Prostaglandin E2 enhances cortical bone mass and activates intracortical bone remodeling in intact and ovariectomized female rats.

To assess the efficacy of prostaglandin E2 (PGE2) in augmenting cortical bone mass, graded doses of PGE2 were subcutaneously administered for 30 days to seven-month old sham-ovariectomized (SHAM) and ovariectomized (OVX) rats. Both groups were operated at three months of age. Histomorphometric analyses of double fluorescent labeled tibial shafts were performed on basal control, OVX, and SHAM rats treated with 0, 0.3, 1, 3, and 6 mg PGE2/kg/d for 30 days. Baseline aging data showed increased cortical tissue and cortical bone area and reduced bone formation parameters at the periosteal and endoc

Non-linear explicit micro-FE models accurately predict axial pull-out force of cortical screws in human tibial cortical bone.

Screws are the most frequently used implants for treatment of bone fractures and play an essential role in determining fixation stability. Robust prediction of the bone-screw interface failure would enable development of improved fixation strategies and implant designs, ultimately reducing failure rates and improving outcomes of bone fracture treatments. This study aimed to compare the accuracy of micro-computed tomography image based bone volume measures, linear micro-finite element (FE) and non-linear micro-FE simulations in predicting pull-out force of 3.5 mm screws in human cadaveric tibia

Biomechanical evaluation of a fully cortical-threaded screw for modified cortical bone trajectory fixation: Combined experimental and finite element study.

Although fully cortical-threaded screws are introduced as a design adaptation tailored to the cortical-dominant load path of modified cortical bone trajectory (MCBT) fixation, the biomechanical consequences of this design across the screw-bone interface and fusion construct remain insufficiently defined. Therefore, this study compared fully cortical-threaded MCBT screws with clinically used dual-threaded screws through an integrated experimental and finite element (FE) framework, spanning in vitro ovine vertebral biomechanical test and specimen-specific L4 vertebra and L1-S1 fusion models. Old Blood, Young Bones: Identification of Middle-Aged Myeloid Cells That Limit Cortical Bone Loss.

Although studies support disrupted bone remodelling within geriatric populations, mid-life changes are understudied. To investigate this, we performed bone marrow transplantation assays using either 8- or 40-week-old mice. Micro-CT analyses of lethally irradiated 8-week-old mice transplanted with 40-week-old bone marrow exhibited increased mid-shaft femoral cortical bone mass and thickness. Intensive bone marrow regeneration mirrors hematopoietic development in that erythro-myeloid progenitors (EMPs) first expand to support blood production before definitive hematopoietic stem cell (HSC) produ

A proposal for the combined analysis of bone quantity and quality of human cortical bone by quasi-brittle fracture mechanics.

Quasi-brittle fracture mechanics is used to evaluate fracture of human cortical bone in aging. The approach is demonstrated using cortical bone bars extracted from one 92-year-old human male cadaver. In-situ fracture mechanics experiments in a 3D X-ray microscope are conducted. The evolution of the fracture process zone is documented. Fully developed fracture process zone lengths at peak load are found to span about three osteon diameters. Crack deflection and arrest at cement lines is a key process to build extrinsic toughness. Strength and toughness are found as size-dependent, not only for

In vivo monitoring of bone architecture and remodeling after implant insertion: The different responses of cortical and trabecular bone.

The mechanical integrity of the bone-implant system is maintained by the process of bone remodeling. Specifically, the interplay between bone resorption and bone formation is of paramount importance to fully understand the net changes in bone structure occurring in the peri-implant bone, which are eventually responsible for the mechanical stability of the bone-implant system. Using time-lapsed in vivo micro-computed tomography combined with new composite material implants, we were able to characterize the spatio-temporal changes of bone architecture and bone remodeling following implantation i

Cortical bone thickness and bone depth of the posterior palatal alveolar process for mini-implant insertion in adults.

