Passive mechanical properties of the left ventricular myocardium and extracellular matrix in hearts with chronic volume overload from mitral regurgitation.

Cardiac volume overload from mitral regurgitation (MR) is a trigger for left ventricular dilatation, remodeling, and ultimate failure. While the functional and structural adaptations to this overload are known, the adaptation of myocardial mechanical properties remains unknown. Using a rodent model of MR, in this study, we discern changes in the passive material properties of the intact and decellularized myocardium. Eighty Sprague-Dawley rats (350-400 g) were assigned to two groups: (1) MR (n = 40) and (2) control (n = 40). MR was induced in the beating heart by perforating the mitral leaflet

Proteomic evaluation and comparison of native and decellularized human saphenous vein extracellular matrices.

BACKGROUND: The growing prevalence of cardiovascular diseases (CVDs) has created an increasing demand for alternative biologically functional vascular grafts. Among the explored tissue engineering strategies, decellularization provides a means to generate suitable acellular scaffolds from donor tissues with preserved complex extracellular matrix (ECM) architecture and vascular basement membrane (BM), both of which are critical for graft endothelialization and function. To assess decellularization efficiency and quality, proteomic profiling offers high-resolution characterization of tissue comp

Biological Matrices Promote Odontoblastic Markers Expression in Stem Cells from Apical Papilla: An In Vitro Study.

INTRODUCTION: Odontoblastic differentiation is a cellular process that requires signaling molecules and extracellular matrix components provided by the dentin-pulp microenvironment. Several scaffolds have been proposed to induce odontoblast differentiation for pulp regeneration. This study evaluated the ability of collagen sponge and decellularized dental pulp, along with dentin, to promote the expression of odontoblastic markers in stem cells from the apical papilla (SCAPs). METHODS: Experimental groups consisted of 2 different biological matrices: (1) human decellularized dental pulp seeded

Fabrication of a Bioactive Human Adipose Extracellular Matrix Allograft Using Supercritical Carbon Dioxide.

PURPOSE: Autologous fat grafting is widely used for soft tissue augmentation but hindered by unpredictable volume retention and donor site morbidity. Decellularized adipose extracellular matrix (DAM) represents a promising alternative. Herein we evaluate an injectable, human-derived DAM decellularized via supercritical carbon dioxide (scCO2, sDAM), promising a gentler, more eco-friendly isolation of matrix components than conventional chemical decellularization. METHODS: Human adipose tissue was homogenized, delipidated, then underwent scCO2 cleansing, two cycles of scCO2 decellularization, c

Temporal regulation of ascorbic acid synergizes with matrix microenvironment directs chondrogenic commitment and limits hypertrophic differentiation.

The avascular and alymphatic nature of articular cartilage severely limits its intrinsic repair capacity. Even when spontaneous healing occurs, it inevitably culminates in fibrocartilage formation, which lacks the biomechanical functionality of native hyaline cartilage. Mesenchymal stem cells (MSCs) have emerged as a promising therapeutic candidate owing to their regenerative potential. However, direct intra-articular transplantation exposes MSCs to a hostile microenvironment characterized by excessive reactive oxygen species and pro-inflammatory cytokines, leading to extensive apoptosis and s

Greener Decellularization of Porcine Auricular Cartilage Using Supercritical Technology and Different Pretreatments for Application in Tissue Engineering.

Scaffolds for tissue engineering can be obtained from synthetic or natural materials, with decellularized tissues being particularly attractive. Among these, porcine auricular cartilage is of special interest because of its availability, similarity to the human extracellular matrix (ECM), and cost-effectiveness. Decellularization of animal tissues yields extracellular matrices (ECM) rich in collagen, elastin, and glycosaminoglycans (GAGs), which are essential for providing mechanical support and creating a favorable environment for cell adhesion and tissue development. Traditional decellulariz

Functional acellular matrix for tissue repair.

In view of their low immunogenicity, biomimetic internal environment, tissue- and organ-like physicochemical properties, and functionalization potential, decellularized extracellular matrix (dECM) materials attract considerable attention and are widely used in tissue engineering. This review describes the composition of extracellular matrices and their role in stem-cell differentiation, discusses the advantages and disadvantages of existing decellularization techniques, and presents methods for the functionalization and characterization of decellularized scaffolds. In addition, we discuss prog

Repairing gastric ulcer with hyaluronic acid/extracellular matrix composite through promoting M2-type polarization of macrophages.

The treatment of gastric ulcer and perforation using synthetic and biomaterials has been a clinical challenge. In this work, a drug-carrying layer of hyaluronic acid was combined with a gastric submucosal decellularized extracellular matrix called gHECM. The regulation of macrophage polarization by the extracellular matrix's components was then investigated. This work proclaims how gHECM responds to inflammation and aids in the regeneration of the gastric lining by altering the phenotype of surrounding macrophages and stimulating the body's whole immune response. In a nutshell, gHECM promotes

A Natural Xenogeneic Endometrial Extracellular Matrix Hydrogel Toward Improving Current Human in vitro Models and Future in vivo Applications.

