Dynamic frontoparietal flexibility and cognitive dysfunction in schizophrenia: disentangling the roles of symptom burden and childhood trauma.

BACKGROUND: Working memory (WM) impairment is a core cognitive deficit in schizophrenia, associated with dysfunction of large-scale brain networks, particularly the triple-network system comprising the default mode, frontoparietal, and salience networks. Given the role of environmental risks like childhood trauma (CT) in cognitive deficits, we investigated whether trauma relates to altered triple-network flexibility and WM in schizophrenia. METHODS: We enrolled 190 patients with schizophrenia (SZ) and 117 healthy controls (HCs). Among them, 162 SZ and 99 HCs underwent n-back task-based functi

Impaired Cognitive Flexibility With Preserved Learning in an Amyloid Precursor Protein Knock-In Mouse Model of Amyloidopathy.

Alzheimer's disease is a debilitating neurodegenerative condition characterized by amyloid beta plaques and tau neurofibrillary tangles, which leads to progressive cognitive decline. Several new mouse models of fast amyloid deposition have been generated with compound mutations, but how these affect high-level cognitive function is still not fully understood. Four cohorts of a second-generation amyloid precursor protein knock-in mouse model, AppNL-G-F/NL-G-F, which develops aggressive amyloidopathy, were compared with two different control groups that do not produce plaques (AppNL/NL and wildt

Effects of chronic social defeat stress on social behavior and cognitive flexibility for early and late adolescent.

This study investigated the risk to social behavior and cognitive flexibility induced by chronic social defeat stress (CSDS) during early and late adolescence (EA and LA). Utilizing the "resident-intruder" stress paradigm, adolescent male Sprague-Dawley rats were exposed to CSDS during either EA (postnatal days 29-38) or LA (postnatal days 39-48) to explore how social defeat at different stages of adolescence affects behavioral and cognitive symptoms commonly associated with psychiatric disorders. After stress exposure, the rats were assessed for anxiety-like behavior in the elevated plus maze

Cognitive flexibility and emotion regulation in eating disorder patients with comorbid generalized anxiety and posttraumatic stress symptoms.

Research has found that difficulties in emotion regulation negatively impact mental health, whereas cognitive flexibility may promote stress resilience and positive mental health. Little is known about cognitive flexibility and emotion regulation in people with comorbid eating disorder (ED) and anxiety and stress disorders. A transdiagnostic ED population (N = 227) at an outpatient ED treatment facility completed several self-report instruments that measured cognitive flexibility, emotion regulation difficulties, posttraumatic stress disorder (PTSD) symptoms, and generalized anxiety disorder (

Ubiquitination of the GluA1 Subunit of AMPA Receptors Is Required for Synaptic Plasticity, Memory, and Cognitive Flexibility.

Activity-dependent changes in the number of AMPA-type glutamate receptors (AMPARs) at the synapse underpin the expression of LTP and LTD, cellular correlates of learning and memory. Post-translational ubiquitination has emerged as a key regulator of the trafficking and surface expression of AMPARs, with ubiquitination of the GluA1 subunit at Lys-868 controlling the post-endocytic sorting of the receptors into the late endosome for degradation, thereby regulating their stability at synapses. However, the physiological significance of GluA1 ubiquitination remains unknown. In this study, we gener

Investigating differences in cognitive flexibility, clinical perfectionism, and eating disorder-specific rumination across anorexia nervosa illness states.

INTRODUCTION: Cognitive inflexibility, clinical perfectionism, and eating disorder (ED)-specific rumination are common characteristics reported in anorexia nervosa (AN) and may contribute to the maintenance of the illness. It is suggested that clinical perfectionism and rumination may mediate the relationship between cognitive flexibility and AN pathology; however, research to date has not investigated all these factors together. The aim of the current study was to explore the relationships between these factors and how they may relate to ED symptoms in AN. METHODS: Participants included 15 w

Impact of abstract vs. concrete processing on state rumination: An exploration of the role of cognitive flexibility.

BACKGROUND AND OBJECTIVES: Abstract and concrete modes of rumination may be associated with different mental health outcomes; whereby concrete rumination strategies increase the individual's tendency to seek solutions to problems and thus leading to significant improvements in the mood state as well as reductions in the intensity of ruminative thoughts. However, related studies also are suggestive of other variables that are potentially effective in the outcome of abstract and concrete processing. The current study aimed to examine how abstract and concrete processing are associated with state

Impaired Cognitive Flexibility Induced by Chronic Cerebral Hypoperfusion in the 5XFAD Transgenic Mouse Model of Mixed Dementia.

Cerebrovascular lesions are widely prevalent in patients with Alzheimer's disease (AD), but their relationship to the pathophysiology of AD remains poorly understood. An improved understanding of the interaction of cerebrovascular damage with AD is crucial for the development of therapeutic approaches. Herein, we investigated the effects of chronic cerebral hypoperfusion (CCH) in a 5XFAD transgenic (Tg) mouse model of AD. We established CCH conditions in both Tg and non-Tg mice by inducing unilateral common carotid artery occlusion (UCCAO). Cognitive performance in mice was evaluated, and thei

Peroxisome proliferator-activated receptor alpha plays a crucial role in behavioral repetition and cognitive flexibility in mice.

BACKGROUND/OBJECTIVES: Nuclear peroxisome proliferator activated receptor-α (PPAR-α) plays a fundamental role in the regulation of lipid homeostasis and is the target of medications used to treat dyslipidemia. However, little is known about the role of PPAR-α in mouse behavior. METHODS: To investigate the function of Ppar-α in cognitive functions, a behavioral phenotype analysis of mice with a targeted genetic disruption of Ppar-α was performed in combination with neuroanatomical, biochemical and pharmacological manipulations. The therapeutic exploitability of PPAR-α was probed in mice using

A neurocognitive comparison of cognitive flexibility and response inhibition in gamblers with varying degrees of clinical severity.