INTRODUCTION: Cortical bone thickness and bone depth are important factors when placing an orthodontic mini-implant. The objective of this study was to investigate both variables for the palatal alveolar process. METHODS: Thirty dry human skulls were imaged by using cone-beam computed tomography technology. Two-dimensional slices bisecting the posterior interdental sites were generated, and cortical bone thickness and bone depth were measured at 4, 8, and 12 mm from the alveolar crest. Two-way repeated measures analysis of variance (ANOVA) was used to analyze the measurements. RESULTS: Inter

Detection of high-energy phosphates in cortical bone as an indicator of bone healing and remodelling: use of a rabbit model.

UNLABELLED: PURPOSE; To study high-energy phosphates in cortical bone through experiments on inbred white New Zealand rabbits. METHODS: Tibial fractures were induced in 80 rabbits and then stabilised by screw osteosynthesis. After 3 (group A; n=40) or 7 days (groups B; n=40), the defective tissue was covered by local muscle flaps. At increasing intervals (from 1 to 16 weeks), the screws were removed and the animals were euthanised (n=8 per group). The bone was removed and analysed histomorphologically; adenosine triphosphate (ATP) levels were determined by high-performance liquid chromatograp

The Impact of Bone Cement on Bone Healing in Revision Hip Arthroplasty for Periprosthetic Femur Fractures and Cortical Osteotomies.

Background It is widely acknowledged that bone cement may infiltrate the fracture site during the implantation of a cemented hip stem for a periprosthetic fracture, potentially leading to non-union. This study sought to examine this hypothesis through a radiological analysis of patients who underwent cemented femoral stem implantation to stabilize a periprosthetic femur fracture or after a cortical osteotomy for stem extraction. Methodology A retrospective study was conducted from 2015 to 2020 at a specialist center in the United Kingdom. Patients over 18 years old receiving cemented femoral s

Insulin Resistance Is Associated With Smaller Cortical Bone Size in Nondiabetic Men at the Age of Peak Bone Mass.

CONTEXT: In type 2 diabetes mellitus, fracture risk is increased despite preserved areal bone mineral density. Although this apparent paradox may in part be explained by insulin resistance affecting bone structure and/or material properties, few studies have investigated the association between insulin resistance and bone geometry. OBJECTIVE: We aimed to explore this association in a cohort of nondiabetic men at the age of peak bone mass. DESIGN, SETTING, AND PARTICIPANTS: Nine hundred ninety-six nondiabetic men aged 25 to 45 years were recruited in a cross-sectional, population-based siblin

A statistical shape model of the human sternum including cortical bone thickness.

INTRODUCTION: Understanding population-level variation in bone size and shape is important for general biomechanics, clinical assessments, and for developing heterogeneous human body models used in safety restraint design. Thoracic injuries are common in severe motor vehicle crashes, and although rib fractures are frequently studied, sternal fractures also occur in these scenarios. This study aimed to investigate how demographic factors-sex, age, stature, and BMI-relate to variations in sternal size, shape, and cortical thickness, all important factors for fracture tolerance. METHODS: Compute

Developmental roots of cortical bone distribution and mandibular robusticity in early hominins.

Mandibular morphology in early hominins reflects both developmental processes and functional demands, yet the ontogenetic emergence of robust mandibular traits remains poorly understood. Here, we present cross-sectional analyses of cortical bone development during early mandibular ontogeny across Paranthropus robustus (N = 7), Paranthropus boisei (N = 1), Australopithecus africanus (N = 1), early Pleistocene Homo erectus (N = 1) and Homo habilis (N = 1), Pan troglodytes (N = 29), Pan paniscus (N = 34), and Homo sapiens (N = 24). We examined the cortical thickness and the structural properties

Bicomponent Mapping of Cortical Bone Using a New Interleaved UTE Imaging Sequence.