Decellularization techniques support the creation of biocompatible extracellular matrix hydrogels, providing tissue-specific environments for both in vitro cell culture and in vivo tissue regeneration. We obtained endometrium derived from porcine decellularized uteri to create endometrial extracellular matrix (EndoECM) hydrogels. After decellularization and detergent removal, we investigated the physicochemical features of the EndoECM, including gelation kinetics, ultrastructure, and proteomic profile. The matrisome showed conservation of structural and tissue-specific components with low amou

A nonhuman primate model of lung regeneration: detergent-mediated decellularization and initial in vitro recellularization with mesenchymal stem cells.

Currently, patients with end-stage lung disease are limited to lung transplantation as their only treatment option. Unfortunately, the lungs available for transplantation are few. Moreover, transplant recipients require life-long immune suppression to tolerate the transplanted lung. A promising alternative therapeutic strategy is decellularization of whole lungs, which permits the isolation of an intact scaffold comprised of innate extracellular matrix (ECM) that can theoretically be recellularized with autologous stem or progenitor cells to yield a functional lung. Nonhuman primates (NHP) pro

Advances in cell-derived extracellular matrix for 3D tumor modelling.

Cell-derived decellularized extracellular matrix (cdECM) is being increasingly explored as a supportive biomaterial for bioengineering biomimetic 3D tumor models. The potential to represent the biomolecular diversity of the extracellular matrix in a tumor-specific mode and its production using human cell cultures renders in vitro produced matrix a particularly attractive alternative to non-human counterparts. In this review, we describe well-established and emerging strategies to modulate human ECM production in vitro, as well as showcase recent advances in decellularization and processing met

Challenges and Strategies for Endothelializing Decellularized Small-Diameter Tissue-Engineered Vessel Grafts.

Vascular diseases are the leading cause of ischemic necrosis in tissues and organs, necessitating using vascular grafts to restore blood supply. Currently, small vessels for coronary artery bypass grafts are unavailable in clinical settings. Decellularized small-diameter tissue-engineered vessel grafts (SD-TEVGs) hold significant potential. However, they face challenges, as simple implantation of decellularized SD-TEVGs in animals leads to thrombosis and calcification due to incomplete endothelialization. Consequently, research and development focus has shifted toward enhancing the endothelial

Effect of ECM nanostructures in decellularized small intestine on differentiation of intestinal epithelial model cells.

Caco-2 cells are derived from human colon cancer and have the ability to differentiate into human intestinal epithelial-like cells. The 2D in vitro intestinal model of Caco-2 cells cultured on a semi-permeable membrane is widely used in drug development and the evaluation of absorption functions. However, these intestinal models lack the structural characteristics of the small intestine in vivo, and the cell behavior is not properly controlled. Previous studies have reported that the microstructure of the villi and crypts on a small intestine-mimicking scaffold promotes Caco-2 differentiation;

In Vitro and In Vivo Testing of Decellularized Lung and Pancreas Matrices as Potential Islet Platforms.

The treatment of type 1 diabetes through pancreatic islet transplantation faces significant limitations, including donor organ shortages and poor islet survival due to post-transplantation loss of extracellular matrix support and inadequate vascularization. Developing biocompatible scaffolds that mimic the native islet microenvironment could substantially improve transplantation outcomes. This study aimed to create and evaluate decellularized (DCL) matrices from porcine organs as potential platforms for islet transplantation. Porcine lung and pancreatic tissues were decellularized using four d

Development of decellularized scaffolds for stem cell-driven tissue engineering.

Organ transplantation is an effective treatment for chronic organ dysfunctioning conditions. However, a dearth of available donor organs for transplantation leads to the death of numerous patients waiting for a suitable organ donor. The potential of decellularized scaffolds, derived from native tissues or organs in the form of scaffolds has been evolved as a promising approach in tissue-regenerative medicine for translating functional organ replacements. In recent years, donor organs, such as heart, liver, lung and kidneys, have been reported to provide acellular extracellular matrix (ECM)-bas

Development of a Vaginal Extracellular Matrix Hydrogel for Combating Genitourinary Syndrome of Menopause.

Genitourinary syndrome of menopause (GSM) is initiated by the hypoestrogenic state consequent to cessation of ovarian function and involves changes in all vaginal layers. These alterations include decreased epithelial proliferation and differentiation, impaired extracellular matrix (ECM) maintenance, pro-inflammatory immune milieu, and smooth muscle atrophy. Patient satisfaction with existing treatments for GSM is low, due in part to their lack of therapeutic effect throughout all vaginal layers and reliance on hormonal treatments. Decellularized ECM therapeutics are known to facilitate soft t

Employing a GelMA/Decellularized ECM Biomaterial to Enhance In Vitro Modeling Approaches in Reproductive Biosciences.

Climate change poses a significant threat to global health, with pregnant women and their offspring being particularly vulnerable to heat stress. This rising concern has created an urgent need for advanced in vitro models to study how climate change-associated heat stress affects maternal and offspring health, focusing on how in utero heat stress impacts life course health. The tissue that mediates this maternal stress to the offspring is the endometrium - the innermost lining of the uterus and is comprised of a larger number of phenotypically diverse cells. This work develops a biomaterial-ba
