Facilitation of bone resorption activities in synovial lavage fluid patients with mandibular condyle fractures.

The aim of this study was to investigate the bone resorption effect of the mediators delivered in joint cavity of patients with mandibular condyle fractures by detecting osteoclast markers using cellular biochemistry methods, and by analysing bone resorption activities via inducing osteoclast differentiation of the infiltrated cells from arthrocentesis. Sixteen joints in 10 patients with mandibular condyle fractures were evaluated. The control group consisted of synovial fluid (SF) samples from seven joints of four volunteers who had no clinical signs or symptoms involving the temporomandibula

Detection of root resorption using dentin and bone markers.

OBJECTIVES - To test the hypothesis that during root resorption, organic matrix proteins and cytokines from the surrounding bone and dentin are released into the gingival crevice. MATERIAL AND METHODS - Subjects with mild (<2 mm loss) and severe root resorption (>2 mm) were identified. Control group subjects with no loss of root structure or undergoing orthodontic treatment were also identified. Gingival crevicular fluid (GCF) was collected non-invasively from the mesial and distal sides of each of the four upper incisors by using filter paper strips. The eluted GCF was used for analysis using

Hyperglycemia simultaneously induces initial caries development and enhances spontaneous occlusal surface wear in molar teeth related to parotid gland disorder in alloxan-induced diabetic rats.

Diabetes and salivary gland dysfunction are major factors that induce dental caries in experimental animals, but there are no reports analyzing the association of dental caries and salivary glands in an animal model of diabetes mellitus (DM). To clarify the initial development of dental caries and preceding salivary gland disorder, we performed a histopathological analysis on teeth and salivary glands in diabetic Wistar rats 7 weeks after alloxan treatment (DM group) in comparison with nondiabetic rats (Non-DM group) and functional analysis on saliva secretion during the experimental period. The antimicrobial peptide, human β-defensin-1, potentiates in vitro osteoclastogenesis via activation of the p44/42 mitogen-activated protein kinases.

Previous studies have demonstrated increased expression and raised levels of human β-defensin (hBD)-1 in gingival tissue and crevicular fluid of patients with chronic periodontitis and peri-implantitis, oral bone-resorbing diseases caused by enhanced osteoclastogenesis. Therefore, we aimed to investigate the effect of hBD-1 on osteoclast formation and function and to elucidate the involved signaling pathway in vitro. Human peripheral blood mononuclear cells (PBMCs) were first incubated with various doses of hBD-1 and cell viability was assayed by MTT. PBMCs were treated with macrophage-colony

A novel root canal irrigant enabling rapid formation of a tannic acid-silver nanocoating for sustained antibacterial therapy in root canal infections.

Continuous intracanal disinfection facilitates the complete elimination of residual pathogenic microorganisms and prevents the recurrence of root canal infections. Current prolonged disinfection strategies rely on calcium hydroxide or antibiotic pastes; however, these agents require thorough subsequent removal, which extends the treatment cycle and increases procedural complexity. Herein, we report a two-component irrigation system based on tannic acid (TA) solution and silver nitrate (AgNO3) solution that rapidly forms a long-lasting antibacterial coating (≤3 min) without the need for subsequ

Identification of the bacterial phylotypes Bifidobacterium dentium, Actinomyces naeslundii, and Propionibacterium species in deep dental caries using fluorescence in situ hybridization.

BACKGROUND AND OBJECTIVES: Dental caries is a progressive irreversible microbial disease. Different regions of the carious dentin contain different bacteria in various proportions. With the recent advent of new techniques in molecular biology, our understanding of the microbial agents associated with dental caries has grown tremendously. It is essential to identify and quantify the organisms from the diseased sites quickly and reliably. Fluorescent in situ hybridization is one such technique. Thus, the present study was aimed at estimating the prevalence of the bacteria Bifidobacterium dentium

Metabolomic signatures distinguish extracellular vesicles from osteoclasts and odontoclasts.

AIMS: Pathological dental root resorption and alveolar bone loss are often detected only after irreversible damage. Biomarkers in the gingival crevicular fluid or saliva could provide a means for early detection; however, such biomarkers have proven elusive. We hypothesize that a multiomic approach might yield reliable diagnostic signatures for root resorption and alveolar bone loss. Previously, we showed that extracellular vesicles (EVs) from osteoclasts and odontoclasts differ in their protein composition. In this study, we investigated the metabolome of EVs from osteoclasts, odontoclasts an

The effect of nano chitosan and calcium hydroxide intracanal medications on sealer penetration & bond strength in radicular dentin.

BACKGROUND: A three-dimensional obturation that provides a fluid tight seal of the root canal system is essential for long-term treatment success. Intracanal medicaments may hinder sealer penetration and/or affect its ability to bond with dentin. This study aimed to evaluate the sealer penetration and push-out bond strength after using Calcium hydroxide and a 2% nano-chitosan gel (CSNPs). METHODOLOGY: Twenty-four extracted single-canal lower premolars were randomly divided, according to the intra canal medicament used, into two groups: Group A, Ca(OH)2, and Group B, 2% CSNPs. After medication