PURPOSE: Ultrashort echo time (UTE) MRI enables direct imaging of cortical bone and quantification of its water compartments via bicomponent T2* modeling. However, conventional approaches require multiple separate dual-echo scans due to limitations in gradient power. This approach is prone to inter-scan inconsistencies such as motion and signal drift, which degrade fitting accuracy. This study proposes an interleaved dual-echo acquisition sequence that acquires multiple echo time (TE) images in a single scan to improve bicomponent T2* quantification in cortical bone. METHODS: The proposed UTE

Heterogeneity of ultrasonic wave properties in ex vivo cortical bone of racehorse.

Despite the prevalence of lower limb disease in racehorses, outdoor bone evaluation equipment is lacking. Consequently, a secure and cost-effective field screening system for bones is required to identify early-stage leg illness. Elastic stiffness cannot be measured using x-ray techniques and is closely associated with bone quality, which is recognized as a significant determinant of fracture risk. Quantitative ultrasound has garnered attention as a viable option, offering insights into elastic stiffness. Understanding the characteristic behaviors of ultrasonic wave properties (velocity, atten

Dietary supplementation with nacre reduces cortical bone loss in aged female mice.

Aging is associated with detrimental bone loss leading to fragility fractures in both men and women. Notably, a majority of bone loss with aging is cortical, as well as a large number of fractures are non-vertebral and at the non-hip sites. Nacre is a product of mollusks composed of calcium carbonate embedded in organic components. As our previous study demonstrated the protective effect of nacre supplementation on trabecular bone loss in ovariectomized rats, we sought to evaluate the effect of dietary nacre on bone loss related to aging in female mice which do not suffer true menopause as obs

Cortical bone trajectory fixation cause low compression force in anterior vertebral column.

BACKGROUND: The cortical bone trajectory (CBT) screws follow a caudocephalad and lateral path from the pedicle to the vertebral body. The bone fusion rate of CBT fixation is equal to or lower than conventional pedicle screw fixation. It remains unclear whether or not CVT screws exert equally compressive forces across the vertebral column. In this study, we intraoperatively examined the insertional torque of CBT screws, and investigated the compression loading and pressure distribution in conventional and CBT fixation using pig bones. METHODS: The insertional torque was measured for a total of

Prediction of mechanical properties of human rib cortical bone using fractal dimension.

A large number of post mortem human subjects was used to investigate the relation between the micro-structure of rib cortical bone and the mechanical properties using Fractal Dimension. Uniaxial tensile tests were performed on coupons of rib cortical bone. Tensile strength, yield stress, Young's Modulus, maximum strain, and work to fracture were determined for each coupon. Fractal dimension was computed using CT images and Digital Image Correlation procedures. A highly significant effect of fractal dimension in the mechanical properties was found. In addition, the variation in mechanical prope

Dietary inflammatory index® and cortical bone outcomes in healthy adolescent children.

UNLABELLED: Diet is thought to modulate inflammation. This study shows no relationships between the dietary inflammatory index (DII) and biomarkers of inflammation or bone after adjusting for covariates. Monocyte chemoattractant protein-1 was inversely associated with peripheral tibia cortical thickness and prospective childhood studies should be conducted to better understand this relationship and to determine if there are long-term consequences in adulthood. INTRODUCTION: Examine the relationships between the DII-scores and bone and biomarkers of inflammation in 290 adolescents, ages 9-13 y

Loss of Lgals3 Protects Against Gonadectomy-Induced Cortical Bone Loss in Mice.

Sex hormone deprivation commonly occurs following menopause in women or after androgen-depletion during prostate cancer therapy in men, resulting in rapid bone turnover and loss of bone mass. There is a need to identify novel therapies to improve bone mass in these conditions. Previously, we identified age- and sex-dependent effects on bone mass in mice with deletion of the gene encoding the β-galactoside binding lectin, galectin-3 (Lgals3-KO). Due to the influence of sex on the phenotype, we tested the role of sex hormones, estrogen (β-estradiol; E2), and androgen (5α-dihydroxytestosterone; D
