To assess the dynamics of noise-induced tissue injury and repair, groups of CBA/CaJ mice were exposed to an octave-band noise for 2 hours at levels of 94, 100, 106, 112, or 116 dB SPL and evaluated at survival times of 0, 12, 24 hours or 1, 2, or 8 weeks. Functional change, assessed via auditory brainstem response (ABR), ranged from a reversible threshold shift (at 94 dB) to a profound permanent loss (at 116 dB). Light microscopic histopathology was assessed in serial thick plastic sections and involved quantitative evaluation of most major cell types within the cochlear duct, including hair c

Medial olivocochlear (MOC) neurones, located in the superior olivary complex, can suppress cochlear gain by their action on the cochlear outer hair cells. Inputs from the contralateral cochlea and the inferior colliculus (IC) have been separately shown to increase activity of MOC neurones. In this study we have investigated in guinea-pigs under barbiturate anaesthesia the interactions between these two inputs by combining electrical stimulation of the IC with acoustic stimulation of the contralateral cochlea. Electrical stimulation of the IC resulted in a significant suppression of the amplitu

OBJECTIVE: "Dead regions" are regions in the cochlea with no functioning inner hair cells (IHCs) and/or neurons. Amplification (using a hearing aid) over a frequency range corresponding to a dead region may not be beneficial and may even impair speech intelligibility. The objective of this article is to illustrate the use of psychophysical tuning curves (PTCs) as a tool for investigating dead regions and to illustrate the variety of audiogram configurations that can be associated with dead regions. We explore the influence of signal level and signal frequency to test the hypothesis that the fr

Transient evoked otoacoustic emissions (TEOAE) were recorded in 21 guinea-pigs undergoing hypothermia. The minimal average body temperature during cooling was 26 degrees C/24.9 degrees C measured orally or rectally, respectively. The animals were subsequently warmed to normal body temperature. A clear influence of body temperature on TEOAE could be documented. During cooling the amplitude and reproducibilities decreased, disappearing completely at a mean temperature below 28.5 degrees C (oral) and 27.3 degrees C (rectal). The emissions reappeared during rewarming at a mean temperature of 30.1

The GDNF family comprises glial cell line-derived neurotrophic factor (GDNF) and the related proteins neurturin, artemin and persephin, which form a subgroup of the TGF-beta superfamily of growth factors. All four neurotrophic factors provide neuronal cell protection and cell survival. GDNF expression was found in the cochlea, and GDNF has been shown to be effective for inner ear protection from drugs and noise-induced insults. As the other members of the GDNF family also provide protective effects on neuronal cells, they may play important roles in the inner ear. We used RT-PCR to examine the

We have estimated the transfer curve relating instantaneous sound pressure in the ear canal to instantaneous receptor current through the outer hair cells (OHCs) in the basal turn of the guinea pig cochlea using the cochlear microphonic (CM) elicited by continuous 200 Hz tones. The transfer curve is well approximated by a Boltzmann activation curve which has been automatically analysed using a custom-built electronic circuit which continuously derives the three parameters defining the curve with a time resolution of seconds. This technique offers a convenient method of monitoring changes in OH

We investigated the effects of noise and salicylate on the guinea pig cochlea by monitoring the acoustic nerve compound action potential (CAP) as well as the 2f1-f2 distortion product in the cochlear microphonics (DP-CM). The alterations of DP-CMs by noise and salicylate revealed an apparent difference, even when the noise-induced and salicylate-induced CAP threshold shifts were equal. This result indicated that noise and salicylate might have an effect on different sites in the cochlea. Combined effects were altered by changing the order in which the two agents were applied. The existing nois

Nerve-fiber regeneration in the chinchilla cochlea following a traumatic noise exposure was systematically described by Bohne and Harding (1992). However, their study did not determine the origin of the regenerated nerve fibers (RNFs). In the present study, 23 chinchillas were exposed for 12 h to a 0.5 kHz octave band of noise at 120 dB SPL. After a 3-month or 1-year recovery period, their right cochleas were incubated to demonstrate acetylcholinesterase (AChE) activity and then briefly counterstained with Neutral Red or OsO4. Their left cochleas were fixed with OsO4 and dissected using a comb

Using the results of Mössbauer measurements, mechanical activity in the cochlea was tested for by comparing the measured basilar membrane (BM) transverse velocity amplitude with that calculated for a lossless mechanically passive system, derived from the measured BM velocity phase. If the cochlea is considered to be a lossless mechanically passive system, then the transverse velocity amplitude can be calculated from the group velocity and the relative variation of stiffness along the BM. The group velocity can be derived from the Mössbauer phase measurements, and the relative variation of stif

