine fever virus (CSFV) to wild boars via an experimental infection using boar-pig hybrids as an alternative to wild boars. We also investigated the effectiveness of a bait vaccine against the CSFV. Naïve boar-pig hybrids and pigs showed clinical signs such as fever, leucopenia, anorexia and conjunctivitis following the experimental infection. In contrast, the boar-pig hybrids administered the bait vaccine did not show any clinical signs. Our data indicated that boar-pig hybrids and domestic pigs have similar susceptibility to the r
