Modulation of the hormone setting by Rhodococcus fascians results in ectopic KNOX activation in Arabidopsis.

The biotrophic actinomycete Rhodococcus fascians has a profound impact on plant development and a common aspect of the symptomatology is the deformation of infected leaves. In Arabidopsis (Arabidopsis thaliana), the serrated leaf margins formed upon infection resemble the leaf phenotype of transgenic plants with ectopic expression of KNOTTED-like homeobox (KNOX) genes. Through transcript profiling, we demonstrate that class-I KNOX genes are transcribed in symptomatic leaves. Functional analysis revealed that BREVIPEDICELLUS/KNOTTED-LIKE1 and mainly SHOOT MERISTEMLESS were essential for the obs

Comparative transcriptome profiling and functional analysis of GmRAMOSA1 as a key regulator of adventitious shoot regeneration in soybean.

This study identifies GmRAMOSA1 as a key regulator of adventitious shoot regeneration in soybean and uncovers its regulatory network via comparative transcriptome profiling, providing new insights into the molecular mechanism of soybean shoot organogenesis. Soybean genetic transformation is strongly constrained by genotype-dependent in vitro regeneration capacity, particularly in adventitious shoot formation. Here, we compared the highly regenerable cultivar Dongnong 50 (DN50) with the low regenerative capacity Sichuan landrace Gongqiudou 5 (GQD5) using a cotyledonary node-based adventitious s

Integrated transcriptomic and hormonomic insights into cadmium tolerance of a Euglena mutabilis fungal-algal-bacterial consortium.

Acidic, metal-contaminated environments harbour specialized microbial consortia adapted to extreme stress. We examined an environmental Euglena mutabilis culture naturally associated with Talaromyces and Acidiphilium acidophilum and exposed it to cadmium (Cd). Integrated transcriptomic, hormonal, structural and taxonomic analyses revealed a coordinated Cd-tolerance strategy. RNA sequencing showed differential regulation of metal transporters consistent with a shift from Cd uptake to intracellular sequestration. Transmission electron microscopy confirmed Cd compartmentalization within chloropla

Transcriptomic analysis of AhAHL23-mediated root development and space-induced mutations in peanut (Arachis hypogaea L.).

Peanut seedling development encompasses four distinct tissues (leaf, stem, hypocotyl, root) with largely uncharacterized transcriptional regulatory networks, so we conducted RNA-seq on the space mutant line ZHM112. Transcriptome analysis showed that differentially expressed genes (DEGs) in peanut seedlings were enriched in basic biosynthesis and physiological metabolism pathways, with photosynthetic metabolism prominent in leaves and hormone metabolism in roots. The weighted gene co-expression network analysis (WGCNA) identified eight modules related to the four tissues. By integrating differe

Demethyleneberberine: A possible treatment for Huntington's disease.

Huntington disease (HD) is a type of neurodegenerative disease that is characterized by presence of multiple repeats (more than 36) of cytosine-adenine-guanine (CAG) trinucleotides and mutated huntingtin (mHtt). This can further lead to oxidative stress, enhancement in level of ROS/RNS, mitochondrial dysfunction and neuroinflammations. Many clinical and preclinical trials have been conducted so far for the effective treatment of HD however, none of the drugs has shown complete relief. The regeneration of neurons is a very complicated process and associated with multiple pathological pathways.

Antifungal triazoles affect key non-target metabolic pathways in Solanum lycopersicum L. plants.

Several 1,2,4-triazoles are widely used as systemic fungicides in agriculture because they inhibit fungal 14ɑ-demethylase. However, they can also act on many non-target plant enzymes, thereby affecting phytohormonal balance, free amino acid content, and adaptation to stress. In this study, tomato plants (Solanum lycopersicum L. var. 'Cherrola') were exposed to penconazole, tebuconazole, or their combination, either by foliar spraying or soil drenching, every week, as an ecotoxicological model. All triazole-exposed plants showed a higher content (1.7-8.8 ×) of total free amino acids than the co

Molecular Mechanisms Underlying Flax (Linum usitatissimum L.) Tolerance to Cadmium: A Case Study of Proteome and Metabolome of Four Different Flax Genotypes.