Chinchillas were exposed to octave band noise, sodium salicylate (300 mg/kg per day intraperitoneally), or the combination of both agents for 15 days. The octave band noise exposure was centered at 500 Hz at an intensity of either 80 or 105 dB sound pressure level. The effects of the experimental treatments were evaluated by determining the number of missing hair cells after recovery as a function of location within the cochlea using a surface preparation technique. Average cochleograms were calculated for each of five experimental groups. Animals given salicylate alone showed little or no hai

The effects of glycerol and mannitol on partial oxygen pressure (PO2) in perilymph were studied with a polarographic technique in the guinea pig cochlea. Following a transient reduction immediately after the intravenous injection of each drug, the oxygen tension increased to reach a peak 5 to 10 minutes later, and then decreased gradually. The magnitude and duration of the PO2 increases were larger after the administration of glycerol than after that of mannitol. These results corresponded well with the clinical experience that glycerol was more effective than mannitol in improving hearing in

Two groups of guinea pigs (the newborn and the adult) were exposed to 80dB SPL white noise for 72 hours. The effects were observed with E CochG, surface preparation, scanning electron microscopy and histochemical techniques. The results showed that: 1. the response threshold of the newborn was elevated after exposure; 2. the rate of OHC loss was higher in cochleas of the newborn than those of the adult; 3. OHC damage was mainly found in the fourth turn of cochlea of the newborn; 4. the LDH activity decreased in the newborn. It seems that the white noise could damage the hair cell function and

A thorough knowledge of the normal range of variation of anatomy and topography of the cochlea is necessary for optimal reproduction of this structure and correct interpretation of the radiographs. Radiographic identification of incomplete cochlear coils is essential in the diagnosis of congenital malformations such as Modini's deformity. Furthermore, a diagnosis of otosclerosis/otospongiosis has to be based on recognition of changes in the otic capsule. The size and shape of the human cochlea and the normal ranges of variation of its dimensions were evaluated in 95 plastic casts, prepared fro

In mammals, the indolealkylamine melatonin is synthesized mainly in the pineal gland and to a lesser degree in a number of extrapineal sites. To obtain more information on its distribution, melatonin was measured in the membranous cochlea of the guinea pig. Moreover, melatonin and other serotonin derivatives were determined in organ cultures after incubation with 14C-labelled serotonin. The results show that melatonin is detectable in the organ of Corti and the basilar membrane, and to a lesser degree also in the cochlear nerve and stria vascularis, including the spiral ligament. In vitro stud

Injection of guinea-pig cytomegalovirus into seronegative guinea pig cochleas results in viral labyrinthitis. The location of infected cells 1-8 days following inoculation was examined using an immunohistochemical assay for viral antigens. Mesothelial cells below the basilar membrane and those lining the scala tympani, especially below the osseous spiral lamina, were the first cells to express antigen. This occurred 2 days following inoculation. Infection then spread rapidly toward the brain and through the perilymphatic duct to involve inflammatory cells, mesothelial cells of Reissner's membr

Examination of pure-tone acoustic damage in the chick basilar papilla revealed that the location and extent of hair cell damage was a function of both the stimulus intensity and the age at which the chicks were exposed. Scanning electron microscopic evaluation of noise-exposed cochleae at post-hatching days 1, 10 and 30 permitted the identification of discrete regions of damage, including hair cells with stereocilia injuries as well as those lost from the epithelium. The hair cell damage was tonotopically distributed along the cochlea according to frequency. However, for each exposure frequenc

The method for isolating and treating the vestibular and cochlear compartments of the inner ear was developed. Also, the procedure of histological treatment of temporal bones was modified. The pathogenesis of vestibulo-cochlear disorders due to acceleration and noise was investigated using histology, electron microscopy and histochemistry methods. It was found that exposure to acceleration of 10 Gx for 5 min and single and repeated (for 7 days) exposures to high-frequency noise of 100 dB produced histological changes and functional-morphological variations in all receptors located in the vesti

This study determines how the magnitude and pattern of cochlear damage is altered when exposure to noise is interrupted by regularly spaced rest periods. Groups of chinchillas were exposed for six hours per session to an octave band of noise with a center frequency of 0.5 kHz. The rest interval between successive exposures varied from 18 to 162 hours. The total energy in these exposures was equal to that in a nine-day continuous exposure at 95 dB sound pressure level. The ears of all animals were prepared for histological study so that the extent of cochlear damage could be determined. The pat

While investigating the possibilities of electrically induced hearing in cases of profound deafness, we observed that very often an appropriate stimulation of the cochlea could result in tinnitus cancellation. We present here a progress report on that phenomenon, including 15 subjects. Such a cancellation was restricted to tinnitus subjectively localized in the ear under test, we observed no effect on noises localized in the other ear, or centrally. This suppression occurred so long as the cochlea was stimulated and stopped when the stimulation ceased. This phenomenon was very reproducible and
