Chronic inflammation, induced by environmental and intrinsic factors, frequently leads to carcinogenesis. In inflammation-associated cancers, such as gastric, colon, and cervical cancers, an important role of epigenetic alterations has been implicated. Such epigenetic alterations include aberrant DNA methylation and histone modifications, some of which can permanently alter cellular characteristics and predispose cells to malignant transformation. Even in normal-appearing tissues, high levels of aberrant DNA methylation can be present, with these levels correlating to future cancer risk. The m

The EXPAH project is a multicentre European study in which biomarkers of exposure, biomarkers of effect, genetic susceptibility and environmental factors were studied in populations exposed to differing levels of carcinogenic polycyclic aromatic hydrocarbons (c-PAHs). We describe here the relationships between the levels of DNA adducts (as biomarkers of exposure), the exposure to air pollution and smoking status. Lymphocyte bulky DNA adducts were significantly correlated with exposure when subjects were classified either by job description or by personal monitor measurements, and both bulky an

DNA methylation in eukaryotes invokes heritable alterations of the of the cytosine base in DNA without changing the underlying genomic DNA sequence. DNA methylation may be modified by environmental exposures as well as gene polymorphisms and may be a mechanistic link between environmental risk factors and the development of disease. In this review, we consider the role of DNA methylation in bone cells (osteoclasts/osteoblasts/osteocytes) and their progenitors with special focus on in vitro and ex vivo analyses. The number of studies on DNA methylation in bone cells is still somewhat limited, n

Growing evidence shows that environmental pollutants pose widespread risks to human health. These pollutants have been detected in multiple human organs, raising concerns about their potential toxic effects. Beyond conventional toxicity pathways, many pollutants disrupt biological systems by perturbing epigenetic machinery. Nucleic acid modifications play crucial roles in gene regulation, and exploring the landscape of environmental pollutant-driven alterations in nucleic acid modifications can yield new insights into their toxicology and health impacts. In this study, we developed a liquid ch

Environmental DNA (eDNA) metabarcoding has emerged as a cost- and time-efficient alternative to traditional morpho-taxonomic methods for benthic foraminiferal inventories. However, the prevalence of soft-walled monothalamous taxa of unknown ecology, which are largely underrepresented in the barcode reference database, limits the accuracy of eDNA studies. Taxonomy-free approaches, which bypass species-level assignment, may overcome these constraints. Here, we use the case study of the Armida gas platform to confirm the potential of the taxonomy-free framework for ecological quality assessment.

The Peace-Athabasca Delta (PAD) is the largest freshwater inland delta in North America. It holds Outstanding Universal Value (OUV) as part of the Wood Buffalo National Park (WBNP) World Heritage Site (UNESCO). The health of the PAD and its OUV was questioned by the Mikisew Cree First Nation in a petition to UNESCO in 2014. In response, UNESCO directed Canada to take a series of actions to determine if WBNP should be listed as a World Heritage in danger. Canada created the WBNP Action Plan (2018), which included a key action to assess the condition of WBNP's OUV with local First Nations. The w

Individuals encounter varying environmental exposures throughout their lifetimes. Some exposures such as smoking are readily observed and have high personal recall; others are more indirect or sporadic and might only be inferred from long occupational histories or lifestyles. We evaluated the utility of using lifetime-long self-reported exposures for identifying differential methylation in an amyotrophic lateral sclerosis cases-control cohort of 855 individuals. Individuals submitted paper-based surveys on exposure and occupational histories as well as whole blood samples. Genome-wide DNA meth

Biodiversity conservation and management in urban aquatic ecosystems is crucial to human welfare, and environmental DNA (eDNA)-based methods have become popular in biodiversity assessment. Here we report a highly overlooked source of significant false positives for eDNA-based biodiversity assessment in urban aquatic ecosystems supplied with treated wastewater - eDNA pollution originating from treated wastewater represents a noteworthy source of false positives. To investigate whether eDNA pollution is specific to a certain treatment or prevalent across methods employed by wastewater treatment

The conventional water filtration approach for collecting environmental DNA (eDNA) has critical limitations. The collection of eDNA via passive eDNA samplers (PEDS) has been proposed as an alternative to the water filtration method. Here, we developed a novel and rapid eDNA sampling approach and evaluated the extent to which this method enhances eDNA sampling efficiency. We drove boats along transects across nine large natural lakes and deployed PEDS either by briefly submerging them at each sampling location ("angling") or towing them in the water ("trolling"). One liter of water was also col

