Crotamine is a polypeptide toxin isolated from rattlesnake venom. Although several studies have been developed identifying many biological effects of isolated crotamine, none of them evaluated its acute toxicity, antinociceptive, and anti-inflammatory activities through oral administration. All in vivo experiments from this study were performed in mice. The up-and-down procedure and hippocratic screening were carried out to evaluate possible pharmacological and toxic effects. Antinociceptive and anti-inflammatory activities of this toxin were evaluated using acetic acid-induced abdominal writh

Salicytamide is a new drug developed through molecular modelling and rational drug design by the molecular association of paracetamol and salicylic acid. This study was conducted to assess the acute oral toxicity, antinociceptive, and antioedematogenic properties of salicytamide. Acute toxicity was based on the OECD 423 guidelines. Antinociceptive properties were investigated using the writhing, hot plate and formalin tests in Swiss mice. Antioedematogenic properties were evaluated using the carrageenan-induced paw oedema model and croton oil-induced dermatitis in Wistar rats. Salicytamide did

Eugenia brasiliensis Lam., a plant from the south of Brazil, is used in the popular medicine for rheumatism treatment. This study reports that topical application of hydroalcoholic extract, fractions and isolated compounds from E. brasiliensis caused an inhibition of ear oedema in response to topical application of croton oil on the mouse ear. For oedema inhibition, the estimated ID50 values (dose reducing the inflammatory response by 50% relative to the control value) for hydroalcoholic extract and fractions (hexane, ethyl acetate and dichloromethane) were 0.17, 0.29, 0.13 and 0.14 mg/ear, re

BACKGROUND: Epizootic lymphangitis is an infectious and chronically debilitating disease of the equines. Histoplasma capsulatum var. farciminosum, a thermally dimorphic fungi, is the causative agent for the disease. In Ethiopia, the disease significantly affects carthorses, posing threats to animal welfare, and resulting in substantial economic losses. Limited availability of widely accessible antifungals in addition to the chronic nature of the disease is the major challenge against management of epizootic lymphangitis. This study aimed to assess the in vitro efficacy of specific local medici

Experiments indicated that the ethanol extract and ether extract from Rhizoma Cynanchi Stauntonii showed obvious antitussive and expectorant effects in mice with stomach administration. The antitussive effect of ethanol extract was stronger than the expectorant effect. The water extract showed a certain expectorant effect but no marked antitussive effect with oral administration. Obvious anti-inflammatory effect was observed on croton oil-caused mouse ear swell with intraperitoneal injection.

Ipomoea stolonifera (I. stolonifera) has been used for the treatment of inflammatory diseases including rheumatism and rheumatoid arthritis in Chinese traditional medicine. However, the anti-inflammatory activity of I. stolonifera has not been elucidated. For this reason, the anti-inflammatory activity of n-butanol extract of I. stolonifera (BE-IS) was evaluated in vivo by using acute models (croton oil-induced mouse ear edema, carrageenan-induced rat paw edema, and carrageenan-induced rat pleurisy) and chronic models (cotton pellet-induced rat granuloma, and complete Freund's adjuvant (CFA)-i

Fungal infections are a major threat to public health care. Cryptococcosis in humans and animals, caused by Cryptococcus neoformans, is a life-threatening disease. In a random antifungal screening of acetone leaf extracts of 400 tree species against Cryptococcus neoformans, the following plant species had good activity: Zanthoxylum capense (Thunb.) Harv. (Rutaceae), Morusmesozygia Stapf (Moraceae), Calodendrum capense (L.f.) Thunb. (Rutaceae), Catha transvaalensis Codd (Celastraceae), Cussonia zuluensis Strey (Araliaceae), Ochna natalitia (Meisn.) Aloe vera at doses of 100 and 300 mg/kg daily for 4 days blocked the wound healing suppression of hydrocortisone acetate up to 100% using the wound tensile strength assay. This response was because of the growth factors present in A. vera masking the wound healing inhibitors such as sterols and certain amino acids. The sterols showed good anti-inflammatory activity (-36%) in reducing the croton oil-induced ear swelling. This activity displayed a dose-response relationship.

A 5-lipoxygenase inhibitor (5-LPI) and a platelet-activating factor antagonist (PAF-A) were studied in dinitrofluorobenzene (DNFB)-induced allergic and croton-oil-induced irritant murine contact dermatitis. Both inhibitors, at 1 and 5% concentrations, significantly reduced the ear swelling in allergic dermatitis while irritant dermatitis was far less affected. This suggests that 5-lipoxygenase-dependent mediators and PAF are involved in allergic contact dermatitis and that these mediators play only a minor role in irritant dermatitis.

The water, ethanol and ether extracts from Cynanchum glaucescems administrated orally showed significant antitussive effect in ammonia-induced cough model in mice. The water and ethanol extracts had obvious expectorant effect. The filtered solution of water decoction injected intraperitoneally could effectively prevent guinea pigs from asthma induced by acetulcholine and histamine mixture, and also inhibit the ear inflammation in mice caused by croton oil. All these effects showed close dose-effect relationship.

Dermal exposure to chemicals may result in allergic or irritant contact dermatitis. In this study, we performed ex vivo local lymph node assay: bromodeoxyuridine-enzyme-linked immunosorbent assay (LLNA: BrdU-ELISA) to compare the differences between irritation and sensitization potency of some chemicals in terms of the 3 end points: lymphocyte proliferation, cytokine profiles (interleukin 2 [IL-2], interferon-γ (IFN-γ), IL-4, IL-5, IL-1, and tumor necrosis factor α [TNF-α]), and ear swelling. Different concentrations of the following well-known sensitizers and irritant chemicals were applied t

Subcellular fractions were prepared from the epidermis of guinea pigs which had been painted with 1-chloro-2,4 dinitrobenzene 14 h previously. The microsomal fraction showed less activity than other fractions in the MIF test. Similarly, the microsomal fraction from the dermis of skin irritated with croton oil before DNCB painting was relatively inactive. However, the microsomal fractions from the epidermis and more especially from the dermis of skin painted with DNCB while undergoing allergic contact dermatitis from pentadecylcatechol showed significant MIF activities.

A methyl salicylate-buffered, croton oil-containing 50% salicylic acid ointment peel, following pretreatment with topical tretinoin and localized 20% trichloroacetic acid, is extremely effective for removal of lentigines, pigmented keratoses, and actinically damaged skin from the dorsum of the hands and forearms. The ease of application, uniform results, decreased risk of scarring, and one-time application of this peel, in comparison with other methods used for treatment of these aging-skin changes, warrants consideration by the dermatologic surgeon.

Assays of steroidal and non-steroidal drugs in an experimental model of dermatitis induced in the ear of the rabbit by the application of a solution of croton oil revealed clearly differentiable inhibitory effects on the rise in skin temperature, the oedema and the increase in tissue mass due to the inflammatory process. The results obtained in this test system fairly accurately reflect the relative therapeutic potencies of known dermatocorticoids and, in conjunction with those found by other methods, afford a more exact characterization of the activity profiles of non-steroidal contra-inflamm