Cadmium is one of the most toxic heavy metal pollutants, and its accumulation in the soil is harmful to agriculture. Plants have a higher cadmium tolerance than animals, and some species can be used for phytoremediation. Flax (Linum usitatissimum L.) can accumulate high amounts of cadmium, but the molecular mechanism behind its tolerance is unknown. Here, we employed four genotypes representing two fiber cultivars, an oilseed breeding line, and a transgenic line overexpressing the metallothionein domain for improved cadmium tolerance. We analyzed the proteome of suspensions and the proteome an

Genome-wide identification, expression and functional analysis of Populus xylogen-like genes.

As an extracellular arabinogalactan protein (AGP) containing a non-specific lipid transfer protein (nsLTP) domain, xylogen mediates the local intercellular communication required for tracheary element (TE) differentiation in Zinnia cell culture. Although XYLP (xylogen-like protein) gene families have been reported in Arabidopsis and rice, no comprehensive analysis has been performed in woody plants. In this work, 31 XYLP genes in five phylogenetic groups were identified from Populus trichocarpa genome and a comprehensive bioinformatic analysis including gene and protein structures, chromosomal

High-efficiency stable transformation of the model fern species Ceratopteris richardii via microparticle bombardment.

Ferns represent the most closely related extant lineage to seed plants. The aquatic fern Ceratopteris richardii has been subject to research for a considerable period of time, but analyses of the genetic programs underpinning developmental processes have been hampered by a large genome size, a lack of available mutants, and an inability to create stable transgenic lines. In this paper, we report a protocol for efficient stable genetic transformation of C. richardii and a closely related species Ceratopteris thalictroides using microparticle bombardment. Indeterminate callus was generated and m

Characterization of HoMADS 1 and its induction by plant hormones during in vitro ovule development in Hyacinthus orientalis L.

To understand the molecular mechanism of ovule development, a MADS box gene, HoMADS 1 , has been isolated from the ovule tissues of Hyacinthus . Sequence comparison showed that HoMADS 1 is highly homologous to both class C and D genes. Furthermore, phylogenetic analysis suggests that HoMADS 1 is most likely a class D MADS box gene. RNA hybridization revealed that HoMADS 1 was exclusively expressed in the ovules. Over-expressing HoMADS 1 in transgenic Arabidopsis plants produced ectopic carpelloid structures, including ovules, indicating that HoMADS 1 is involved in the determination of carpel

Phytohormones in fungi: inter-kingdom modulators or fungal self-controlling elements?

SUMMARYLeveraging data from innovative experimental approaches, omics technologies, and bioinformatics, we offer new insights into how fungi communicate with and perceive their environment to achieve ecological success. By integrating comparative data from both the fungal and plant kingdoms, we critically reassessed the evolutionary, biochemical, and functional landscape of phytohormones in fungi, challenging the conventional notion that these molecules serve exclusively as plant regulators or as means of communication with them. Our analysis demonstrates that fungi not only synthesize a diver

Physiological and transcriptomic analysis uncovers salinity stress mechanisms in a facultative crassulacean acid metabolism plant Dendrobium officinale.

Dendrobium officinale is a precious medicinal Chinese herb that employs facultative crassulacean acid metabolism (CAM) and has a high degree of abiotic stress tolerance, but the molecular mechanism underlying the response of this orchid to abiotic stresses is poorly understood. In this study, we analyzed the root microstructure of D. officinale plantlets and verified the presence of chloroplasts by transmission electron microscopy. To obtain a more comprehensive overview of the molecular mechanism underlying their tolerance to abiotic stress, we performed whole-transcriptome sequencing of the

Interplay of phytohormones facilitate sorghum tolerance to aphids.