BACKGROUND: As a behavioral addiction with clinical and phenomenological similarities to substance addiction, recreational and pathological gambling represent models for studying the neurobiology of addiction, without the confounding deleterious brain effects which may occur from chronic substance abuse. METHOD: A community sample of individuals aged 18-65 years who gamble was solicited through newspaper advertising. Subjects were grouped a priori into three groups (no-risk, at-risk, and pathological gamblers) based on a diagnostic interview. All subjects underwent a psychiatric clinical inte

A preliminary investigation of cognitive flexibility for emotional information in major depressive disorder and non-psychiatric controls.

Clinical research suggests that individuals with major depressive disorder (MDD) are cognitively inflexible, exhibiting ruminative, rigid, and automatic thoughts within a negative schema. However, existing neuropsychological research on cognitive flexibility in this population has not employed emotional stimuli. Because research suggests that the performance of individuals with MDD is modulated when emotional stimuli are used, this study investigates the impact of emotional stimuli on cognitive flexibility performance through a novel emotional modification of the Wisconsin Card Sorting Test. The relationship between teachers' cognitive flexibility and perceptions of curriculum change: the mediating role of innovative behavior.

BACKGROUND: Curriculum reforms require teachers not only to understand new curricular expectations but also to adapt them to classroom practice. In this process, teachers' cognitive resources and professional behaviors may be associated with how they interpret and respond to curriculum change. Although cognitive flexibility, innovative behavior, and perceptions of curriculum change have been examined separately in previous research, limited attention has been paid to how these constructs are jointly related within a single explanatory model. AIM: This study aims to examine the mediating role

Depletion of Neurocan in the Prefrontal Cortex Impairs Temporal Order Recognition, Cognitive Flexibility and Perisomatic GABAergic Innervation.

The condensed form of the neural extracellular matrix (ECM), known as perineuronal nets (PNNs), is predominantly associated with parvalbumin-expressing (PV+) interneurons in the cortex and hippocampus. PNNs are enriched in lecticans, including neurocan (Ncan). Genetic variation in the human Ncan gene has been linked to alterations in hippocampus-dependent memory, structural differences in the prefrontal cortex (PFC), and increased risk for schizophrenia and bipolar disorder. However, the specific contribution of Ncan to PFC-dependent cognitive functions and synaptic organization remains incomp

Are Metacognitive Beliefs and Cognitive Flexibility Associated With Cannabis Use Disorder In Adolescents? A Case-Control Study.

OBJECTIVE: Cannabis is among the most commonly used substances by adolescents and is frequently associated with the development of addiction. Various individual, environmental and psychological factors are associated with the onset and maintenance of cannabis use disorder (CUD). Our study aimed to compare levels of depression, anxiety, stress, perceived cognitive flexibility and metacognitive beliefs in adolescents diagnosed with CUD with those in a healthy control (HC) group. Another objective was to investigate the relationship between CUD severity and these parameters within the CUD group.

Long-Term Flywheel Resistance Training Enhances Cognitive Flexibility in Older Women: A 36-Week Randomized Controlled Trial.

Background: Executive functions (EFs) decline with aging, impacting functional independence. While resistance training (RT) is a promising non-pharmacological intervention, the optimal modality and duration for cognitive benefits remain unclear. Flywheel RT (FRT) imposes greater motor control and attentional demands than traditional RT (TRT), potentially enhancing EFs. Objective: To compare the effects of a 36-week FRT versus TRT program on executive function and serum insulin-like growth factor 1 (IGF-1) levels in healthy older women. Methods: This parallel-group randomized controlled trial a

Cognitive Flexibility Mediates the Associations Between Perceived Stress, Social Camouflaging and Mental Health Challenges in Autistic Adults.

Autistic people are at an elevated risk of experiencing co-occurring anxiety and depression. The contributors to this are likely multifaceted and complex and remain poorly understood. Cognitive flexibility, social camouflaging, and perceived stress provide useful indices of the interacting neurocognitive, behavioral, and environmental factors that have been associated with anxiety and depression in autistic individuals. Here, we test if cognitive flexibility, as the factor most closely related to individual differences in thinking styles, mediates the relationships between social camouflaging,

Sex differences in interactions between the muscarinic-1 and orexin-1 receptors on cognitive flexibility in rats.

Previous research has shown that cognitive flexibility, the ability to adjust to different rules, is affected by manipulations of orexin and acetylcholine (ACh). Because there are strong neural connections between orexin and ACh, these two neurotransmitter systems may interact to impact cognitive flexibility. In the present experiment, the effects of pharmacological manipulations of orexin-1 and muscarinic-1 receptors in rats were tested on cognitive flexibility. Rats received intraperitoneal injections of VU0453595, a positive allosteric modulator for the M1 subtype of ACh receptors, for seve

Middle-school students who consumed school breakfast perform better on tasks assessing cognitive flexibility and executive function.

Breakfast improves nutrient adequacy, weight management, and cognitive performance in children; however, less is known regarding the quality, type, and location of breakfast, particularly in adolescents. The purpose of this cross-sectional study was to examine whether consuming breakfast at home or school elicits greater cognitive performance compared to skipping breakfast in 236 urban middle-school students from low-income households. During a single school day, 3 hours after the start of school, students completed questionnaires assessing breakfast consumption followed by a 20-minute compute