High Throughput DNA Sequencing (HTS) revolutionized the field of paleomicrobiology, leading to an explosive growth of microbial ancient DNA (aDNA) studies, especially from environmental samples. However, aDNA studies that examine environmental microbes routinely fail to authenticate aDNA, examine laboratory and environmental contamination, and control for biases introduced during sample processing. Here, we surveyed the available literature for environmental aDNA projects—from sample collection to data analysis—and assessed previous methodologies and approaches used in the published microbial

ObjectiveSeveral studies investigated the link between agricultural occupational exposures and DNA damage, in an attempt to bring elements of biological plausibility to the increased cancer risk associated with them. However, only a few of these studies focused on females.MethodsThe comet assay was performed on PBMC (Peripheral Blood Mononuclear Cells) samples from 245 females working in open field farming and cattle raising, located in the Normandy area of France. Individual questionnaires on tasks performed were administered at the time of sampling to directly assess exposures. Environmental

BACKGROUND: Studies in non-human mammals suggest that environmental factors can influence spermatozoal DNA methylation, and some research suggests that spermatozoal DNA methylation is also implicated in conditions such as subfertility and imprinting disorders in the offspring. Together with an increased availability of cost-effective methods of interrogating DNA methylation, this premise has led to an increasing number of studies investigating the DNA methylation landscape of human spermatozoa. However, how the human spermatozoal DNA methylome is influenced by environmental factors is still un

DNA methylation in AXL, a receptor tyrosine kinase relevant in cancer and immune function, is reportedly highly heritable. We present evidence to suggest that heritability of DNA methylation in AXL is variable, dependent on population characteristics and cell type studied. Moreover, environmental exposures in utero, particularly exposure to maternal smoking, contributes to variation in DNA methylation of select CpG loci that can affect calculations of heritability. Children exposed to maternal smoking in utero had a 2.3% increase (95 % CI 0.3, 4.2) in DNA methylation in AXL, which was magnifie

Pancreatic cancer is one of the most common cancers of the gastrointestinal tract, alongside stomach and colon cancers, yet remains among the least studied. Due to its non-specific symptoms, late diagnosis, and limited treatment options, it is associated with a poor prognosis and high mortality. Major risk factors for pancreatic cancer include smoking, alcohol consumption, pancreatitis, obesity, and type 2 diabetes. These environmental factors can damage DNA through various mechanisms and, if not properly repaired, may initiate carcinogenesis. DNA repair is one of the key mechanisms in cancer

Extracellular pH has been assumed to play little if any role in how bacteria respond to antibiotics and antibiotic resistance development. Here, we show that the intracellular pH of Escherichia coli equilibrates to the environmental pH following treatment with the DNA damaging antibiotic nalidixic acid. We demonstrate that this allows the environmental pH to influence the transcription of various DNA damage response genes and physiological processes such as filamentation. Using purified RecA and a known pH-sensitive mutant variant RecA K250R we show how pH can affect the biochemical activity o

Epidemiologically related cheese and environmental strains and epidemiologically unrelated strains of Listeria monocytogenes serotype 4b were examined by restriction enzyme analysis of chromosomal DNA with a total of 10 restriction enzymes. The DNA fingerprint patterns generated from each restriction enzyme digest of total DNA of all strains were classified. The restriction enzyme patterns of seven strains recovered from cheese and environmental samples in the same plant were identical to each other, but differed from those of seven epidemiologically unrelated strains. Previous studies examining the reproductive health of alligators in Florida lakes indicate that a variety of developmental and health impacts can be attributed to a combination of environmental quality and exposures to environmental contaminants. The majority of these environmental contaminants have been shown to disrupt normal endocrine signaling. The potential that these environmental conditions and contaminants may influence epigenetic status and correlate to the health abnormalities was investigated in the current study. The red blood cell (RBC) (erythrocyte) in the alligator is nucleated

Because of their sessile lifestyle, plants are inescapably exposed to various kinds of environmental stresses throughout their lifetime. Therefore, to regulate their growth and development, plants constantly monitor the environmental signals and respond appropriately. However, these environmental stress factors, along with some endogenous metabolites, generated in response to environmental stress factors often induce various forms of DNA damage in plants and thus promote genome instability. To maintain the genomic integrity, plants have developed an extensive, sophisticated and coordinated cel

Epigenetic modifications to DNA are proposed to underpin plastic responses to environmental change, and the manner in which DNA methylation contributes to plasticity likely differs among tissues. However, few studies have investigated tissue-specific DNA methylation responses to ecologically relevant environmental stressors in natural settings. Here, we used reduced representation bisulfite sequencing to examine the influence of in situ microhabitats on DNA methylation in gill and foot of juvenile California mussels (Mytilus californianus), a foundation species with widespread dispersal and li
