Opiates, prolactin, and the dopamine receptor.

The administration of a dopamine antagonist, chlorpromazine, and two opiates, morphine and methadone, resulted in a significant rise in serum PRL within 90-150 min. Prior administration of dopamine receptor agonists (apomorphine, levodopa, aand bromocriptine) blocked this effect. In contrast, cyproheptadine, a serotonin antagonist, did not. We suggest that the opiates induce hyperprolactinemia in man via dopamine receptor blockade.

Involvement of neurotransmitters in the action of apelin-13 on passive avoidance learning in mice.

The widespread distribution of apelin-13 and apelin receptors in the brain suggests an important function of this neuropeptide in the brain that has not been explored extensively so far. In the present work, apelin-13 was found to facilitate the consolidation of passive avoidance learning in mice. In order to assess the possible involvement of transmitters in this action, the animals were pretreated with the following receptor blockers in doses which themselves did not influence the behavioral paradigm: phenoxybenzamine (a nonselective α-adrenergic receptor antagonist), propranolol (a β-adrene

Scleroderma and the carcinoid syndrome.

The follow up of a case of the carcinoid syndrome complicated by scleroderma is reported, in which progress of the disease may have been halted by treatment with a combination of cyproheptadine, parachlorophenylalanine and prednisolone. Impairment of tryptophan and 5-hydroxytryptamine (serotonin) metabolism appears central to the development of skin fibrosis in the carcinoid syndrome and may be indicative of important mechanisms in the pathogenesis of idiopathic scleroderma.

Inhibition of insulin secretion from the perfused pancreas of the rat by pizotifen.

1 In the perfused rat pancreas the effects of pizotifen on insulin release induced by 20 mM glucose were studied. 2 Pizotifen (10 and 100 muM) significantly reduced the insulin release during a 25 min perfusion period to 49% and 7% of the controls. 3 The same concentrations of the structurally related agents cyproheptadine, doxepin, and chlorpromazine produced a comparable inhibition.

Neuronal involvement in type 1 hypersensitivity reactions in gut epithelia.

1. Using a number of 5-hydroxytryptamine (5-HT)-antagonists we have compared their activity against the chloride-secretory response in guinea-pig ileal and colonic epithelia when challenged with 5-HT or antigen. Guinea-pigs sensitized to beta-lactoglobulin (beta LG) were used throughout; these were obtained by providing animals with cows' milk for drinking. 2. Of methysergide, ketanserin, cyproheptadine and ICS 205-930, only the latter inhibited both the response to 5-HT and to antigen challenge. Methysergide caused a minor, significant effect on 5-HT but not on beta LG responses. Ketanserin h

Monoamine regulation of prolactin and TSH secretion in hypothyroidism.

Dopaminergic and serotoninergic systems are involved in regulation of prolactin (PRL) and TSH secretion. About 10% of patients presented because of an galactorrhea and hyperprolactinemia (HP) have uncompensated primary hypothyroidism (PH). Two groups of patients were investigated using metoclopramide (MCP, DA antagonist, 10 mg i.v.), cyproheptadine (serotonin antagonist, 4 mg per os) and TRH tests. Group A consisted of 12 females with PH and hyperprolactinemia (HP). Group B presented 4 females with PH, HP and pituitary microadenoma on CT-scan. In control group of healthy euthyroid women basal

Receptor mediation of microvascular responses to serotonin in striated muscle.

Serotonin constricts large arterioles and dilates small arterioles in striated muscle. Our study aimed to identify the receptors that mediate this differential response. Rats were anesthetized with pentobarbital sodium, and the cremaster muscle was prepared for videomicroscopy. Serotonin, applied topically, caused a constriction of large (A1) arterioles that was attenuated by cyproheptadine, methysergide, and LY 53857 but not by MDL 7222, phentolamine, propranolol, or diphenhydramine. The 5-hydroxytryptamine (5-HT)-induced constriction of A1 arterioles was mimicked by alpha-CH3-5-HT but not by

Possible role of central serotoninergic neurons in the development of dental pain and aspirin-induced analgesia in the monkey.

The effects of aspirin or 5-hydroxytryptamine (5-HT)-related drugs on the dental pain induced by electrical stimulation of tooth pulp afferent fibers were assessed in conscious monkeys. The electrical current required for producing jaw opening is referred to as the pain threshold. Both systemic (25 to 75 mg/kg, i.p.) or central (0.5 to 1.5 mg, third cerebral ventricle) administration of aspirin produced analgesia in monkeys. In addition, activation of central 5-HT receptors with central injection of either 5-HT or its precursor, 5-hydroxytryptophan, also produced analgesia. On the other hand,

Fenfluramine-induced hyperthermia and stimulation in the rabbit.

