THE BROAD FISH TAPEWORMS DIBOTHRIOCEPHALUS SPP., THE SEALWORM PHOCANEMA CATTANI, AND OTHER ENDOHELMINTH PARASITES IN THE RETURNING NONNATIVE CHINOOK SALMON, ONCORHYNCHUS TSHAWYTSCHA, IN THE VALDIVIA RIVER BASIN, CHILE.

In the present study, we examined nonnative returning Chinook salmon caught between 2018 and 2023 in the Valdivia River basin, Chile, for endohelminth parasites. Zoonotic endohelminths were identified by morphological and molecular methods, based on a multiplex PCR or PCR and sequencing targeting the cytochrome c oxidase gene of diphyllobothriids or Ascaridoidea mitochondrial DNA. A total of 5,350 larvae of helminths were detected in the salmon, with 3.6% corresponding to the cestodes Dibothriocephalus latus and Dibothriocephalus dendriticus, with freshwater life cycles, and 96% to "Scolex ple

Gastric ulceration caused by genetically identified Anisakis simplex sensu stricto in a harbor porpoise from the Western Pacific stock.

The genus Anisakis is a well-known group of nematodes that parasitize cetaceans as the final host and cause mucosal damage to their stomach. However, little has been done to precisely identify the nematodes recovered from the final hosts, especially in the Western Pacific, because of taxonomic confusion about the discrimination of sibling species and the difficulties of obtaining specimens from cetaceans. We describe the results of genetic identification and histopathological observations of specimens recovered from an ulcerated lesion and stomach contents in the forestomach of a female harbor

Global genetic diversity of Spirometra tapeworms.

Spirometra larvae are etiological agents of human sparganosis. However, the systematics of spirometrid cestodes has long been controversial. In order to determine the current knowledge on the evolution and genetic structure of Spirometra, an exhaustive population diversity analysis of spirometrid cestodes using the mitochondrial gene: cytochrome c oxidase subunit 1 (cox1) was performed. All publicly available cox1 sequences available in the GenBank and 127 new sequencing genes from China were used as the dataset. The haplotype identify, network, genetic differentiation and phylogenetic analysi

Phylogenetic relationships within the Opisthorchis viverrini species complex with specific analysis of O. viverrini sensu lato from Sakon Nakhon, Thailand by mitochondrial and nuclear DNA sequencing.

The liver fluke Opisthorchis viverrini sensu lato causes serious public-health problems in Northeast Thailand and Southeast Asian countries. A hypothesis has been proposed that O. viverrini represents a species complex with varying levels of genetic differentiation in Thailand and Lao PDR. This study aimed to clarify whether O. viverrini populations can be genetically divided into separate taxa. We collected O. viverrini s.l. from eight different locations in Lao PDR and Thailand. The results of nad1, cox1, CF-int6, Pm-int9, ITS2 and 28S rDNA sequence analysis revealed that sub-structuring occ

DNA-based detection of onchocerca volvulus.

During the 1980s, the idea of using deoxyribonucleic acid probes for specific identification and diagnosis of infectious agents became very fashionable. There was therefore an explosion in the development of these tools and one particular group of organisms which received much attention was the parasitic nematodes. This review traces the development and use of such probes with the filarial nematode Onchocerca volvulus, with emphasis on their application to resolving certain 'problems' associated with this parasite, e.g. whether or not strains exist and difficulties in distinguishing the infect

Differentiation of Trichinella genotypes by polymerase chain reaction using sequence-specific primers.

A method was developed to identify species and genotypes within the genus Trichinella using polymerase chain reaction (PCR) and specific primers. Enzymatic amplification of 2 partially conserved and repetitive genomic DNA sequences that have been shown to be variable in length within the different Trichinella genotypes form the basis of this test. Within these regions of the genome, 4 sets of primers were evaluated from which 2 were chosen for their ability to differentiate among the genotypes under stringent primer annealing conditions while maintaining high yields of amplification product. Two New Species of Haematoloechus (Digenea: Plagiorchidae) Parasitizing Rana brownorum (Amphibia: Ranidae) from Southeast Mexico.

In an ongoing investigation on the helminths of amphibians in southeastern Mexico, specimens of 2 undescribed species of Haematoloechus were collected from Rana brownorum. Haematoloechus ceciliae n. sp. is morphologically most similar to Haematoloechus meridionalis, but differs in the shape of the oral sucker, in the nature of the acetabulum, and in the distribution of the glandular cells in the pharyngeal region; Haematoloechus celestunensis n. sp. closely resembles Haematoloechus floedae, but differs in the form and size of the testes and measurements of acetabulum. Molecular evidence for resurrection of Plesiochorus elongatus (Digenea: Gorgoderidae): An urinary bladder parasite of sea turtles.

Trematodes of the genus Plesiochorus were recovered from the urinary bladder of a stranded female adult loggerhead sea turtle, Caretta caretta, on a beach in Rio de Janeiro State, Brazil. Morphological analysis of the specimens revealed characteristics resembling the sub-species Plesiochorus cymbiformis elongatus rather than the recently synonymised Plesiochorus cymbiformis. Molecular phylogenetic analysis of the ITS2 region also showed that P. c. elongatus was distinct from P. cymbiformis and related taxa. Further analysis of the ITS2 revealed substantial differentiation between P. cymbiformi

Toxoplasma CRISPR/Cas9 constructs are functional for gene disruption in Neospora caninum.

