The potential use of low-frequency tones to locate regions of outer hair cell loss.

Current methods used to diagnose cochlear hearing loss are limited in their ability to determine the location and extent of anatomical damage to various cochlear structures. In previous experiments, we have used the electrical potential recorded at the round window -the cochlear response (CR) -to predict the location of damage to outer hair cells in the gerbil. In a follow-up experiment, we applied 10 mM ouabain to the round window niche to reduce neural activity in order to quantify the neural contribution to the CR. We concluded that a significant proportion of the CR to a 762 Hz tone origin

[Evoked otoacoustic emissions in diagnosis of cochlear hearing disorders].

The authors consider the possibility of routine use of evoked otoacoustic emissions (EOE) in diagnosis of sensory hearing loss by comparison of EOE curves with relative curves obtained by pure tone audiometry. 180 ears with sensory hearing loss of different aetiologies were examined. EOE were obtained by application of Bray and Kemp's Evoked Otoacustic Measurement Package (ILO88). The results obtained show a close correlation between presence of EOE and real hearing losses of different frequencies obtained by pure tone audiometric evaluation. We could detect no EOE with hearing losses over 50

[Evoked otoacoustic emissions and their modification by contralateral acoustic stimulation].

The active vibration of the basilar membrane as well as evoked otoacoustic emissions (EOAE) are probably based on the motile properties of cochlear outer hair cells (OHC). In the present study we examined the effects of contralateral acoustic stimulation on ipsilateral EOAE and thereby the active cochlear micromechanics. Contralateral white noise with intensities below 30 dB HL enhanced the EOAE amplitude, whereas higher sound levels reduced the evoked acoustic emissions. Similar effects of contralateral acoustic stimuli on ipsilateral EOAE were observed in patients with a conductive hearing l

Decline in the endocochlear potential corresponds to decreased Na,K-ATPase activity in the lateral wall of quiet-aged gerbils.

The ion transport-mediating enzyme, Na,K-ATPase, is abundantly present in the cochlear lateral wall. This enzyme is essential for the generation and maintenance of the endocochlear potential. Diminished enzyme activity has been observed previously in the lateral wall of quiet-aged gerbils. The present study was designed to investigate the impact of the age-related decline in Na,K-ATPase specific activity upon auditory function. Measures of the resting endocochlear potential value and the level of Na,K-ATPase specific activity were made in cochleae obtained from gerbils aged in quiet conditions

[Investigations of the ototoxicity of dicortinef (ear drops)].

The authors investigated the ototoxic influence of Dicortinef in laboratory animals. Their examinations were performed on 15 guinea pigs (weighing 210-380 g.) after application of this medicine to the fenestra rotunda. The harmful effect of Dicortinef was expressed by the characteristic fall in the microphonic potential (PM) and potential of the acoustic nerve (AP).

Effects of aging on normal hearing loss and noise-induced threshold shift in albino and pigmented guinea pigs.

In a previous investigation into noise-induced hearing loss by comparing 2-month-old albino with pigmented guinea pigs, albinos displayed significantly greater shifts in cochlear microphonic (CM) threshold and less recovery than the pigmented animals 7 days after noise exposure. The present study compared the responses of 14-month-old albino and pigmented guinea pigs to the same noise parameters used previously. Thresholds for the first detectable elicitation of CM for three pure tones were recorded prior to, at 90 min and at 7 days after a 45-min exposure to 126 dB broadband noise. Before exp

Cochlear vascular and electrophysiological effects in the guinea pig to 4 kHz pure tones of different durations and intensities.

Round window cochlear microphonic recordings from 33 guinea pigs were obtained prior to and following exposure to a 4 kHz pure tone at levels ranging from 124 dB to 140 dB for 5 to 80 minutes. When electro-physiological evaluation was complete, the animals were killed and tje cochlear tissues prepared using a soft-surface preparation technique. Experimental and control animals were mixed and randomly assessed without prior knowledge of the groups to which the specimens belonged. Histopathological observations were quantified and computer analysed. Statistical analyses suggest that noise exposu

Effect of emotional stress on hearing.

An experimental model for emotional stress is described. Emotional stress can affect hearing if severe enough or if it lasts long enough. The noxious effect on the ear can be explained by the high level of blood catecholamines and exaggerated activity of the cochlear sympathetic innervation.

[Acoustic nerve adaptation in chronic acoustic trauma].

The adaptation phenomenon of the the cochlear nerve in the highest frequency region was studied by means of electrocochleography in the group of 32 persons exposed to prolonged noise and with noise-induced, mild hearing loss (in the 4--6 kHz region) and in 19 persons under the same conditions of noise-exposure but, without any evidence of the hearing loss. The same investigations were performed in the group of 27 normal-hearing persons and non-exposed to noise. In the whole group of persons exposed to prolonged noise, disregarding their hearing condition the amplitude-decrease of the action po

Chronological changes of electrocochleogram in experimental endolymphatic hydrops. Special reference with AP output potential and hair cell cilia.

Chronological changes of the whole nerve action potential (AP), cochlear microphonics (CM) and summating potential (SP) in experimental endolymphatic hydrops in guinea pigs were studied during a period from 1 week to 13 months after the endolymphatic sac obliteration. Endolymphatic hydrops became extensive in month 3 and persisted thereafter. The threshold of AP increased with the lapse of time but good AP output potential was obtained, being maximum in month 3. The threshold of CM increased with the lapse of time. The CM output potential was the highest around week 3 when endolymphatic hydrop

Cochlear protection from carbon monoxide exposure by free radical blockers in the guinea pig.

