Dental Pulp Stem Cells Orchestrate Macrophage Polarisation in Pulpitis via Mitochondrial Transfer.

AIM: Pulpitis represents a prevalent clinical condition in dentistry. Macrophages play pivotal roles in pulpitis immunopathology, while dental pulp stem cells (DPSCs) serve as key effectors in pulp tissue repair and immune regulation. Although mesenchymal stem cells are known to regulate immunity through mitochondrial transfer, this mechanism remains unexplored in pulpitis. This study investigated how mitochondrial transfer influences pulpitis progression and resolution. METHODOLOGY: To investigate DPSC-macrophage mitochondrial transfer and its role in polarisation of macrophages, Lipopolysac

Historical Perspectives on the Use of Kalium and Natrium on Putrescent, Infected Dental Pulps.

In the late 1800s, subsequent to the identification of the role of oral bacteria in the demise of the dental pulp by Willoughby D. Miller, the dentist was faced with the major challenge of removing gangrenous pulp tissue from the small root canal spaces in the tooth. Under these clinical circumstances, investigators viewed the root canal contents as a test tube filled with gelatinous, decaying matter along with being overwhelmed by a wide range of bacterial species. One of the first attempts to eradicate the root canal contents was achieved through the use of kalium (potassium) and natrium (so

Is herpes simplex virus associated with pulp/periapical inflammation?

OBJECTIVES: This study focuses on the detection of herpes simplex virus (HSV) DNA in dental pulp and inflamed periapical tissue. STUDY DESIGN: Dental pulp tissue (vital and necrotic) and periapical tissue samples were collected under strictly sterile conditions and examined for the presence of HSV DNA. Saliva samples were also examined for the presence of the viral DNA. The polymerase chain reaction assay was used to detect viral DNA. Blood samples were collected, and the enzyme-linked immunosorbent assay (ELISA) for immunoglobulin G (IgG) and immunoglobulin M (IgM) antibodies against HSV was

Blockade of connexin43-containing hemichannel attenuates the LPS-induced inflammatory response in human dental pulp cells by inhibiting the extracellular flux of ATP and HMGB1.

INTRODUCTION: Tissue repair can be promoted by moderate inflammation but suppressed by excessive levels. Therefore, control of excessive inflammation following removal of infection plays a critical role in promotion of pulpal repair. Connexin 43 (Cx43) forms hemichannels (HCs) or gap channels (GJs) to facilitate the delivery of small molecules between cells to regulate both inflammation and repair. Understanding the role of Cx43 in dental pulp may help develop a potential strategy to attenuate the inflammation and promote the formation of reparative dentin in deep caries. METHODS: We firstly

Observation of Inflammation, Oxidative Stress, Mitochondrial Dynamics, and Apoptosis in Dental Pulp following a Diagnosis of Irreversible Pulpitis.

OBJECTIVE: Mitochondrial dynamics play a pivotal role in maintaining the homeostasis of the dental pulp. Inflammation and oxidative stress can trigger changes in mitochondrial dynamics, leading to cell death in the dental pulp. This study aimed to investigate inflammation, oxidative stress, mitochondrial dynamic alterations, and cell death in inflamed pulpal tissues compared to healthy pulp tissues. METHODS: Pulpal tissues were collected (n=15 per group) from: 1) healthy people as the control and 2) people with clinically diagnosed irreversible pulpitis. Proteins indicating inflammation, oxid

Endodontic treatment of mandibular molars with concrescence.

Endodontic treatment of a permanent mandibular first molar and a second molar presumably joined by cementum is presented. This case illustrates the difficulty in interpreting the vitality testing in such a situation. It also suggests possible irreversible pulp injury of a vital healthy tooth caused by extension of the inflammatory periapical pathosis of the adjacent tooth.

Knowledge, attitude, and practice regarding standardized treatment protocol for pulp therapy in deciduous dentition among general dental practitioners of Vadodara, Gujarat, India.

INTRODUCTION: Dental caries is the most prevailing disease not only affecting permanent dentition, but also involving deciduous dentition. Deciduous teeth are considered a best space maintainer and preserve arch integrity for eruption of succedaneous permanent teeth. General dental practitioners (GDPs) seem to be less interventionist in their investigation and treatment planning and many times, the treatment done remains incomplete either due to the lack of knowledge of the dentist or due to noncooperation of the child and/or parents. PURPOSE: The aim of this study was to assess the knowledge

Influence of maturation status and tooth type of permanent teeth upon electrometric and thermal pulp testing.

The threshold values of developing teeth were determined for two commonly used electrometric pulp testers (Siemens Sirotest and Bofors Pulp Tester) and a new thermal test based on carbon dioxide snow (Odontotest). The teeth examined were divided into 7 stages according to maturation status. This study showed that teeth in Stages 3-5, i.e. until completion of root formation, show an increased electrometric threshold value. The Siemens tester appeared to be equal to the Bofors tester with regard to reliability. The Odontotest was the most reliable method examined, giving a consistent positive re

[Exposit in a biological test].

The biocompatibility of the composite resin materials Exposit and Evicrol was tested by functional studies of the rat incisor and the biological test of the dental pulp of pigs. Exposit shows mainly reversible reactions of the vascular connective tissue both in the vital-microscopical examination of the immediate reaction and in short-term tests (24 hours, 7 days), where positive adaptation reactions (new vessels and revascularization) are to be observed. Evicrol causes distinct and irreversible damages in the pulp of the rat incisor. The results of the vital-microscopical examination are prov

Classification and incidence of middle mesial canals of mandibular first molars in a northern Chinese subpopulation: a cone-beam computed tomographic study.

BACKGROUND: The middle mesial canal (MMC) represents a complex anatomical variation of mandibular first molars (MFMs). The association between MMC occurrence and anatomical landmarks remains controversial in cone-beam computed tomography (CBCT) studies. MATERIALS AND METHODS: CBCT imaging data and data regarding age, sex, tooth position, the distance between the mesiobuccal (MB) and mesiolingual (ML) orifices, and the presence of radix entomolaris were collected. When MMCs were present, root canal morphology was classified according to the Ahmed classification system. The data were evaluated