Herein we describe, to our knowledge for the first time the use of the clustered regularly interspaced short palindromic repeats/CRISPR-associated gene 9 (CRISPR/Cas9) system for genome editing of Neospora caninum, an apicomplexan parasite considered one of the main causes of abortion in cattle worldwide. By using plasmids containing the CRISPR/Cas9 components adapted to the closely related parasite Toxoplasma gondii, we successfully knocked out a green fluorescent protein (GFP) in an Nc-1 GFP-expressing strain, and efficiently disrupted the NcGRA7 gene in the Nc-Spain7 isolate by insertion of

Anisakis (Nematoda: Ascaridoidea) from Indonesia.

Despite Indonesia's high marine biodiversity, there is a lack of information regarding fish parasites in Indonesian waters. During a sampling of 136 teleost species from Indonesian waters, 22 of them were infected with larvae of Anisakis Dujardin, 1845, a genus with zoonotic potential. We genetically identified 118 worms, provide a revision of all available sequences of the ITS-1-5.8S-ITS-2 marker from Indonesian Anisakis in GenBank (n = 125), and establish 16 new host records. So far, 53 Indonesian teleosts harbour Anisakis spp., 32 of them with known sequence data, increasing the worldwide t

Molecular approaches to Taenia asiatica.

Taenia solium, T. saginata, and T. asiatica are taeniid tapeworms that cause taeniasis in humans and cysticercosis in intermediate host animals. Taeniases remain an important public health concerns in the world. Molecular diagnostic methods using PCR assays have been developed for rapid and accurate detection of human infecting taeniid tapeworms, including the use of sequence-specific DNA probes, PCR-RFLP, and multiplex PCR. More recently, DNA diagnosis using PCR based on histopathological specimens such as 10% formalin-fixed paraffin-embedded and stained sections mounted on slides has been ap

Genetic characterization of atypical Mansonella (Mansonella) ozzardi microfilariae in human blood samples from northeastern Peru.

DNA sequence comparisons are useful for characterizing proposed new parasite species or strains. Microfilariae with an atypical arrangement of nuclei behind the cephalic space have been recently described in human blood samples from the Amazon region of Peru. Three blood specimens containing atypical microfilariae were genetically characterized using three DNA markers (5S ribosomal DNA, 12S ribosomal DNA, and cytochrome oxidase I). All atypical microfilariae were clustered into the Mansonella group and indistinguishable from M. ozzardi based on these DNA markers.

Human cysticercosis and taeniasis: molecular approaches for specific diagnosis and parasite identification.

The construction and antibody screening of Taenia solium cDNA libraries, generated in the Escherichia coli bacteriophage lambda gt11, with the identification of clones putatively expressing antigen B, T. solium-specific and other antigens is described. Lysogens were produced from a number of selected clones and beta-galactosidase fusion peptides ranging in Mr of approximately 135,000-150,000 were demonstrated. These proteins were shown by immunoblotting to be reactive with a pool of sera from cysticercotic patients originally used in the cDNA library screening. We report a method whereby Taeni

First report of Echinococcus granulosus (genotype G6) in a dog in Bamako, Mali.

Cystic echinococcosis is one of the most widespread and important helminthic zoonoses, caused by the larval stage of Echinococcus granulosus sensu lato. However, to date there is little information about the disease in West Africa. Faecal and fur samples from 193 dogs, the main final hosts, were collected in 2010 and 2011 in Bamako, Mali. Taeniid eggs were found microscopically in 28/118 (24%) and 80/223 (36%) faecal and fur samples, respectively. One faecal and one fur sample from the same dog were positive for E. granulosus s. l. DNA. In the remaining 27 faecal (96%) and 77 fur samples (96%)

The erection of two subgenera for Metarhadinorhynchus Yamaguti, 1959 (Acanthocephala: Isthmosacanthida) with a redescription of Metarhadinorhynchus (Metarhadinorhynchus) indicus in India.

BACKGROUND: Adult Metarhadinorhynchus indicus (Bhattacharya & Banerjee, 2003) Kita, Nitta, Kajihara, 2024 were originally described from Megalaspis cordyla (Linn.) in the Chennai coast, Tamil Nadu, India as Gorgorhynchoides indicus Bhattacharya & Banerjee, 2003. The genus Gorgorhynchoides Cable and Linderoth, 1963 was later synonymized with the genus Metarhadinorhynchus Yamaguti 1959. RESULTS: We accept the synonymies involving 15 species of Gorgorhynchoides transferred to Metarhadinorhynchus. Our recognition of the synonymies is partially resolved with the erection of two subgenera: Metarhad

Chromosome fusion and programmed DNA elimination shape karyotypes of nematodes.

An increasing number of metazoans undergo programmed DNA elimination (PDE), where a significant amount of DNA is selectively lost from the somatic genome during development. In some nematodes, PDE leads to the removal and remodeling of the ends of all germline chromosomes. In several species, PDE also generates internal breaks that lead to sequence loss and increased numbers of somatic chromosomes. The biological significance of these karyotype changes associated with PDE and the origin and evolution of nematode PDE remain largely unknown. Here, we assembled the single germline chromosome of t

Phylogeny and systematics of the Proterodiplostomidae Dubois, 1936 (Digenea: Diplostomoidea) reflect the complex evolutionary history of the ancient digenean group.

The Proterodiplostomidae Dubois, 1936 is a relatively small family of diplostomoidean digeneans parasitising the intestines of reptilian hosts associated with freshwater environments in tropical and subtropical regions. The greatest diversity of proterodiplostomids is found in crocodilians, although some parasitise snakes and turtles. According to the most recent revision, the Proterodiplostomidae included 17 genera within 5 subfamilies. Despite the complex taxonomic structure of the family, availability of testable morphology-based phylogenetic hypotheses and ancient hosts, molecular phylogen