Interactions among phytohormones are essential for providing tolerance of sorghum plants to aphids. Plant's encounter with insect herbivores trigger defense signaling networks that fine-tune plant resistance to insect pests. Although it is well established that phytohormones contribute to antixenotic- and antibiotic-mediated resistance to insect pests, their role in conditioning plant tolerance, the most durable and promising category of host plant resistance, is largely unknown. Here, we screened a panel of sorghum (Sorghum bicolor) inbred lines to identify and characterize sorghum tolerance

An improved extraction method enables the comprehensive analysis of lipids, proteins, metabolites and phytohormones from a single sample of leaf tissue under water-deficit stress.

Phytohormones play essential roles in the regulation of growth and development in plants. Plant hormone profiling is therefore essential to understand developmental processes and the adaptation of plants to biotic and/or abiotic stresses. Interestingly, commonly used hormone extraction and profiling methods do not adequately resolve other molecular entities, such as polar metabolites, lipids, starch and proteins, which would be required to comprehensively describe the continuing biological processes at a systematic level. In this article we introduce an updated version of a previously publishe

[Nitric oxide].

Nitric oxide was identified as the relaxing factor derived from the endothelium in 1987. Nitric oxide synthesis allows the vascular system to maintain a state of vasodilation, thereby regulating arterial pressure. Nitric oxide is also found in platelets, where it inhibits adhesion and aggregation; in the immune system, where it is responsible for the cytotoxic action of macrophages; and in the nervous system, where it acts as neurotransmitter. A deficit in endogenous synthesis of nitric oxide contributes to such conditions as essential arterial hypertension, pulmonary hypertension and heart di

Plant growth regulating compounds from microbial resources: Structural diversity, biological activities, and applications in sustainable agriculture.

Plant hormones play important roles in plant growth and development and in response to abiotic and biotic stress. The endogenous phytohormones had been widely applied to improve the productivity and the quality of crop products, and enhance the resistance of plant to abiotic stress. Many microbes produce the endogenous phytohormones, like auxins, cytokinins, gibberellins, abscisic acids, jasmonic acids, ethylene, and their analogues, of which some had been commercialized through fermentation and applied in large scale in crop production and ornamental. Other than the endogenous plant growth re

Panoramic contamination profiling and dietary exposure risk of plant growth regulators in medicinal and edible plants: A data modeling-driven MMSPE-UPLC-MS/MS platform.

Plant growth regulators (PGRs) are extensively deployed in medicinal and edible plants (MEPs) to enhance yields, yet their environmental persistence and potential ecotoxicological effects have raised significant public health concerns. However, current regulatory frameworks remain deficient in PGR-specific residue limits and cross-matrix surveillance tools. Herein, a data modeling-driven broad-spectrum analytical platform was engineered for comprehensive occurrence mapping and dietary exposure assessment of PGRs. Global optimization was achieved through simplex lattice mixture design combined

Methylocystis borbori sp.nov., a novel methanotrophic bacterium from the sludge of a freshwater lake and its metabolic properties.

A novel methanotrophic strain 9NT was isolated from the sludge of a freshwater lake. Cells were aerobic, Gram-stain-negative, non-motile pleomorphic rods with intracytoplasmic membrane systems that appropriate type-II methanotrophs and hemispherical and spherical exocellular formations on the perimeter of the cell wall surface. The novel isolate grows only on methane or methanol as the sole carbon and energy source, at 10-37 °C (optimum 28-30 °C), pH 4.5-9.0 (optimum 7.0-7.5), up to 1% NaCl (optimum 0.3-0.5%). Methanol supported the growth of the strain 9NT in a wide range of concentrations fr

The family of maize D-type cyclins: genomic organization, phylogeny and expression patterns.

Cyclin proteins, associated to cyclin-dependent kinases (CDKs), play fundamental roles in cell cycle control as they constitute a very important driving force to allow cell cycle progression. D-type cyclins (CycDs) are important both for interpreting external mitogenic signals and in the control of the G1 phase. The maize (Zea mays) genome appears to contain at least 17 different CycD genes, and they fall into the subgroups previously described for other plants. Maize CycDs have been named according to identity percentages of the corresponding orthologs in rice and Arabidopsis. In silico analy