Acute carbon monoxide exposure produces a significant impairment in high-frequency auditory sensitivity that can be prevented using the N-methyl-D-aspartate receptor blocker MK-801. This finding suggests an excitotoxic component to carbon monoxide ototoxicity and establishes the potential for free radical formation. Free radical scavengers and inhibitors are protective in many organs, including the brain and cochlea, during hypoxic events such as ischemia/reperfusion and, in the cochlea, during noise exposure. This study evaluated the protection afforded by two such agents, phenyl-n-tert-butyl

Hearing loss in experimental cytomegalovirus infection of the guinea pig inner ear: prevention by systemic immunity.

Guinea pig cytomegalovirus (GPCMV) has been used to establish a reproducible model of viral labyrinthitis and hearing loss. Cochlear function was assessed by electrophysiological recordings of cochlear microphonic (CM) and eighth nerve N1 compound action potential (AP) thresholds prior to and up to eight days following inoculation of the scala tympani. Inner ear inoculation of seronegative subjects with live GPCMV produced profound elevations in CM and AP thresholds: 70% of these subjects had their thresholds raised to the limits of the sound system throughout the tested frequency range of 0.1

Hearing loss and visco-elasticity of middle ear fluid.

It has been suggested that the physical properties of middle ear effusion--its viscosity and elasticity--affect the degree of conductive impairment. Exogenous mucus having substantial visco-elasticity was instilled in the bullae of guinea pigs, and the resulting hearing loss was determined by measurements of the cochlear microphonic potential. In this preparation the hearing loss induced by mucus did not differ significantly from that found when the bulla was filled with saline. We find no evidence of a relationship between visco-elasticity of middle ear fluid and amount of conductive impairme

Caffeine and ryanodine demonstrate a role for the ryanodine receptor in the organ of Corti.

The hypothesis that the release of Ca(2+) from ryanodine receptor activated Ca(2+) stores in vivo can affect the function of the cochlea was tested by examining the effects of caffeine (1-10 mM) and ryanodine (1-333 microM), two drugs that release Ca(2+) from these intracellular stores. The drugs were infused into the perilymph compartment of the guinea pig cochlea while sound (10 kHz) evoked cochlear potentials and distortion product otoacoustic emissions (DPOAEs; 2f(1)-f(2)=8 kHz, f(2)=12 kHz) were monitored. Caffeine significantly suppressed the compound action potential of the auditory ner

Electrocochleographic study of Ménière's disease.

Auditory nerve action potential (AP), cochlear microphonics (CM), and summating potential (SP) were recorded from 32 patients with Ménière's disease. Patients were classified into three groups according to the degree of their hearing impairment. The group of patients that had a mild to moderate hearing loss and whose hearing was reversible showed notably satisfactory responses for AP, CM, and negative SP (-SP), and also showed a high incidence of positive SP (+SP). Low responses for AP and CM with an increase of detection thresholds and a high -SP/AP ratio were obtained from patients with mode

Evolution of recruitment at different frequencies during the development of endolymphatic hydrops in the guinea pig.

The study of human temporal bones has identified endolymphatic hydrops as a common feature of several diseases. In particular it is systematically found in those bones removed from patients with premortem Menière's disease. Menière's disease is known to induce sensorineural pathology with recruitment, which changes with the evolution of the hearing loss, and is suspected to induce a cochlear conductive loss by a possible increase in static pressure of endolymph. Amplitude/intensity functions of sensorineural responses can reflect recruitment and/or conduction loss. Experimentally induced hydro

The influence of transducer operating point on distortion generation in the cochlea.

Distortion generated by the cochlea can provide a valuable indicator of its functional state. In the present study, the dependence of distortion on the operating point of the cochlear transducer and its relevance to endolymph volume disturbances has been investigated. Calculations have suggested that as the operating point moves away from zero, second harmonic distortion would increase. Cochlear microphonic waveforms were analyzed to derive the cochlear transducer operating point and to quantify harmonic distortions. Changes in operating point and distortion were measured during endolymph mani

Inputs from the cochlea and the inferior colliculus converge on olivocochlear neurones.

Medial olivocochlear (MOC) neurones, located in the superior olivary complex, can suppress cochlear gain by their action on the cochlear outer hair cells. Inputs from the contralateral cochlea and the inferior colliculus (IC) have been separately shown to increase activity of MOC neurones. In this study we have investigated in guinea-pigs under barbiturate anaesthesia the interactions between these two inputs by combining electrical stimulation of the IC with acoustic stimulation of the contralateral cochlea. Electrical stimulation of the IC resulted in a significant suppression of the amplitu

Identification of the nonlinearity governing even-order distortion products in cochlear potentials.

In order to characterize the cochlear transducer nonlinearities which are involved in the generation of the summating potential (SP), we investigated the effect of a change in the electrical operating point of the cochlear transducer on the SP. The electrical operating point of the cochlear transducer was affected by suppressing reversibly the endocochlear potential (EP). This was realized by intravenous injection of furosemide in guinea pig. A differential recording technique was used in the basal turn of the cochlea to measure locally generated even-order distortion products: the SP and the