Fenfluramine produces dose-related hyperthermia and behavioral stimulation in rabbits following intravenous administration. Both responses were antagonized by pretreatment with cinanserin, cyproheptadine, or fluxetine. Methergoline likewise abolished the stimulatory effect of fenfluramine and produced a partial blockade of the hyperthermia. Haloperidol was ineffective as a fenfluramine antagonist. This study demonstrates that fenfluramine-induced hyperthermia and stimulation in the rabbit result from an indirect action upon serotonergic receptors.

Anorexia nervosa. Current perspectives in research.

This article is a thorough update of drug treatment studies and basic research. In their presentation of drug interventions, the authors discuss the efficacy of such psychotropic medications as neuroleptics, tricyclic antidepressants, lithium carbonate, cyproheptadine, and delta 9-tetrahydrocannabinol. An overview of new research perspectives on two new anorectogenic substances--AP, isolated from the urine of "hypothalamic" anorectics, and satietin, purified from normal serum--is presented.

Treatment of cold urticaria.

In a single patient with severe essential acquired cold urticaria, the following drug regimes were assessed: diphenhydramine, cyproheptadine, hydroxyzine, terbutaline, cimetidine, hydroxyzine plus cimetidine, and hydroxyzine plus terbutaline. Each regime showed favourable results, but a significance level of P = 0.01 was reached only for (a) hydroxyzine plus cimetidine in the suppression of erythema, and (b) the histamine H1 antagonists in the suppression of wealing.

Pharmacological investigation of the nociceptive response and edema induced by venom of the scorpion Tityus serrulatus.

In this study we characterized the nociceptive response and edema induced by the venom of the scorpion Tityus serrulatus in rats and mice and carried out a preliminary pharmacological investigation of the mechanisms involved in these responses. Intraplantar injection of the venom (1 or 10mug) induced edema and a marked ipsilateral nociceptive response, characterized by thermal and mechanical allodynia and paw licking behaviour. The nociceptive response was inhibited by previous intraperitoneal administration of indomethacin (4mg/kg), dipyrone (200mg/kg), cyproheptadine (10mg/kg) or morphine (5

Effect of benzodiazepines on central serotonergic neuron systems.

Intracerebral (i.c.) injection of serotonin (5-HT) into mice induced head twitches in a dose-dependent manner at 10 min after injection. The head twitches induced by 5-HT (i.c.) were potentiated by the pretreatment of isocarboxazid (3 mg/kg i.p.), and inhibited by cyproheptadine (0.3 mg/kg i.p.), a 5-HT antagonist. Benzodiazepines such as fludiazepam and diazepam potentiated the head twitches induced by 5-HT (i.c.) in a dose-dependent manner. Mescaline (50 mg/kg i.p.) also induced head twitches in mice at 15 and 30 min after injection. Benzodiazepines potentiated the head twitches induced by m

Effect of a serotonin antagonist on delay in grip muscle relaxation for persons with chronic hemiparetic stroke.

OBJECTIVE: To investigate if, following stroke, sustained involuntary activity after voluntary contraction (e.g., grip) of the long finger flexor muscles of the paretic hand is attributable to augmented serotonin release from brainstem pathways, affecting excitability of spastic motoneurons. METHODS: This single-dose placebo-controlled study examined whether a serotonin receptor (5-HT2) antagonist, cyproheptadine hydrochloride, could reduce delay in muscle relaxation of a key paretic long finger flexor muscle immediately after grip for persons with stroke. Time to initiate the long finger fle

Lack of evidence that the central serotoninergic system plays a role in the activation of prolactin secretion following inhibition of dopamine synthesis or blockade of dopamine receptors in the male rat.

Administration of alpha MT to inhibit catecholamine synthesis or dopamine (DA) receptor blockade with spiroperidol had no effect on the hypothalamic concentration of 5HT or 5HIAA. Fluoxetine to block serotonin uptake had no influence on the elevation of serum prolactin levels induced by alpha MT or DA receptor blockers and conversely alpha MT did not influence the prolactin-releasing action of 5HTP alone or in combination with fluoxetine. Depletion of brain serotonin stores with p-chlorophenylalanine did not affect the prolactin-releasing action of alpha MT or DA receptor blockers. In contrast
